Effects of Electroacupuncture and Electrical Stimulation on VEGF Expression After Muscle Crush Injury in Rats (전침 자극과 전기 자극의 적용이 근육압좌손상이 유발된 흰쥐의 골격근 내 혈관내피성장인자 발현에 미치는 영향)

  • Skeletal muscle injury occurs frequently in sports medicine and is the most general form of injury followed by physical impact. There are growth factors which conduct proliferation, differentiation, and synthesis of myogenic prodromal cells and regulate vascular generation for the continued survival of myocytes. The purpose of the present study was to confirm the effects of electroacupuncture (EA) and electrical stimulation (ES) on muscle recovery processes according to vascular endothelial growth factor (VEGF) expression. Eighteen Sprague-Dawley rats were separated into 2 experimental groups and a controlled group. All animals had suffered from crush damage in the extensor digitorum longus for 30 seconds and were killed 1, 3, and 7 days after injury. 30 Hz and 1 mA impulsion for 15 minutes was applied to the EA experimental groups Zusanli (ST36) and Taichong (LR3) using electroacupuncture and the same stimulation was applied to the ES group using an electrical node. Hematoxyline-Eosin staining and VEGF immunohistochemistry were used to ascertain the resulting muscle recovery. There were few morphological differences between the EA and ES groups, and both groups were observed to have tendencies to decrease atrophy as time passed. In the controlled group, gradually diminishing atrophy could be observed, but their forms were mostly disheveled. There were few differences in VEGF expression between the EA and ES groups, and tendencies to have an increased quantity of VEGF with the lapse of time were observed in both groups. In the controlled group, a little VEGF expression could be observed merely 7 days after injury. In conclusion, EA and ES contributed to muscle recovery processes and could be used for the treatment of muscle injury.

Base Metal's Price Hike and Chinese Economic Growth (Base Metal 가격상승과 중국의 경제성장)

  • With the financial crisis from USA had negative impacts on the real economy, base metals price on LME was downward in 4 quarter of 2008. Following the deepest global downturn in recent history, economic growth solidified and broadened to advanced countries and simultaneously the price of base metal on LME showed a rising curve over 2009. There are three factors supported an upward tendency of base metal's price. The First factor is the US economy recovery, the second factor is the weak dollar, the third factor is the chinese base metal demand. Among the factors, the last one is a major factor. Therefore, this study analyze the factor of the movement of price of base metal with linear regression analysis. The result of analysis show that the chinese GDP growth has effect on the recent upward base metal price. Despite the result, the upward movement is difficult to be sustained without the full recovery of advanced economies.

Establishment of a Standard Nitrogen Application Rate for Zoysia matrella Using Growth Responses to Various Fertilization Level (질소 시비 수준별 금잔디의 생육과 질소 시비량 설정)

  • BACKGROUND: Nitrogen (N) is an important element for turfgrass (Zoysia matrella) growth; however, standard N application rate for turfgrass is not established yet. This study was conducted to evaluate the effect of N application rates on the growth and quality of turfgrass for establishment of standard N application rate. METHODS AND RESULTS: Treatments were as follows; control (0 N g/m2/month), 1N (1 N g/m2/month), 2N(2 N g/m2/month), 3N (3 N g/m2/month), 4N (4 N g/m2/month), and 5N (5 N g/m2/month). N application improved visual turfgrass quality. Compared with the control, clipping yield of all N treatments increased by 90~194%. The grass shoot weight of 3N, 4N, and 5N treatments increased by 52%, 43%, and 111%, respectively, and the stolon weight of 4N and 5N treatments increased by 412% and 201%, respectively, compared to the control. The N uptake amount and N recovery rate were estimated to be 4.10~6.28 g/m2 and 14~58%, respectively. CONCLUSION(S): These results indicate that considering visual quality, clipping yield, N uptake amount, and N recovery, the application rate of 2~3 N g/m2/month was suggested to be suitable for Z. matrella production.

  • In the previous study, we reported the result to prepare lithium carbonate powder from various lithium-contained solution. Therefore, using the lithium hydroxide solution, it is conformed that the reaction could occur thermodynamically, and the recovery rate of lithium was 89.4 %. In this study, we carried out the experiment to prepare lithium carbonate powder through gas-liquid reactions with lithium hydroxide solution and CO2 gas using ultrasound energy. In case ultrasonic energy is applied to the reaction of lithium carbonate, the recovery rate of lithium at room temperature was approximately 83.8 %, and the recovery rate of lithium was greatly increased to approximately 99.9 % at 60℃ reaction temperature. And when ultrasonic energy is not applied, the particle size of lithium carbonate powder was 7.7 ㎛ in D50. But the particle size of lithium carbonate powder was significantly reduced to 8.4 ㎛ in D50 under the influence of ultrasonic.

