Effects of Scarification and Soaking Treatment on Germination of Sword Bean Seed (작두콩 종자의 종피파상과 침지처리가 발아에 미치는 영향)

  • Seed coat of sword bean (Canavalia gladiata) is very thick and hard, it is difficult to absorb water during germination and it requires much time that cotyledon come out from seed coat since seminal root appearanced. Therefore this experiment was carried out to increase the germination rate by easing water absorption through mechanical scarification on seed coat. Non-scarification seed did not germinated at 7 days after treatment but scarification seed showed germination rate of 98%. Therefore mechanical scarification of seed coat was increased germination rate. Non-scarification seed absorbed less than 10% water of dry weight at 8 hours after treatment but scarification seed absorbed about 90% water at 7 hours after treatment and more than 150% water at 8 hours and swelled about 1.4-1.5 times in length and width of seed. Germination rate of scarification seed was high under high temperature and it was 96% and 93% at $25^{\circ}C$ and 3$0^{\circ}C$, respectively. The rate of cotyledon emergence on clay loam soil was the highest among bed soils. Soaked seed after mechanical scarification in distilled water was germinated faster than non-soaked seed on early in bed soil. The rate of cotyledon emergence was more than 92% at 9 days after sowing but non-scarification and soaked seed was germinated late and showed very low germination rate as 67%.

Present Status and Prospects of Horticultural Seed Industry (원예종묘산업의 현황과 발전전망)

  • The horticultural seed industry has made great progress in the last fifty years of its history after the independence of this country. The industry has accomplished self-sufficiency in major vegetable seeds and has even gained international competitive edge in certain crop seeds, particularly in hybrid cultivar development. However, the industry is facing a crisis at present coming from excessive competition among the domestic seed companies as well as the foreign currency crisis of the country. Several major seed companies have already been acquired by multinational corporations. Many people in the country as well as agriculturists are concerned about this situation. Although it is true that the industry is undergoing hard times, this crisis can be turned into a chance for making new progress depending on the joint efforts of the groups responsible, i.e., private companies, public institutions, and policy makers. Therefore, we have to turn this crisis into an opportunity for self-reform and progress. We should not be discouraged or give up. We Propose some ideas for the Progress of the seed industry. Public institutions and policy makers should do such things in cooperation as 1) training human resources for future breeders, 2) securing a wide range of genetic resources and improvement of the management system, 3) basic science research including biotechnology, 4) releasing breeding stocks from germplasm enhancement through use of exotic accessions, 5) support for breeding region- or use-specific cultivars, and 6) collection of foreign agricultural information for breeding cultivars for overseas markets. Eventually the responsible group for the final development of commercial cultivars, production, sales and management is private companies. Therefore, private companies also should have to do their best to develop competitive cultivars with a missionary spirit and endeavor for both domestic and overseas markets in cooperation with public institutions. Management based on competition, professionalism and rationalism is also urged. We are going to conclude with optimism that we can make a new vault of progress once the private and public sectors closely cooperate for the development of Korean horticultural seed industry by overcoming this crisis.

Change of Seed Dormancy and Viability of Chinese Milk Vetch (Astragalus sinicus L.) in Rice Field (토양속에서 자운영 종자의 휴면성 및 종자활력 변화)

