  • Pseudomanas sp. EL-04J was previously isolated from phenol-acclimated activated sludge. This bacterium was capable of degrading phenol and cometabolizing trichloroethylene (TCE). After precultivation in the mineral salts medium containing phenol as a sole carbon source, Pseudomonas EL-04J degraded 90% of TCE $25 \mu\textrm{M}$ within 20 hours. Thus, phenol-induced Pseudomonas sp. EL-04J cells can bdegrade TCE. Followsing a transient lag period, Pseudomonas sp. EL-04J cells degraded TCE at concentrations of at least $250 \mu\textrm{M}$ with no apparent retardation in rate, but the transformance capacity of such cells was limited and depended on the cell concentration. The degradation rate of TCE followed the Michaelis-Menten kinetic model. The maximum degradation ratio ($V_{max}$) and saturation constant ($K_{m}$) were $7nmo {\ell}/min{\cdot}mg$ cell protein and $11 \mu\textrm{M}$, respectively. Cometabolism of TCE by phenol fed experiment was evaluated in $50m {\ell}$ serum vial that contained $10m {\ell}$ of meneral sals medium supplemented with $10 \mu\textrm{M}$ TCE degradation was inhibited in the initial period of 1 mM phenol addition, but after that time Pseudomonas sp. EL-04J cells degraded TCE and showed cell growth.

  • This study provides first-hand proteomic data on the survival strategy of Anabaena sp. PCC 7120 when subjected to long-term iron-starvation conditions. 2D-gel electrophoresis followed by MALDI-TOF/MS analysis of iron-deficient Anabaena revealed significant and reproducible alterations in ten proteins, of which six are associated with photosynthesis and respiration, three with the antioxidative defense system, and the last, hypothetical protein all1861, conceivably connected with iron homeostasis. Iron-starved Anabaena registered a reduction in growth, photosynthetic pigments, PSI, PSII, whole-chain electron transport, carbon and nitrogen fixation, and ATP and NADPH content. The kinetics of hypothetical protein all1861 expression, with no change in expression until day 3, maximum expression on the $7^{th}$ day, and a decline in expression from the $15^{th}$ day onward, coupled with in silico analysis, suggested its role in iron sequestration and homeostasis. Interestingly, the up-regulated FBP-aldolase, Mn/Fe-SOD, and all1861 all appear to assist the survival of Anabeana subjected to iron-starvation conditions. Furthermore, the $N_2$-fixation capabilities of the iron-starved Anabaena encourage us to recommend its application as a biofertilizer, particularly in iron-limited paddy soils.

  • Effect of Cr contents(Cr: 0.27, 0.45 and 0.65wt.%) on precipitation process has been studied by electrical resistivity measurements, hardness and scanning electron microscope. The first stage of the process consists of the formation of Cr-rich particles, the second stage consists of the competitive growth of these particles. The kinetics of precipitation could be described by Johnson-Mehl-Avrami equation, $f(t)=1-\exp(-kt^n)$. The values of n were found to be in the range from 0.17 to 0.39 at the first stage and from 1.0 to 1.5 at the second stage. The activation energies of Cu-Cr alloys were determined by Cross-Cut method and were 90~136 kJ/mol. The maximum hardness value of $H_RB$ 84 was obtained in Cu-0.65wt.%Cr alloy.

  • The present study is devoted to the validation of a new method for in line electrokinetic characterisation of deposits on membrane surfaces. This method is based upon simultaneous measurements of transversal streaming potential and permeates flux at constant pressure before and during the deposit formation. Dead-end filtration experiments were conducted with negative flat membranes forming a narrow slit channel, negative hollow fiber membranes and mono-dispersed suspensions of (negatively charged) polystyrene latex and (positively charged) melamine particles at various concentrations. It was observed that the overall streaming potential coefficient increased in absolute value with the deposited latex quantity, whereas it decreased and changed of sign during the filtration of melamine suspensions. By considering a resistance-in-series model, the streaming potential coefficient of the single deposit ($SP_d$) was deduced from the electrokinetic and hydraulic measurements. The independence of $SP_d$ with respect to growth kinetics validates the measurement method and the reliability of the proposed procedure for calculating $SP_d$. It was found that $SP_d$ levelled off much more quickly when filtration was performed through the slit channel. This different behaviour could result from a non-uniform distribution of the deposit thickness along the membrane given that the position of measuring electrodes is different between the two cells.

  • The expression kinetics of human lipocortin I (LCI), a potential anti-inflammatory agent, was studied in the shake-flask and fermenter cultures of Saccharomyces cerevisiae carrying a galactose-inducible expression system. The cell growth, expression level of LCI, and the plasmid stability were investigted under various galactose induction conditions. The expression of LCI was repressed by the presence of a very small amount of dextrose in the culture medium, but it was induced by galactose after dextrose became completely depleted. The optimal ratio of dextrose to galactose for lipocortin I production was found to be 1.0 (10 g/l dextrose and 10 g/l galactose). With optimal D/G ratio of 1.0 and the addition of galactose prior to dextrose depletion, LCI of about 100~130 mg/l was produced. LCI at a concentration of 174 mg/l was porduced in the fed-batch culture, which was nearly a twice as much of that produced in the batch culture. The plasmid stability was very high in all culture cases, and thus was considered to be not an important parameter in the expression of LCI.

