Screening of Antioxidative Effect of Combined Medicinal Plants on Oxidative Stress (산화적 스트레스에 대한 복합 한약재의 항 산화활성 검색)

  • Reactive oxygen species (ROS) are known to cause oxidative modification of DNA, proteins, lipids and small cellular molecules and are associated with tissue damage and are the contributing factors for diabetes, inflammation, aging, cancer, arteriosclerosis, and hypertension. We screened the anti-oxidative effect on V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with eleven extracts of combined medicinal plants. Dancheonhwankakambang and Samikangyabtang were found to show the scavenging activities of DPPH radical and intracellular reactive oxygen species, which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA).

Yeast hydrolysate as a low-cost additive to serum-free medium for the production of human thrombopoietin in suspension cultures of Chinese hamster ovary cells

  • To enhance the performance of a serum-free medium (SFM) for human thrombopoietin (hTPO) production in suspension cultures of recombinant Chinese hamster ovary (rCHO) cells, several low-cost hydrolysates such as yeast hydrolysate (YH), soy hydrolysate, wheat gluten hydrolysate and rice hydrolysate were tested as medium additives. Among various hydrolysates tested, the positive effect of YH on hTPO production was most significant. When 5 g/L YH was added to SFM, the maximum hTPO concentration in batch culture was 40.41 ${\mu}g/mL$, which is 11.5 times higher than that in SFM without YH supplementation. This enhanced hTPO production in YH-supplemented SFM was obtained by the combined effect of enhanced $q_{hTPO}$ and increased culture longevity. In addition YH supplementation did not affect in vivo biological activity of hTPO. Taken together, the results obtained demonstrate the potential of YH as a medium additive for hTPO production in serum-free suspension cultures of rCHO cells.

  • This study was done to investigate the effects of Gamichihyo-san and Gamiijung-tang on airway mucus secretion. After administer Gamichihyo-san(GCHS) and Gamiijung-tang(GIJT) extract to Golden Syrian Hamster for 8-10 weeks, we examined mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Following results were obtained; GCHG significantly stimulated mucin release from cultured HTSE cells, with minute cytotoxicity GIJT did not affect mucin release and have no cytotoxicity; GCHG and GIJT did not affect contractility of isolated tracheal smooth muscle. These results suggest that Gamichihyo-san might be usefully applied for airway mucus secretion.

  • 본 연구는 Chinese hamster ovary (CHO-KI) 배양세포에서 S-15분획에 의해 활성화된 bezo(a) pyrene (BP)과 3-methylcholanthrene(MC)에 의해 유발된 DNA단사절단과 DNA복제억제 및 그 회복과정에 미치는 한국산 인삼추출물 saponin의 영향을 조사하여 다음과 같은 결과를 얻었다. S-15분획으로 활성화된 10-5 M의 BP 또는 MC를 1.0-10ng/ml의 saponin과 함께 처리할 경우 BP와 MC단독처리군에 비해 DNA 단사절단률이 감소하였다. DNA 합성률은 활성화된 10-5M의 BP와 10-6 M의 MC에 의해 각각 50% 및 75% 억제되었으나, 0.1-10ng/ml의 saponin을 동시에 처리할 경우 DNA 합성억제률이 약 10% 이상 둔화되었다.

Effect of Erythromycin on Basal and Stimulated Mucin Release from Cultured Hamster Tracheal Surface Epthelial Cells

  • In the present study, we investigated whether lipopolysaccharide induce mucin release and erythro-mycin affect basal and adenosine triphosphate-induced (stimulated mucin release, from airway goblet cells. Confluent primary hamster tracheal surface epithelial cells were metabolically radiolabeled and chased for 30 min or 24 hr in the presence of varying concentrations of lipopolysaccharide or erythromycin to assess the effects on $^3H$-mucin release. The results were as follows : 1) Lipopolysaccharide failed to induce mucin release, 2) Erythromycin showed no effect on both basal and stimulated mucin release during 30 min of 24 hr treatment period. We conclude that lipopolysaccharide and erythromycin can not affect mucin release by direct acting on airway mucin-secreting cells.

  • 2- [(4- Cyanophenyl)amino] -3-chloro-1, 4- naphthalenedione (NQ-Y15) was asssayed for its genotoxic potential by using Salmonella typhimurium reversion assay and in vitro chromosome aberration test on Chinese hamster lung cells. In the Ames test, NQ-Y15 induced his + revertants of Salmonella typhimurium TA 98 and TA1537, reaching levels twice the negative control values. But, NQ-Y15 induced only his+ revertants of Salmonella typhimurium TA1537 more than twice the control values under the condition with metabolic activation system. In the cytogenetic test on chinese hamster lung cells. NQ -Y15 showed significant chromosomal aberrations, but the incidence was significantly reduced in the presence of metabolic activation.

Identification and Characterization of Rodent Germ Cells-Specific Hyaluronidases

  • Germ cell-specific hyaluronidases such as sperm adhesion molecule 1 (SPAM1) and hyaluronoglucosaminidase 5 (Hyal5) are in part responsible for dispersal of the cumulus cell mass, which is a critical step in establishing fertilization in mammals. In this study, we identified two testis-hyaluronidases, SPAM1 and Hyal5, in hamster and rat. These two genes were expressed specifically in the testis. At the protein level, hamster SPAM1 and Hyal5 display 78.7% and 75.4% identity with mouse SPAM1 and Hyal5. Further, the activity of the enzymes with respect to cumulus cell dispersion did not differ, although we observed that the enzymatic activity differed in pH range. These studies suggest that different sperm hyaluronidases are capable of dispersing the cumulus cell mass despite differences in enzyme activity.

Protective effect of enzymatic hydrolysates from highbush blueberry (Vaccinium corymbosum L.) against hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast cell line

  • Blueberry was enzymatically hydrolyzed using selected commercial food grade carbohydrases (AMG, Celluclast, Termamyl, Ultraflo and Viscozyme) and proteases (Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex) to obtain water soluble compounds, and their protective effect was investigated against $H_2O_2$-induced damage in Chinese hamster lung fibroblast cell line (V79-4) via various published methods. Both AMG and Alcalase hydrolysates showed higher total phenolic content as well as higher cell viability and ROS scavenging activities, and hence, selected for further antioxidant assays. Both AMG and Alcalase hydrolysates also showed higher protective effects against lipid peroxidation, DNA damage and apoptotic body formation in a dose-dependent fashion. Thus, the results indicated that water soluble compounds obtained by enzymatic hydrolysis of blueberry possess good antioxidant activity against $H_2O_2$-induced cell damage in vitro.
