Inhibition of Tyrosinase by Metabolites Originating from Thrichoderma atroviride (Thrichoderma atroviride 배양액의 tyrosinase 억제제에 관한 연구)

  • In today's society, functional whitening cosmetics are important to beauty. Fungi are known to produce a variety of whitening-related metabolites. In this study, we searched for tyrosinase inhibitors with metabolic products derived from Trichoderma atroviride supernatant in order to apply a material for whitening functional cosmetics. In addition, the inhibitory effect was compared to arbutin, which has already been approved as a whitening raw material by the Korea Ministry of Food and Drug Safety (KMFDS). The metabolites from the T. atroviride supernatant showed higher tyrosinase inhibitory activity than that of arbutin. Some of the tyrosinase inhibitors were stable to heat, whereas some were unstable. The heat unstable material was exhibited in the case of samples treated with little amounts, such as 0.02~0.2%. They were very unstable in acidic and alkali pHs, especially under acidic conditions. However, it was found that a weakly-acidic to neutral pH range was the optimal working pH, especially neutral pH. Since the activity of the inhibitory substances in the T. atroviride supernatant was maintained regardless of proteinase K treatment, it was assumed that the metabolites, but not the bioactive peptides, were involved in the activity. In summary, we propose that the metabolites derived from T. atroviride supernatant have strong potential as whitening raw material.

Microbiological and Physicochemical Changes of Vegetable Juices (Angelica keiskei and Brassica loeracea var. acephala) Treated by UV Irradiation (UV 조사 신립초 및 케일 녹즙의 저장기간에 따른 미생물 및 이화학적 변화)

  • A fresh juice has become a new functional food available for dieting and health. However, the shelf-life of vegetable juice is very short because of the absence of heat pasteurization process. To elongate the shelf-life of vegetable juices, such as Angelica keiskei and Brassica loeracea var. acephala, the changes of microbiological, chemical and sensory property by UV irradiation were investigated. The total aerobic bacterial numbers of A. keiskei and B. loeracea var. acephala vegetable juices were $3.2{\times}10^5$ and $7.0{\times}10^4\;CFU/mL$, respectively, after wring process. However, the numbers were $3.6{\times}10^3{\sim}9.7{\times}10^3$ and $3.7{\times}10^3{\sim}2.7{\times}10^4\;CFU/mL$ after UV treatment on wring juice, and this lower microbial number was maintained during storage. The number of coliform bacteria also reduced significantly by UV treatment, and the bactericidal effect was higher when the flow rate is slower. The increase of lightness and yellowness, and decrease of redness were observed after treatment of UV on both vegetable juices, but the differences were not significant between flow rates. The ascorbic acid contents of vegetable juices were reduced by UV irradiation regardless of flow rate, and storage. Overall acceptance in sensory analysis revealed that there was no significant difference between the control and vegetable juice irradiated UV at 0 days, but sample with UV treatment showed higher score at 3 days. Therefore, UV treatment on vegetable juice can elongate the shelf-life without any problems in flavor and color.

  • In previous paper(Lee et al., 1984), preparation formula and processing conditions of the fish meat (mackerel) paste using dielectric heating were described, that included the proper shape and size of product and the conditions of dielectric heating, hot air dehydration, and heating with electric heater to yield the minimum expansion and case hardening during heating and to controll the final rater activity of 0.86 to 0.83 accompanying with a complete reduction of viable cells and good texture. In present study, changes in VBN, pH, total plate count, water activity, texture, the loss of available lysine, color indexes, TBA value, and the content of TI were determined to assess the quality stability and shelf-life of the product during the storage for 35 days at $5^{\circ}C\;and\;25^{\circ}C$, respectively. And the effect of vacuum sealing and hot water treatment before storage on the storage stability of product was also mentioned. As the product was vacuum packed in K-flex film bag, heat treated in boiling water for 6 minutes, and stored, water activity was maintained 0.86 to 0.84 for 35 days regardless of storage temperature, and the increase of total plate count was negligible in case of $5^{\circ}C$ storage while tended to gain slightly after 25 days at $25^{\circ}C$ storage. Changes in VBN was also minimum with an increase of 1.5 mg/100g at $5^{\circ}C$ and 7.0mg/100g at $25^{\circ}C$, but in case of unpacked sample, it was 24.5mg/100g at $5^{\circ}C$ and 42.4 mg/100g at $25^{\circ}C$ even after 7 days. In textural property hardness tended to increase after 28 days and folding test score was down to A or B from AA grade. The loss of available lysine was $7.5\%\;at\;5^{\circ}C$ and $17.0\%\;at\;25^{\circ}C$ but brown color was not deeply developed as the color index score indicated. TBA value was not increased at $5^{\circ}C$ while it tended to increase rapidly after 30 days at $25^{\circ}C$. Changes in TI content was not obvious except that it showed a tendency of increase at the end of storage as well as in the change of lysine and TBA value. It is concluded from the results that the quality of the product, pasteurized and water activity controlled by dielectric heating, and vacuum packed in K-flex film would be stable for more than 35 days at $5^{\circ}C$ and at least 25 days even at room temperature.

