Methoxychlor Produces Many Adverse Effects on Male Reproductive System, Kidney and Liver by Binding to Oestrogen Receptors

  • Methoxychlor (MXC) was developed to be a replacement for the banned pesticide DDT. HPTE [2,2-bis (p-hydroxyphenyl)-1,1,1-trichloroethane], which is an in vivo metabolite of MXC, has strong oestrogenic and anti-androgenic effects. MXC and HPTE are thought to produce potentially adverse effects by acting through oestrogen and androgen receptors. Of the two, HPTE binds to sex-steroid receptors with greater affinity, and it inhibits testosterone biosynthesis in Leydig cells by inhibiting cholesterol side-chain cleavage enzyme activity and cholesterol utilisation. In a previous study, MXC was shown to induce Leydig cell apoptosis by decreasing testosterone concentrations. I focused on the effects of MXC on male mice that resulted from interactions with sex-steroid hormone receptors. Sex-steroid hormones affect other organs including the kidney and liver. Accordingly, I hypothesised that MXC can act through sex-steroid receptors to produce adverse effects on the testis, kidney and liver, and I designed our experiments to confirm the different effects of MXC exposure on the male reproductive system, kidney and liver. In these experiments, I used pre-pubescent ICR mice; the puberty period in ICR mice is from postnatal day (PND) 45 to PND60. I treated the experimental group with 0, 100, 200, 400 mg MXC/kg b.w. delivered by an intra-peritoneal injection with sesame oil used as vehicle for 4 weeks. At the end of the experiment, the mice were sacrificed under anaesthesia. The testes and accessory reproductive organs were collected, weighed and prepared for histological investigation. I performed a chemiluminescence immune assay to observe the serum levels of testosterone, LH and FSH. Blood biochemical determination was also performed to check for other effects. There were no significant differences in our histological observations or relative organ weights. Serum testosterone levels were decreased in a dose-dependent manner; a greater dose resulted in the production of less testosterone. Compared to the control group, testosterone concentrations differed in the 200 and 400 mg/kg dosage groups. In conclusion, I observed markedly negative effects of MXC exposure on testosterone concentrations in pre-pubescent male mice. From our biochemical determinations, I observed some changes that indicate renal and hepatic failure. Together, these data suggest that MXC produces adverse effects on the reproductive system, kidney and liver.

Effect of Ethanol Extract of Yukmijiwhang-Whan on Trabecular Bone Area in OVX Rats (육미지황환(六味地黃丸) 에탄올 추출물이 난소제거 흰쥐의 경골 소주골에 미치는 영향)

  • Bone is continuously remodeled during adult life with the resorption of old bone by osteoclasts and its subsequent replacement by osteoblast. Bone homeostasis is maintained by a balance between activities of osteoblasts and osteoclasts, but an imbalance between resorption and formation results in bone diseases including osteoporosis. Osteoblasts line up on the bone surface, especially regions of new bone formation, lay down bone matrix (osteoid) in orderly lamellae and induce its mineralization. Thus, the increased activity of osteoblasts is helpful to treat and prevent osteoporosis. In this study, we examined whether 80% EtOH extract of yukmijiwhang-whan is capable of affecting osteoblast proliferation using human osteoblast-like cell line, MG-63 and Saos-2. In an in vivo experiment, extract of yukmijiwhang-whan was administered for 9 weeks to ovariectomized (OVX) rats. At necropsy, uterus weights were measured, and trabecular bone areas (TBAS) of tibia and the sixth lumbar vertebra were measured by bone histomorphology. The maximum cell proliferation of MG-63 caused by extract of yukmijiwhang-whan at $5\;{\times}\;10^{-6}\;mg/ml$ was approximately 115% compared with control. In Saos-2, cell proliferation was approximately 145% of control at $5\;{\times}\;10^{-4}\;mg/ml$ and maximum alkaline phosphatase (ALP) activity was approximately 143% of control at $5\;{\times}\;10^{-5}\;mg/ml$. In animal study, however, the tibia and lumbar TBAS of the yukmijiwhang-whan group did not increased than the OVX control group. In conclusion, the 80% EtOH extract of yukmijiwhang-whan increased proliferation of osteoblasts but did not prevent bone loss in OVX rats.

