The fucoidan from Laminaria relicollected at Wando in Korea was purified with the yield of 2.3% in mass. The monosaccharide composiof the purified fucoidan was nearly identical to that of the commercial standard: fucose 63.71 %, xylose 22.98%, galactose 6.62%, mannose 0.24%, and uronic acid 3.26%. Microorganisms capable of degrading the purified fucoidan were isolated from the colonies on the minimal medium containing 0.2% of purified fucoidan as a sole carbon source. Of these isolates, a strain showing a relatively higher capability to degrade fucoidan, up to 63%, was partially characterized as a Gram positive, aerobic, moderately halophilic marine bacterium.
The pathogenic Vibrio genus contains halophilic bacteria that are distributed in marine and freshwater environments. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are potent human pathogens and leading causes of septicemia, wound infection, and seafood-borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrio species in seawater off the west coast of Korea. Sixty-four seawater samples were obtained from different sites in Gomso Bay and Byeonsan from April 2018 to November 2018. Pathogenic Vibrio species were detected using a combination of most probable number (MPN)-polymerase chain reaction methods. V. cholerae, V. vulnificus, and V. parahaemolyticus were found in 0.0%, 20.3%, and 65.6% of seawater samples, respectively. Quantitative results revealed 3.6-23 MPN/100 mL of V. vulnificus, and 3.6-930 MPN/100 mL of V. parahaemolyticus in the samples. Overall, these results provide new insight into the necessity for seawater sanitation in Gomso Bay and Byunsan; they also provide evidence that will help reduce outbreaks of seafood-borne illness caused by pathogenic Vibrio species.

This amylase was purified by acetone fractionation, DEAG-Cellulose column chromatography, 1st Sephadex G-75 gel filtration, CM-Cellulose column chromatography and 2nd Sephadex G-75 gel filtration. The amylase was purified about 98.64 fold with a yield of 11.75%. The molecular weight of amylase was estimated to be about 43,000and 40,000 by gel filtration and SDS-polyacrylamide gel electrophoresis, respectively, suggesting that the enzyme was a monomer. Amylase had an optimal temperature of 4$0^{\circ}C$, and an optimum pH of 7.0, and the thermal stability was observed the above 50% at 10$0^{\circ}C$ after 1 hour, and the stable range of pH was 6.0 to 8.0. The enzymatic activity was increased in the presence of 10 mM 2-mercaptoethanol, slightly by 10 mM SnCl2.2H2O.FeCl2.4H2O.CuCl2.2H2O.HgCl2.6H2O and SDS. End products from soluble starch were glucose, maltose and maltotriose, and Km value for soluble starch was 2.5mg/ml.
염장미역은 염농도 감소와 저장조건의 완화를 위한 연구의 일환으로서 2~4kGy의 감마선을 조사하고 ${30{\pm}2^{\circ}C와\;10{\pm}1^{\circ}C$에 저장하면서 미생물적, 관능적 품질을 평가한 결과는 다음과 같다. 염장미역의 변질에 관련된 원인 미생물은 호염성 세균과 효모로 확인되었으며, 2~4kGy의 감마선 조사는 초기의 미생물 수준을 1~2 log orders 감소시켰으나 저장 기간 중 증식에는 뚜렷한 영향이 없었다. 관능적 품질에서도 감마선 조사에 의한 미역조직의 연화현상으로 비조사군보다 낮은 기호도를 나타내어 저장 중 품질 안정성 향상에 별 효과가 없는 것으로 나타났다.
