Characterization of In vitro Propagated Plants Via Somatic Embryo Formation from Old Wild Panax ginseng

  • Wild Korean ginseng has been recognized as highly precious medicine since ancient times. Nowadays, the population of wild ginseng in the forest of Korean peninsula is very rare due to indiscreet harvest. In this work, we investigated the plant regeneration via somatic embryogenesis from embryogenic callus of old wild ginseng (more than 50 years-old) and compared the features of plants regenerated from 5-years old and 50 years-old ginseng. Induction of embryogenic callus from adventitious roots of 50 year-old wild ginseng required 83 weeks of culture, but only 10 weeks were sufficient for 5 year-old ginseng. Height and width of plants derived from the old wild ginseng was smaller and slender compared to the plantlets derived from 5 year-old ginseng. Total chlorophyll contents was 2-6 time lower in plantlets regenerated from 50 year-old wild ginseng than those from 5 year-old ginseng, but anthocyanin content was higher in 50 year-old ginseng. Our results revealed that plants regenerated from old wild ginseng have different morphological and physiological characters probably due to age-dependent phenomenon.

Forage Productivity of Phragmites communis according to Harvest Management in Autumn (가을철 갈대의 수확관리에 따른 이듬 해 조사료 생산성 비교)

  • This study was carried out to determine the harvest management during autumn on the forage production, quality and regrowth of following year of Phragmites communis as native grass. Field experiments were conducted in Ansan and in Cheonan from 2010 to 2011. Forages were cut at 3 cm, 15 cm and 25 cm in height as compared to control (no harvest) in Ansan plots, and forages grown in Cheonan plots were cut at 10 cm and 20 cm in height with control (no harvest). The forages were harvested one time on Oct 7, 2011 in Ansan, and were harvested twice on July 7 and on October 7 in Cheonan. In Ansan, dry matter (DM) yield of control plot the following year were higher than those from harvested plots. In vitro digestible DM (IVDDM) yields, however, were the same: 1,236 kg/ha from control, 1,234 kg from 15 cm cut and 1,241 kg from 25 cm cut plots, except that lower IVDDM yield from 3 cm cut plot than these were observed. Forage quality of control plot was poorer than those of harvested ones. In vitro DM digestibility (IVDMD) and relative feed value (RFV) of control were 39.98% and 65.3, respectively as compared to harvested plots (42.36~46.40% IVDMD and 67.8~72.5 RFV). Yield and quality from Cheonan plots were similar to those from Ansan plots. Annual yield in DM and in CP from control plot were a little higher than those from harvested plots. However, there were no difference found in IVDDM yield between 20 cm cut (5,354 kg/ha) and control (5,178 kg). But IVDDM of 10 cm cut forages was less (4,531 kg). Forage quality scores were better from 20 cm cut, and 10 cm cut plot than control, in order (p<0.05). The quality grades were the 5th and 6th for forages from harvested plots and control plot, respectively. Regrown Phragmites communis from all Cheonan plots ranked the 4th in forage quality. More than 70% of annual yield was observed from the first grown forages. One time harvest per year was desirable for following regrowth and long time stable production of Phragmites communis. We recommended that Phragmites communis be harvested once for more forage yield with higher quality, and that optimum cutting height is 15~20 cm.

Estimation of Optimal Harvest Volume for the Long-term Forest Management Planning using Goal Programming (장기산림경영계획의 목표수확량 산출을 위한 목표계획법의 적용)

  • To facilitate the sustainable forest management, Forest Service in Korea has assigned 2.9 million hectare forests as 'intensive management forests' and encouraged local governments to develop a strategic management plan for their forests. One of problems for the sustainable forest management in Korea is the skewed distribution of forest age classes. Currently the majority of forestlands in Korea is occupied by age classes III and IV. In this study, we intended to find an optimum harvest volume, which enable one to make the intensive management forest in Youngdong-Gun evenly distributed for the age classes and allow an even harvest volume through a 50 year time horizon. To develop an optimization model, we applied the goal programming technique which is adequate for a multi-purpose management planning. The results indicated that it is necessary to harvest 1.2 million cubic meters in each decade to achieve the most stable distribution of age classes for the study site. The harvest volume target resulted from this study would be used in a management planning or an associated policy making process in the future.

