가온 재배 시 '피오네' 포도(Vitis vinifera × V. labrusca)의 엽과비에 따른 과실 특성 (Fruit Characteristics Based on Leaf to Fruit Ratio in 'Pione' Grapevine (Vitis vinifera × V. labrusca) during Cultivation with Heating)

  • BACKGROUND: Defoliation in grapevine cultivation is practically used to improve light environment within the canopy and thereby fruit quality. Effects of defoliation in five-year-old 'Pione' grapevine during cultivation with heating were investigated to find out optimum ratio of leaf area to fruit cluster weight (L/F). METHODS AND RESULTS: The grapevines were defoliated with berry-thinning 20 days after full bloom to provide various levels of L/F. At harvest, total leaf area values of fruit bearing branches were between 0.23 and $0.60m^2$. With increasing L/F, soluble solids and anthocyanin contents curvilinearly increased ($R^2=0.76^{**}$). At L/F over $0.6m^2/kg$, soluble solids content (SSC) leveled off. With increasing L/F, titratable acidity (TA) linearly decreased ($R^2=0.87^{**}$), but the ratio of SSC to TA linearly increased ($R^2=0.86^{**}$). Anthocyanin content was significantly correlated with SSC and the ratio of SSC to TA ($R^2=0.80^{**}$ and $0.82^{**}$, respectively). When total leaf area per fruit bearing branch was maintained $0.40m^2$, soluble solids and anthocyanin contents linearly decreased ($R^2=0.79^{**}$ and $0.85^{**}$, respectively), but TA linearly increased with increasing fruit cluster weight ($R^2=0.70^{**}$). Fruit was low in quality when the L/F was below $0.6m^2/kg$. CONCLUSION: L/F is recommended to be maintained at least $0.6m^2/kg$ in 'Pione' grapevine during cultivation with heating to produce higher-quality fruits.

생육단계별 포도 잎의 생리활성 성분 및 항산화능 (Physiologically Active Components and Antioxidant Capacity of Grapevine Leaves at Growth Stages)

  • 본 연구에서는 포도 잎의 품종별, 재배시기별 생리활성 효과를 분석하여 포도 잎의 자원화에 기여할 수 있는 기초자료를 제공하고자 하였다. 사용된 포도 잎은 캠벨얼리와 로자리오비앙코 품종으로 생육단계별로 전엽기, 개화기, 결실기, 착색기 및 성숙기로 분류하여 포도 잎의 생리활성의 차이를 분석하였다. 수분과 조단백질 함량은 생육단계별로 유의적으로 감소하였으나 섬유소의 함량은 시기가 지남에 따라 유의적으로 증가하였다. 총 플라보노이드 함량은 캠벨얼리와 로자리오비앙코 품종 모두에서 전엽기에서 높게 나타났으며, 생육단계가 진행됨에 따라 감소되었다. 총 페놀 함량은 캠벨얼리 품종이 로자리오비앙코 품종에 비해 높은 경향이었다. Hydroxyl radical 소거능은 전엽기에서 가장 높게 나타났으며 생육단계가 지남에 따라 점차 낮은 소거능을 보였다. 포도 잎의 전자공여능은 개화기까지 캠벨얼리 품종이 로자리오비앙코 품종에 비해 높았으며, 총 항산화능은 성숙기를 제외하고는 생육단계별로 캠벨얼리 품종이 로자리오비앙코 품종에 비해 높은 수준을 나타내었다. 본 연구 결과로 미루어 볼 때 특히 개화기 이전의 포도 잎은 폴리페놀 함량이 높으며 강력한 항산화 효능을 가진 것으로 나타나 이 시기의 포도 잎은 건강기능식품의 소재로 산업화하는데 유용한 자원으로 이용될 수 있을 것으로 생각된다.

RAPD를 이용한 한국 포도 품종의 계통유연관계 분석 (Analysis of Genetic Relationships of Grapevine Cultivars (Vitis ssp.) in Korea Using RAPD Markers)

  • 본 실험은 29개 포도 품종(Vitis spp.)의 효율적 유전학적 유연관계를 분석함으로써 포도품종 육성을 위한 기초자료로 이용하고자 RAPD(Random Amplified Polymorphic DNA) 방법을 이용하여 분석하였다. 총 60개의 프라이머를 이용하여 558개의 다형성 밴드를 얻었다. 획득된 다형성 밴드를 사용하여 UPGMA(unweighted pair-group method arithmetic average) clustering 분석 결과, 포도 품종은 유사도 0.588을 기준으로 나누었을 때 6개 그룹으로 분류되었다. 가장 높은 유사도 값(0.980)을 나타낸 것은 국내 육성 품종인 "흑보석"과 "수옥" 품종이었고, 가장 낮은 유사도 값(0.404) 을 나타낸 것은 프랑스 품종인 "S. 9110"과 일본 품종인 "Super Hamburg"로 나타났다. "Super Hamburg"는 첫 번째 그룹(I)에 구성되었다. 두 번째 그룹(II)은 "원교 라-23"과 "Muscat Hamburg" "Tano Red" 그리고 "탄금추"가 포함되어 있었다. 세 번째 그룹(III)은 "Alden" "원교 라-21", "원교 라-30" 그리고 "Dutchess" 품종이 속해있었다. 네 번째 그룹(IV)은 14개 포도 품종이 포함되었으며("흑구슬", "흑보석", "수옥", "원교라-29", "원교 라 -22", "거봉", "Pione", "홍이두", "Golden Muscat", "진옥", "두누리", "캠벨얼리", "Delaware", "Schuyler"), 거봉 계열의 5 품종과 캠벨얼리 계열의 4 품종 그리고 이들의 교배조합에 사용된 품종이 포함되어 있었다. 다섯 번째 그룹(V) 3개 품종으로 구성되었다(홍단', "탐나라", "홍이슬", "Himrod seedless"). 여섯 번째 그룹(VI)은 "청수"와 모본인 "S. 9110"이 포함되었다. 본 실험의 결과는 DNA 수준에서 포도의 교배육종에 이용할 양친의 선정이나 육종연구에 기초 자료가 될 수 있을 것으로 기대된다.

