Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level.   • This study is to find out effects of hot water soluble extract from green tea, one of the Korean favorites, on the calcium metabolism and bone strength in body. To do so, calcium, phosphate, creatinine concentration and ALP activity in blood and the content of calcium and ash in the organ, the length, weight, strength in bone were measured. In addition, to find the calcium metabolism, the level of calcium intake, excretion, retention were measured. Twenty male Sprague-Dawley rats were divided into two groups and isoloated soy protein was provided as the source of protein and CaCO₃ was provided as the source of calcium. 0.5% hot water soluble extract from green tea was provided to the green tea groups and for the control group deionized water was provided. The results are as follows ; 1. There is no difference between the experimental groups in diet intake, weight gain, and the feed intake. 2. Feed efficiency ratio was low in the group which hot water soluble extract from green tea was provided. 3. There is no difference between groups the level of calcium, phosphorus, creatinine and ALP activity in serum. 4. There is no difference between groups weight, contents of ash and calcium in kidney and liver. 5. There is no difference between groups in calcium intake, absorption, excretion, and retention. 6. There is no difference between groups weight, length and strength in bone. In summary, when hot water soluble extract from green tea was provided with the amount of 150-200mg, which is taken when people generally drink as favorite tea, weight gain was reduced due to the decrease of feed efficiency ratio. However, it did not affect the availability of calcium in body at all. Thus, even if a big quantity of green tea powder or solid of hot green tea extract is not provided, the quantity obtained when people drink green tea lowers the feed efficiency ratio without reducing availability of calcium in body.

Antimicrobial Activities of Commercially Available Tea on the Harmful Foodborne Organisms (식품유해균에 대한 차류 추출물의 항균효과)

  • Use of chemical preservative for controlling harmful microorganisms in food products has been debated due to public concerns about food quality because of perceived toxic and carcinogenic potential. Thus, use of non toxic natural antimicrobial agents has become essential. This study was investigated to determine the antimicrobial activity of water or ethanol extract of commercially available tea, and of solvent fractionated ethanol extracts obtained from steamed green tea. Both of water and ethanol extracts of green tea(steamed or roasted), oolong tea and black tea exhibited strong antimicrobial activity against gram positive and negative bacteria, but not effective against yeast and mold. Also, antimicrobial activity of ethanol extract of 4 different kinds of tea was stronger than that of water extract. Among 4 different tea, ethanol extract of steamed green tea was further fractionated. One thousand g/disk buthanol extract had the strongest antimicrobial activity against bacteria and mold. The concentration of the antimicrobial activity of buthanol extract in tested microorganisms ranged from 125~1000 g/disk except for Rhizopus javanicus. Antimicrobial activity of buthanol extract of steamed green tea was not destroyed by heating at 100oC for 60 min and at 121oC for 15 min, which is very stable over heat treatment. The inhibitory effect of the buthanol extract on the growth of Listeria monocytogenes and Staphylococcus aureus was investigated. Growth of both strains was started in the presence of 250 and 500 g/ml after 12 and 24 hour respectively, whereas complete inactivation of both strains was occurred in the presence of 1000 g/ml.

A Convenient Manufacturing Method for Mass Production of EGCG Rich Green Tea Extract (Epigallocatechin Gallate 고함유 녹차추출물의 제조공정 개선)

  • A facile and convenient method was developed for the mass production of epigallocatechin gallate (EGCG) rich green tea extract (Er-GTE). The Er-GTE was successfully obtained from the crude water extract of green tea by the combination of two step purification, i.e., a simple adsorption process on the cation exchange resins (Trilite SCR-B) followed by the chromatography with Diaion HP-20 resins. The green tea extract produced by water extraction under $45^{\circ}C$ was subjected to adsorb on the strongly acidic cation exchange resin, Trilite SCR-B. The eluate passed through the resin was reabsorbed on Diaion HP-20 resin, which was subjected to elute with a mixture of water and alcohol by conventional chromatographical manner. The EGCG content in Er-GTE was estimated above 97% by HP-LC analysis and the newly developed method was regarded as the most suitable and appropriate process for the mass production of epigallocatechin gallate rich green tea extract (Er-GTE).

Effects on blood pressure and heart rate in normal cats by administration of Korean native green tea (한국산 녹차가 가축의 혈압 및 심박수에 미치는 영향)

  • This study was undertaken to investigate changes in blood pressure and heart rate in normal cats by administration of Korean native green tea. The chemical components of Korean native green tea which determined are water(2.2%), water extract(32.7%), vitamin C(480%), caffeine(2.6%), tannin(32.7%) and amino acid of water soluble content 5.8%. Effect on blood pressure in administration 100mg/kg, 300mg/kg, 500mg/kg of extract of green tea, the mean depressor response is 44.8$\pm$3.3mmHg, 60.5$\pm$3.6mmHg, and 65.0$\pm$3.3mmHg in normal cats. Effect on heart rate in administration 100mg/kg, 300mg/kg of extract of Korean native green tea, the mean decreased heart rate is 2.8$\pm$4.5 beats/min, 15.2$\pm$6.4beats/min and 19.1$\pm$4.0 beats/min.

