잉어류 바이러스성전신괴사증바이러스 (VSNCV) 백신 투여에 대한 잉어의 면역반응 (Immune responses th the vaccines of viral systemic necrosis of carp virus (VSNCV) of comom carp, Cyprinus carpio L.)

  • 양식 잉어류에서 심각한 피해를 야기하고 있는 VSNCV에 대한 백신을 제작하여 잉어에 투여한 후 특이적 및 비특이적 면역반응을 조사하였다. 백신은 포르마린처리백신 (FKV)과 가온처리백신 (HKV)를 제작하여 건강한 잉어에 0.2㎖씩 복강주사 하였으며, 2주 후 동일한 방법으로 boost 처리하였다. 백신 투여 후 비특이적 면역반응 중 혈청 라이소자임 활성과 대식세포의 chemiluminescent (CL) reponses은 1차 주사 후 및 boost 처리 후 2일째부터 7일째까지 유의적인 증가를 나타내었으며, 이후 대조구 수준으로 감소하였다. ELISA법으로 항체가를 조사한 결과 대조구를 제외한 FKV와 HKV 투여구에서 주사 후 2주 째부터 boost 처리 후 6주 째까지 유의적으로 증가하였으며, 그 중에서도 boost 처리 후 2주 째 최고치를 나타내었다. 각 시험구별로 boost 처리 후 2주 째에 VSNCV로 공격 실험한 결과 FKV 투여구에서는 20%의 누적폐사율을 나타내어 방어력이 인정되었으나, HKV 투여구에서는 70%의 누적폐사율이 나타나 방어효과가 거의 없는 것으로 나타났다.

  • A total of 100 swelled, springered or flippered canned meat and fish products were studied the degree of contamination with clostridias and serological relationships to Hobbs'13 "heat resistant" types, heat resistance of spores and susceptibility of Clostridium perfringens isolates to several antibiotics. Samples examined in this study were collected from Seoul area from June to October, 1975 and prepared in Korea. Clostridias were isolated from 46(46%) of these samples; 19 strains of Cl. perfringens, 9 strains of Cl. oedematiens A, B, 5 strains of Cl. sordelli, each 3 strains of Cl. chauvoei, Cl, oedematiens C.E, and Cl. difficile, 2 strains of Cl. sporogenes. The highest percentage of contamination by Cl. perfringens was found in beef products(26.5%), and the following(5.2%) in mackerel pike and none in baitop shell. whale, manna brand. and top shell. One of 19 isolates of Clostridium perfringens found in meat products was shown to produce heat resistant spores which resist $100^{\circ}C$ for 60 minutes and others were heat labile strains which is killed at $90^{\circ}C$ for 30 minutes. The distribution of Hobbs' serotype of 19 isolates were each 4 strains of type 6, 8, and 11, 1 strain of type 13 and others untypable. 19 Strains of Cl. perfringens were shown a marked susceptibility to cefamezin, lincomycin and minocin and relatively sensitive to vibraimycin, geopen, and chloramphenicol. A marked resistance to kanamycin, colimycin, and gentamycin were shown. Aerobic enteropathogens from samples were not recovered.

Micronized and Heat-Treated Lactobacillus plantarum LM1004 Stimulates Host Immune Responses Via the TLR-2/MAPK/NF-κB Signalling Pathway In Vitro and In Vivo

  • Although nanometric dead Lactobacillus plantarum has emerged as a potentially important modulator of immune responses, its underlying mechanism of action has not been fully understood. This study aimed to identify the detailed biochemical mechanism of immune modulation by micronized and heat-treated L. plantarum LM1004 (MHT-LM1004, <$1{\mu}m$ in size). MHT-LM1004 was prepared from L. plantarum LM1004 via culture in a specifically designed membrane bioreactor and heat treatment. MHT-LM1004 was shown to effectively induce the secretion of $TNF-{\alpha}$ and IL-6 and the mRNA expression of inducible nitric oxide synthase (iNOS). MHT-LM1004 enhanced the expression of TLR-2, phosphorylation of MAPKs (ERK), and nuclear translocation of $NF-{\kappa}B$ in a dose-dependent manner. Oral administration of MHT-LM1004 ($4{\times}10^9$ or $4{\times}10^{11}cells/kg$ mouse body weight) increased the splenocyte proliferation and serum cytokine levels. These results suggested that MHT-LM1004 effectively enhances early innate immunity by activating macrophages via the TLR-2/MAPK/$NF-{\kappa}B$ signalling pathway and that this pathway is one of the major routes in immune modulation by the Lactobacillus species.