Effects of Aqueous Extract of Achyranthes Japonica on Functional Recovery in Sciatic Nerve after Crushed Sciatic Nerve Injury in Rats (우슬 추출물이 흰쥐 좌골신경 손상 후 좌골신경의 기능회복에 미치는 영향)

  • Objectives : Peripheral nerve injuries are commonly encountered clinical problem and often result in severe functional deficits. The aim of this study is to evaluate the effects of aqueous extract of Achyranthes japonica(AJ) on functional recovery in sciatic nerve after crushed sciatic nerve injury. Methods : In the present study, the animals in the AJ-treated groups received the aqueous extract of AJ at the respective doses orally for 13 consecutive days. In order to assess the effects of the aqueous extract of AJ on function recovery in crushed sciatic nerve injury, sciatic functional index(SFI) was performed. c-Fos expression in the paraventricular nucleus(PVN) and ventrolateral periaqueductal gray(vIPAG), and neurofilament, and the expressions of brain-derived neurotrophic factor(BDNF), nerve growth factor(NGF) following crushed sciatic nerve injury in rats were investigated. For this, immunohistochemistry and western blot were performed. Results : In the present study, crushed sciatic nerve injury showed characteristic gait changes showing decrease of SFI value and treatment with the aqueous extract of AJ significantly enhanced the SFI value. Neurofilament expression in the sciatic nerve was decreased by crushed sciatic nerve injury and treatment with the AJ increased neurofilament expression. The expressions of BDNF and NGF in the sciatic nerve were increased following crushed sciatic nerve injury and treatment with the AJ significantly controlled the sciatic nerve injury-induced increment of BDNF and NGF expressions. c-Fos expressions in the PVN and vIPAG were increased following crushed sciatic nerve injury and treatment with the AJ significantly suppressed the sciatic nerve injury-induced increment of c-Fos expressions. Conclusions : These results suggest that AJ treatment after crushed sciatic nerve injury is effective in the functional recovery by enhancing axonal regeneration and suppressing of pain.

Effects of Haein-tang(Hairen-tang) Extract on Functional Recovery in Sciatic Nerve and c-Fos Expression in the Brain after Crushed Sciatic Nerve Injury in Rats (해인탕 추출물이 흰쥐 좌골신경 손상 모델에서 기능회복과 뇌의 c-Fos 발현에 미치는 영향)

Neurofilament immunohistochemistry were performed on neurofilament regeneration. 4. Western blot were performed on brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF) expression. Results : 1. Haein-tang(Hairen-tang) extract significantly enhanced the SFI value in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 2. Haein-tang(Hairen-tang) extract significantly suppressed the sciatic nerve injury-induced increment of c-Fos expressions in the PVN and vIPAG in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 3. Haein-tang(Hairen-tang) extract significantly increased neurofilament expression in the sciatic nerve injury and 50 mg/kg, 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 4. Haein-tang(Hairen-tang) extract significantly controled the sciatic nerve injury-induced increment of BDNF and NGF expressions in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. Conclusions : These results suggest that Haein-tang(Hairen-tang) treatment after sciatic nerve injury is effective for the functional recovery by enhancing of axonal regeneration and suppressing of pain.

Reduction of Inflammation and Enhancement of Motility after Pancreatic Islet Derived Stem Cell Transplantation Following Spinal Cord Injury

