  • Fermentation strategies for recombinant protein production in Pichia pastoris have been investigated and are reviewed here. Characteristics of the expression system, such as phenotypes and carbon utilization, are summarized. Recently reported results such as growth model establishment, app58lication of a methanol sensor, optimization of substrate feeding strategy, DOstat controller design, mixed feed technology, and perfusion and continuous culture are discussed in detail.

잠재성장모형과 성장혼합모형을 이용한 도서관 대출권수의 종단적 분석 (A Longitudinal Analysis of the Number of Checked-out Books Using Latent Growth Model and Growth Mixture Modeling)

  • 본 연구의 목적은 도서관 이용에 영향을 미치는 요인을 종단적으로 분석하는 것이다. 도서관 이용의 지표 중의 하나인 대출권수의 통계적 분석을 위해 잠재성장모형과 성장혼합모형을 적용하였다. 국가도서관통계시스템에서 제공하는 2014년부터 2019년까지의 공공도서관 통계정보를 수집하고 846개 도서관을 대상으로 분석을 수행했다. 분석결과, 대출권수는 감소하는 경향이 나타났지만 감소세는 점차 완화되고 있었다. 다음으로 대출권수에 영향을 미치는 요인들을 통제한 상태에서 분석한 결과, 장서수와 독서프로그램 참여자수가 중요한 요인으로 나타났다. 마지막으로 성장혼합모형을 적용하여 분석된 5개의 계층들에서 사서수의 증가는 감소 계층에 비해 증가 계층에 속할 가능성을 높이는 것으로 나타났다.

  • 컴퓨터 그래픽스 기술의 발달로 영화, 게임 등의 엔터테인먼트 산업에서 장면의 사실성이 증대됨에 따라, 다양한 소재의 콘텐츠 개발을 위한 디지털 콘텐츠 기술의 요구가 증가되고 있다. 지상에서의 생태 및 환경 변화에 대한 연구 및 시뮬레이션 기술 개발에 비해, 해양 생태 및 환경 변화 등을 콘텐츠화하기 위한 기술 개발은 아직 미비한 상태이다. 본 논문에서는 가상 해저 환경의 변화에 따른 해양 객체의 성장과정을 효과적으로 시뮬레이션할 수 있는 모델링 방법과 객체의 움직임에 따른 효율적인 변형 방법을 제안하고자 한다. 제안 방법은 비등방성 메타큐브를 이용하여 해양 생물들을 모델링함으로써 가상 해저 환경에서 물고기의 성장과 모양 변화가 가능하게 해준다.

Effects of Dissolved Oxygen and Agitation on Production of Serratiopeptidase by Serratia Marcescens NRRL B-23112 in Stirred Tank Bioreactor and its Kinetic Modeling

  • The effects of the agitation and aeration rates on the production of serratiopeptidase (SRP) in a 5-L fermentor (working volume 2-l) were systematically investigated using Serratia marcescens NRRL B-23112. The dissolved oxygen concentration, pH, biomass, SRP yield, and maltose utilization were all continuously measured during the course of the fermentation runs. The efficiencies of the aeration and agitation were evaluated based on the volumetric mass transfer coefficient ($K_La$). The maximum SRP production of 11,580 EU/ml with a specific SRP productivity of 78.8 EU/g/h was obtained with an agitation of 400 rpm and aeration of 0.075 vvm, which was 58% higher than the shake-flask level. The $K_La$ for the fermentation system supporting the maximum production (400 rpm, 0.075 vvm) was 11.3 $h^{-1}$. Under these fermentor optimized conditions, kinetic modeling was performed to understand the detailed course of the fermentation process. The resulting logistic and Luedeking-Piret models provided an effective description of the SRP fermentation, where the correlation coefficients for cell growth, SRP formation, and substrate consumption were 0.99, 0.94, and 0.84, respectively, revealing a good agreement between the model-predicted and experimental results. The kinetic analysis of the batch fermentation process for the production of SRP demonstrated the SRP production to be mixed growth associated.

성인의 만성질환 유형에 따른 우울변화에 영향을 미치는 요인에 관한 연구 (Factors Affecting Depression Changes in Adults According to Chronic Disease Type)

  • Objectives : The purpose of this study was to investigate the depressive change and predictors of chronic illnesses and to examine whether depressive change predictors are different according to chronic disease types. Methods : Data from the 1st to the 10th year of the Korean Welfare Panel of 5,023 adults over 20 years of age who were treated for chronic diseases for more than three months were examined. We assessed demographic, sociological, and socioeconomic factors including depression predictors of chronically ill patients, and analyzed the data using latent growth modeling and multiple group analysis. Results : Depression symptoms of chronically ill patients gradually decreased, showing a linear relationship across time. In addition, multiple group analysis was performed for each type of chronic disease. Conclusions : We described the phenomenon of depression in chronically ill patients, and discussed the practical and policy interventions required to improve depression in chronically ill patients.