  • It has been reported that some plants have the potential to metabolize the 2,4,6-trintroluene (TNT) in contaminated soils, sediments and natural water. In this study, the effects of TNT on germination and early seedling development of Abutilion avicennae was characterized in a germination test. Concentration up to 80 mg/L TNT did not affect germination but root and shoot growth, and fresh biomass decreased as TNT concentration increased. A series of axenic hydroponical batch culture of Abutilion avicennae at various initial TNT concentration was used to determine its transformation kinetics, to identify products formed, and to evaluate phytotoxic effects on the TNT transformation process. At higher initial TNT concentrations, TNT removal rate constant decreased, however, total amount of TNT removed was increased in the culture media. Reductive transformation products of TNT were not detected in the plant culture media but higher concentrations of reduced metabolites were detected in the root and stem extracts of plant material at the 7 days of batch incubation. From these results we concluded that Abutilion avicennae has an intrinsic capacity for taking up and transforming TNT.

  • The ankylosis of temporomandibular joint occured by several causes directly developes TM joint disfunction, In cases with prolonged condylar ankylosis, especially at growing age the condylar ankylosis evokes malfunction of growth center area, and then developes the abnormal facial morphology and malocclusion. Therefore it must be cured. Almost authors have agreed to the necessity of surgical correction of the TMJ anylosis. but they did not decide the one surgical method to get the best result. The Tx. method suggested by many authors are the using interposition after resection of condyle to remain a lever of 3rd class in Mn. kinetics, the autogenous condylar graft and the alloplastic condylar graft. Some authors have got the satisfied results only with the condylectomy of the involved TMJ. This study also operated only the detachment of fibrous adhesion on ankylosed condylar side and then established occlusion in the case with the unilateral TMJ ankylosis and fibrous joint adhesion and facial asymmetry evoked after the fracture of condylar head at early age. This study got a improved mouth opening and a stable postsurgical result after 1 year. Also, this study reviewed many author's study about the chanracteristics, etiology, diagnosis and Tx. method for the ankylosis of TMJ.

  • A model to explain the observed kinetics in a whole process of Cheddar-cheese ripening has been developed. It includes growth and lysis of cells in the cheese matrix, cell-wall bound protelnases and intracellular dipeptidases that are released into cheese upon cell lysis, and the production of dipeptides and amino acids from casein in cheese. Model simulations have been conducted to figure out the crucial factors in the process of the cheese ripening. The influential factors have been found to be the cell numbers and the dipeptidase activity at the beginning of the cheese ripening, and the cell-lysis rate of cheese starters. The simulation results have also suggested the use of a mixed culture as well as the experimental screening for a more suitable organism as a cheese starter hence, the model shows how to accelerate the cheese ripening.

  • Singlet oxygen ($^1O_2$) is highly reactive form of molecular oxygen that may harm living systems by oxidizing critical cellular macromolecules. The oxyR gene product regulates the expression of the enzymes and proteins that are needed for cellular protection against oxidative stress. In this study, the role of oxyR in cellular defense against a singlet oxygen was investigated using Escherichia coli oxyR mutant strains. Upon exposure to methylene blue and visible light, which generates singlet oxygen, the oxyR overexpression mutant was much more resistant to singlet oxygen-mediated cellular damage when compared to the oxyR deletion mutant in regard to growth kinetics, viability and protein oxidation. Induction and inactivation of major antioxidant enzymes, such as superoxide desmutase and catalase, were observed after their exposure to a singlet oxygen generating system in both oxyR strains. However, the oxyR overexpression mutant maintained significantly higher activities of anticxidant enzymes than did the oxyR deletion mutant. These results suggest that the oxyR regulon plays an important protective role in singlet oxygen-mediated cellular damage, presumably through the protection of antioxidant enzymes.

  • A seaweed biofilter/production system of being developed to reduce the environmental impact of marine fish farm effluent in coastal ecosystems as a part of an integrated aquaculture system. Several known seaweed taxa and their cultivars have been considered as candidate biofilter organisms based on their species-specific physiological properties such as nutrient uptake kinetics and their economic value. Porphyra is an excellent cadidate and shows efficient nutrient extraction properties. Rates of ammonium uptake were maintained at around 3 ${\mu}moles{\cdot}g{\cdot}dw^{-1}{\cdot}min^{-1}$ at 150 ${\mu}M$ inorganic nitrogen at $10^{\circ}C$. Ulva is another possible biofilter candidate with an uptake rate of 1.9 ${\mu}moles{\cdot}g{\cdot}dw^{-1}{\cdot}min^{-1}$ under same conditions. A simple uptake/growth and harvest model was applied to estimate the efficiency of the biofilter/production system. The model was deterministic and used a compartment model structure based on difference equations. The efficiency of Porpyra filter was estimated over 17% of ${NH_4}^+$ removal from the contimuous supply of 100 ${\mu}mole{\cdot}l^{-1}\;{NH_4}^+\;at\;100l{\cdot}sec^{-1}$ flow rate.
