Coenzyme Q10(CoQ10)은 피부를 포함한 모든 생체조직에 존재하는 널리 알려진 조효소이다. 전자전달에 관여하는 퀴논링은 세포에서 에너지를 생성하기 위한 매우 중요한 기능을 가지고 있다. CoQ10은 피부에서 항산화제로서 연구되어 왔으며, 최근 외용제로써 노화억제와 주름개선작용에 대해 보고된 바 있다. 이런 보고들은CoQ10이 항산화제로서 산화-환원작용을 통해 피부의 방어기능에 중요한 역할을 한다는 점을 시사하며, 일반적으로 산화-환원작용은 피부에서 흑화과정의 조절에도 많은 영향을 미친다. 따라서 본 연구자들은 CoQ10 이 피부의 색소조절기능이 있는지 알아보고자 하였다. 인체 정상 색소세포에CoQ10을 0.05-0.5 mM 처리한 결과 0.5, 0.25mM에서 멜라닌의 생합성을 약 50% 저해하였으며 이는 알려진 미백제인 Kojic acid나 vitamin C와 유사한 수준이었다. 또한, CoQ10은 인체 정상 색소세포에서 자외선이나 세포내 cAMP 증가 유도물질에 의한 멜라닌 생성을 억제하였다. 그러나 tyrosinase inhibitor인 kojic acid와는 달리, in vitro tyrosine hydroxylase의 억제효과는 보이지 않았다. CoQ10을 자외선으로 tanning을 유도시킨 brown guinea pig에 4주간 도포하고 육안 및 chromameter를 이용하여 미백효과를 측정한 결과, vehicle처리군에 비해 미백효과가 있음을 확인할 수 있었다. 이상의 결과에서 coenzyme Q10 은 in vitro및 in vivo에서 미백효과를 지닌 물질임을 확인할 수 있었다.
Kojic acid, antimelanogenic agent, has been widely used in cosmetics to lighten the skin color. However, it has skin irritancy and instability against pH, temperature and light. To overcome these problems and optimize the molecular structure of kojic acid (KA), a prodrug, kojic acid monostearate(KMS), has been synthesized to modify the topical drug delivery in the point of sustained release of the parent drug via enzymatic hydrolysis during skin absorption. The prodrug was tested for enzymatic hydrolysis with cytosolic fraction of hairless mouse, skin. From the in vitro skin permeation study through hairless mouse skin, we found that KMS was retained in the skin and generated KA continuously by the skin esterase cleavage. In addition, topical formulations of o/w type creams and polyolprepolymer-containing cream were further tested for whitening effects using in vivo yellow skin guinea pig model.
In traditional medicines, Dansam-samul-tang and Samwhang-sasim-tang extracts were known to effects of antioxidant and antimicrobial activity. To investigate the safety evaluation of skin, these extracts were measured to skin safety testing such as primary skin irritation test, eye irritation test and skin sensitization test. The results of the primary skin irritation test and eye irritation test in vitro showed that these extracts included in the nonirritating area. Skin sensitization test results by Guinea Pig Maximization (GPMA) indicated that there was no allergy reaction. Therefore, Dansam-samul-tang and Samwhang-sasim-tang extracts were very safe in every safety test of skin. These results suggests that Dansam-samul-tang and Samwhang- sasim-tang extracts in 1 % concentration can be useful cosmetic ingredients.
본 연구에서는 H. pylori의 감염을 예방하고 치료보조제로 사용할 목적으로 포유동물을 통한 피동면역용 항체를 생산하고자 하였다. 따라서 anti-H. pylori 항체를 함유한 면역우유 생산용 백신개발에 기초자료를 얻고자 H. pylori의 면역성과 면역독성에 관한 실험을 수행하였다. 백신을 반복해서 투여했을 때 야기될 수 있는 알레르기 및 과민반응을 예측하고, 페니실린 쇼크와 같은 심각한 부작용 및 독성유발 가능성을 검색하기 위하여 백신의 면역독성을 평가하였다. 전신성 anaphylaxis 쇼크반응의 유무를 평가하기 위해 각 군당 5마리의 guinea pig에 감작투여한 후 최종감작 1주와 2주째에 귀정맥(ear vein)으로 야기항원을 투여하였다. anaphylaxis 쇼크 반응 시험에서 5마리 중 1마리가 양성반응을 나타낸 경우 의약품의 안전성 평가라는 관점에서는 양성으로 판정한다[11]. H. pylori의 WC항원에 대한 1차 및 2차 야기항원 투여 후 전신성 anaphylaxis 쇼크반응에 대한 관찰 결과는 다음과 같다. H. pylori의 WC에 대한 anaphylaxis 쇼크반응은 WC (H) $60\;{\mu}g/100\;{\mu}l$의 항원농도에서 1차, 2차 야기항원 투여 모두 경증의 증상을 나타내었고, WC (L) $20\;{\mu}g/100\;{\mu}l$의 항원농도에서는 아무런 anaphylaxis 쇼크 증상이 관찰되지 않았다. 그리고 crude urease에 대한 anaphylaxis 쇼크반응은 항원농도가 $20\;{\mu}g/100\;{\mu}l$의 urease (L)와 $60\;{\mu}g/100\;{\mu}l$의 urease (H) 모두에서 아무런 증상도 관찰되지 않았다. Guinea pig-rat를 이용한 PCA 시험에서는 WC (H), WC (L), urease (H), urease (L) 투여군 모두에서 양성반응이 나타나지 않았다. 피부감작성 시험에서는 항원농도에 따라 각각 $80\;{\mu}g/100\;{\mu}l$, $40\;{\mu}g/100\;{\mu}l$, $20\;{\mu}g/100\;{\mu}l$, $20\;{\mu}g/100\;{\mu}l$ 일 때 피부이상 증상 즉, 피부 트러블이 발생하지 않는 최고의 항원농도는 $40\;{\mu}g/100\;{\mu}l$인 것으로 관찰되었다. 결론적으로 항원성 시험에서 H. pylori로부터 분리된 urease 항원이 WC 항원보다 면역독성 측면에서 좀 더 안전할 것으로 조사되었다.