Effect of Seed Mixture and Organic Fertilizer Application on Productivity, Botanical Composition and Forage Quality in Middle Mountainous Pasture (중부지역 초지에서 혼파조합 및 유기질 비료 시용이 목초 수량, 식생 구성 및 사료가치에 미치는 영향)

  • This experiment was carried out to determine the effect of organic fertilizer and seed mixture on productivity, botanical composition and forage quality in middle areas(Geumsan, 250m sea level) pasture. Total six experimental pastures (orchardgrass, timothy and tall fescue based seed mixture and with or without organic fertilizer) were established in autumn, 2014 and evaluated productivity and agronomic characteristics from 2015 to 2016. Plant heigh was higher in orchardgrass based mixture and orchardgrass and tall fescue based mixture were higher in $2^{nd}$ harvest. There was not found the significant difference among treatment(P<0.05) in dry matter (DM) content. Botanical composition in $1^{st}$ harvest was higher weed portion in orchardgrass based mixture and legumes was disappeared at $3^{rd}$ harvest in all treatments. Fresh and dry matter yield was higher in $2^{nd}$ year than $1^{st}$ year. In $1^{st}$ year, orchardgrass based mixture with organic fertilizer plot was the highest(P<0.05) DM yield, but tall fescue based mixture with organic fertilizer was the highest in $2^{nd}$ year. Content of crude protein (CP) was the highest in tall fescue based mixture and Timothy based mixture showed low in ADF (acid detergent fiber) and NDF (neutral detergent fiber) content. But the content if in virto dry matter digestibility was higher than others. In $2^{nd}$ year, the nutritive value of $2^{nd}$ and $4^{th}$ harvested grasses were higher and average RFV (relative feed value) value was the highest at $4^{th}$ harvest. According to this result, orchardgrass and tall fescue based mixture would be recommendable for farm who considering productivity and timothy based mixture would be recommendable for considering forage quality in middle areas of Korea.

The saponin contents of cultivated Anemarrhena asphodeloides B$_{UNGE}$ in Korea (재배지모의 saponin 함량에 관한 연구)

  • Optimum condition for cultivation of Anemarrhena asphodeloides B$_{UNGE}$ and seasonal variation of saponin and sapogenin contents in the rhizome were investigated. It is confirmed that the best harvest time is 3rd year. Average contents of saponin in rhizome was 8.5% and 5.0%, in fibrous roots, being highest in winter. The facotrs of saponin content variation were seemed to be temperature and humidity. It was also proved that asphonin was the mixture of timosaponin A-III, timosaponin A-I and mangiferin. Mangiferin and isomangiferin were isolated from the flower.

Early Effect of Environment-friendly Harvesting on the Dynamics of Organic Matter in a Japanese Larch (Larix leptolepis) Forest in Central Korea (중부지역 일본잎갈나무림의 친환경벌채가 산림 내 유기물 변화에 미치는 초기 영향)

  • Environment-friendly harvesting is practiced to maintain ecosystem, landscape, and forest protection functions. The present study was conducted at Simgok-ri, Sinbuk-myeon, Pocheon, Gyeonngi-do, where a 41-50-year-old Japanese larch forest was harvested in an environment-friendly manner from 2017 to 2019. The dynamics of organic matter in this forest were investigated at three years after the harvest. Specifically, organic matter content was measured on the forest floor and in overstory biomass, litterfall, and soil up to 30 cm in depth from June 2020 to January 2021. Owing to the harvest, the amount of overstory biomass of the Japanese larch stands decreased from 142.22 to 44.20 t ha-1. On the forest floor, the amount of organic matter was 32.87 t ha-1 in the control plots and 23.34 t ha-1 in the harvest plots. Annual litterfall was 4.43 t ha-1 yr-1 in the control plots and 1.16 t ha-1 yr-1 in the harvest plots. Soil bulk density in the B horizon was 0.97 g cm-3 in the control plots and 1.06 g cm-3 i n the harvest plots. Soil organic matter content was 11.5% in the control plots and 12.8% in the harvest plots. The total amount of soil organic matter did not differ significantly between the control plots (245.21 t ha-1) and harvest plots (263.92 t ha-1), although the amount of soil organic matter tended to be higher in the harvest plots. The total amount of organic matter in the forest was estimated to be 406.48 t ha-1 in the control plots and 338.21 t ha-1 in the harvest plots. In the harvest plots, the ratio of aboveground organic matter decreased to 13.1% and soil organic matter increased to 78.0%, indicating that the distribution of organic matter changed significantly in these plots. Overall, the carbon accumulated in aboveground biomass was substantially reduced by environment-friendly harvesting, whereas the soil carbon level increased, which played a role in mitigating the reduction of system carbon in the forest. These results highlight one possible resolution for forest management in terms of coping with climate change. However, given that only three years of environment-friendly harvesting data were analyzed, further research on the dynamics of organic matter and tree growth is needed.

