Identification of Staphylococcus hyicus subsp hyicus of swine and poultry origin by API STAPH-IDENT system (API STAPH-IDENT system에 의한 돼지 및 닭유래(由來) staphylococcus hyicus subsp hyicus의 동정(同定))

  • The API STAPH-IDENT system was evaluated as a means for identifying Staphylococcus hyicus subsp hyicus straints isolated from swine and poultry. Of 80 strains from swine, 68 (85%) were correctly identified by the API STAPH-IDENT system alone after 5 h of incubation. When results were determined after 24 h of incubation, the accuracy of this system alone was 93.8%. By additional tests in conjunction with the API STAPH-IDENT system, however, all 80 strains could be correctly identified. Of 120 strains from poultry, 87 (72.5%) required additional testing to achieve a correct identification, and 33 (27.5%) were incorrectly identified by this system after 5 h of incubation. After 24 h of incubation, 99 of 120 (82.5%) avian strains were incorrectly identified as Staph epidermidis owing to false-negative mannose and trehalose utilizations. Seventy-seven (96.3%) of swine strains were positive for ${\beta}-glucuronidase$, whereas all 120 strains recovered tram poultry were negative for it.

Effects of pH and Potassium Chloride in Solvent System of High-Speed Countercurrent Chromatography (pH 및 염화칼륨 첨가가 고속역류크로마토그래피의 용매시스템에 미치는 영향)

  • Effects of the physical properties of solvent system such as pH and polarity change by salt addition in solvent system were investigated by using high speed countercurrent chromatography apparatus (Model CCC-1000, Pharm-Tech Research Corp. USA). The changes of pH and interfacial tension in solvent system of high speed countercurrent chromatography did not significantly affect on retention of stationary phase, but induced remarkable changes in the partition coefficient of ginkgo flavonoids, kaempferol, quercetin and isorhamnetin. The partition coefficients of ginkgo flavonoid standard increase with an increased pH of solvent system and quercetin sharply increased at pH 10.0. Retention of stationary phase decreases with an increased concentration of KCl in butanol of solvent system. Interfacial tension between two phase in solvent system of hexane increases with an increased concentration of KCl. The polarity of solvent system significantly changes the partition coefficients of ginkgo flavonoid.

Oxidative Damage of DNA Induced by the Cytochrome c and Hydrogen Peroxide System

  • To elaborate the peroxidase activity of cytochrome c in the generation of free radicals from $H_2O_2$, the mechanism of DNA cleavage mediated by the cytochrome c/$H_2O_2$ system was investigated. When plasmid DNA was incubated with cytochrome c and $H_2O_2$, the cleavage of DNA was proportional to the cytochrome c and $H_2O_2$ concentrations. Radical scavengers, such as azide, mannitol, and ethanol, significantly inhibited the cytochrome c/$H_2O_2$ system-mediated DNA cleavage. These results indicated that free radicals might participate in the DNA cleavage by the cytochrome c and $H_2O_2$ system. Incubation of cytochrome c with $H_2O_2$ resulted in a time-dependent release of iron ions from the cytochrome c molecule. During the incubation of deoxyribose with cytochrome c and $H_2O_2$, the damage to deoxyribose increased in a time-dependent manner, suggesting that the released iron ions may participate in a Fenton-like reaction to produce $\cdot$OH radicals that may cause the DNA cleavage. Evidence that the iron-specific chelator, desferoxamine (DFX), prevented the DNA cleavage induced by the cytochrome c/$H_2O_2$ system supports this mechanism. Thus we suggest that DNA cleavage is mediated via the generation of $\cdot$OH by a combination of the peroxidase reaction of cytochrome c and the Fenton-like reaction of free iron ions released from oxidatively damaged cytochrome c in the cytochrome c/$H_2O_2$ system.

Performance Test of Cooling System for the KEPCO HTS Power Cable (한전 초전도전력케이블 냉각시스템 성능시험)

  • As a power transmission line supplying power to a densely populated city, the high temperature superconducting (HTS) cable is expected to one of the most effective cables with a compact size because of its high current density. The verification of HTS power cable system have been progressed by KEPRI. A cooling system for a 3-phase 100m HTS power cable with 22.9kV/1.25kA was installed and tested at KEPCO's Gochang power testing center in Korea. The system consists of a liquid nitrogen decompression cooling system with a cooling capacity of 3kW and a closed circulation system of subcooled liquid nitrogen. Several performance tests of the cable system with respect to the cooling such as cooling capacity, heat load and temperature stability, were performed at several temperatures. Thermal cycle test, cool-down to liquid nitrogen temperature and warm-up to room temperature, was also performed to investigate thermal cycle influences. The outline of the installed cooling system and performance test results are presented in this paper.

Hybrid Control of an Active Suspension System with Full-Car Model Using H$_{}$$\infty$/ and Nonlinear Adaptive Control Methods

  • This paper presents hybrid control of an active suspension system with a full-car model by using H$\sub$$\infty$/ and nonlinear adaptive control methods. The full-car model has seven degrees of freedom including heaving, pitching and rolling motions. In the active suspension system, the controller shows good performance: small gains from the road disturbances to the heaving, pitching and rolling accelerations of the car body. Also the controlled system must be robust to system parameter variations. As the control method, H$\sub$$\infty$/ controller is designed so as to guarantee the robustness of a closed-loop system in the presence of uncertainties and disturbances. The system parameter variations are taken into account by multiplicative uncertainty model and the system robustness is guaranteed by small gain theorem. The active system with H$\sub$$\infty$/ controller can reduce the accelerations of the car body in the heaving, pitching and rolling directions. The nonlinearity of a hydraulic actuator is handled by nonlinear adaptive control based on the back-stepping method. The effectiveness of the controllers is verified through simulation results in both frequency and time domains.