  • Fibrous dysplasia is a developmental tumor-like condition that is characterized by replacement of normal bone by an excessive proliferation of cellular fibrous connective tissue intermixed with irregular bony trabeculae. Craniofacial lesions may cause facial pain, headache, cranial asymmetry, facial deformity, tooth displacement and visual or auditory impairment. In this case, a 2-year-9-month old boy who was diagnosed as the fibrous dysplasia showed delayed eruption on affected area. Teeth of left lateral dentition group have erupted completely but teeth of right lateral dentition group are erupting slowly. Eruption and maturation of affected teeth are in progress, so continuous observation is required presently. If the eruption state stops, surgical opening or forced eruption of the impacted teeth will be considered.

Analysis of Risk Factors Affecting Breast Cancer Incidence : Breast Ultrasonography (유방암 발생에 영향을 미치는 위험인자 분석: 유방초음파 검진자 대상으로)

  • In general, a number of studies have been conducted on factors affecting breast cancer development, but systematic investigations of risk factors are rare. The purpose of this study was to investigate the factors involved in breast cancer screening before breast ultrasound diagnosis and the risk factors associated with breast cancer screening by ultrasound. Self-administered questionnaire was performed on 417 patients who underwent breast ultrasonography and classified as benign and malignant. Breast cancer was associated with age, BMI, and type of medication(p<0.05). Multivariate analysis showed that the odds ratio was 4.93 times higher in the 50s compared to the less than 50s, 2.43 times higher in the obese group than in the normal group, 0.14 times and 0.16 times lower in hormonal replacement therapy(p<0.05). Therefore, as age increases, periodical examination of health and appropriate weight management are needed. So this study is expected to provide basic data for identification of risk factors affecting breast cancer development.

The Prevalence of Urinary Incontinence of The Women in Daegu (대구지역 성인 여성의 요실금 유병률에 관한 연구)

  • Background: The aim of this study was to estimate the prevalence of urinary incontinence and its correlation to the underlying diseases of women in Daegu. Materials and Methods: Urinary incontinence questionnaire regarding age, body weight, height, body mass index (BMI), parity, delivery mode, menopausal status, history of hormonal replacement therapy, abortion history, and any underlying diseases were administered from May to November, 2001 to 412 women over 20 who had been randomly selected from the Outpatient Department of Obstetrics and Gynecology of Yeungnam University. The clinical characteristics of women who experience, and those who do not experience urinary incontinence were compared by means of the Student's t-test for continuous variables and by the Pearson's Chi-square test or Fisher's exact test for categorical variables. A p value of <0.05 was considered statistically significant. Results: The mean age of the urinary incontinent group (N=193) was 44.2 years. There was a significant increase in the prevalence of incontinence with the increase of age (p=0.000). The prevalence of urinary incontinence was significantly related to the number of deliveries and the mode of delivery (p=0.007, p=0.001) No significant relationship was demonstrated between urinary incontinence and BMI (body mass index); the number of abortions; hormonal status; or any underlying diseases such as thyroid disease, diabetes mellitus, and chronic respiratory disease. Also, most of the urinary incontinent women didn't recognize their incontinence as pathological and consequently, didn't consult a physician. Conclusion: Our study indicates that the prevalence of urinary incontinence is significantly correlated to age, parity, and the mode of delivery. Most of the middle-aged women who suffered from urinary incontinence didn't recognize their incontinence a pathological. Those results suggest that women in this age group need more information and more education about urinary incontinence.

Effect of Fermented Food Wastes on the Growth Performance and Feeding Efficiency in Pig (남은 음식물의 고온숙성 발효사료가 돼지의 성장 및 사료효율에 미치는 영향)

  • The effect of the fermented food wastes (FFW) on growth performance and feeding efficiency war investigated in growing-finishing pigs. Four treatments with different mixing rates [0% FFW (100% commercial diet: control) ; 25% FFW ; 50% FFW ; and 100% FFW] were tried. Twenty heads of cross-bred barrows ($Duroc\;{\times}\;Landrace$, average live weight : 28.5 kg) were divided into 4 treatments with 5 replicates. During entire feeding trial (10 to 27 week), average daily gain increased by 29.4% and 13.9% in 25% FFW and 50% FFW, but decreased by 38.2% in 100% FFW compared to control. Daily feed intake was the highest in 25% FFW and it greatly decreased in 50% and 100% FFW feeding. The amount of feeds required for weight gain was similar with control in 25% FFW or significantly decreased in 50% FFW, while greatly increased in 100% FFW feeding. Cost of feeds per kg weight gain was decreased by 28.6%, 49.4% and 32.6% in 25%, 50% and 100% FFW, respectively. The results showed that replacement up to 50% with fermented food wastes was advantageous with respect to feed efficiency and feed cost, but 100% FFW feeding was disadvantageous because of a very low feed efficiency.