Development of an efficient genotyping-by-sequencing (GBS) library construction method for genomic analysis of grapevine

  • Genotyping-by-sequencing (GBS) is an outstanding technology for genotyping and single nucleotide polymorphism (SNP) discovery compared to next generation sequencing (NGS) because it can save time when analyzing large-scale samples and carries a low cost per sample. Recently, studies using GBS have been conducted on major crops and, to a greater extent, on fruit crops. However, many researchers have some problems due to low GBS efficiency resulting from low quality GBS libraries. To overcome this limitation, we developed an efficient GBS library construction method that regulates important conditions such as restriction enzymes (RE) digestion and a PCR procedure for grapevine. For RE digestion, DNA samples are digested with ApeKI (3.6U) at $75^{\circ}C$ for 5 hours and adapters are ligated to the ends of gDNA products. To produce suitable PCR fragments for sequencing, we modified the PCR amplification conditions; temperature cycling consisted of $72^{\circ}C$ (5 min), $98^{\circ}C$ (30 s), followed by 16 cycles of $98^{\circ}C$ (30 s), $65^{\circ}C$ (30 s), $72^{\circ}C$ (20 s) with a final extension step. As a result, we had obtained optimal library construct sizes (200 to 400 bp) for GBS analysis. Furthermore, it not only increased the mapping efficiency by approximately 10.17% compared to the previous method, but also produced mapped reads which were distributed equally on the19 chromosomes in the grape genome. Therefore, we suggest that this system can be used for various fruit crops and is expected to increase the efficiency of various genomic analysis performed.

Optimization of Agrobacterium-mediated transformation procedure for grapevine 'Kyoho' with carrot antifreeze protein gene

  • We report an Agrobacterium-mediated transformation procedure optimized for 'Kyoho' that is a major table grapevine cultivar in Korea, and its transgenic plants with antifreeze protein gene of carrot (DcAFP). The full length of DcAFP coding region in accordance with the previous report was isolated from young leaves of carrot and recombined into a plant transformation vector. Ethylene inhibitors such as silver nitrate and aminoethoxyvinylglycin (AVG) supplemented in a co-cultivation medium distinctly increased frequency of shoot regeneration when explants were sub-cultured in a selection medium: particularly ten-fold higher in treatment with 0.1 mg/L AVG than one without ethylene inhibitor. Among various antibiotics and their concentrations, the combination of 150 mg/L cefotaxime plus 150 mg/L $Clavamox^{TM}$ was selected for elimination of Agrobacterium cells in addition to minimization of adverse effect on shoot regeneration, while 50 mg/L kanamycin monosulfate effectively suppressed regeneration of non-transgenic shoots. Applying the elucidated culture condition, we finally obtained a total of 5 transgenic 'Kyoho' plantlets with DcAFP, of which integration with the grapevine genome and transcription was confirmed by nucleic acid analyses.

기온자료에 근거한 주요 포도품종의 휴면해제 및 발아시기 추정 (Prediction of Dormancy Release and Bud Burst in Korean Grapevine Cultivars Using Daily Temperature Data)

  • An accurate prediction of dormancy release and bud burst in temperate zone fruit trees is indispensable for farmers to plan heating time under partially controlled environments as well as to reduce the risk of frost damage in open fields. A thermal time-based two-step phenological model that originated in Italy was applied to two important grapevine cultivars in Korea for predicting bud-burst dates. The model consists of two sequential periods: a rest period described by chilling requirement and a forcing period described by heating requirement. It requires daily maximum and minimum temperature as an input and calculates daily chill units (chill days in negative sign) until a pre-determined chilling requirement for rest release is met. After the projected rest release date, it adds daily heat units (anti-chill days in positive sign) to the chilling requirement. The date when the sum reaches zero isregarded as the bud-burst in the model. Controlled environment experiments using field sampled twigs of 'Campbell Early' and 'Kyoho' cultivars were carried out in the vineyard at the National Horticultural Research Institute (NHRI) in Suwon during 2004-2005 to derive the model parameters: threshold temperature for chilling and chilling requirement for breaking dormancy. The model adjusted with the selected parameters was applied to the 1994-2004 daily temperature data obtained from the automated weather station in the NHRI vineyard to estimate bud burst dates of two cultivars and the results were compared with the observed data. The model showed a consistently good performance in predicting the bud burst of 'Campbell Early' and 'Kyoho' cultivars with 2.6 and 2.5 days of root mean squared error, respectively.