Effect of ${\alpha}-Amylase$ Treatment of Brown Rice(Goami) Alcohol Fermentation By-Product (현미(고아미) 알코올발효 부산물의 ${\alpha}-amylase$처리 효과)

  • To utilize the non-heat treated alcoholic by-products of brown rice(Goami) as food sources, the quality characteristics change according to the treatment conditions of ${\alpha}-amylase$ were evaluated. It resulted that the increase of hydrolysis temperature correspondingly increased the soluble solids, total dietary fiber and total sugar in the by-products of Goami, and the highest reducing sugar content was observed at $80^{\circ}C$. The free amino acids contents were tended to slowly decrease by the hydrolysis temperature more than $70^{\circ}C$, and the highest content of oligosaccharides were detected at the hydrolysis temperature of $80^{\circ}C$. The soluble solid according to the ${\alpha}-amylase$ concentration resulted to increase with the increase of the enzyme concentration and the total dietary fiber revealed similarly showing approximately 0.65%. The high content of reducing sugars was observed at the enzyme concentration around 0.08%(v/w). Total sugars and oligosaccharides contents tend to increase as the concentration of enzyme increased, and the content of oligosaccharides acquired at the enzyme concentration more than 0.10%(v/w) maintained to show rather similar contents. The soluble solids and total dietary fiber by hydrolysis time were found to show 6.66% and 0.65%, respectively at more than 60 min of hydrolysis, and the reducing sugars and total sugars were found to be 3,600 and 4,800 mg% in all treatment groups showing no significant difference. The content of oligosaccharides was increased with the increase of hydrolysis time, and the content was similar at more than 90 min of hydrolysis by ranging around 2,100 mg%. Based upon these results, the by-products of Goami are expected to be used as various food sources showing the highest dietary fiber and oligosaccharides contents by the hydrolysis at $80^{\circ}C$ for 90 min with the addition of 0.10%(v/w) of ${\alpha}-amylase$.

Changes in Characteristics of Brown Rice (Goami) Alcohol Fermentation By-Product by Cellulase (Cellulase처리에 따른 현미(고아미) 알코올발효 부산물의 특성 변화)

  • To utilize non-heat treated alcoholic by-products of brown rice (Goami) as food sources, the quality characteristics changes according to the treatment conditions of cellulase were evaluated. Results showed that the increase of hydrolysis temperature correspondingly increased the soluble solids and total sugar amounts in the by-products of Goami, and total dietary fiber amount was found to be around 0.67% Reducing sugar concentration was the highest at the hydrolysis temperature of $70^{\circ}C$. Maltooligosaccharides amounts were detected to be the highest at the hydrolysis temperature of $80^{\circ}C$ and were also, maltopentose and maltopentose were found. In the soluble solid, total dietary fiber, reducing sugar and total sugar according to the cellulase concentration, the content of hydrolysates with enzyme were higher than control, and the content of hydrolysates with enzyme was similar (6.30 and 0.69% 3,600 and 5,500 mg% respectively). The content of maltooligosaccharides was increased with the increase of enzyme concentration, and the content was similar at more than 0.6%(w/w) of enzyme concentration. The soluble solids and total dietary fiber by hydrolysis time were found to be 6.25% and 0.70%, respectively at more than 60 min. of hydrolysis. The content of reducing sugar, total sugar and maltooligosaccharides were increased with the increase of hydrolysis time, and the content was similar at more than 120min. of hydrolysis (3,800, 5,680 and 1,950 mg% respectively). Based upon these results, the byproducts of Goami are expected to be valuable as various food sources showing the highest dietary fiber and maltooligosaccharides contents by the hydrolysis at $80^{\circ}C$ for 120 min. with the addition of 0.6%(w/w) of cellulase.