  • Experiments were conducted to investigate seed persistence of Chinese milk vetch(CMV) in naturally reseeded rice field in 2007~2009. The seed and pods with seeds were buried in rice field at 0, 5 and 10 cm depths and retrieved from the field at one to three month intervals from July to the following year March and determined change of seed dormancy and viability. In the second experiment, persistence of the CMV seeds in the naturally reseeded rice field at different tillage methods and soil depths were also investigated after rice harvest in autumn. Burial depths and durations affected recovery rate, dormancy and viability of CMV seed. The viability loss was faster and greater in the seed than the pod with seeds and on the soil surface than the 5 or 10 cm burial depths. The recovery rate of CMV seed was decreased starting from one month as seed burial and it was significantly decreased to 52~65% for the seed in September. However, unlike the seed burial, the nearly 100% CMV seeds were recovered for burial as pod with seeds even after four months burial in both 0 and 5 cm depths. However, the recovery rate was sharply declined to below 30% at October in 2007 in both seed and pods with seeds and in the 2008/2009 experiment. the 15~47% of CMV seeds still remained even after October. The CMV had high seed dormancy of 95%, showing only 4~5% germination at the beginning in June but the seed germination increased to 25 to 35% in seed and 55 to 61% in pod with seeds in September due to breakage of hard seed dormancy. The viability loss was faster in the seed than in the pod with seeds regardless of depths of placement in the soil base on decayed seeds. Also the seed placed on the soil surface lost viability faster than the 5~10 burial depths. On the other hand, field observation in the naturally reseeded CMV rice field showed that as many as 917~2,185 CMV seeds $m^2$ were from the 0~15 cm soil depth in the rotary tillage and 250~10,105 CMV seeds in minimum tillage treatmints. The recovered seed germinated 25~33%, 23~43% but still had high percentage of hard seed having 64~72% and 51~77% even after rice harvest in autumn. These results indicate that freshly harvested CMV seeds had high level of primary dormancy and the dormancy was gradually broken in soil with time during rice cultivation periods and appreciable number of CMV seeds remained even 4 month after burial in soil. CMV plant regenerated naturally from the remained seed bank at rice harvest time in autumn. The CMV seedling still emerged even after 2 years of continuous destructive killing of emerged CMV plant by rotary tillage in naturally reseeded CMV plant in rice field, indicating that CMV seeds do persistent as least two years in soil.

Growth and Root Yield in Progeny the Derived from Different Bolting Years in Angelica gigas Nakai. (참당귀(當歸) 추태년차별(抽苔年次別) 채종종자(採種種子)의 후대생육(後代生育) 및 수양(收量))

  • This studies were conducted to basic information on growth characteristics and root yield in progeny the derived from different bolting years and to establish optimum seed production time in Angelica gigas. Plant height of early and mid-growing stages had decreased with each year increase in bolting year, but showed no significance in late growing stage at different bolting years. Root charac­teristics including root length, root diameter and dry root weight was slightly good and it had high root/ shoot ratio in third year seed production. respectively. Bolting rate showed that first year seed 38.1%, second year seed 10.4% and third year seed 1.2%, respectively. Therefore, optimum seed production time revealed third year seed for reduce bolting rate. Root yield at different bolting years showed average 1,690kg per hectare as fresh root weight on first year seed, 2,860kg on second year and 2,940kg on third year seed, respectively. A significant positive correlation was appeared between bolting rate and plant height in mid-growing stage. On the other hard, there was highly significant negative correlation between bolting rate and root yield.

The Effects of Cone Harvesting on the Regeneration of Korean Pine and the Life of Animals in Mt. Changbai Nature Reserve

  • Based on the former researches, this article studied the influence of cone harvesting of Korean pine on the regeneration of Korean pine trees and the life of animals in Mt. Changbai Nature Reserve. When the cone matures, scales of the cone do not open so the seeds can not be released automatically. And the seeds, if left inside, are hard to germinate and can not grow further into seedlings. The seeds of Korean pine have deep dormancy characteristics. Natural regeneration of Korean pine is very poor under mother trees. Hoarding behavior of dispersing animals not only helps animals for food shortage period but also contributes to the dispersion of seeds of Korean pine. Among those hoarding animals, squirrel and Eurasian nutcrackers are found to be the most important dispersing agents for the seeds of Korean pine. After cone harvesting, the number of those dispersers reduced a lot since the seeds of Korean pine are very important food for them. Seed quantity of Korean pine on surface layer became very few and most of them only showed single distribution. Most of the seeds were buried under litter layer and showed a single or 2-4 seeds/cluster distribution. The case of more than 4 seeds in one cluster was few. The seed quantity of Korean pine forest on steep slopes of the research area was only 0.3% of the seed quantity in 1980 for the same forest type. If seed source of Korean pine are not protected, Korean pine forest in Mt. Changbai Nature Reserve would not maintain present feature in the future.

