We aimed to analyze the microbiological quality of the ready-to-eat (RTE) side dishes collected from traditional markets, supermarkets, and cafeterias in Gyeonggi-do in 2019. A total of 108 samples were analyzed for total aerobic bacterial counts, coliforms and foodborne pathogens depending on place of purchase and cooking methods. Results show that Bacillus cereus was detected in 14 (12.9%) out of 108 samples of side dishes, while no other foodborne pathogens were detected. The mean detected level (range) of total aerobic bacteria depending on place of purchase was 5.8 log CFU/g (3.0 to 8.2 log CFU/g) for traditional markets, 4.3 log CFU/g (2.4 to 7.8 log CFU/g) for supermarkets, and 3.80 log CFU/g (0.0 to 6.8 log CFU/g) for cafeterias, indicating that there was a significant (P<0.05) difference in total aerobic bacterial counts among places of purchase. Among the samples, the highest counts of total aerobic bacteria and coliforms were detected in saengchae (raw vegetables), followed by namul (seasoned herbs, vegetables), bokkeum (stir-fried foods), and jorim (foods cooked in soy sauce). The growth of total aerobic bacteria in seasoned soybean sprouts was inhibited when the sprouts were stored at 4℃ up to 24 h, whereas bacteria rapidly grew at 20 and 35℃ after 3 and 6 h, respectively. These results reveal that storage temperature might play a significant role for the microbiological quality of seasoned soybean sprouts when they are sold in markets. Thus, this study suggests that RTE side dishes should be stored at refrigerated temperature when being sold at markets as well as after purchasing to improve their microbiological quality.
Scuticociliatosis has badly settled one of most damaging diseases during the seedling production process of olive flounder. Paralichthys olivaceus in Korea. We isolated a new type of Scuticociliate from flounder. The parasite metamorphoses to ciliate and cyst phases with each other by environmental changes and survive for a relatively long span. The ciliate was measured average 41.8 ${\mu}m$ in length and 21.0 ${\mu}m$ in width, and cyst was 17.0 ${\mu}m$ and 13.5 ${\mu}m$, respectively. Nutritional condition was determined as a major parameter of metamorphosing between ciliate and cyst stages. The ciliate transforms to a cyst stage because of food shortage, and the cyst returns to a ciliate stage with a favorite environmental condition and shows active growth and reproduction. The ciliate multiplied at the maximal density of $2.9 {\times} 10^5 {m\ell}^{-1}$cells in vitro cultivation at $15 ^{\circ}C$temperature using MS BHI medium and bacterial food sources. The ciliate could be proliferated at a 2.5 to $30 ^ \circ}C$ temperature range, pH 6 to 9, and 1 to 55 ppt salinity. Particularly, it survived over one week at $0 ^{\circ}C$temperature showing a high resistance against unfavorable environmental conditions. And the cyst survived for 320 days in the condition of $5 ^{\circ}C$with no feeding, but its survival period was markedly shortened in higher temperature conditions. The chemotherapeutants (formalin and hydrogen peroxide) were clarified as effective chemicals against the ciliate during in vitro trials, but the effect of therapeutants differed in proportion, depending upon the density and the bathing time of chemical compounds.

In this study, the effects of alfalfa hay included in different ratios of 60%(A), 50%(B), 40%(C) and 30%(D) for the diets of young weaned Korean native goats were investigated on feed intake, digestibilities of nutrients, nitrogen retention, average daily gain and feed efficiency. Crude protein contents of experimental diets were 18% in all treatments. The contents of ADF, NDF and crude ash were higher in A as 26.1, 37.0 and 9.0% compared to the lowest values in D of 20.3, 30.4 and 6.6%. However the contents of non-structural carbohydrates was in the opposite tendency. Daily feed intake (DM) was highest in A (414.4g) and B (417.7g) and significantly (p<0.05) lower in D(362.6g). Dry matter intake per metabolic basal weight (DM g/kg of $BW^{0.75}$ ) were highest in A(73.9g) and lowest in D(64.0g). Average daily gain and feed efficiencies were in the range of 33.3~48.7g and 8.1~13.4% respectively with no statistical differences, however with the increasing trends according to higher percent of roughage in the diets. Digestibilities of dry matter (DM), organic matter (OM), crude protein (CP), ADF, NBF, crude ash (CA) and non-structural carbohydrates (NSC) were in the tendency of becoming higher according to higher ratios of concentrate but becoming lower according to higher ratio of alfalfa hay in the diets fed to Korean native goats. Nitrogen retention was lowest in A (1.8g, 15.5%) and highest in D (2.7g, 25.7%). In overall, inclusion of 30~40% alfalfa hay as a roughage in the diets of weaned Korean native goats was superior to the higher rates of inclusion of alfalfa hay in aspects of average daily gain and feed efficiency. The fixed 18% of crude protein content in the diets was efficient for the growth of weaned goats.
