Catabolic Plasmid-Mediated Heavy Metal Resistance in Herbicide Diuron-Degrading Pseudomonas species

  • Three Pseudomonas strains (Bk8, Bk9, Bk10) selected from soil for their ability to degrade herbicide diuron were tested for their heavy metal resistance. The growth of these catabolic strains on a minimal medium with various concentrations of $Cd^{2+},\;Zn^{2+},\;Ni^{2+}$, and $Hg^{2+}$ revealed a minimal effect on the carbon source for the inhibitory effect of the metals. One of these strains, namely, Bk8, exhibited a high resistance to the heavy metals as compared to the two other strains. This strain harbors plasmid pBk8 (110 kb) and contains at least fur determinants encoding heavy metal resistance. Nickel and zinc resistance are encoded by genes located on the chromosome, while cadmium and mercury resistance are on plasmid pBk8. Accordingly, the characteristics of strain Bk8 suggest that it would be useful in the bioremediation of aromatic compounds in the presence of toxic heavy metals as co-contaminants.

하천에서 분리한 비소 내성세균의 유전적 특성 (Genetic Characteristics of Arsenic Compounds-Resistant Bacteria Isolated from Stream Water)

  • Several arsenic compound-resistant bacteria were isolated from Jung-Rang stream. The isolates, D-3, D-12, and D-14 were characterized phenotypically and genetically, and identified as Serratia liquefaciens, Klebsiella oxytoca, and Klebsiella pneumoniae, respectively. A plasmid of 67kb was found in Klebsiella oxytoca D-12 and designated as pMH12. Transfer of this plasmid from D-12 to E. coli HB101 was occurred, and the resulting transconjugant strains expressed the same level of heavy metal resistance as the donor strain. The physical presence of this plasmid in transconjugant was detected with agarose gel electrophoresis. Arsenite-sensitive derivatives of isolate D-12 were obtained with Mitomycin C treatment which cured pMH12. Antibiotics and heavy metal resistances were also examined to be used as a proper marker for the isolates in gene cloning. Isolate D-12 has resistance to several heavy metal ions such as $Cd^{2+}$ , $Zn^{2+}$ and $Hg^{ 2+}$ Also, all the other arsenite resistant isolates showed resistance to several heavy metal ions and antibiotics.

약수에서 分離한 大腸菌群의 일부 중금속 및 抗生劑耐性에 관한 연구 (Antibiotic and Heavy Metal Resistance of Coliform Bacteria Isolated from Mineral Water)

  • The purposes of this study were to find out the heavy metal and antibiotic resistant coliform bacteria from mineral water and the resistant factors. For the experiment, mineral water samples were taken from A area and B area during the period from march to July, 1988. The results of the experiment were as follows 1. From mineral water, eleven resistant coliforms and one susceptible coliform were isolated. 2. All resistant isolates harbored diverse plasmids of ranged ca. 14-54kb. 3. Susceptible coliform harbored a only plasmid of ca. 2.8 kb. 4. All resistant isolates harbored common size of plasmid of ca. 14kb. 5. As a result of the transformation and agarose gel electrophoresis experiments, resistant factor was R-plasmid. In conclusion, It is suggested that heavy metal contamination of mineral water is the selective pressure for the plasmid encoding the tolerance. Heavy metal resistance, in some case, is present with antibiotic resistance. Therefore, heavy metal contamination of mineral water induces antibiotic resistant bacteria.

Incidence of Multiple Heavy Metal Resistance in a Bacillus Species

  • A new strain of Bacillus capable of growing upto pH 11 was isolated from a local pond polluted with detergents. This strain elicited unimpaired growth in media supplemented with heavy metals such as As, Cu, Cd, Hg, Ni and Zn. A MIC value of 200, 3, 1.60, 2.25, 7.50 and 3.70 mM was noticed for As, Cu, Cd, Hg, Ni and Zn respectively. Analysis of total DNA revealed the presence of a plasmid of 26 kb. This plasmid was lost by acriflavine treatment to the cultures. Such cured strains were found sensitive to heavy metals. Our findings suggest that incidence of heavy metal resistance is widely distributed and resistant strains could be isolated from heavy metal unpolluted sites.

  • Microorganisms play a significant role in bioremediation of heavy metal contaminated seawater. In this study, we reported an effective removal of Cu and Zn in marine envionment by using Desulfovibrio desulfuricans (D. desulfuricans) which belong to sulfate reducing bacteria. D. desulfuricans showed stable growth characteristics in high salt concentration and had resistance to heavy metals. Cu and Zn was removed not only by physical adsorption on the surface of bacteria but also by precipitation reaction of microbial metabolism by D. desulfuricans in seawater. In case of different heavy metal concentration, Cu was effectively removed 85% at 25 ppm and 60% at 50 ppm and Zn was effectively removed 54% at 50 ppm and 46% at 200 ppm, respectively.