Effects of Environment and Storage Condition on Germination of Astragalus membranaceus (환경 및 저장조건이 황기 종자발아에 미치는 영향)

  • This experiment was conducted to investigate the effects of temperature, light, seed harvesting time, storage temperature, and period on its germination of Astragalus membranaceus. Optimum germination temperature was ${20{\sim}25^{\circ}C}$ and it was found to be light insensitive seed. The germination rates were not significantly different between 1-year and 2-year old plants, and seed harvesting times. Seeds stored at such cold temperatures as ${-4^{\circ}C\;and\;-20^{\circ}C}$ for 15 months showed over 80% in germination rate at the 6th day after placement. On the other hand, low germination rate of 60% at the 11th day was observed from the seeds stored at $4^{\circ}$ and room temperature.

  • The powder characteristics of seeded alumina prepared from alkoxide by sol-gel method were studied. When ${\alpha}$-Al2O3 seeded powders used, these ${\alpha}$ phase transformation temperatures decreased than those of unseeded powders by 110$^{\circ}C$ and fine powders under 0.1 $\mu\textrm{m}$ could be obtained. When Fe-nitrate added powders used, fast transformation rate resulted from ionic effects of Fe3+, but hard aggregated morphology exhibited. When ${\alpha}$-Al2O3 and Fe nitrate simultaneously added, these powders represented lower transformation temperature but resulted in microstructure with aggregated particles.

  • Solomon's seal (Polygonatum odoratum) seedlings raised through its seeds can replace the rhizomes impelling higher cost for transplanting, This experiment was done to determine the seed characteristics and the germinating processes to give some information on bulk production of seedlings using the seeds. The external or internal morphology of seeds or seedlings grown in lab. or greenhouse was examined mainly with stereomicroscope. The external shape of Solomon's seal seed was hard seed-coat and orthotropous ovule with linear type embryo stretching to the center of seed. Germination proceeded through the several steps. The lower part of seed embryo having the primordia of bulbil and roots first grew before the bulbil and roots was developed from the primordia. The lower part of embryo was enlarged toward the endosperm of seed as soon as seed germinated. Then epicotyl was formed on the apex of bulbil. The epicotyl was elongated after at least 6-week chilling treatment for breaking its dormancy and the first leaf shape was affected by light intensity given during seedling emergence. The bulbil was the first organ of the rhizome used as tea or herb medicine.

  • Seed coat of hairy vetch is very thick and hard, and difficult to absorb water during germination. It requires much time that cotyledon comes out from seed coat. Therefore this experiment was carried out to increase the germination rate by enhancing water absorption through water-soaking of seed, chemical scarification by sulfuric acid, and mechanical scarification on seed coat. Water-soaking for 5 hours seemed to be highly effective in enhancing germination rate. Although not significant, water-soaking resulted in increase of germination rate by 9%. Effect of soaking temperature was not significant, but early germination rate was increased to be 5.0%, 31.7% and 48.3% at $10^{\circ}C$, $20^{\circ}C$ and $30^{\circ}C$, respectively. Mechanical scarification of seed coat led to a germination rate of 97% whereas intact seed showed that of 65%. Chemical scarification by sulfuric acid for 10min, 20min and 30min resulted in a germination rate of 76.7%, 74.7% and 96.0% respectively. It is clear that scarification increased germination rate.

Application of Seed Vigor Test for Predicting Field Emergence in Azuki Bean (Vigna angularis Wight) (팥 포장출현력 예측을 위한 종자세 검사)

  • Field emergence of Azuki bean is poor due to hard seed coat as compared to other legumes. In this study, an attempt was made to develop prediction method with regression analysis based on various seed vigor tests in laboratory for field emergence of azuki bean. Azuki bean seeds artificially aged to provide various levels of seed quality were evaluated by the standard germination test (SGT), cold germination test (CT), cool germination test (CGT), complex stressing vigor test (CSVT), tetrazolium(TZ) vigor test and electroconductivity test. The SGT was suitable for predicting the field emergence in the unaged high vigor seeds. The abnormal seedling percentage and shoot length in the CGT were highly correlated with field emergence of moderate vigor seeds artificially aged for 2 days. Electroconductivity, seed viability in the CSVT, and vigor and predicted germinability in the tetrazolium vigor test were also useful for predicting field emergence. Percent of ungerminated seed in the CSVT was correlated with field emergence in the low vigor seeds artificially aged for 4 days. In a stepwise multiple regression analysis, seed viability in the SGT, normal seedling percentage and dry matter weight in the CGT accounted for 86.9% of the predicted value of field emergence in azuki bean.