This study was conducted to get basic information of dry matter production and agronomic characteristics for increasing grain ripening in rice cultivar with large grains. Three cultivars which have different grain size were used. Of the two large grain cultivars, SR7796-3-1-5-1-l(SR7796F$_{7}$) had long culm and SR11269-13-3-4-2(SR11269F$_{6}$) had short culm. On the other hand, Tamjinbyeo was recommended cultivar had medium size of grain and culm. LAI was highest in Tamjinbyeo, followed by the SR11269F$_{6}$ and SR7796F$_{7}$, LAI decrement was higher in large grain cultivars than that of Tamjinbyeo after 20days after heading. Total dry weight(TDW) of areal part was highest in SR11269F$_{6}$, followed by the Tamjinbyeo and SR7796F$_{7}$. Dry weight (DW) of stem (leaf sheath+culm) at harvesting stage was decreased 15%, 12% in Tamjinbyeo, SR11269F$_{6}$, respectively, but was increased 5% in SR7796F$_{7}$ compared to heading stage. In distribution rate of each organ to TDW, distribution rate of leaf blade was low in large grain cultivar, but that of stem in Tamjinbyeo, SR7796F$_{7}$ and SR11269F$_{6}$ was 36%, 38% and 42%, respectively. In canopy architecture leaf blade weight was the highest 30~40cm, 40~50cm and 70~8Ocm above the ground in SR11269F$_{6}$, Tamjinbyeo and SR7796F$_{7}$, respectively. Range of panicle distribution in the canopy was 40cm in Tamjinbyeo, and was 70cm in large-grain cultivars. The results suggest that it has to take a growing interest in developing culture method for increasing LAI during growth stage, especially, around heading, in keeping green leaf till ripening stage and also in growing unformly plant within rice hill in large grain cultivars.cultivars.ltivars.

Chemical properties and heavy metal concentrations of forest soils of four abandoned coal mine lands affected by coal mining activities in the Mungyeong area were investigated to provide basic information for revegetation of abandoned coal mine lands. Soil pH in abandoned coal mine lands ranged from 5.30 to 6.76 it in the control site was 5.23. Contents of organic matter and total N in abandoned coal mine lands were $4.46\~7.19\%\;and\;0.07\~0.15\%$, respectively. Available P contents were 6.54 for A (Samchang), 6.52 for B (Bongmyeong),3.94 fur C (Kabjung), 5.45 mg/kg for D (Danbong coal mine land) and 5.25 mg/kg for the control site, which had a positive correlation with soil pH. Contents of exchangeable Ca, Mg, K and Na in abandoned coal mile lands averaged 196.1, 88.7, 88.2 and $10.2cmol^+/kg$, with a range of $132.1\~242.1,\;24.2\~138.\; 64.9\~120.8\;and\;8\~12.2cmol^+/kg$, respectively. Those of the control site were 192.8, 95.8, 104 and $21.2 cmol^+/kg$, respectively. Heavy metals such as Al, As, Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn of forest soil in abandoned coal mine lands have a larger content than those of the control site. Al, Mn and fb content was especially high in abandoned coal mine lands. The Al content of forest soil in abandoned coal mine lands ranged from 397 to 917 ppm, which was considered to be high enough to inhibit tree growth. Therefore, it is suggested that soils of abandoned coal mine lands contaminated by mining activities need to be properly treated for remediation of environmental problems.

