Lactobacillus sakei 생육저해활성 보유 Leuconostoc mesenteroides가 생산하는 Bacteriocin의 특성 (Characterization of the Bacteriocin Produced by a Leuconostoc mesenteroides Strain Inhibiting the Growth of Lactobacillus sakei)

  • 김치발효 후기의 우점종으로 알려진 Lactobacillus sakei의 생육을 저해하는 Leuconosotc 속 23균주가 김치로부터 분리되었고, 이들 중 가장 높은 생육저해활성을 나타낸 Leuconostoc mesenteroides CK0122 균주가 생산하는 생육 저해물질의 특성을 검토하였다. CK0122 균주의 배양 상징액을 분자량 3,000 Da을 기준으로 분획하여 동결건조한 분자량 3,000 Da 이하의 조항균물질로부터 Lb. sakei 생육저해가 확인되었고, proteinase K 처리에 의해 항균력이 소실되었기 때문에, CK0122가 생산하는 항균물질은 분자량 3,000 Da 이하의 단백질성 항균물질(bacteriocin)으로 추정된다. 분자량 3,000 Da 이하의 조항균물질은 pH 4에서 7 정도의 영역에서 최적활성을 나타내었고, $121^{\circ}C$, 15분 살균처리에 의해서도 활성이 유지되는 높은 열안정성을 나타내었으며, Bacillus cereus, Listeria monocytogenes, Salmonella typhimurium, Alcaligenes xylosoxydans, Staphylococcus aureus 및 Flavobacterium sp.에 대하여 항균 활성을 나타내어 Gram 양성균 및 음성균의 제어에 적용 가능한 것으로 나타났다.

세균에 의한 남조 Anabaena cylindrica의 분해에 대한 연구 (A Study on the Lysis of a Bluegreen Alga Anabaena cylindrica by a Bacterium)

  • A Gram (-), rod-shaped bacterium in size of 1.6-2.8 $\times$ 0.4 μm was isolated from a eutrophic reservoir, which exhibited growth-inhibiting effect against a bluegreen alga (Anabaena cylindrica). This isolate showed positive reactions for catalase and oxidase, and optimal conditions of 35-40°C and pH 9.0. This isolate was designated AC-1 in this manuscript. In a mixed-culture of A. cylindrica and AC-1, their growth patterns were inversely correlated and the bluegreen algal vegetative cells completely disappeared within 24-36 hours. AC-1 showed similar lytic activity in natural water as in an artificial medium. The lytic activity of AC-1 was dependent on the photosynthetic activity of A. cylindrica. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas heterocysts have not been lysed.

Relationship between Chemical Structure and Antimicrobial Activities of Isothiocyanates from Cruciferous Vegetables against Oral Pathogens

  • We evaluated the potentials of 10 isothiocyanates (ITCs) from cruciferous vegetables and radish root hydrolysate for inhibiting the growth of oral pathogens, with an emphasis on assessing any structure-function relationship. Structural differences in ITCs impacted their antimicrobial activities against oral pathogens differently. The indolyl ITC (indol-3-carbinol) was the most potent inhibitor of the growth of oral pathogens, followed by aromatic ITCs (benzyl ITC (BITC) and phenylethyl ITC (PEITC)) and aliphatic ITCs (erucin, iberin, and sulforaphene). Sulforaphene, which is similar in structure, but has one double bond, showed higher antimicrobial activity than sulforaphane. Erucin, which has a thiol group, showed higher antimicrobial activity than sulforaphane, which has a sulfinyl group. BITC and iberin with a short chain exhibited higher antimicrobial potential than PEITC and sulforaphane with a longer chain, respectively. ITCs have strong antimicrobial activities and may be useful in the prevention and management of dental caries.

Effect of Probiotic Clostridium butyricum NCTC 7423 Supernatant on Biofilm Formation and Gene Expression of Bacteroides fragilis

  • Enterotoxigenic Bacteroides fragilis (ETBF) is the main pathogen causing severe inflammatory diseases and colorectal cancer. Its biofilm plays a key role in the development of colorectal cancer. The objective of this study was to determine the antagonistic effects of cell-free supernatants (CFS) derived from Clostridium butyricum against the growth and biofilm of ETBF. Our data showed that C. butyricum CFS inhibited the growth of B. fragilis in planktonic culture. In addition, C. butyricum CFS exhibited an antibiofilm effect by inhibiting biofilm development, disassembling preformed biofilms and reducing the metabolic activity of cells in biofilms. Using confocal laser scanning microscopy, we found that C. butyricum CFS significantly suppressed the proteins and extracellular nucleic acids among the basic biofilm components. Furthermore, C. butyricum CFS significantly downregulated the expression of virulence- and efflux pump-related genes including ompA and bmeB3 in B. fragilis. Our findings suggest that C. butyricum can be used as biotherapeutic agent by inhibiting the growth and biofilm of ETBF.

