식이 셀레늄 수준과 식이 지방산 조성이 쥐의 지질과 산화 상태와 조직의 지방산 조성에 미치는 영향 (Effect of Dietary Selenium and Fish oil on Lipid Peroxidation and Fatty Acid Profile in the Rat)

  • 식이 셀레늄(Se)의 수준과 식이지방산의 조성이 조직(혈장,동맥,간)의 지질과산화 상태와 조직의 지방산 조성에 미치는 영향에 대해 연구하였다. Sprague Dawley종 수컷쥐들을 식이 Se 수준(0.005mg/kg diet, 0.5mg/kg diet)과 식이 지방산의 주요 급원(옥수수유, 어유)에 따라 4군으로 나누어 8주간 실험하였다. 식이에 함유된 vitamin E의 함량은 117IU정도이다. Se 영양상태의 지표인 glutathione peroxidase 활성도와 Se 수준은 Se이 결핍된 식이를 공급시키면 식이지방 종류에는 상관없이 낮게 나왔다. 어유공급은 간과 동맥의 MDA를 증가시키나 Se을 공급시킨 쥐의 간 MDA수준은 감소되었다. 어유를 공급시키면 혈장 총치질, 동맥 과 간의 인지질에서 20:5(n-3), 22:5(n-3), 22:6(n-3), 20:3(n-6) 수준은 증가하였으며 20:4(n-6)수준은 감소되었다. 비록 Se공급이 n-3 지방산 수준 [22:6(n-3)]을 증가시키나 어유공급 효과에 비하면 그 효과가 적었다. 본 실험결과에 의하면 vitamin E가 현재 권장되는 양보다 충분히 많게 공급되어도 Se이 지질과산화를 방지하는데 중요한 역활을 하고 있으므로 적절한 Se공급이 바람직하다.

Effects of Moist Extruded Full-fat Soybeans on Gut Morphology and Mucosal Cell Turnover Time of Weanling Pigs

  • Ten barrows, weaned at 28 days (7.2$\pm$0.1 kg BW), were used to evaluate the effects of feeding extruded full-fat soybeans on intestinal morphology and mucosal cell turnover time. All pigs were fed corn-based diets with half of the pigs receiving diets supplemented with 15.5% soybean meal and 3% soybean oil and the remaining pigs fed a diet in which the soybean meal and oil were replaced by 18.5% extruded full-fat soybeans. The pigs were individually placed in $80{\times}150cm$ metabolic cages and fed twice daily an amount approximately equal to their ad libitum intake for a period of 14 days. On day 14, pigs were weighed and then injected intraperitoneally with $^3$H]thymidine ($100{\mu}Ci/kg$ of BW, specific activity 20 Ci/mmol) 6 h after the morning meal. A pig from each treatment was killed 1, 4, 8, 16, or 24 h postinjection and intestinal tissues were collected. Daily gains for pigs fed the soybean diet and extruded full-fat soybean diet were 0.24 and 0.31 kg/day (p=0.05) with feed conversions of 1.58 and 1.39 (p=0.05), respectively. In comparison with pigs fed soybean meal, pigs fed moist extruded full-fat soybeans had a decreased crypt depth in their duodenum and cecum (p<0.1), while the villus height in the mid jejunum and ileum and the total height (villus height plus crypt depth) of the ileum and mid jejunum increased (p<0.05). The villus width in the duodenum and mid jejunum decreased (p<0.05). The number of crypt epithelial cells in the upper jejunum increased but decreased in the ileum, colon and cecum (p<0.05). The number of villus epithelial cells in the ileum and the upper and mid jejunum increased (p<0.05). The time for migration of epithelial cells in the crypt-villus column decreased (p<0.05) in all sites except the upper jejunum, ileum and cecum. The mucosal turnover rate for all intestinal sites except the upper jejunum, colon and cecum decreased (p<0.05). From these data, we conclude that inclusion of moist extruded full-fat soybeans in weanling pig diets can improve the intestinal morphology and slow the migration rate and turnover time of epithelial cells of the small intestine, especially in the mid jejunum compared with soybean meal.