Modeling Age-specific Cancer Incidences Using Logistic Growth Equations: Implications for Data Collection

  • Large scale secular registry or surveillance systems have been accumulating vast data that allow mathematical modeling of cancer incidence and mortality rates. Most contemporary models in this regard use time series and APC (age-period-cohort) methods and focus primarily on predicting or analyzing cancer epidemiology with little attention being paid to implications for designing cancer registry, surveillance or evaluation initiatives. This research models age-specific cancer incidence rates using logistic growth equations and explores their performance under different scenarios of data completeness in the hope of deriving clues for reshaping relevant data collection. The study used China Cancer Registry Report 2012 as the data source. It employed 3-parameter logistic growth equations and modeled the age-specific incidence rates of all and the top 10 cancers presented in the registry report. The study performed 3 types of modeling, namely full age-span by fitting, multiple 5-year-segment fitting and single-segment fitting. Measurement of model performance adopted adjusted goodness of fit that combines sum of squred residuals and relative errors. Both model simulation and performance evalation utilized self-developed algorithms programed using C# languade and MS Visual Studio 2008. For models built upon full age-span data, predicted age-specific cancer incidence rates fitted very well with observed values for most (except cervical and breast) cancers with estimated goodness of fit (Rs) being over 0.96. When a given cancer is concerned, the R valuae of the logistic growth model derived using observed data from urban residents was greater than or at least equal to that of the same model built on data from rural people. For models based on multiple-5-year-segment data, the Rs remained fairly high (over 0.89) until 3-fourths of the data segments were excluded. For models using a fixed length single-segment of observed data, the older the age covered by the corresponding data segment, the higher the resulting Rs. Logistic growth models describe age-specific incidence rates perfectly for most cancers and may be used to inform data collection for purposes of monitoring and analyzing cancer epidemic. Helped by appropriate logistic growth equations, the work vomume of contemporary data collection, e.g., cancer registry and surveilance systems, may be reduced substantially.

Cybernetic Modeling of Simultaneous Saccharification and Fermentation for Ethanol Production from Steam-Exploded Wood with Brettanomyces custersii

  • The simultaneous saccharification and fermentation (SSF) process consists of concurrent enzymatic saccharification and fermentation. In the present cybernetic model, the saccharification process, which is based on the modified Michaelis-Menten kinetics and enzyme inhibition kinetics, was combined with the fermentation process, which is based on the Monod equation. The cybernetic modeling approach postulates that cells adapt to utilize the limited resources available to them in an optimal way. The cybernetic modeling was suitable for describing sequential growth on multiple substrates by Brettanomyces custersii, which is a glucose- and cellobiose-fermenting yeast. The proposed model was able to elucidate the SSF process in a systematic manner, and the performance was verified by previously published data.

Toward precise and accurate modeling of matter clustering in redshift space

  • This dissertation presents the results on two-dimensional Redshift space distortion (hereafter RSD) analyses of the large-scale structure of the universe using spectroscopic data and on improvement of modeling of the RSD effect. RSD is an effect caused by galaxies' peculiar velocity on their clustering feature in observation along the line of sight and is thus intimately connected to the growth rate of the structure in the universe, from which we can test the origin of cosmic acceleration and Einstein's theory of gravity at cosmic scales in the end. However, there are several challenges in modeling precise and accurate RSD effect, such as non-linearities and the existence of an exotic component, e.g. massive neutrino. As part of endeavors for modeling more precise and accurate galaxy clustering in redshift space, this dissertation includes a series of works for this issue. Clustering Asian and North African Countries According to Trend of Colon and Rectum Cancer Mortality Rates: an Application of Growth Mixture Models

  • Background: Colorectal cancer is the second most common cause of cancer death with half a million deaths per year. Incidence and mortality rates have demonstrated notable changes in Asian and African countries during the last few decades. In this study, we first aimed to determine the trend of colorectal cancer mortality rate in each Institute for Health Metrics and Evaluation (IHME) region, and then re-classify them to find more homogenous classes. Materials and Methods: Our study population consisted of 52 countries of Asia and North Africa in six IHME pre-defined regions for both genders and age-standardized groups from 1990 to 2010.We first applied simple growth models for pre-defined IHME regions to estimate the intercepts and slopes of mortality rate trends. Then, we clustered the 52 described countries using the latent growth mixture modeling approach for classifying them based on their colorectal mortality rates over time. Results: Statistical analysis revealed that males and people in high income Asia pacific and East Asia countries were at greater risk of death from colon and rectum cancer. In addition, South Asia region had the lowest rates of mortality due to this cancer. Simple growth modeling showed that majority of IHME regions had decreasing trend in mortality rate of colorectal cancer. However, re-classification these countries based on their mortality trend using the latent growth mixture model resulted in more homogeneous classes according to colorectal mortality trend. Conclusions: In general, our statistical analyses showed that most Asian and North African countries had upward trend in their colorectal cancer mortality. We therefore urge the health policy makers in these countries to evaluate the causes of growing mortality and study the interventional programs of successful countries in managing the consequences of this cancer.

한국 아동패널 2008-2012 자료를 활용한 자녀 출산 후 5년간의 부부의 결혼만족도 변화궤적 (Trajectories of Marital Satisfaction of Married Couples From Childbirth to 5 Years Postpartum: The Panel Study of Korea Children 2008-2012)

  • Objective: This study aimed to explore marital satisfaction changes of married couples from childbirth to 5 years postpartum using the data from the Panel Study of Korean Children 2008-2012. Methods: A total of 2,072 coupes were followed longitudinally during the first 5 years after childbirth. Their repeatedly measured marital satisfaction were analyzed with growth modeling analyses. Results: Husbands and wives had different marital satisfaction trajectories regarding the rate of change and initial marital satisfaction scores. Further, the facets of postpartum marital satisfactions change varied depending on the couples' first, second, and third (or more) birth(s). Conclusion: In general, Korean married couples' marital satisfaction has a decreasing tendency after childbirth. Fathers' marital satisfaction is generally higher but more rapidly decreases compared to mothers' marital satisfaction. Also, couples after first child birth experience a faster decline of marital satisfaction compared to couples after second or third (or more) birth(s).