칠엽일지화(七葉一枝花)의 항종양(抗腫瘍) 효과(效果) (Study on Antitumor Activity of Paridis Rhizoma)

  • To evaluate the antitumor activity of Paridis Rhizoma(PR), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against MCF-7, SK-OV-3, HCT15, concentration inhibiting cell growth up to below 50% of control was recognized at $100-200{\mu}g/m{\ell}$ of PR and also against A549, XF498 was recognized at $50-100{\mu}g/m{\ell}$. 2. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $50-100{\mu}g/m{\ell}$ of PR. 3. The concentration inhibiting adhesion of A549 and SK-OV-3 to complex extracellular matrix up to below 50% of control was recognized at $10-100{\mu}g/m{\ell}$ of PR. 4. The T/C% was 137.9 in PR-treated group in S-180 bearing ICR mice. From above results it was concluded that PR could be usefully applied for the prevention and treatment of cancer.

Purification and Characterization of an Antibacterial Substance from Aerococcus urinaeequi Strain HS36

  • A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.

Growth-Inhibiting Effect of Bufadienolides on Cultured Vascular Endothelial Cells

  • We found that bufalln, one of the prominent components of the bufadlenolides in the Chinese medicine chan'su, has the potent inhibitory effects on growth and proliferation of the cultured bovine aortlc endothelial (BAE) and human umbilical vein endothelial (HUVE) cells. All naturally-occuring bufadienolides used in this study inhibited the cell growth in a dose-dependent manner. Particularly, bufalin among the bufadienolides showed the strongest inhibitory activity for the cell growth. The order of growth inhibition by bufadienolides on BAE cells was as follows: bufalin > gamabufotalln > bufotalln > cinobufagin > cinobufotalin > resibufogenin. The $IC_50$ values (50% inhibition of cell growth) of bufalin as determined by XTT assay were the range of 1-10 nM in BAE and HUVE cells. Bufalin exhibited a higher sensitivity towards cultured bovine aortic endothelial cells than human umbilical vein endothelial cells.

Growth and Chlorophyll Biosynthesis of Vigna angularis under Lead Stress

  • The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna anguluris seedlings. Expose to 50 $\mu$N Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of AA reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. 5-Aminolevulinic acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.

Antimicrobial Activity of Quinoline Derivatives Isolated from Ruta chalepensis Toward Human Intestinal Bacteria

  • The growth responses of Ruta chalepensis leaf-derived materials toward human intestinal bacteria were examined. The biologically active constituent of the R. chalepensis extract was characterized as quinoline-4-carboxaldehyde($C_{10}H_{7}NO$). The growth responses varied depending on the bacterial strain, chemicals, and dose tested. At 0.25 and 0.1 mg/disk, quinoline-4-carboxaldehyde strongly inhibited the growth of Clostridium perfringens and weakly inhibited the growth of Escherichia coli without any adverse effects on the growth of three lactic acid bacteria. Furthermore, at 0.05 and 0.025 mg/disk, this isolate showed moderate activity against C. perfringens. In comparison, chloramphenicol at as low as 0.01 mg/disk significantly inhibited the growth of all bacteria tested, and cinnamaldehyde at 0.25 mg/disk did not inhibit Bifidobacterium bifidum, B. longum, E. coli, and Lactobacillus acidophilus, with the exception of C. perfringens. The structure-activity relationship revealed that quinoline-3-carboxaldehyde had strong growth inhibition against C. perfringens, but quinoline, quinoline-3-carboxylic acid, and quinoline-4-carboxylic acid did not inhibit the growth of B. bifidum, B. longum, C. perfringens, E. coli, and L. acidophilus. These results indicate that the carboxyl aldehyde functional group of quinolines seems to be required for growth-inhibiting activity against C. perfringens, thus indicating at least one of the pharmacological actions of R. chalepensis leaf.