As an effort to find a potential biopreservative, we isolated bacterial strains producing bacteriocin from fermented foods. A strain was finally selected and characteristics of the bacteriocin were investigated. The selected strain was identified as Bacillus subtilis E9-1 based on the 16S rRNA gene analysis. The culture supernatant of B. subtilis E9-1 showed antimicrobial activity against Gram-positive bacteria. Subtilisin A, ${\alpha}$-chymotrypsin, trypsin and proteinase K inactivated the antimicrobial activity, which means its proteinaceous nature, a bacteriocin. The bacteriocin activity was fully retained at the pH range from 2.0 to 8.0 and stable at up to $100^{\circ}C$ for 60 min. Solvents such as ethanol, isopropanol and methanol had no effect on the antimicrobial activity at the concentration of 100% but acetone and acetonitrile reduced the activity at up to 100% concentration. Cell growth of four indicator strains was dramatically decreased in dose-dependent manner. Listeria monocytogenes was the most sensitive, but Enterococcus faecium was the most resistant. Bacillus cereus and Staphylococcus aureus showed the medium sensitivity. The bacteriocin showed its antimicrobial activity against B. cereus and L. monocytogenes via bactericidal action. The number of viable cells of L. monocytogenes started to reduce after addition of bacteriocin to the minced beef. The bacteriocin was purified through acetone concentration, gel filtration chromatography and RP-HPLC. The whole purification step led to a 6.82 fold increase in the specific activity and 6% yield of bacteriocin activity. The molecular weight of the purified bacteriocin was determined to be 3.3 kDa by MALDI-TOF/TOF mass spectrometry.

A unicellular green alga was axenically isolated from a tidal pool on Ulleung-do, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Auxenochlorella protothecoides. The current study is the first record of this species in Korea. The microalgal strain was named as A. protothecoides MM0011 and its growth, lipid and pigment compositions, and biomass properties were investigated. The strain is able to thrive in a wide range of temperatures ($5{\sim}35^{\circ}C$) and to withstand up to 1.5 M NaCl. The results of GC/MS analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids (PUFAs). Its major fatty acids were linoleic acid (27.6%) and ${\alpha}-linolenic$ acid (39.6%). Thus, this indigenous microalga has potential as an alternative source of ${\omega}3$ and ${\omega}6$ PUFAs, which currently come from fish and plant oils. Also, the HPLC analysis revealed that the value-added antioxidant, lutein, was biosynthesized as the accessory pigments by the microalga. A proximate analysis showed that the volatile matter content was 85.6% and an ultimate analysis indicated that the gross calorific value was $20.3MJ\;kg^{-1}$. Since 40.5% of total nitrogen and 27.9% of total phosphorus were removed from the medium, respectively, it also has potential as a feedstock for biofuel applications which could be coupled to wastewater treatment. In addition, the biomass may also serve as an excellent animal feed because of its high protein content (51.4%). Therefore, A. protothecoides MM0011 shows promise for application in production of microalgae-based biochemicals and as a biomass feedstock.

To assess the effect of environmental factors on the sustainability of cultured production shellfish, we investigated the habitat characteristics of tidal flat (Namhae-po in Taean). We measured the physiochemical parameters (temperature, salanity, pH, dissolved oxygen and nutrients) and the geochemical characteristics (chemical oxygen demand, ignition loss, C/N ratio and C/S ratio). Surface sediments were collected from several site of tidal flat to examine the geochemical characteristics of both the benthic environment and heavy metal pollution. The grain size for research area of tidal flat were similar at the ratio of silt and clay in comparison with the other site of it. The C/N ratio was more than 5.0, reflecting the range arising from the mix of marine organism and organic matter. The C/S ratio (about 2.8) showed that survey area had anoxic or sub-anoxic bottom conditions. The enrichment factor (Ef) and index of accumulation rate (Igeo) of the metals showed that those research areas can be classified as heavily polluted, heavily to moderately polluted, or more or less unpolluted, respectively. Adult surf clam (Mactra veneriformis) density was highest at St. 2 (middle part of the Namhae-po), on the other hand, surf clam spat density was highest at St. 3 (lower part of the Namhae-po). Heavy rain, terrigenous suspended clay with fresh water from neighboring agricultural land, and severe high air temperature during summer could be thought as detrimental causes of spat and adult mortality in Namhae-po tidal flat. We suggested that the growth of shellfish in the tidal flat was effected by the various environmental conditions, so an improvement in the cultured method was needed.

