Adsorption Characteristics of Organophosphorus and Carbamate Pesticides in Four Soils and the Evaluation for Their Leaching Potential Using Two Screening Models (유기인계 및 카바메이트계 농약의 토양흡착성과 간이선발모형을 이용한 용탈 잠재성 평가)

  • Soil adsorption study was carried out to define the mobility of pesticides and to evaluate leaching potential in soils. Nine pesticides including metolcarb, molinate, fenobucarb, isazofos, diazinon, fenitrothion, dimepiperate, parathion and chlorpyrifos-methyl were subjected to adsorption experiment for four types of soils, such as upland, paddy, forest and volcanic ash soil. Based on Koc values, metolcarb and molinate were classified as mobile, fenobucarb as mobile or moderately mobile isazofos as moderately mobile, diazinon, fenitrothin, dimepiperate, and parathion as slightly or moderately mobile and chlorpyrifos-methyl as slightly mobile. Two evaluation methods, Groundwater Ubiquity Score (GUS) index and standard indices of soil-chemical adsorption and biodegradation (half-life), were used for the estimation of pesticide leaching potential. Leachabilities of metolcarb, molinate and fenobucarb were evaluated as high, and isazofos, dimepiperate and diazinon as a little potential, while fenitrothion, parthion and chlorpyrifos-methyl showed very low leaching potential. The leaching potential of pesticides was determined on the basis of intrinsic properties of the pesticides and the soil properties. Among the soil properties, organic matter gave a great influence on the leachability in soil. Therefore, leachabilities of the pesticides used were expected less in Sineom soil with relatively higher organic matter than Gangseo, Jungdong and Yesan soil with lower organic matter.

Adsorption and Leaching Characteristics of the Insecticide Imidacloprid in Paddy Soils (살충제 Imidacloprid의 논토양 중 흡착 및 용탈 특성)

  • In order to elucidate the adsorption and leaching characteristics of the insecticide imidacloprid in two types of paddy soils near Suwon, this experiment was carried out with $[^{14}C]$imidacloprid as a radiotracer. In an adsorption study conducted using $[^{14}C]$imidacloprid in 2 test soils, the adsorption coefficient was higher in soil A $(K_f\;2.6)$ than that in soil B $(K_f\;1.7)$. As calculated from Freundlich constant, distribution coefficients and half lives in soils, GUS indices showed low leachabilities of imidacloprid treated on the paddy soils into the groundwater. The amount of imidacloprid leached from the soil columns during the 4 weeks of leaching was less than 2% of the originally treated $^{14}C$. In the leaching test the amounts of $^{14}C$ activities distributed in the soil layer of 0-10 cm were more than 80% of the originally tented $^{14}C$ and those in rice plants were less than 3% of the originally treated $^{14}C$, suggesting that imidacloprid has very low teachability and bioavailability.

  • Soil adsorption study was carried out to define the mobility of pesticides or to evaluate leaching potential in soils. Five pesticides including ethoprophos, procymidone, iprobenfos, isoprothiolane, and butachlor were subjected to optimized adsorption experiment protocol for three types of cultivation soils. Freundlich adsorption coefficients (K) were ranged $0.35{\sim}0.95$ for ethoprophos, $0.98{\sim}2.2$ for iprobenfos, $1.2{\sim}4.3$ for procymidone, $1.5{\sim}3.5$ for isoprothiolane and $7.9{\sim}19$ for butachlor in three soils. Based on Koc values, ethoprophos was classified as mobile, iprobenfos, isoprothiolane and procymidone as moderately mobile and butachlor as slightly mobile. Two evaluation methods, Groundwater Ubiquity Score (GUS) index and standard indices of soil-chemical adsorption and biodegradation, were used for the estimation of pesticide leaching potential. Leachability of isoprothiolane and iprobenfos were evaluated as moderate, ethoprophos as a little potential, while butachlor and procymidone showed very low leaching potential. Among the soil properties, organic matter gave a great influence on the leachability of soils. Therefore, leachabilities of pesticides were expected less in loam with relatively higher organic matter than clay loam with lower organic matter.