Antioxidative Effect on the Green Tea and Puer Tea Extracts (녹차와 보이차 추출물의 항산화 효과)

  • To see the antioxidative ability of puer tea against green tea, antioxidative effects of water and methanol extracts were compared and researched each. Water extracts from green tea and puer tea contain similar percentage of total polyphenol content while methanol extract from puer tea has about $58\%$ of total polyphenol content of that from green tea. Water extracts from both teas contain similar percentage of the electron donating ability while methanol extract from puer tea has about $54\%$ of the electron donating ability of that from green tea. Solvents made a big difference in SOD-like activities. The activations in water extracts were from $13.46\%$ to $48.93\%$, which didn't give much difference between green tea and puer tea. However, the activations in methanol extracts were $59.63\%$ in green tea and $48.93\%$ in puer tea, which was $3\~4$ times higher than that in water extracts and green tea activated better. The nitrite scavenging activity was similar in water extracts from both teas and the methanol extract from puer tea had a bit higher nitrite scavenging activity than that from green tea. The ACE inhibitory activity in water extract from puer tea was about $38\%$ higher than that from green tea while the activity of methanol extract from green tea was higher than that from puer tea. These results indicate that the antioxidative ability of water extract from puer tea is similar to that from green tea but the ACE inhibitory activity of puer tea reveals higher value and antioxidative ability of methanol extract from puer tea was lower than that from green tea while the nitrite scavenging activity of puer tea was higher.

A Study on Characteristics of Soybean Sprouts Cultured with Green Tea Extracted (녹차 물로 재배한 콩나물의 생육 특성)

  • This study examined growth and sensory characteristics of soybean sprout cultured at $25\pm1^{\circ}C$ for 4 days with distilled water(control) and green tea extract(0.03 and $0.05\%$). Initial germination rate was higher when soaking in green-tea extract than in the control. The proper concentration of green-tea extract was $0.03\%$. The growth of soybean sprout in $0.05\%$ green-tea extract was better than the control for the hypocotyl thickness and mot hair. Soybean sprout grown with green-tea extract were better than that with control inappearance, flavor, texture and over-all acceptabilitty.

  • It was reported that green tea extract has antioxidant and antimicrobial activities. To improve the quality of seasoned squid, green tea water extract was sprayed on seasoned squid (water content 25%) and water content was adjusted to 27% and 29%. During storage at $30^{\circ}C$, TBA value, total plate count, browning were analyzed. Browning were analysed by 3 different method (absorbance of water solubilized color, 5% KOH-methanol solubilized color and color difference meter). Water and 5% KOH-methanol solubilized color showed good correlation, but color index detected by color difference meter was not. Green tea water extract slowed TBA value increment and reduced total plate count, so it was supposed that green tea water extract acts positively in manufacturing seasoned squid.

Effects of Green Tea Extract Diet on the Phospholipid Content of Aluminum-Induced-Damaged Cerebral Tissue of Old Rats (노령 흰쥐의 대뇌 조직에서 알루미늄 투여에 대한 녹차 추출물이 인지질 함량에 미치는 효과)

  • This study was designed to investigate the effects of green tea extract on aluminum-induced damage to phospholipid content in old rat cerebral tissue. The aim of this study was to investigate the possibility that aluminum is the cause of Alzheimer's disease. Forty Sprague-Dawley old male rats weighing 350$\pm$10 g were divided into four groups, consisting of a control group (CON), 40 ppm aluminum sulfate group (Al-40), green tea water extract group (GTWE), and 40 ppm aluminum sulfate and green tea water extract groups (Al-40+GTWE) and kept on their respective diets for 12 weeks. In order to discover the influence of aluminum on cerebral tissue of old male rats, the serum aluminum concentration and phospholipid composition were compared between the aluminum-treated group and the normal group. The results showed that the serum aluminum concentration was higher in the aluminum sulfate-treated group than in the normal group. The cerebral tissue phospholipid concentration decreased significantly in the aluminum sulfate treated group as compared to the normal group. The results of this experiment show that increase of aluminum concentration in experimental animals causes the rise of serum aluminum and phospholipid concentrations, phenomena that are very similar to those shown in Alzheimer's disease., The results of this experiment, together with reports that aluminum is a cause of neurofibrillary tangles in cerebral tissue, therefore demonstrate the possibility that aluminum is the cause of Alzheimer's disease. Green tea water extract is also shown to be an effective therapeutic candidate for the treatment of Alzheimer's disease.

  • In this study we isolate lactic acid bacteria from kimchi, which were identified as pediococcus pentosaceus and Lactobacillus bevis by using API 50 CHL Kit, some morphological and physiological tests, The water extract of green tea(50, 100, 500, 1000ppm) inhibieted the growth of pediococcus pentosaceus and Lactobacillus brevis in broth system. Furthermore, their time were prolonged to about 4hours at 500 to 1000ppm. The leakage amount of intracellular materials and Mg++ in tested bacteria cells increased with increasing concentration of green tea water extract.