REGENERATION OF THE ALVEOLAR BONE AND TRANSPLANTED ROOTS INTO THE PERIODONTALLY INVOLVED EXTRACTION SOCKETS IN DOGS;I : EFFECT OF ROOT PLANING PROCEDURE (성견치주질환 이환 발치와내 이식 치근과 발치와 치조골 재생에 대한 연구;I. 치근활택술의 영향)

  • The authors have transplanted periodontally involved roots which had been root planed into healthy and periodontally involved extraction sockets, and studied the root resorption patterns as well as its effect on new bone formation and wound healing. Alveolar bone around mandibular premolars of 6 adult dogs has been surgically removed, followed by ligation of orthodontic elastic wires for 8 weeks inducing chronic periodontal disease. After removing the crown portions, roots were extracted, and notches were made on the root surfaces discriminating healthy and periodontally involved areas using burs. Controls and experimental groups were divided as follows. Control I : Transplantation of periodontally involved root into healthy extraction sockets. Control II : Transplantation of periodontally involved root into diseased extraction sockets. Experimental group I : Transplantation of root planed roots into healthy extraction sockets. Experimental group II : Transplantation of root planed roots into diseased extraction sockets. Extraction sockets were sutured after transplantations, completely submerging the roots. Healing progress was histologically observed at 2nd, 8th, 12th, and 20th weeks, and the results were as follows ; 1. No inflammation or infection within the extraction sockets had been observed in all groups throughout the experimental period. 2. Reversal lines were observed at week 2 in all groups, clearly discriminating socket walls and new bone, and numerous blood vessels were observed in the new bone trabeculae. 3. Experimental groups showed markedly less root resorption compared to the controls at week 2, but as time progressed, severe resorptions were present in all groups. 4. Localized areas of new bone ankylosis were observed, and the rest of the areas showed collagen fiber insertion with new bone formation at its periphery. 5. No clear differences were found in healing and alveolar bone regeneration between healthy and diseased extraction sockets. 6. The amount of root resorption and ankylosis had increased up to week 8 and 12, showing ankylosis of new bone and the roots. However, no further increase in ankylosis was observed at week 20. 7. Most of the cementum on healthy roots was directly ankylosed to new bone at week, 2, and were gradually resorbed and replaced by new bone thereafter. These results appear to indicate that root planing may inhibit early root resorption of transplanted roots, but gradual replacement by alveolar bone and collagen fibers eventually occur. Condition of the roots or presence of disease in extraction sockets do not appear to make marked differences in alveolar bone regeneration process.

Durability of Xenograft Cardiac Valves (이종조직 판막의 내구성)

