Corynespora Leaf Spot of Balsam Pear Caused by Corynespora cassiicola in Korea

  • Corynespora leaf spot occurred severely on balsam pear (Momordica charantia) at Changwon, Gyeongnam province in Korea in November and December 2003. The causal fungus isolated from infected leaves of the plants grew well on potato dextrose agar showing gray to brown color. Solitary or catenary conidia of the fungus were obclavate to cylindrical in shape, and pale olivaceous brown or brown in color. The number of isthmus pseudosepta ranged from 4 to 20 and measured 36~186${\times}$8~19 ${\mu}m$ in size. Conidiophores were pale to light brown in color and measured 94~648${\times}$3~8 ${\mu}m$ in size. Optimal temperature for mycelial growth was $30{\circ}C$. On the basis of mycological characteristics and pathogenicity, the fungus was identified as Corynespora cassiicola (Berk. & Curt.) Wei. This is the first report on the corynespora leaf spot of M. charantia caused by C. cassiicola in Korea.

  • A corynespora leaf spot of pepper, which has not been reported previously in Korea, occurred severely at the major pepper cultivation area of Chinju, Gyeongnam province in 2001. Infection rate ranged from 48.2 to 84.7% in eight fields surveyed. The causal fungus was identified as Corynespora cassicola based on the following cultural and morphological characteristics. The fungi grew well on potato dextrose agar, showing gray to brown color with cultural age. Conidia formed solitary or catenary were obclavate to cylindrical in shape, and pale olivaceous brown or brown in color. They had 420 pseudoseptate and isthmus, and measured 42.7-197.6 x 9.3-$23.5\mu\textrm{m}$. Septate conidiophores were pale to light brown in color, and measured 116.5-836.0 x 4.2-$11.0\mu\textrm{m}$. Conidia germinated as a bi-polar type. Optimal temperature for mycelial growth and conidial germination was $30^{\circ}$ and 25-$30^{\circ}$, respectively. The fungus showed strong pathogenicity to pepper plant, and the symtoms on pepper by the artificial inoculation were similar to those observed in the field. This is the first report on the corynespora leaf spot on pepper (Capsium annuum) caused by Corynespora cassicola in Korea.

Alternaria tenuissima에 의한 칡 점무늬병 (Leaf Spot of Kudzu (Pueraria lobata) Caused by Alternaria tenuissima)

  • 2002년 7월 전북지방에 야생하는 칡의 잎에 점무늬병이 발병하였다. 잎에 처음에는 작은 점무늬증상이 나타나고. 이 점무늬 주변에 황색의 halo가 형성되는 것이 전형적인 특징이었다. 병반들이 점차 확대되어 융합되어 잎 전체가 황색으로 변색되며 결국 낙엽이 된다. 병원균이 분리되었으며 본 병원균의 균사집락은 PDA 배지에서 밝은 회녹색에서 암갈색이었다. 분생포자는 대부분 단생이며 드물게 3-8개 포자가 연쇄상으로 형성되었다. 분생포자는 암갈색이며 긴 난형 또는 곤봉형이고 크기는 20-60${\times}$10-25 $\mu\textrm{m}$이었다. 격막은 4-8개의 횡격막과 1-2개의 종격막을 형성하였다. 칡의 잎에 발생한 병징과 본 병원균의 균학적 특징을 검정한 결과, 이 병을 Alternaria tenuissima에 의한 칡 점무늬병으로 명명하고자 제안한다.

  • A disease causing severe leaf spots in pepper plants has been observed in northern Gyeongbuk and Gangwon provinces in Korea since 1994. The current study diagnosed the disease as gray leaf spot caused by Stemphylium solani Weber and S. lycopersici (Enjoji) Yamamoto, both of which are pathogenic in pepper and tomato plants. Although the disease has been found in almost all areas where peppers are grown, it is more severe in mountain terrains where the nights are cool. Both species of pathogenic fungi were found to sporu-late profusely on V-8 juice agar in plastic or Pyrex glass Petri dishes, although not in domestically-produced glass Petri dishes, when cultured at $20^{\circ}C$ under irradi-ation from a daylight fluorescent lamp with a 12-hour light and dark alternation. The domestically-produced glass Petri dishes, which are made of window glass, were found to block near ultraviolet wavelengths, around and below 300 nm, which explained why the fungi did not sporulate. However, sporulation decreased at above $25^{\circ}C$ and most isolates failed to sporulate above $27^{\circ}C$. The worst level of disease was obtained when the inoculated plants were incubated with a $15^{\circ}C$ night and $20^{\circ}C$ day temperature regime relative to 4 night/day temperature combinations (15/20, 20/25, 25/30, and 30/35$^{\circ}C$).

Morphological and Molecular Characterization of Pseudocercospora chionanthi-retusi Causing Leaf Spot on Chionanthus retusus in Korea

  • Leaves of Chionanthus retusus were found to be damaged by leaf spot disease associated with a fungus in Iksan, Korea. Leaf spots were angular to irregular, vein-limited, scattered, 1-8 mm diameter, brownish-gray to dark brown when dry, with heavy fructification. The pathogen causes premature defoliation of C. retusus plant and was identified as Pseudocercospora chionanthi-retusi based on morphological and molecular-phylogenetic analyses. The phylogenetic tree was constructed using multi-locus DNA sequence data of partial actin (actA), partial translation elongation factor 1-alfa (tef1), partial DNA-directed RNA polymerase II second largest subunit (rpb2) genes, and internal transcribed spacer regions. Current study provides detail morphological description of P. chionanthi-retusi on C. retusus in Korea, with supports of phylogenetic analysis and pathogenicity test.

