In this study, a total of 217 samples were collected from five grapevine cultivars in Chungcheongbuk-do and Gyeongsangbuk-do in 2021. These samples were tested for the presence of five grapevine viruses using specific primers through polymerase chain reaction. The results indicated that grapevine fleck virus was confirmed in 126 samples (58.06%), grapevine leafroll-associated virus (GLRaV)-3 in 112 samples (51.61%) and GLRaV-1 in four samples (1.84%). The infection rate was high in Shine Muscat and Alexandria cultivars, and by region, the Gyeongbuk region showed a high infection rate. However, the presence of grapevine red blotch associated virus and grapevine berry inner necrosis virus were not detected.
Grape fruit quality can be deteriorated with leaf chlorosis caused by magnesium (Mg) deficiency during fruit coloring season in several grapevine varieties. The occurrence of leaf chlorosis and soil Mg states for four grapevine varieties (Campbell Early, Muscat bailey A (MBA), Sheridan, and Kyoho) in the vineyards of Gyeongsan and Youngcheon were surveyed. The relationships between leaf chlorosis and fruit qualities were also investigated. Leaf chlorosis was more widely found and the symptom was stronger in Campbell Early in comparison to the other varieties. Sugar content and Hunter values (L, a, b) of grape fruit were significantly lower in the trees of chlorosis when compare to healthy grapevine trees. Soil Mg contents in the vineyards where leaf chlorosis was found were lower than the optimum level for grapevine. Also Mg content in the petioles of chlorosis grapevines was significantly lower than healthy grapevines. Application of Mg through foliar spray and soil fertigation was quite effective in correcting deficiencies of Mg in grapevine. Proper management of soil Mg availability and K/Mg ratio is strongly recommended to prevent Mg deficiency in grapevine.

Grapevine leafroll-associated 1 virus (GLRaV-1) and Grapevine leafroll-associated 3 virus (GLRaV-3), member of the genus Ampelovirus, are important viral disease of grapevine in the world. these viruses transmitted only dicotyledonous host by vectors such as mealybugs and there is no suitable herbaceous host for virus. The diseased leaves turn yellowish or reddish depending on cultivars and viruses. Viruses are existed at low concentration and ununiformly distribution in grapevine. Using small-scale double-stranded RNA (dsRNA) extraction method, reverse transcription and polymerase chain reaction (RT-PCR) product of 1Kb long which encoded of coat protein (CP) gene for both viruses was successfully amplified with a specific primers. The RT-PCR product was cloned into the plasmid vector and its nucleotide sequences were determined from selected recombinant cDNA clones. Sequence analysis revealed that the CP of GLRaV-1 consisted of 969 nucleotide, which encoded 323 amino acid residues and CP of GLRaV-3 consisted of 942 nucleotide, which encoded 314 amino acid residues. The CP of GLRaV-1 and GLRaV-3 has 93.8% and 98.7% amino acid sequence identities, respectively.

Grapevine leafroll-associated 3 virus (GLRaV-3) and Grapevine fanleaf virus (GFLV) are important viral diseases of grapevine in the world. In this study, the most reliable woody indicator plants were selected for virus indexing. Two grapevines, LN33 (Couderc 1613x vitis berlandieri) and Vitis riparia Gloire, were selected for CLRaV-3 and CFLV graft indexing, respectively. The specific characteristics of Closterovirus isolated from grapevines cultivated in Korea were identified. filamentous virus-like particles only existed in the phloem parenchyma cell. In particular, the vesiculation of mitochondria was observed. This mitochondrial vesicu-lation was considered to be one of the most reliable cytopathic features of Closterovirus. During observation of GFLV-infected Chenopodium quinoa sections, virus-like particles arranged consistently were found forming several layers in cytoplasm. Moreover, virus-like particles in tubules were observed and were associated with plasmodesmata in cytoplasm. This is the first report on cytopathological characteristics of Closterovirus and Nepovirus identified from grapevines in Korea.
Prospecting of local grapevine (Vitis vinifera L.) germplasm revealed that Tunisia possesses a rich patrimony which presents diversified organoleptic characteristics. However, viral diseases seriously affect all local grapevine cultivars which risk a complete extinction. Sanitation programs need to be established to preserve and exploit, as a gene pool, the Tunisian vineyards areas. The presence of the Grapevine leafroll associated virus-3 (GLRaV-3), Grapevine stem pitting associated virus (GRSPaV) and Grapevine virus A (GVA), were confirmed in a Tunisian grapevine cultivar using serological and molecular analyses. The association between GRSPaV and GVA viruses induces more rugose wood symptoms and damages. For this reason the cleansing of the infected cultivar is highly advisable. Direct and recurrent somatic embryos of cv. 'Hencha' were successfully induced from filament, when cultured on $Ch{\acute{e}}e$and Pool (1987). based-medium, enriched with $2mg1^{-1}$ of 2,4-dichlorophenoxyacetic acid and $2.5mg1^{-1}$ of Thidiazuron, after 36 weeks of culture. After six months of acclimatization, RT-PCR carried on 50 somaplants confirmed the absence of GVA, GRSPaV as well as GLRaV-3 viruses in all somaplants. Ampelographic analysis, based on eight OIV descriptors, was carried out on two years acclimated somaplants, compared to the mother plant. Results demonstrated that the shape and contours of 46 somaclones leaves are identical to mother plant leaves and four phenotypically off-type plants were observed. The healthy state of 100% 'Hencha' somaclones and the high percentage of phenotypically true-to-type plants demonstrate that somatic embryogenesis is a promising technique to adopt for grapevine viruses elimination.