  • Binary trans-activation (pOp/LhG4) system is one of the regulatory systems of transgene expression. The target gene expression is achieved by crossing the reporter plants with an activator in this system. In this study, we used the features of this system in Chinese cabbage as a way to protect genetic resources and new varieties. To establish pOp/LhG4 system in Chinese cabbage, we designed an activator (35SLhG41300), and reporter constructs (pOpGUSBart) and co-transformed using Agrobacterium. The transgenic plants were selected by antibiotics and the functional activity of pOp/LhG4 system was confirmed by GUS expression. To induce the tissue-specific function, we constructed pOp/LhG4 system (795LhGBart) using female tissue specific promoter (ProAt1g26795) of Arabidopsis. Co-transformed transgenic plants clearly showed tissue specific expression in Arabidopsis. The results suggest the possibility of the system's application of $F_1$ generation can be restricted by expressing the target gene to protect a new variety and genetic resource in Chinese cabbages.

  • 최근 뛰어난 성능의 모바일 기기들이 많이 사용되고 있으며 특히나 GIS를 통해 다양한 공간 질의의 서비스 사용이 많아지고 있다. 본 논문에서는 GUS 서버의 전반적인 시스템 구조와 데이터 관리기의 설계 및 구현, 서버 및 PDA 기반 클라이언트의 사용자 인터페이스 등에 대해 소개하고자 한다. 데이터 관리기는 데이터를 객체화하거나 바이트 배열화하고 데이터의 안전한 백업을 보장하는 등의 중요한 역할을 한다. 또한 사용자가 보다 쉽고 자유롭게 질의를 작성할 수 있는 사용자 인터페이스를 구현하였다.

Strength Map of Presidential Candidates 2019 in Indonesia Based on a NodeXL Analysis of Big Data from Twitter

  • Leading up to the 2019 presidential election in Indonesia, campaigns have emerged through social media, particularly Twitter, using various hashtags, such as #2019GantiPresiden (2019 Change President) and #TetapJokowi (Always Jokowi). This paper tries to understand the presidential candidates' power map in forming opinions and influencing voter behavior by analyzing Twitter from August 6, 2018 to September 15, 2018, just before the beginning of the official campaign period, by searching for the keyword "pemilihan presiden RI Tahun 2019" (RI presidential election in 2019). According to our NodeXL's analysis, there were 1,650 active Twitter users talking about the 2019 presidential election. The 1,650 Twitter users have formed a communication network of 46,750 relationships formed from messages in the form of tweets, comments, and retweets. Our analysis found that those mentioning "pilihan presiden 2019" form large communication networks around four clusters: one for each of the two candidates (Jokowi and Prabowo) and two for opinion leaders who are undecided about the election (Gus Mus and Mas Piyu). GusMus is a religious leader, as an official of the PBNU Rais Syuriah (an Islamic organization) and has a large following both on and off Twitter. "MasPiyu" is an unidentified Twitter user; he only has a large following on Twitter, but does not have support offline.

Optimization of different factors for an Agrobacterium-mediated genetic transformation system using embryo axis explants of chickpea (Cicer arietinum L.)