Harmful algal blooms (HABs), commonly known as red tides, are aquatic phenomena caused by the rapid growth and accumulation of certain microalgae, which can lead to marked discoloration of surface waters, and severe impacts on public health, commerce, and the environment. In South Korea, the red tides have been a serious and recurrent problem, especially along the south coast. Plenty of yellow loess was used to control an outbreak of the red tides for 15 years from 1996 until now. Yellow loess was almost sprayed in the vicinity of a large fish farming industry. In this research, the distribution characteristics and density distribution of zooplankton were investigated in autumn (Oct. 2008) and spring (Apr. 2009) using volume backscattering strength (SV) calculated by the zooplankton collected with north pacific standard (NORPAC) net and the echo intensity measured with ADCP at stations on the study area in the spraying ocean of yellow loess (SOYL), and the non-spraying ocean of yellow ocean (NOYL) by the red tide generating every year. The species number and the individuals per unit volume of the zooplankton collected in NOYL was high and it which was collected in SOYL was low. As a result of comparing the volume backscattering strength ($SV_c$) calculated by species and length of the zooplankton collected with NORPAC net with the volume backscattering strength ($SV_m$) calculated by the echo intensity measured with ADCP at stations on the study area, although $SV_c$ and $SV_m$ of NOYL were generally in agreement, $SV_m$ of SOYL was higher than $SV_c$ 4.3dB, i.e. ADCP is greatly influenced by suspended solid in SOYL. The horizontal distribution map of $SV_m$ at the study area in autumn (Oct. 2008) and spring (Apr. 2009) was drawn. $SV_m$ of SOYL is higher than NOYL and autumn is higher than spring. $SV_m$ can suppress the overestimate or underestimate of $SV_c$.
The purpose of this study is to provide the basic data such as distribution status, growth characteristics, and the structural characteristics of forest stands for the systematic conservation and management of relict forest trees (stem girth of 300cm or larger) established naturally in Gangwon Province, Korea. The survey showed that 434 individuals of 19 species (conifers: 228 individuals of 4 species, broad-leaved trees: 206 individuals of 15 species) were distributed in Gangwon Province, and Taxus cuspidata was the most abundant among them with 203 individuals or about 46.7 % of the total. The stem girth was average of 404cm (conifers: 373cm, broad-leaves: 421cm), and Tilia amurensis with multi-stemmed growing on Sorak mountain range had the largest stem girth at 1,113cm. The average height and the crown width of relict forest trees were 15.4m and 10.0m, respectively. Although the environments of relict forest trees showed a slight difference by species, the relative appearance frequencies of most trees were high in the environments where the altitude was higher than 1,000 m, slope degree was greater than $25^{\circ}$, the slope faced north, and microtopography was at the upper of slopes. Regarding the stand characteristics of relict forest trees per unit area ($/100m^2$), the average total coverage was 294% (max. 475%), the total average number of species was 36 species (max. 60 species), the average species diversity index (H') was 2.560 (max. 3.593), the average canopy closure was 84.8% (max. 94.6%), and the average basal area (/ha) was $52.7m^2$ (max. $116.4m^2$, relict trees $30.0m^2$, and other trees $22.7m^2$). The analysis of the dynamics of the forest stands where relict forest trees were growing showed four types of the maintenance mechanisms of relict forest trees depending on the supply pattern of succeeding trees: "Low-density but persistent type (Quercus mongolica, Abies holophylla, Tilia amurensis, and Pyrus ussuriensis)," "Long ago stopped type (Pinus densiflora)," "Recently stopped type (Abies nephrolepis, Quercus variabilis, and Betula schmidtii)," and "Periodically repeated types of supply and stop (Salix caprea and Quercus serrata).".