Inhibitory Effects of Plant Extracts on Adjuvant-induced Arthritis

  • Twenty seven plant extracts were selected on the basis of ancient literature search for rheurnatoid arthritis or similar syndrome. Methanol extract of each plant was prepared and administered orally to rats everyday at a dose of 200 mg/kg/day. Experimental arthritis was induced by subplantar injection of heat-killed Mycobacterium butyricum to right hind paw of rats. This treatment provoked swelling of the treated paw in two phases, acute primary swelling and secondary arthritic swelling. An inhibition of secondary swelling was considered to be antiarthritic activity. Several plant methanol extracts such as Akebia quinata (caulis), Ephedla sinica (herba) and 5ophorae subprostratd (radix) were found to show significant inhibitory activity against secondary swelling at the dose tested. Our results strongly suggested an antiarthritic potential of these plant extracts.

  • Antitumor activity of Lactobacillus casei YIP 9018(LC9018) was studied in mice by using sarcoma 180(S-180) and Lewis lung carcinoma (3LL). Following the eatablishment of in vivo tumor models such as ascites form S-180, solid form S-180 and 3LL for estimating antitumor activity of Lactobacilli, optimal dose and injection route of heat-killed LC9018 for supperssion of local tumor were examined. Administration of 100ng/mouse of LC9018 significantly inhibited the growth of ascites form S-180, solid form S-180 and 3LL.

  • Crude enzymes of Trichoderma viride and Phanerochaete chrysosporium were evaluated for their effect to brightness and physical properties of recycled pulp from newspaper and corrugated container board. The brightness of recycled pulp from newspaper increased with crude enzymes from Trichoderma viride and Phanerochaete chrysosporium. The brightness of recycled pulp of corrugated container board increased with crude enzyme from Phanerochaete chrysosporium, but decreased with crude enzyme from Trichoderma viride. The Δbrightness with chemical bleaching of crude enzyme treated pulp was lower than that of heat killed enzyme treated pulp, but the final brightness of pulp was increased. Modification of recycled paper with crude enzymes can result in a substantial increase in physical properties with little no loss in pulp freeness.

  • The development of delayed-type hypersensitivity(DTH) reaction to Staphylococcus aureus in mice was studied, Mice received 3 injections of $10^8$ viable S. aureus subcutaneously showed a marked footpad swelling when mice were challenged with $10\;{\mu}g$ staphylococcal protein antigen into footpad(The percent increase of footpad thickness at 24 h after challenge wsa 35% approximately). Histological observation of footpad of immunized mice showed a marked thickness of subcutaneous tissue due to edematous reaction and massive infiltration of lymphocytes and neutrophils which are characteristic cells in DTH reaction. Intensity of DTH reaction of mice immunized with viable bacteria was much higher than that of mice immunized with staphylococcal protein or heat-killed bacteria. The DTH reaction to S. aureus could be transferred to normal recipient mice by both spleen cells and lymph node cells.

건조온도 및 아황산처리가 건조마늘의 품질에 미치는 영향 (Effects of drying temperature and sulfiting on the qualities of dried garlic slices)

  • 세절 마늘 건조중 갈변물질의 형성속도는 건조온도가 증가함에 따라서 증가하였다. 건조마늘의 pyruvate함량은 건조온도가 증가하거나 건조시간이 경과함에 따라서 감소하였다. Sodium metabisulfite는 세절된 건조마늘의 비효소적 갈변을 줄일 수 있었고, pyruvate 함량의 감소를 억제하였다. 아황산 처리는 antimicrobial activity를 갖는데, $65^{\circ}C$에서 대조구의 세균수가 $4.6{\times}10^5$이던 것을 $5.2{\times}10^4$으로 줄일 수 있었다.

  • CD1 분자는 결핵균 유래 지질항원 발현하는 단백질이며, 특히 수지상세포(dendritic cells)가 결핵균 감염시에 발현이 점차 감소함을 관찰하였다. 이는 결핵균의 사균이나 항원만으로는 관찰되지 않는 결과였다. 2차원 전기영동(2D electrophoresis)을 통하여 CD1b 의 인산화를 관찰하였고 이러한 현상이 식세포작용과 연관됨을 확인하였다.

Lethal Temperature for the Black Timber Bark Beetle, Xylosandrus germanus (Coleoptera: Scolytidae) in Infested Wood Using Microwave Energy

  • The thermal death kinetics of the Black timber bark beetle, Xylosandrus germanus, was investigated to develop a heat treatment for control of infested wood packing materials used to export goods. To determine the feasibility of microwave irradiation as an alternative control method, laboratory experiments irradiating wooden blocks of Douglas fir ($200{\times}200{\times}250mm$), which were artificially infested with adults, with 2.45 GHz of microwave energy. All (100%) Ambrosia beetle adults were killed by both hot water treatments and microwave irradiation at $52^{\circ}C$ and $58^{\circ}C$, respectively. Probit analyses estimated the internal wood temperature required to produce Probit (0.99) efficacy to be $64.7^{\circ}C$ (95% CI $62.4-69.9^{\circ}C$) at one minute after microwave treatment.