The development of a wear resistant hard-metal tappet in diesel engines

  • Diesel engines have many sliding parts with solid body contact. For example, a piston-ring and a cylinder bore, a valve and a valve-seat, a cam and a valve tappet. These parts have a severe wear problem. during engine life times. During these times, the valve tappet has abnormal wear such as scuffing and pitting due to a high hertzian contact stress between the cam and the tappet. Excessive wear problems frequently occur to both the cam and the tappet. To solve these problems, we developed an advanced wear resistant tappet. The developed tappet consisted of a hard-metal wear part and a steel body. To increase a bonding strength, those two parts, were directly bonded to each other. Also to decrease a bonding temperature, we developed the composition of Ni-binder materials in the hard metal. To estimate the wear characteristics of the newly developed tappet, we performed wear tests and engine dynamo tests in order to compare them with a conventional Fe-base tappet. As a result, the newly developed tappet has better wear characteristics than those of the conventional tappet. In addition, we performed a 100,000km field-test, and the newly developed tappet showed much improved wear resistance.

  • 납, 아연, 카드뮴에 의한 미생물 생장 저해를 변형된 Tris minimal medium을 사용하여 측정하였다. Escherichia coli 균주에 대한 독성의 세기는 아연 > 카드뮴 > 납 순으로 나타났고 Streptomyces coelicolor A3(2)의 철 함유 superoxide dismutase 2를 과발현하는 E. coli 균주는 중금속 저항성이 증가되었음을 알 수 있었다.

Construction and Characterization of Multiple Heavy Metal-Resistant Phenol-Degrading Pseudomonads Strains

  • Metal ions contamination may inhibit microorganisms involved in the biodegradation of organic compounds and affect biodegradation rates. Therefore, it is likely that bioremediation of xenobiotics-contaminated soils and waste will require inoculation with efficient biodegrading microbial communities, with capabilities of being resistant to heavy metals as well. Two different transconjugants (Pseudomonas sp. KMl2TC and P. aeruginosa TC) were constructed by conjugation experiments. Results on MIC, induction and growth inhibition strongly indicated that arsenic-resistant plasmid, pKM20, could be mobilized, and the newly acquired phenotype of pKM20 was not only expressed but also well regulated, resulting in newly acquired resistances to $As^{5+},\;As^{3+},\;and\;Sb^{3+} in\;addition\;to\;Cd^{2+},\;Zn^{2+},\;and\;Hg^{2+}$. The phenol- degradation efficiencies of Pseudomonas sp. KMl2TC were maintained significantly even at high heavy metal concentrations at which these efficiencies of P. aeruginosa TC were completely impaired. The results in this study on the effects of heavy metals on phenol degradation, especially after conjugation, are the first ever reported. All the results described in this study encourage to establish a goal of making "designer biocatalysts" which could degrade certain xenobiotics in the area contaminated with multiple heavy metals.

Siderophore-producing rhizobacteria reduce heavy metal-induced oxidative stress in Panax ginseng Meyer

  • Background: Panax ginseng is one of the most important medicinal plants and is usually harvested after 5 to 6 years of cultivation in Korea. Heavy metal (HM) exposure is a type of abiotic stress that can induce oxidative stress and decrease the quality of the ginseng crop. Siderophore-producing rhizobacteria (SPR) may be capable of bioremediating HM contamination. Methods: Several isolates from ginseng rhizosphere were evaluated by in vitro screening of their plant growth-promoting traits and HM resistance. Subsequently, in planta (pot tests) and in vitro (medium tests) were designed to investigate the SPR ability to reduce oxidative stress and enhance HM resistance in P. ginseng inoculated with the SPR candidate. Results: In vitro tests revealed that the siderophore-producing Mesorhizobium panacihumi DCY119T had higher HM resistance than the other tested isolates and was selected as the SPR candidate. In the planta experiments, 2-year-old ginseng seedlings exposed to 25 mL (500 mM) Fe solution had lower biomass and higher reactive oxygen species level than control seedlings. In contrast, seedlings treated with 108 CFU/mL DCY119T for 10 minutes had higher biomass and higher levels of antioxidant genes and nonenzymatic antioxidant chemicals than untreated seedlings. When Fe concentration in the medium was increased, DCY119T can produce siderophores and scavenge reactive oxygen species to reduce Fe toxicity in addition to providing indole-3-acetic acid to promote seedling growth, thereby conferring inoculated ginseng with HM resistance. Conclusions: It was confirmed that SPR DCY119T can potentially be used for bioremediation of HM contamination.

Effects of Rare Earth Metal Addition on the Cavitation Erosion-Corrosion Resistance of Super Duplex Stainless Steels

  • Austenitic stainless steels such as AISI 316L have been used in equipment in which fluid flows at high speeds which can induce cavitation erosion on metallic surfaces due to the collapse of cavities, where the collapse is caused by the sudden change of local pressure within the liquid. Usually AISI 316L is susceptible to cavitation erosion. This research focuses on developing a better material to replace the AISI 316L used in equipment with high speed fluid flow, such as impellers. The effects of Rare Earth Metal (REM) additions on the cavitation erosion-corrosion resistance of duplex stainless steels were studied using metallographic examination, the potentiodynamic anodic polarization test, the tensile test, the X-ray diffraction test and the ultrasonic cavitation erosion test. The experimental alloys were found to have superior mechanical properties due to interstitial solid solution strengthening, by adding high nitrogen (0,4%), as well as by the refinement of phases and grains induced by fine REM oxides and oxy-sulfides. Corrosion resistance decreases in a gentle gradient as the REM content increases. However, REM containing alloys show superior corrosion resistance compared with that of other commercial alloys (SAF 2507, AISI 316L). Owing to their excellent mechanical properties and corrosion resistance, the alloys containing REM have high cavitation erosion-corrosion resistance.