  • In this study, we developed a reliable and efficient Agrobacterium-mediated genetic transformation system by applying sonication and vacuum infiltration to six chickpea cultivars (ICCV2, ICCV10, ICCV92944, ICCV37, JAKI9218, and JG11) using embryo axis explants. Wounded explants were precultured for 3 days in shoot induction medium (SIM) before sonication and vacuum infiltration with an Agrobacterium suspension and co-cultivated for 3 days in co-cultivation medium containing 100 µM/l of acetosyringone and 200 mg/l of L-cysteine. Responsive explants with putatively transformed shoots were selected using a gradual increase in kanamycin from 25 mg/l to 100 mg/l in selection medium to eliminate escapes. Results showed optimal transformation efficiency at a bacterial density of 1.0, an optical density at 600 nm wavelength (OD600), and an infection duration of 30 min. The presence and stable integration of the β-glucuronidase (gusA) gene into the chickpea genome were confirmed using GUS histochemical assay and polymerase chain reaction. A high transformation efficiency was achieved among the different factors tested using embryo axis explants of cv. JAKI 9218. Of the six chickpea cultivars tested, JAKI9218 showed the highest transformation efficiency of 8.6%, followed by JG11 (7.2%), ICCV92944 (6.8%), ICCV37 (5.4%), ICCV2 (4.8%), and ICCV10 (4.6%). These findings showed that the Agrobacterium-mediated genetic transformation system will help transfer novel candidate genes into chickpea.

Highly efficient production of transgenic Scoparia dulcis L. mediated by Agrobacterium tumefaciens: plant regeneration via shoot organogenesis

  • Efficient Agrobacterium-mediated genetic transformation of Scoparia dulcis L. was developed using Agrobacterium tumefaciens strain LBA4404 harboring the binary vector pCAMBIA1301 with ${\beta}$-glucuronidase (GUS) (uidA) and hygromycin phosphotransferase (hpt) genes. Two-day precultured leaf segments of in vitro shoot culture were found to be suitable for cocultivation with the Agrobacterium strain, and acetosyringone was able to promote the transformation process. After selection on shoot organogenesis medium with appropriate concentrations of hygromycin and carbenicillin, adventitious shoots were developed on elongation medium by twice subculturing under the same selection scheme. The elongated hygromycin-resistant shoots were subsequently rooted on the MS medium supplemented with $1mg\;l^{-1}$ indole-3-butyric acid and $15mg\;l^{-1}$ hygromycin. Successful transformation was confirmed by PCR analysis using uidA- and hpt-specific primers and monitored by histochemical assay for ${\beta}$-GUS activity during shoot organogenesis. Integration of hpt gene into the genome of transgenic plants was also verified by Southern blot analysis. High transformation efficiency at a rate of 54.6% with an average of $3.9{\pm}0.39$ transgenic plantlets per explant was achieved in the present transformation system. It took only 2-3 months from seed germination to positive transformants transplanted to soil. Therefore, an efficient and fast genetic transformation system was developed for S. dulcis using an Agrobacterium-mediated approach and plant regeneration via shoot organogenesis, which provides a useful platform for future genetic engineering studies in this medicinally important plant.

Development of Seashore Paspalum Turfgrass with Herbicide Resistance

  • Seashore Paspalum (Paspalum vaginatum Swartz) is a warm season grass and indigenous to tropical and subtropical regions of coastal areas worldwide. The species is used as feed for cattle and horses and has been very successful for golf courses worldwide. One of the most outstanding characteristics of seashore paspalum is its tolerance to saline soils compared to other warm season turfgrasses. The development of new seashore paspalum cultivars with improved traits could be facilitated through the application of biotechnological strategies. The purpose of this study was to product for herbicide resistant seashore paspalum using Arobacterium-mediated transformation and this study is the first report on transformation and herbicideresistant transgenic plants in seashore paspalum. Embryogenic calli were induced from the seeded variety of pseashore paspalum. Embryogenic calli were transformed with Agrobacterium tumefaciens strain EHA105 carrying the binary vector pCAMBIA3301 with two genes encoding gusA and bar. Transformed calli and plants were selected on medium containing 3 mg/l PPT. PCR detected the presence of the gusA and bar gene, indicating both genes are integrated into the genome of seashore paspalum. A chlorophenol red assay was used to confirm that the bar gene was expressed. By application of herbicide BASTA, the herbicide resistance in the transgenic seashore paspalum plants was confirmed.