Effects of Prunellae Spica Extract on LT4-induced Hyperthyroidism in Rats through the Regulation of Heat and Cold Imbalance (하고초추출물의 갑상선기능항진증 랫트모델에서의 한열조절작용에 의한 개선효능 연구)

  • Objective : This study was intended to examine the effects of water extract of Prunellae Spica (PS), which is a herb with 'cold' nature based on hot and cold theory of traditional Korean medicine. Methods : Hyperthyroidism was induced in SD rats by LT4 (0.5 mg/kg, i.p.) daily for four weeks. After 2 weeks of LT4 injection, rats were divided randomly into four groups; normal, LT4-induced hyperthyroid control, PS extract (500 mg/kg, p.o.)-treated group, and propylthiouracil (PTU, 10 mg/kg, s.c.)-treated positive group. After 2 weeks of drug treatment, all rats were sacrificed and harvested blood samples and thyroid tissues. The changes of body weight, food and water intake, and body temperature were measured weekly. Serological markers were analyzed in sera using an enzyme-based assay, and thyroid tissues were stained with Hematoxylin & Eosin (H&E). Brain and dorsal root ganglion (DRG) tissues were isolated and analyzed the expression of transient receptor potential (TRP) channels by Western blot. Results : PS extract administration attenuated the loss of body weight and the increase of body temperature in LT4-induced hyperthyroidism rats. PS extract increased the level of thyroid stimulating hormone (TSH) and decreased tiiodothyronine (T3) and tetraiodothyronine (T4). In action mechanism, PS extract regulated the expression of transient receptor potential channel subfamily V member 1 (TRPV1) and transient Receptor Potential channel subfamily M member 8 (TRPM8), the thermoregulators. Conclusion : To conclude, PS extract can improve the symptoms of hyperthyroidism through regulation of the thyroid hormones imbalance and thermoregulation via TRP channels.

Evaluation of Immune Enhancing Activity of Luthione, a Reduced Glutathione, in RAW 264.7 Macrophages (RAW 264.7 대식세포에서 환원형 glutathione인 luthione의 면역 증강 활성 평가)

  • Although glutathione (GSH) has been shown to play an important role in the prevention of oxidative damage as an antioxidant, studies on immune regulation by it have not been properly conducted. In this study, we investigated whether luthione®, a reduced GSH, has an immune enhancing effect in murine macrophage RAW 264.7 cells. The results of flow cytometry and immunofluorescence experiments indicated that luthione increased phagocytic activity, a representative function of macrophages, compared to the control cells. According to the results of the cytokine array, the expression of interleukin (IL)-5, IL-1β, and IL-27 was significantly increased in the luthione-treated cells. Luthione also enhanced the production of tumor necrosis factor-α and IL-1β through increased expression of their proteins, and increased release of the immune mediators such as nitric oxide (NO) and prostaglandin E2 was associated with increased expression of inducible NO synthase and cyclooxygenase-2. In addition, the expression of cluster of differentiation 86, an M1 macrophage marker, was dramatically enhanced in RAW 264.7 cells treated with luthione. Furthermore, as a result of heat map analysis, we found that cytokine signaling 1/3-mediated signal transducer and activator of transcription/Janus tyrosine kinase signaling pathway was involved in the immunomodulatory effect by luthione. In conclusion, our data suggested that luthione could act as a molecular regulator in M1 macrophage polarization and enhance immune capacity by promoting macrophage phagocytic function.