The purpose of this study is to evaluate the skin whitening effect of Scirpi rhizama ethanol extract (SREE) in an animal model. For the experiment of the study, three brown guinea pigs weighing about 450 g to 550 g were exposed to ultraviolet-B rays on the backs at 500 $mJ/cm^2$ once a week, three consecutive weeks and the total quantity of light was 1,500 $mJ/cm^2$. The artificial tanning spots were divided into six different groups including normal (N), control (C), vehicle control (VC), positive control (PC), experimental 1 (E1, 1% SREE), experimental 2 (E2, 2% SREE) groups. Then, 30 ${\mu}L$ of SREE was transdermaly applied on the artificial tanning spots twice a day and 5 days a week for 8 weeks. With the result of a gross observation, it was found that the degree of pigmentation became apparently thinner in the group applied with E2, compared to the control or the vehicle control group. The melanin index of E2 group was significant lower than the control or the vehicle control group. In the observation with a light microscope, it was found that the degree of melanin pigmentation and S-100 protein expression considerably decreased in the groups applied with SREE, compared to the control or the vehicle control group. With the numerical analysis of melanin pigmentation and S-100 protein expression by using image-analysis software, it was found that the tendency was coincide with the results of microscopic observation.
Chitosan cross-linked collagen-glycosaminoglyan (CCGM) is an artificial skin substitute made to form a sponge like dimensional matrix. It can be used to facilitate reconstruction of dermal tissue when applied on large wounds such as severe burns. In order to study the toxicological effects of CCGM the cytotoxicity, local irritation and skin sensitization test were carried out according to the standards of ISO 10993. In the cytotoxicity test utilizing LDH and MTT test, both the CCGM and its extract had no toxicity of Balb/c 3T3 cells. The local irritatioin test on rabbit skin demonstrated that CCGM did not promote any harmful when directly applied on skin. In addition, it did not elicit any allergic reaction in the guinea pig maximization test. Based on these results, it is suggested that CCGM is a material without cytotoxicity, local irritation and allergenicity.
The general pharmacological effects of a new anthracycline anticancer agent, DA-125 $[7-0-(2,\;6-dideoxy-2-fluoro-{\alpha}-L-talopyranosyl)-adriamycinone-14-{\beta}-alaninate{\cdot}HCI]$ were investigated in mice, rats, guinea pigs, rabbits and dogs. Intravenous administration of DA-125 presented no significant effects on the central and peripheral nervous systems of ICR mice except a decrease in the numbers of acetic acid-induced writhing response at a dose of 10 mg/kg. In anesthetized rats and dogs, DA-125 produced a transient depression of blood pressure and an increase in heart rate, but did not affect the peripheral blood flow in the isolated ear vessels of rabbits and the mechanical functions of the isolated hearts of guinea pigs. No significant effects were observed on the gastrointestinal functions and the contractilities of smooth muscle preparations obtained from guinea pig trachea, rabbit ileum, pregnant and non-pregnant uterus and vas deferens of rats. DA-125 Increased the contractility of the isolated ileum of guinea pigs in a dose range of $10^{-6}{\sim}10^{-9}g/ml$, and also increased, but weaker than adriamycin, the vascular permeability in rat skin. DA-125 had no effect on the kallikrein-induced increase in permeability and the permeability of the visceral organs. DA-125 did not adversely affect the liver function and the blood coagulation system, and did not induce hemolysis in vitro. It is concluded from the results that the general pharmachological effects of DA-125 are similar to or weaker than those of adriamycin, and that little adverse effects are anticipated with a therapeutic dose range.