We investigated the physiological activities of extracts from Campbell Early and Muscat Bailey A (MBA) grapevine leaves. Total phenol and flavonoid contents were highest in ethanol extracts from MBA grapevine leaves compared to extracts from Campbell Early grapevine leaves. Specific polyphenols higher in ethanol extracts from MBA grapevine leaves include gallic acid, epicatechin, caffeic acid, naringin, and resveratrol. Resveratrol content from MBA grapevine leaves increased when extracted for more than two hours in ethanol and water. The hydroxyl radical scavenging ability of ethanol extracts was higher than the water extract from both strains of grapevine leaves. DPPH and total antioxidants were highest in ethanol extracts from MBA grapevine leaves among the other extracts. Therefore, these results suggest that ethanol extracts from MBA grapevine leaves are a highly valuable resource for the development of natural functional foods.

Green shoot cuttings of 10 different grapevine species were inoculated with Agrobacterium vitis to find disease-related metabolites in the grapevine. Crown galls formed 60 days after inoculation varied in gall severity (GS) evaluated by gall incidence (GI) and gall diameter (GD), which were classified into three response types as RR (low GI and small GD), SR (high GI and small GD), and SS (high GI and large GD), corresponding to resistant, moderately resistant, and susceptible responses, respectively. In this, 4, 4, and 2 Vitis species were classified into RR, SR, and SS, respectively. Gas chromatography mass spectrometry (GC-MS) analysis of the grapevine stem metabolites with A. vitis infection showed 134 metabolites in various compound classes critically occurred, which were differentially clustered with the response types by the principal component analysis. Multivariate analysis of the metabolite profile revealed that 11 metabolites increased significantly in relation to the response types, mostly at post-inoculation stages, more prevalently (8 metabolites) at two days after inoculation than other stages, and more related to SS (7 metabolites) than RR (3 metabolites) or SR (one metabolite). This suggests most of the disease-related metabolites may be rarely pre-existing but mostly induced by pathogen infection largely for facilitating gall development except stilbene compound resveratrol, a phytoalexin that may be involved in the resistance response. All of these aspects may be used for the selection of resistant grapevine cultivars and their rootstocks for the control of the crown gall disease of the grapevine.

This study was conducted to investigate the genetic diversity and to develop a technique for cultivar identification using SSR markers in grapevine. Thirty Korean bred and introduced grapevine cultivars were evaluated by 28 SSR markers. A total of 143 alleles were produced ranging from 2 to 8 alleles with an average of 5.1 alleles per locus. Polymorphic information contents (PIC) were ranged from 0.666 (VVIp02) to 0.975 (VVIn33 and VVIn62) with an average of 0.882. UPGMA (unweighted pair-group method arithmetic average) clustering analysis based on genetic distances using 143 alleles classified 30 grapevine cultivars into 7 clusters by similarity index of 0.685. Similarity values among the tested grapevine cultivars ranged from 0.575 to 1.00, and the average similarity value was 0.661. The similarity index was the highest (1.00) between 'Jinok' and 'Campbell Early', and the lowest (0.575) between 'Alden' and 'Narsha'. The genetic relationships among the 30 studied grapevine cultivars were basically consistent with the known pedigree. The three SSR markers sets (VVIn61, VVIt60, and VVIu20) selected from 28 primers were differentiated all grapevine cultivars except for 'Jinok' and 'Campbell Early'. Five cultivars ('Narsha, 'Alden', 'Dutchess', 'Pione', and 'Muscat Hamburg') were identified by VVIn61 at the first step. Then 21 cultivars including 'Hongsodam' by VVIt60 at the second step and 2 cultivars ('Heukbosuck' and 'Suok') by VVIu20 at the third step were identified. These markers could be used as a reliable tool for the identification of Korean grapevine cultivars.
Grapevine rupestris stem pitting-associated virus (GRSPaV) is a worldwide-distributed pathogen in grapevines with a high genetic variability. Our study revealed differences in the complexity of GRSPaV population in a single host. A single-variant GRSPaV infection was detected from the SK30 grapevine plant. On the contrary, SK704 grapevine was infected by three different GRSPaV variants. Variant-specific RT-PCR detection protocols have been developed in this work to study distribution of the three different variants in the same plant during the season. This study showed their randomized distribution in the infected SK704 grapevine plant. Comparative analysis of full-length genome sequences of four Slovak GRSPaV isolates determined in this work and 14 database sequences showed that population of the virus cluster into four major phylogenetic lineages. Moreover, our analyses suggest that genetic recombination along with point mutations could play a significant role in shaping evolutionary history of GRSPaV and contributed to its extant genetic diversification.

Agrobacterium vitis is a causal agent of crown-gall disease on grapevine. In Korea, grapevine variety (GeoBong) have severely been infected by the bacteria since stems of the variety were buried in soil for overwintering. Infection ratio over 70-80% was observed on 7 years old GeoBong grapevine in Ansung and Cheonan. PCR specific primers for A. vitis strains were designed using nucleotide sequences of vir A gene in Ti-Plasmid, pheA gene in chromosomal DNA and a URP-PCR polymorphic band. Three hundred bacterial strains were isolated from the different 80 galls formed on GeoBong grapevine in Cheonan and Ansung of Korea and were screened to identify A. vitis using the three specific PCR primers for Agrobacterium vitis. Twenty-four bacterial strains that are detected by the primers were further confirmed by pathogenicity and biochemical methods. To investigate the genomic diversity of the bacterial strains, twenty primers of 20 mer referred to universal rice primers (URP) were applied for PCR fingerprinting, Of them, URP2R and URP2F primers could effectively be used to detect polymorphism within the bacterial strains.