Saline soils comprise more than half a billion hectares worldwide. Thus, they warrant attention for their efficient, economical, and environmentally acceptable management. Halophytes are being progressively utilized for human benefits. The halophyte microbiome contributes significantly to plant performance and can provide information regarding complex ecological processes involved in the osmoregulation of halophytes. Microbial communities associated with the rhizosphere, phyllosphere, and endosphere of halophytes play an important role in plant health and productivity. Members of the plant microbiome belonging to domains Archaea, Bacteria, and kingdom Fungi are involved in the osmoregulation of halophytes. Halophilic microorganisms principally use compatible solutes, such as glycine, betaine, proline, trehalose, ectoine, and glutamic acid, to survive under salinity stress conditions. Plant growth-promoting rhizobacteria (PGPR) enhance plant growth and help to elucidate tolerance to salinity. Detailed studies of the metabolic pathways of plants have shown that plant growth-promoting rhizobacteria contribute to plant tolerance by affecting the signaling network of plants. Phytohormones (indole-3-acetic acid and cytokinin), 1-aminocyclopropane-1-carboxylic acid deaminase biosynthesis, exopolysaccharides, halocins, and volatile organic compounds function as signaling molecules for plants to elicit salinity stress. This review focuses on the functions of plant microbiome and on understanding how the microorganisms affect halophyte health and growth.
The pathogenic Vibrio genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. V. cholerae, V. vulnificus, and V. parahaemolyticus are the main species that can be potent human pathogens and the leading cause of septicemia, wound infections, and seafood borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrios in seawater. We obtained a total of 80 seawater samples from the Geum River estuary area in the west coast of Korea from April to December 2021. Pathogenic Vibrios was determined using a combination of the most probable number-polymerase chain reaction (MPN-PCR) methods. The detection levels of V. cholerae, V. parahaemolyticus, and V. vulnificus in the seawater samples were 7.5%, 68.8%, and 30.0%, respectively. The quantitative results were as follows: 3.6-3.6 MPN/100 mL in V. cholerae, 3.6-3,400 MPN/100 mL in V. parahaemolyticus, and 3.6-4,300 MPN/100 mL in V. vulnificus. Overall, these results provide novel insight into the necessity for seawater sanitation in the Geum River estuary area, and could help reduce the risk of seafood-borne outbreaks caused by pathogenic Vibrios.
비브리오균은 해양 및 하구 환경에서 상재하는 균으로 그람 음성 간균, 호염성, 비포자형성의 특징을 가지고 있다. 질병과 관련된 균주로는 Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio cholera 등이 있다. 매년 어패류의 섭취 등을 통해 식중독 사고가 빈번하게 발생하고 있으며, 이에 본 연구에서는 서해안 6곳에서 패류를 채취하여 비브리오균의 분포도를 모니터링하고, 이들의 항생제 내성을 분석하였다. 총 120건의 패류를 TCBS 평판배지와 API 20E kit 기법으로 동정하였을 때 23.3%의 시료에서 비브리오균이 검출되었다. 이들 분리균주들의 항생제 내성을 16종의 항생제에 대해 분석한 결과 82.1%가 각각 vancomycin과 ampicillin에 대해 내성을 보였고, rifampin (71.4%)과 cephalothin (53.6%)에 대한 내성비율도 다른 항생제들에 비해 상대적으로 높은 것으로 나타났다. 분리균주들의 항생제 감수성은 chloramphenicol (92.9%), sulfamethoxazole/trimethoprim (92.9%)과 tetracycline (96.4%)에 대해 가장 높았다. 분리균주들의 약 71.4%가 vancomycin과 ampicillin을 포함하여 3개 이상의 항생제에 대해 다중내성을 보였다.
낙동강 하구언 건설에 따른 하구생태계의 변화를 파알하기 위하여 축조 전후의 환경인자와 종속 영양세균의 생리적 특성을 요인분석에 의해 비교하였다. 하구언 축조 후 해수유입의 영향이 급격히 감소하였으며, 축조 전 50.7% 를 차지하던 영양물질의 과다와 관련된 환경변수가 축조후 77.1%로 증가하였다.하구언 축조 전 환경변수와 종속영양세균의 생리적 특성 사이에 밀접한 관련성을 나타냈으며, 축조 후에도 종속영양세균의 생리적 특성이 영양물질의 과다와 관련된 형질로 대부분 해석되어 환경변화와 분포 미생물의 생리적 특성 사이의 관련성을 나타내었다. 그러나 하구언 축조에 따른 해수유입의 차단에도 불구하고 분리균주의 생리적 특성에 의한 요인분석의 3번째 쥬요인이 호염성 형질로 해석되어 작동강 하구의 환경변화에 따른 종속영양세균의 형질이 아직 완전 안정화되지 않은 것으로 사료된다.