Guinea pigs were cutaneously inoculated with Trichophyton verrucosum var album, that is a common causative fungus of dermatophytosis in cattle. The developmental process of lesions, clinical and histopathological changes and reisolations of the fungi were studied to evaluate the pathogenicity of Trichophyton verrucosum var album in guinea pigs. Results obtained through the experiments were summarized as follows : 1. The incidence of infection of the clipping group was 13(86%) of 15 animals, and that of the plucking group was 14(93%) of 15 animals. In both of the clipping and plucking groups, visible cutaneoas lesions were developed between 4 and 7 days post inoculation(p.i.). The spreading and the climax stages persisted for 4 to 11 and 6 to 12 days, respectively. 2. In macroscopic observations, formation of various degree of erythemas and scales over the inoculated skin sites were observed in the spreading stage. In the climax stage, exudative changes and dark red crusts were formed as typical circular lesions. In the healing stage, the lesions revealed shedding of crust, alopecia and hair regrowth. 3. In histopathological observations, infiltration of inflammatory cells, hyperplasia, microabscesses and keratinous-hyaloid materials of epidermis were observed in the spreading stage. Hyphal invasion was primarily observed at the level of epidermis and pilosebaceous ducts. In the climax stage, the infected epidermis was thick with severe hyperplasia, hyperkeratosis and acanthosis. The microabscesses with fungal hyphae, folliculitis and hyperplasia of external root sheath were observed in the dermis. The fungal hyphae were observed only in the tissues of hair follicles, that were internal root sheath, cuticle, the keratinized portions of cortex and medulla 4. In reisolation of the inoculated fungus, all trials for ten animals showed positive cultures until 25 days p.i.. Afterward, the reisolation rates were gradually decreased, showing all negative after 40 days p.i..
The present studies, the investigations were undertaken for an attemp to analyze time course change of biogemic amines in respons to bite injury in rats and guinea pigs, and obtained the following results that might be applicable to the forensic medicine and Forensic Odontology. The histamine concentrations measured at rat abdominal skins were decreased during the first 8 hours and recovered to control levels after 12 hours. However the serotonin concentrations were decreased during the first hours and increased during 4-8 hours, and then returned to control levels thereafter. In contrast with these results, both histamine and serotonin concentrations were tended to increase in guinea pig skins, but the concetration of histamine showed maximum rate of increase immediately after injury and those of sertonin showed maximum rate of increase at 30 minutes. The differencd in the response of histamine and serotonine after injury between rats and ginea pigs revealed that there is specials deffrence in the response of biogenic amines with regard to external stimuli. These results suggest that the determinition of biogenic amines in skin lesion of bite marks could be applied to the estimation of the elapsed time after antemortem injury.
본 논문은 백지 에탄올추출물의 미백효능을 조사하기 위하여 melan-a cell, 브라운 기니피그 및 HMB-45 염색을 사용하였다. melan-a 세포에 시료를 6.25, 12.5, 25, 50, ${\mu}g/m{\ell}$ 농도로 처치하여 세포 생장율의 변화 관찰 및 멜라닌세포의 형태학적 변화를 관찰하였다. 또한 brown guinea pig (450~500 g)등 부위에 1500 mJ/$cm^2$ 광량의 ultraviolet B조사로 유발시킨 인공색소반 (${\Phi}$ 2 mm)에 1일 2회, 주 5일, 매회 30 ${\mu}{\ell}$씩 총 5주간 시료를 도포하여 주 1회 육안적 변화를 관찰 하였고, 실험 5주째 되는 날 실험동물은 염산 케타민으로 마취 후 시료를 도포한 인공색소반 부위를 biopsy punch로 절취하여 HMB-45 염색으로 멜라닌소체 내 glycoprotein(gp100) 단백질의 조직학적 변화를 관찰 하였다. 세포생장율 측정에서 시료처치 6.25~50 ${\mu}g/m{\ell}$에서의 세포생장율은 높았고, 멜라닌세포의 형태학적 변화에서는 시료처치 농도가 증가할수록 멜라닌합성 및 수지상 돌기가 적게 관찰 되었다. 동물 실험 육안적 관찰에서 시료 도포군이 용매대조군에 비해 멜라닌 색소침착 정도가 확연하게 밝아졌고, gp100 단백질의 조직학적 관찰에서 시료 도포군은 gp100 단백질 발현 정도가 현저하게 줄어들었으며, 영상분석에서도 시료 도포군이 용매대조군에 비해 유의하게(p<0.001) 감소하였는데 이러한 결과는 현미경 관찰 결과와 일치하였다. 이상의 결과를 종합하면 백지 에탄올추출물은 우수한 미백효과가 있는 것으로 판단된다.