Alternaria mali에 의한 아로니아 점무늬낙엽병 (Alternaria Leaf Spot Caused by Alternaria mali on Black Chokeberry in Korea)

  • 2014년 6월 초 충남 예산과 금산의 농가포장에서 재배 중인 아로니아에서 점무늬낙엽병으로 의심되는 증상이 관찰되었다. 초기에는 잎에 갈색의 작은 점무늬가 생기고 이것이 확대되면서 둥근 겹무늬를 형성하고, 암갈색으로 변하였다. 그리고 병세가 심해지면 잎 전체에 병반이 확대되면서 떨어지는 증상이 관찰되었다. 병든 아로니아 잎에서 분리한 병원균은 PDA에 배양하였을 때 균체는 올리브 회색과 진한 회색으로 다양한 색을 나타내었다. 포장에서 감염된 잎과 PDA에서 분리한 분생포자의 크기는 각각 $19-50{\times}5-9{\mu}m$, $20-59{\times}8-13{\mu}m$이고, 길거나 짧은 부리를 가지며, 분생자경의 끝에 1-3개의 분생포자가 형성되었으며, 3-8개의 횡경막을 보이고 종경막은 없거나 1-3번째 횡경막 내에 1개 형성되었다. 병원성 검정 결과, 접종 7일 후 아로니아와 사과 모두에서 자연발생한 병징과 동일하게 관찰되었다. 이상 병원균의 형태적 특성, 병원성 검정과 ITS rDNA 염기서열 비교분석 결과를 토대로 아로니아에서 분리한 병원균은 A. mali로 동정하였고, 이 병을 아로니아 점무늬낙엽병으로 명명하였다.

Incidences of Leaf Spots and Blights on Kiwifruit in Korea

  • Various kinds of leaf spots and blights were found in kiwifruit (Actinidia deliciosa) orchards on 2006 in Korea. Disease incidences were quite variable between open-field and rain-proof shelter. Rate of diseased leaves was recorded as about 70% at open-field orchards in late season but use of rain-proof vinyl shelters alleviated the disease incidences by 20%. Angular leaf spots appeared at early infection stage on June and several other symptoms were also recognized as the disease developed afterward. On September, brown leaf blights were the most frequent, followed by grayish brown ring spots, silvering gray leaf blights, zonate leaf blights, dark brown ring spots and angular leaf spots at open-field orchards. Four fungal species were frequently isolated from the disease symptoms. Phomopsis sp. was the most predominant fungus associated with the leaf spot and blight symptoms on kiwifruit, followed by Glomerella cingulata, Alternaria alternata and Pestalo-tiopsis sp. Phomopsis sp. was commonly isolated from angular leaf spots, silvering gray leaf blights, and zonate brown leaf blights. G. cingulata, A. alternata and Pestalotiopsis sp. were isolated from grayish brown ring spots (anthracnose), brown ring spots and zonate dark brown leaf blights. Typical symptoms appeared on the wounded and unwounded leaves, which were inoculated by each of Phomopsis sp., G. cingulata, and Pestalotiopsis sp., but A. alternata caused symptoms only on the wounded leaves.

Leaf Rot and Leaf Ring Spot Caused by Rhizoctonia solani in Chinese Cabbage

  • This study was conducted to determine the occurrence of leaf rot and leaf ring spot, caused by Rhizoctonia solani in Chinese cabbage under seedling nursery and cultivation greenhouses. Symptoms of leaf rot and leaf ring spot were found in three Chinese cabbage cultivars, Brassica campestris subsp. pekinensis, 'Ryeokgwang', 'Daetong', and 'CR mat'. In Hwacheon, the disease incidence was 73.8% in the seedling stage of the Chinese cabbage. In Icheon, the symptoms were observed on the upper leaves of the Chinese cabbage cultivar, 'Norangmini' with 20.5% of disease incidence. The symptoms appeared as primary lesions consisting of small, circular necrotic ring spots with gray color, 1.4-3.0 mm in diameter, accompanied by secondary rot lesions with large irregular borders of leaves. The color of mycelial mat of 20 isolates was dark brown and light brown. The average hyphal diameter of all the isolates was within 5.01-11.12 ${\mu}m$. Among the 20 strains isolated from Chinese cabbage, 16 isolates and four isolates anastomosed with the AG-1 (IB) and AG-1 (IC), respectively. Twenty isolates tested were only virulent on foliage parts of Chinese cabbage leaves but were avirulent on stem parts of the plants. Based on the mycological characteristics and pathogenicity test on host plants, the fungus was identified as Rhizoctonia solani.

  • Yellow spot or blotch symptoms on the upper surface of leaf, without the production of velvet-like fungi on the lower surface of leaf as in the gray mold of tomato caused by Cladosporium fulvum, were observed in tomato (cv. Seokwang) plants in May, 1995, in a vinyl-house of the experimental plot of Gyeongnam Provincial Rural Development Administration, Chinju, Gyeongnam, Korea. We identified this disease as powdery mildew of tomato caused by Oidiopsis taurica (L v.) Arnaud (=Leveillula taurica), which was new to Korea. Conidia of the fungus were borne on uni- or bi-septated conidiophores which were developed through the stomata of the tomato leaf. The conidia were slender, clavate and variable in size (31~111.6$\times$13.1 ${\mu}{\textrm}{m}$). The fungal conidia isolated from tomato leaves were inoculated to tomato plants, and the occurrence of the same disease was confirmed based on the symptomatology and the morphology of the pathogen reisolated.