Soil salinization refers to the buildup of salts in soil to a level toxic to plants. The major factors that contribute to soil salinity are the quality, the amount and the type of irrigation water used. The presented review discusses the different sources and causes of soil salinity. The effect of soil salinity on biological processes of plants is also discussed in detail. This is followed by a debate on the influence of salt on the nutrient uptake and growth of plants. Salinity decreases the soil osmotic potential and hinders water uptake by the plants. Soil salinity affects the plants K uptake, which plays a critical role in plant metabolism due to the high concentration of soluble sodium ($Na^+$) ions. Visual symptoms that appear in the plants as a result of salinity include stunted plant growth, marginal leaf necrosis and fruit distortions. Different strategies to ameliorate salt stress globally include breeding of salt tolerant cultivars, irrigation to leach excessive salt to improve soil physical and chemical properties. As part of an ecofriendly means to alleviate salt stress and an increasing considerable attention on this area, the review then focuses on the different plant growth promoting bacteria (PGPB) mediated mechanisms with a special emphasis on ACC deaminase producing bacteria. The various strategies adopted by PGPB to alleviate various stresses in plants include the production of different osmolytes, stress related phytohormones and production of molecules related to stress signaling such as bacterial 1-aminocyclopropane-1-carboxylate (ACC) derivatives. The use of PGPB with ACC deaminase producing trait could be effective in promoting plant growth in agricultural areas affected by different stresses including salt stress. Finally, the review ends with a discussion on the various PGPB activities and the potentiality of facultative halophilic/halotolerant PGPB in alleviating salt stress.
굴(Crassostrea virginica)과 Weakfish(Cynoscion regalix)를 냉장(6$^{\circ}C$), 빙장($0^{\circ}C$), 부분동결저장(-4$^{\circ}C$) 및 동결저장(-2$0^{\circ}C$)온도에서 45일간 저장하면서 생균수 와 microflora의 변화를 조사하였다. 각 온도에서 저온저장중 굴로부터 255주, Weakfish로부터 240주, 합계 495주를 분리하여 microflora의 변화를 조사하였다. 저장직전의 생선에서 생균수는 굴이 4.9$\times$10/ sup 5/CFU/g, Weakfish가 $1.5\times$$10^4$CFU/$cm^2$였다. 저장직전의 굴에서는 Pseudomonas ll1III/IV가 67%, Vibrio가 20%를 차지하였다. Weakfish에서는 Acinetobacter가 40% Moraxella가 33%로서 주종을 이루었으며 Pseudomonas와 Vibrio는 아주 적은 비율을 차지하였다. 굴의 저온저장중 microflora는 저장온도에 큰 관계 없이 모든 저장온도에서 Pseudomonas lIII/IV-H가 전체 균주의 67.4%, Flavobacterium/Cytophaga가 9.3%, 다음으로는 Vibrio가 6.3%를 차지하였고 약 15%의 세균은 동정하지 못하였다. Weakfish의 저온저장중 microflora는 냉장, 빙장, 부분동결저장한 경우에 비호염성균인 Pseudomonas III/IV-NH가 전체 균주의 60~100%를 차지하였으며 동결저장한 Weakfish에서는 Moraxella가 전체 분리 균주의 40~60%를 차지하였다. 전체적으로 굴에서는 호염성 균주(pseudomonas III/ IV-H와 Vibrio)가 우세하였고 구균류가 분리되지 않았으나 Weakfish에서는 비호염균주(PseudomonasIII/IV -NH와 Moraxella가 우세하였으며 구균류가 4.6% 검출되었다. 저온저장한 굴과 Weakfish에서 Vibrio의 검출률은 굴에서 Weakfish보다 3배 높았으며 Listeria spp. 는 검출되지 않았으나 강한 용혈작용을 가진 균들이 각각 9주, 8주씩 분리되어 주의가 요망된다.