Net Portal Fluxes of Nitrogen Metabolites in Holstein Steers Fed Diets Containing Different Dietary Ratios of Whole-crop Corn Silage and Alfalfa Hay

  • The objectives of the present study were to investigate the effects of different dietary ratios of whole-crop corn silage and alfalfa hay on nitrogen (N) digestion, duodenal flow and metabolism across the portal-drained viscera (PDV) of growing beef steers, and to elucidate their relationships. Four steers (236${\pm}$7 kg BW) fitted with duodenal cannulae and chronic indwelling catheters into the portal and mesenteric veins and abdominal aorta were used in a 4${\times}$4 Latin square design. Animals were fed (at 12-h intervals) the 4 diets consisting of whole-crop corn silage (C) and alfalfa hay (A) in 80:20 (C8A2), 60:40 (C6A4), 40:60 (C4A6) and 20:80 (C2A8) ratios of which dietary crude protein (CP) was 10.5, 12.0, 13.5 and 15.0% of dry matter (DM), respectively. Feeding level was restricted to 95% of ad libitum intake to measure N digestion, blood flow and net flux of N across the PDV. Digestibility of DM and neutral detergent fiber and digestible energy intake linearly increased as the ratio of alfalfa hay increased. The N intake, duodenal flow and intestinal disappearance increased linearly with increasing alfalfa hay. Arterial and portal concentrations of ${\alpha}$-amino N showed a quadratic response to increasing levels of alfalfa hay and were the highest in steers fed the C6A4 diet. The net PDV release of ${\alpha}$-amino N and ammonia N increased linearly with increasing alfalfa hay, but urea N uptake by PDV did not differ among diets. As a percentage of apparently digested N in the total gut, net PDV release of ${\alpha}$-amino N linearly decreased from 66 to 48% with increasing alfalfa hay. Conversely, net PDV recovery of ${\alpha}$-amino N to intestinal N disappearance varied with increasing alfalfa hay accounting for 49, 50, 58 and 61% on C8A2, C6A4, C4A6 and C2A8 diets, respectively. Net PDV uptake of urea N, relative to apparently digested N, linearly decreased from 81 to 25% as alfalfa hay increased from 20 to 80% of DM intake. Considering PDV uptake of urea N, microbial efficiency and conversion of total tract digested N to PDV ${\alpha}$-amino N net supply, a diet consisting of 80% whole-crop corn silage and 20% alfalfa hay (10.5% CP) was the best, while considering the quantities of intestinal N disappearance and ${\alpha}$-amino N absorption, a diet of 20% whole-crop corn silage and 80% alfalfa hay (15% CP) would be preferred. The proportion of ${\alpha}$-amino N recovered by PDV relative to the intestinal N disappearance may vary with energy intake level of mixed forage diets.

Association between interstitial cells of Cajal and anti-vinculin antibody in human stomach

  • Interstitial cells of Cajal (ICC) are known as the pacemaker cells of gastrointestinal tract, and it has been reported that acute gastroenteritis induces intestinal dysmotility through antibody to vinculin, a cytoskeletal protein in gut, resulting in small intestinal bacterial overgrowth, so that anti-vinculin antibody can be used as a biomarker for irritable bowel syndrome. This study aimed to determine correlation between serum anti-vinculin antibody and ICC density in human stomach. Gastric specimens from 45 patients with gastric cancer who received gastric surgery at Kangwon National University Hospital from 2013 to 2017 were used. ICC in inner circular muscle, and myenteric plexus were counted. Corresponding patient's blood samples were used to determine the amount of anti-vinculin antibody by enzyme-linked immunosorbent assay. Analysis was done to determine correlation between anti-vinculin antibody and ICC numbers. Patients with elevated anti-vinculin antibody titer (above median value) had significantly lower number of ICC in inner circular muscle (71.0 vs. 240.5, p = 0.047), and myenteric plexus (12.0 vs. 68.5, p < 0.01) compared to patients with lower anti-vinculin antibody titer. Level of serum anti-vinculin antibody correlated significantly with density of ICC in myenteric plexus (r = -0.379, p = 0.01; Spearman correlation). Increased level of circulating anti-vinculin antibody was significantly correlated with decreased density of ICC in myenteric plexus of human stomach.