  • The durability of the xenograft cardiac substitute valves is of a great concern on the clinical grounds. Four groups of tc tal and consecutive patients to the end of study operated on between 1976 and 1984 were Group ISM, 291 patients of MVR, ISA, 65 patients of AVR, and ISMA, 107 patients of MVR+AVR with the standard Ionescu-Shiley bovine pericardial valve, and H, 163 patients of valve replacement with the Hancock porcine aortic vlave. Operative mortality was 5.2%[ISM], 10.8%[ISA], 7.5%[ISMA] and 6.1%[H]. Early survivors were followed up for a total of 1148.3 patient-years[pt-yrs] [ISM], 271.2 pt-yrs [ISA], 488.1 pt-yrs[ISMA] and 822.9 pt-yrs[H]. Linearized late mortality was 2.1% /pt-yr [ISM], 1.l%/pt-yr[ISA], 1.8%/pt-yr[ISMA] and 1.8% /pt-yr[H]. Thromboembolic complication was experienced at the linearized rate of 1.045% /pt-yr [ISM], 1.475%/pt-yr[ISA], 0.615%/pt-yr[ISMA] and 1.822%/pt-yr[H], and bleeding complication at the rate of 0.871% /pt-yr[ISM], 0.63% /pt-yr[ISA], 0.205% /pt-yr [ISMA] and 0.729%a /pt-yr[H], respectively. Prosthetic valve endocarditis occurred at the rate of 0.610% /pt-yr[ISM], 1.475% /pt-yr[ISA], 1.639% /pt-yr[ISMA] and 0.972% /pt-yr[H]. The linearized annual incidence of primary tissue failure was 1.655%/pt-yr[ISM], l. 475%/pt-yr[ISA], 1.639% /pt-yr[ISMA], 2.187% /pt-yr[H] and 1.785% /pt-yr[Group HM : MVR with Hancock valve]. The incidence of tissue failure was significantly high in the patients younger than 30 years of age compared with the older patients. The actuarial survival was 87.7$\pm$2.5% at 10 years[ISM], 94.3$\pm$3.2% at 11 years[ISA], 89.6$\pm$3.4% at 10 years[ISMA] and 81.3$\pm$6.6% at 12 years[HM], The freedom from thromboembolism was 93.2$\pm$2.0% at 10 years[ISM], 90.6$\pm$4.6% at 11 years[ISA], 95.8$\pm$2.6% at 10 years[ISMA] and 80.9$\pm$11.1% at 12 years[HM], And, the freedom from primary tissue failure was 84.2$\pm$3.8% and 28.1$\pm$23.0% at 9 and 10 years[ISM], 60.4$\pm$16.9% at 11 years[ISA], 62.3$\pm$12.7 at 10 years[ISMA] and 65.6$\pm$9.8% at 12 years[HM]. In conclusion, the standard Ionescu-Shiley and the Hancock bioprosthetic valves are excellent in their antithrombogenicity and long-term survival. However, the features of the structural failure with the prolonged follow-up beyond 10 years appear to be guarding, and the clinical indications of these bioprostheses seem to be quite limited.

Quality Characteristics of the Hasuo (Polygoni multiflori Radix) Muffin Prepared with Different Types of Sweeteners (하수오 머핀 제조시 당 종류가 품질에 미치는 영향)

  • The purpose of this study was to develop a functional hasuo (Polygoni Multiflori Radix) muffin prepared via the replacement of sucrose (SM) with oligosaccharide (fructooligosaccharide, isomaltooligosaccharide, galactooligosaccharide). The fructooligosaccharide-added hasuo muffin showed the highest weight while the volume and specific volume were lower in the oligosaccharide-added group than in the sucrose-added sample. The pH of the sucrose-added sample was higher than that of the oligosaccharide-added sample. The lowest lightness value was found in the sucrose-added sample, but the highest redness and yellowness values were found in the sucrose-added sample. The springiness and cohesiveness were highest in the fructooligosaccharide-added sample. The DPPH-radical-scavenging activity of the hasuo muffin including sucrose was 38.67, while that including the oligosaccharide group ranged from 42.87 to 65.95. In the sensory evaluation of the hasuo muffin, the fructooligosaccharide-added sample obtained the highest score in after-swallow, flavor, taste, and texture. The hasuo muffin with fructooligosaccharide and isomaltooligosaccharide obtained high scores in overall acceptability. These results suggest that hasuo muffin with fructooligosaccharide and isomaltooligosaccharide have the potential to become commercially successful muffins.

5 Year Cumulative Survival Rate of Composite Resin Restorations in Permanent First Molars (제1대구치 복합레진 수복의 5년 누적 생존율)

  • The purpose of this retrospective study was to evaluate the survival rate of composite resin restorations in permanent first molars in pediatric patients focusing on the influence of risk factors related to patients and teeth. 172 patients (6 - 12 years old) who had their permanent first molars restored with composite resin from July 2010 to July 2012 were investigated. From the dental records, the influence of the risk factors on the survival of restorations was assessed. Location of teeth, classification of cavities, patients' age and caries risk were included as risk factors in this study. The caries risks of patients were evaluated by the value of the decayed-missing-filled teeth index with the records of patients taken at 5 years old. Among 354 restorations, 272 restorations retained and 82 restorations were replaced. The overall cumulative survival rate at 5 years was 73.9%. The main reason for replacement of restorations was secondary caries (81.7%). Patients with older age group and with lower caries risk group showed higher survival rate of restorations. No statistically significant influence was detected between the survival rates and the possible risk factors : location of teeth, patients' age and caries risk. The survival rate of restorations was significantly affected by the classification of the cavities (p = 0.002).