  • Objective : Spinal cord injury (SCI) is a very serious health problem, usually caused by a trauma and accompanied by elevated levels of inflammation indicators. Stem cell-based therapy is promising some valuable strategies for its functional recovery. Nestin-positive progenitor and/or stem cells (SC) isolated from pancreatic islets (PI) show mesenchymal stem cell (MSC) characteristics. For this reason, we aimed to analyze the effects of rat pancreatic islet derived stem cell (rPI-SC) delivery on functional recovery, as well as the levels of inflammation factors following SCI. Methods : rPI-SCs were isolated, cultured and their MSC characteristics were determined through flow cytometry and immunofluorescence analysis. The experimental rat population was divided into three groups : 1) laminectomy & trauma, 2) laminectomy & trauma & phosphate-buffered saline (PBS), and 3) laminectomy+trauma+SCs. Green fluorescent protein (GFP) labelled rPI-SCs were transplanted into the injured rat spinal cord. Their motilities were evaluated with Basso, Beattie and Bresnahan (BBB) Score. After 4-weeks, spinal cord sections were analyzed for GFP labeled SCs and stained for vimentin, $S100{\beta}$, brain derived neurotrophic factor (BDNF), 2',3'-cyclic-nucleotide 3'-phosphodiesterase (CNPase), vascular endothelial growth factor (VEGF) and proinflammatory (interleukin [IL]-6, transforming growth factor $[TGF]-{\beta}$, macrophage inflammatory protein [MIP]-2, myeloperoxidase [MPO]) and anti-inflammatory (IL-1 receptor antagonis) factors. Results : rPI-SCs were revealed to display MSC characteristics and express neural and glial cell markers including BDNF, glial fibrillary acidic protein (GFAP), fibronectin, microtubule associated protein-2a,b (MAP2a,b), ${\beta}3$-tubulin and nestin as well as anti-inflammatory prostaglandin E2 receptor, EP3. The BBB scores showed significant motor recovery in group 3. GFP-labelled cells were localized on the injury site. In addition, decreased proinflammatory factor levels and increased intensity of anti-inflammatory factors were determined. Conclusion : Transplantation of PI-SCs might be an effective strategy to improve functional recovery following spinal cord trauma.

Interactive Effects of Ozone and Light Intensity on Platanus occidentalis L. Seedlings

  • Sycamore (Platanus occidentalis L.) seedlings were grown under low light intensity and ozone treatments to investigate the role of the light environment in their response to chronic ozone stress. One-year-old seedlings of Platanus occidentalis L. were grown in pots for 3 weeks under low light (OL, $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and high light (OH, $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) irradiance in combination with 150 ppb of ozone fumigation. After three weeks of ozone and light treatment, seedlings were placed in ozone free clean chamber for 3 weeks for recovery from ozone stress with same light conditions to compare recovery capacity. Ozone fumigation determined an impairment of the photosynthetic process. Reduction of leaf dry weight (14%) and shoo/root ratio (17%) were observed in OH treatment. OL treatment also showed severe reductions in leaf dry weight and shoot/root ratio by 48% and 36% comparing to control, respectively. At the recovery phase, OH-treated plants recovered their biomass, whereas OL-treated plant showed reduction in leaf dry weight (52%) and shoot/root ratio (49%). OH-treated plants reached similar relative growth rate (RGR) comparing to control, whereas OL-treated plants showed lower RGR in stem height. However, there were no significant differences in response to those treatments in stem diameter RGR at the recovery phase. Ozone treatment produced significant reduction of net photosynthesis in both high and low light treatments. Carboxylation efficiency and apparent quantum yield in OL-treated plants showed significant reductions rate to 10% and 45%, respectively. At the recovery stage, ozone exposed seedlings under high light had similar photosynthetic capacity comparing to control plants. Antioxidant enzymes activities such as superoxide dismutase (SOD), ascorbate peroxidase (APX), and glutathione reductase (GR) were increased in ozone fumigated plants only under low light. The present work shows that the physiological changes occur in photosynthesis-related parameters and growth due to ozone and low light stress. Thus, low light seems to enhance the detrimental effects of ozone on growth, photosynthesis, and antioxidant enzyme responses.

Improvement of Hydrocarbon Recovery by Two-Stage Cell-Recycle Extraction in the Cultivation of Botryococcus braunii

  • In situ extraction by organic solvent was studied in order to improve the recovery yield of hydrocarbon from the culture of Botryococcus braunii, a green colonial microalga. When the solvent mixture of octanol as an extractive solvent and n-octane as a biocompatible solvent was added to a two-phase column, the algal growth was seriously inhibited, even at a low concentration of polar octanol. Therefore, a two-stage cell-recycle extraction process was proposed to improve the contact area between the organic phase and the aqueous phase. The hydrocarbon recovery with in situ cell-recycle extraction showed a three-fold increase (57% of cell) in yield over that with two-phase extraction. In addition, over 60% of the hydrocarbon could be recovered without serious cell damage by downstream separation when this process was applied to the culture broth after batch fermentation.

Alkaline Protease of a Genetically-Engineered Aspergillus oryzae for the Use as a Silver Recovery Agent from Used X-Ray Film

  • Aspergillus oryzae U1521, which was a genetically engineered strain, produced 1,000,600 U per g . glucose of extracellular alkaline protease within 72 h in a submerged fermentation. However, the alkaline protease was not detected during the first 24 h. Northern blot analysis indicated that the enzyme synthesis was repressed at the transcriptional level during the lag period. Both catabolite repression and pH of the growth medium significantly affected the enzyme production. Use of this enzyme as a silver recovery agent from used X-ray film was confirmed by experiments in the shake-flask scale.