보양환오탕(補陽還五湯) 경구투여 후 면역 활성화에 의한 암 전이 억제 효과 (Anti-metastatic Effect on Cancer cell and Immune System Activation by Orally Administered Boyanghwano-tang)

  • Objectives: This study was designed to investigate intestinal immune system activation and anti-metastatic effect on cancer cells by orally administered extracts of Boyanghwano-tang. Methods: To observe immunomodulating effects of Boyanghwano-tang on Peyer's patch cells, we measured cytokines GM-CSF, IL-4. In addition to observing effects of Boyanghwano-tang on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in intestinal content and serum was measured to observe the effect of orally administered Boyanghwano-tang on mucosal immune system. After administering ovalbumin (OVA) with Boyanghwano-tang, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Anti-metastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. Results: The amounts of GM-CSF and IL-4 in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. The proliferation of bone marrow cell was significantly up-regualted with Boyanghwano-tang. These results indicate that oral administration of Boyanghwano-tang enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-4 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation. After orally administering OVA with Boyanghwano-tang, IgA induction and Proliferation of peyer's patch cell was up-regulated with Boyanghwano-tang. These results means orally administered Boyanghwano-tang activates intestinal immune system and has an inhibitory effect on tumor metastasis. In addition, We found that orally administered Boyanghwano-tang significantly inhibited tumor metastasis in vivo. Conclusions: Orally administered Boyanghwano-tang appears to have considerable activity on the anti-metastasis by activation of immune system.

오적산(五積散) 경구투여에 의한 면역활성과 종양 전이 억제 효과 (Activation of Immune System & Antimetastatic Effects of Ojeok-san by Oral Administration)

  • Objectives: This study was designed to investigate intestinal immune system activation and antimetastatic effect of Ojeok-san on cancer cells by oral administration. Methods: Cell viability of Ojeok-san was tested with colon 26-M3.1 carcinoma cells and Peyer's patch cells in vitro. Antimetastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. To observe immunomodulating effects of Ojeok-san on Peyer's patch cells, we measured interleukin (IL)-4, GM-CSF. In addition to observing effects of Ojeok-san on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in serum and intestinal content was measured to observe the effect of orally administered Ojeok-san on mucosal immune system. After administering Ovalbumin (OVA) with Ojeok-san, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Results: in vitro cytotoxicity analysis, the inhibitory concentration $(IC)_{50}$ of the colon 26-M3.1 carcinoma cell was $890{\mu}g/ml$. $IC_{50}$ of the Peyer's patch cells with LPS was $990{\mu}g/ml$. We found that orally administered Ojeok-san significantly inhibited tumor metastasis in vivo. In addition, the amounts of IL-4 and GM-CSF in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. The proliferation of bone marrow cell was significantly up-regulated with Ojeok-san. 어휘 의미론에서 대두되는 전일주의적 의미관 (Holistische Bedeutungsauffassung in der lexikalischen Semantik)