Changes of Indicative Substances According to Heat Treatment of Milk (우유의 가열처리에 따른 지표물질의 변화)

  • This study was carried out to analyze the physicochemical properties of bovine milks, which were heated with LTLT, HTST, UHT pasteurization and UHT sterilization methods and to compare the heat intensity among the heating methods and samples. The mean HMF values per liter milk were measured as 0.66~1.62 $\mu$M (LTLT), 0.9~1.78$\mu$M (HTST), 3.53$\mu$M(UHT pasteurized) and 7.43~8.97$\mu$M (UHT sterilized) in samples, re- sportively. The available Iysine contents per 100ml milk showed 293.2 mg (Raw), 289.2~291.2 mg (LTLT), 298.4~292.4mg (HTST), 272.4~261.6mg (UHT pasteurized) and 279.0mg (UHT sterilized), respectively. The rates of whey protein denaturation were 9.5~11.4% (LTLT), 9.5~17.1% (HTST), 89.3~95% (UHT pas-tsterilized) and 62.7% (UHT sterilized), respectively. The contents of SH groups per g protein were determined as 2.86$\mu$M (Raw) and 2.95~3.15$\mu$M (LTLT), 3.08~3.18$\mu$M (HTST), 3.26~3.42$\mu$M (UHT Pasteurized) and 3. 36$\mu$M (UHT sterilized), respectively, The SS groups Contents per g protein were 28.93$\mu$M (Raw), 25.72~26. 51 $\mu$M (LTLT), 26.93~26.79$\mu$M (HTST), 23.65~23.04 $\mu$M (UHT pasteurized) and 24.69$\mu$M (UHT sterilized), respectively. The ascorbic acid contents per liter milk were measured 6.05mg (Raw), 1.47~1.65mg (LTLT), 2.50~3.85mg (HTST), 2.87~3.69mg (UHT pasteurized) and 4.50mg (UHT sterilized). The changes of some in-dices in milk samples depend on the heating temperature and time ; the HMF values, SH groups, whey protein denaturation rates increased, while the available lysine contents and SS groups decreased in LTLT, HTST, UHT pasteurized and UHT sterilized milks. No remarkable differences were found in heating indicators between LTLT and UHT milks.

Antimicrobial Activity of Soy Protein Hydrolysate with Asp. saitoi Pretense (콩 단백 효소 가수분해물의 항균활성)

  • Soy protein was hydrolyzed by 5 different pretenses and determinated antimicrobial activity of each hydrolysate. The soy protein hydrolysate treated by pretense from Aspergillus saitoi showed the highest antimicrobial activity among the protease studied and was used for further analysis. Soy protein hydrolysate was fractionated by ultrafiltration for M.W. 10,000,3,000 and 1,000. The M.W 1,000∼3,000 showed the highest antimicrobial activity. The minimum inhibition concentrations of obtained fraction were 0.5∼0.8 mg/mL for gram positive and negative microbials, and its activity was even observed after heating at 121$^{\circ}C$ for 10 min, suggesting that hydrolyzed protein having antimicrobial activity is quite heat-stable. Reverse-phase HPLC was further applied to separate the fraction and 8 peaks were found. Each 8 peaks were separated and pooled and measured antimicrobial activity. Among them, retention time of peak at 16.02 min showed the prominent antimicrobial activity.

Desmutagenic Effects of Maillard Reaction Products against Mutagenic Heterocyclic Amines (변이원성 Heterocyclic Amine에 대한 Maillard 반응생성물의 변이원성 억제효과)

  • Each molecular weight (Mw) fraction of melanoidins prepared from a D-glucose and glycine system, i. e., Mw below 1,000, Mw between 1,000 to 5,000 and Mw above 5,000 and nondialyzable and ozone-treated melanoidins were reacted with heat-induced mutagens such as Trp-P-1, Trp-P-2, Glu-P-1, Glu-P-2 and IQ at $37^{\circ}C$ for 30 min. The inhibitory effects of the melanoidins on the mutagens increased with increasing molecular weight. The reducing ability ana antioxidative activity of melanoidins also increased in proportion to the increase in molecular weight, whereas the mutagenic inhibitory effect decreased on reduction of the melanoidins with sodium borohydride. It was also observed that a part of Trp-P-1 was adsorbed to melanoidin molecules. On modification of amino groups of these mutagens with carbonyl compounds derived through the Maillard reaction such as diacetyl and glyceraldehyde, their mutagenic activities were remarkably suppressed. Accordingly, it is speculated that the mutagenic inhibitory action of melanoidins is due to their reducing ability and antioxidative activity, and electrostatic binding and carbonyl groups of the melanoidin molecules.