A Population ecological study of the hen clam(Mactra chinensis) in the Dong-li self-regulatory community of Busan (부산 동리어촌계 개량조개, Mactra chinensis의 자원생태학적 연구)

  • This study was performed to estimate biomass and to provide management plan through population ecological characteristics, including growth parameters, survival rate, instantaneous coefficient of natural and fishing mortalities, and age at first capture of hen clam, Mactra chinensis, in the Dong-li self-regulatory community of Busan. The von Bertalanffy growth parameters estimated from a non-linear regression were $SH_{\infty}=86.24mm$, K=0.12/year, and = -1.37year. Survival rate(s) of the hen clam was 0.515. The instantaneous coefficients of natural mortality(M) was estimated to be 0.232/year and fishing mortality(F) 0.432/year for hen clam. The current biomass of the hen clam in the study area was estimated to be 713mt and the acceptable biological catch(ABC) was estimated under various harvest strategies based on $F_{0.1}$ and $F_{40%}$.

Changes of Root Yield and Essential Oil Content by Cultivated Years in Asaram siebold Mio (세신의 재배년차에 따른 근수량 및 정유성분 함량변화)

  • This experiment was conducted to investigate the change of root yield and essential oil contents by cultivated year in Asaram siebold Mio. Growth of aerial part such as plant height, leaf length and leaf width increased rapidly in two to three-year-old. Root length and root weight per plant were increased as cultivation year passed. Especially the speed of development was very fast in two and three-year-old alike aerial part. Root yield was the highest at five-year-old. The content of essential oil was decreased gradually as cultivation year passed. Methyleugenol content was increased, while safrole content was decreased by cultivated years. The increasing of methyleugenol content in four to five-year-old was not higher than three to four-year-old. In the result, it concluded that the optimum harvest time of Asarum sieboldi MiO is five-year-old cultivated based on root yield and effective medical components.

Effects of Different Pruning Time on Bush Growth and Fruit Production of Southern Highbush Blueberry 'Scintilla' Cultivated in a Heated Plastic House (전정시기가 가온재배 남부하이부쉬 블루베리 '신틸라'의 신초 생장과 과실 생산에 미치는 영향)

  • Appropriate pruning is very important for southern highbush blueberries cultivated in a heated plastic house to control their severe crowding within the bush canopy after harvest. Pruning treatments at different times were evaluated to find out an appropriate pruning time to southern highbush 'Scintilla' cultivation with heating. Seven-year-old (2018) bushes, grown in 180-L containers, were summer-pruned on May 20 (35-39 days after harvest) and June 20, or dormant-pruned on December 20 (5 days before flowering), consecutively in both 2018 and 2019 removing 30% of the total woods. May pruning activated occurrence of shoots the following years, increasing number of shoot by 17 to 49% and total shoot length by 18 to 32% compared with those of the dormant pruning. Fruit characteristic was not significantly affected by different pruning times the previous year. The first year pruning treatment did not influence the yield the following year, but the second year consecutive May pruning significantly increased yield per bush by 7% compared with the dormant pruning. The results indicated that summer pruning in May could be favorable to promote shoot growth and to maintain stable yield.