  • Was die Konzeption von Wortbedeutung angeht, so ist festzustellen, $da\ss$ sich in der lexikalischen Semantik Konzeptionen von sprachlicher Wortbedeutung in zwei Hauptauffassungen teilen lassen. D.h. eine komponentielle Bedeutungsauffassung, nach der Wortbedeutungen in einzelne semantische Komponenten zerlegbar sind, $l\"{a}{\ss}t$ sich von einer holistischen Bedeutungskonzeption, die Wortbedeutungen als ganzheitlich und nicht weiter zerlegbar begreift, unterscheiden. Holistische $Ans\"{a}tze$ im allgemeinen Sinn erfassen ein $Ph\"{a}nomen$ ganzheitlich. Bezogen auf sprachwissenschaftliche und psychologische $Ans\"{a}tze$ bedeutet aber 'holistisch' eine nichtkomponentielle Beschreibung. Bestimmt man den Begriff 'holistisch' als die Leugnung klar abgrenzbarer $partikul\"{a}rer$ Einheiten, so $erh\"{a}lt$ 'holistisch' einen spezifischen Sinn in holistischen $Ans\"{a}tzen$. Im Hinblick darauf $k\"{o}nnen$ der kognitionspsychologische Prototypenansatz von Rosch und der philosophische Stereotypenansatz von Putnam bedeutsam werden. Entweder ist damit die Ablehnung einer definitorisch verfahrenden Merkmalsemantik verbunden, oder es werden Merkmale nun lediglich als stereotypisch-definitorische uminterpretiert. Eine ausgeformte Prototypensemantik bzw. Stereotypensemantik gibt es bisher nur als Postulat eines zur definitorisch verfahrenden Merkmalsemantik altemativen Bedeutungskonzepts. Diese holistischen $Ans\"{a}tze$ versuchen sich gerade in der Kontroverse zur Merkmalsemantik zu etablieren, kommen aber selbst nicht ohne das Konzept des Merkmals aus. Sie sind auch nicht auf aile $W\"{o}rter$ unseres Wortschatzes gleich gut anzuwenden, weil es nicht $f\"{u}r$ alle $W\"{o}rter$ Stereotype gibt und die Existenz digital struktuierter Kategorien, die mithilfe von hinreichenden und notwendigen Bedingungen beschrieben werden $k\"{o}nnen$, nicht geleugnet wird. Die holistischen $Ans\"{a}tze$ sind als Versuch zu beurteilen, der $Realit\"{a}t$ in ihrer Komplexitat $m\"{o}lichst$ nahe zu kommen. Doch bleibt das Problem, wie Holismus im Bereich der Sprachbeschreibung $ad\"{a}quat$ wiederzugeben ist. Auch wenn wir $f\"{u}r$ die Annahme der psychologischen $Realit\"{a}t$ eines holistischen Bedeutungsbegriffs $pl\"{a}dieren$, so bestreiten wir damit nicht die $N\"{u}tzlichkeit$ der komponentiellen Bedeutungsanalyse als leicht handhabbare Beschreibungsnotation $f\"{u}r$ die Bedeutung von Wortern mit wohlunterscheidbaren Referenten. In diesem Sinn $k\"{o}nnten$ vielleicht beide methodischen $Ans\"{a}tze\;in\;einer\;komplement\"{a}en$ Sicht integriert werden.

Hugo $H\"{a}ring$의 '신건축(Neues Bauen)' 사상과 그 이론 발전의 상징론적 측면에 대한 고찰 (A study on Hugo $H\"{a}ring's$ Theory of 'Neues Bauen' and its Symbolic Meaning)

  • Hugo $H\"{a}ring$(1882-1952) belonged to that special generation of architects born in the 1880s which became responsible for the establishment of Modern Movement in the 1920s as W. Gropius, Mies van der Rohe and Le Corbusier, etc. Although he have been overlooked by many historian, He was a key figure of the Modern architecture and as the main theorist for Organic stream in German architecture. He is well-known for his theory of 'Neues Bauen(New Building)', the organic functionalism that is epitomized as the design process from the inside outwards, starting with the life-processes of dwelling. So he argued that the builder must become aware of the life process his building is to serve, and he should not impose a form but try to find the form. These concepts are expressed well in his key-words, the 'Organwerk(organ-work)' and 'Leistungsform(form as achievement)'s. $H\"{a}ring's$ theory can be found in the short early essay, 'Wege zur Form(approaches to form)' of 1925. But His concept of 'function' is based on the speciality and individual identity that concerned him from the start, not purely pragmatic aspects. After 1940s his theory moved increasingly in this direction. He defined this as the transition from 'Organwerk' to 'Gesetaltwerk', from mere anatomy to essence, being, personality, life. It suggest that Hugo Haring's idea of Gestalt is a dimension of mystical or symbolic meaning. This paper Is about the way in which this theoretical transition can be parallel with contemporary philosophers as E. Cassirer's philosophy of symbolic forms and M. Heldegger's phenomenology. And the key example of this viewpoint is (1921-1926) near Lubuk in Germany, with its 'cowshed' of pear shaped plan devised around the requirements and rituals of farm. This study presents the symbolic conception of Hugo $H\"{a}ring's$ theory can propose the ability of a symbolic intuition as a view that re-integrate technical thinking with knowledge of other kinds beyond the immediate material.

광합성 섬모류 Mesodinium rubrum MR-MAL01의 응용성(1) 이매패류 종묘생산을 위한 먹이생물 유용성 (Applicability of a Photosynthetic Ciliate, Mesodinium rubrum MR-MAL01 -Usefulness as a Live Prey Species for the Marine Aquaculture of Bivalves-)

  • 최근, 곰소만에서 채집한 시료로부터 최초의 온대역산 광합성 섬모류 Mesodinium rubrum MR-MAL01 배양체를 분리ㆍ배양하였다. 이 섬모류를 이용하여 이매패류의 섭식실험을 수행한 결과, 비단가리비와 바지락과 같은 이매패류가 섬모류를 직접 섭식 하였음을 확인하였는데, 개체 당 최대 섭식률은 비단가리비 9,590 cells $min^{-1}$, 바지락 23,200 cells $min^{-1}$으로 나타났다. 또한 비단가리비의 소화관에서 완전한 M. rubrum세포를 확인하지 못했으나, 파괴된 세포로 생각되는 작은 입자들을 관찰하였다. 바지락 치패에 섬모류인 M. rubrum과 미소조류인 Isochrysis galbana (KMCC H-2)를 공급하면서 28일 동안 사육한 결과, 작은 치패 그룹(각장 0.46 mm)에서는 I. galbana를 공급한 실험구가 좋은 성장을 보였던 반면, 큰 치패 그룹(각장 0.84 mm)에서는 M. rubrum을 공급한 실험구가 I. galbana를 공급한 실험구에 비해 성장이 좋았다. 따라서 이 광합성 섬모류는 이매패류의 치패 및 모패와 입이 작은 자어의 먹이생물, rotifer의 영양강화제 및 어류의 천연 착색제로도 활용할 수 있는 잠재력이 있어 해양 생물공학적 활용을 위한 생물재료로서 개발 가능성을 제안한다.

Utilization of Dietary Nutrients, Retention and Plasma Level of Certain Minerals in Crossbred Dairy Cows as Influenced by Source of Mineral Supplementation

  • Nutrient utilization and plasma level of minerals were studied in lactating crossbred cows fed diet supplemented from different source of minerals. Twelve crossbred cows of first to third lactation during their mid stage of lactation were distributed equally under two groups and were fed concentrate mixture, green fodder (para grass) and finger millet straw as per requirement. Cows in group I were fed concentrate mixture supplemented with 1% mineral mixture whereas cows in group II were not supplemented with mineral mixture in the concentrate mixture but were offered additional quantity of green fodder (1 kg DM/cow/day) to compensate for the requirement of minerals. Balance study conducted towards the end of 120 days of feeding experiment indicated that the total dry matter intake in both the groups did not differ significantly. The cows in group II offered additional quantity of green fodder consumed significantly (p<0.05) more of green fodder (5.11 vs. 4.51 kg /animal/day) and the cows in group I consumed significantly (p<0.01) more of finger millet straw (1.71 vs. 0.92 kg/animal/day). The digestibility of major nutrients did not differ between the groups except for ether extract which was significantly (p<0.05) lower in cows fed additional green fodder. The total daily intake of P, Cu, Fe and Co did not differ significantly in both the groups whereas significantly higher intakes of Ca, Mg, Zn and Mn were observed in cows supplemented with inorganic source of minerals. However, supplementation through both the sources could meet the mineral requirement in group I and group II. The gut absorption (%) of all the minerals was comparable between the groups except for Mg which was significantly (p<0.05) higher in cows supplemented mineral mixture. The net retention of all the minerals was significantly more in the group supplemented with inorganic source of minerals. Except for P, Mg and Co the retention as percentage of total intake was comparable for all minerals in both the groups. Irrespective of source of mineral supplementation the average monthly blood plasma mineral levels (Ca, P, Mg, Cu, Zn, Fe) were within the normal range and comparable between the groups. Lower level of Cu observed at the start of the experimental feeding in both the groups increased with the advancement of supplemental feeding. The plasma Zn and Fe content in cows of both the groups did not vary. The blood plasma level of some minerals (Ca, P, Mg and Cu) was significantly higher towards the end of experimental feeding as compared to the initial values due to the reduction in milk yield with advancement of lactation or due to supplemental effect of minerals. It could be concluded that supplementation of minerals through inorganic source is better utilized in terms of retention as compared to green fodder (para grass), which is a moderate source of most nutrients.
