신갈나무 잎의 용매분획별 항균 및 항산화 효과 (Antimicrobial and Antioxidative Activities of Solvent Fractions of Quercus mongolica Leaf)

  • The ethanol extract of Quercus mongolica leaf was fractionated by various organic solvents, and their antimicrobial and antioxidative activities were investigated against several microorganisms. The ethanol extract of Quercus mongolica leaf at two thousand $\mu\textrm{g}$ per disc showed 17~21mm inhibition zone against Gram postive and Gram negative bacteria. Among the various solvent fractions from ethanol extract of Quercus mongolica leaf, the hexane fraction showed the strongest antimicrobial activity. Minimal inhibitory concentration (MIC) of hexane fraction was 250$\mu\textrm{g}$/mL against Bacillus cereus, 250~500$\mu\textrm{g}$/mL against Listeria monocytogenes, 500$\mu\textrm{g}$/mL against Staphylococcus aureus, Salmonella typhimurium, Escherichia coli O157, and Pseudomonas aeruginosa. Also, the hexane and chloroform fraction had the similar antioxidative activity compared to that of butylated hydroxy toluene(BHT).

  • Regulation of pyrimidine nucleotide synthesis has been studied extensively in enteric bacteria and Bacillus species. Varieties of control modes have been proposed for regulation of pyrimidine nucleotide biosynthetic (pyr) genes. In Bacillus caldolyticus and B. subtilis, it has been proved that pyrimidine de novo biosynthetic operon is controlled by a regulatory protein PyrR-mediated attenuation. Another Gram-positive bacteria including Enterococcus faecalis, Lactobacillus plantarum, and wctococcus lactis have been found to constitute a pyr gene cluster containing the pyrR gene. In addition, it has been proposed that the structure of the 5' leader region of the Gram-negative extreme thermophile Thermus strain Z05 pyr operon provides a novel mechanism of PyrR-dependent coupled transcription-translation attenuation. Bacterial genome sequencing projects have identified the PyrR homologues in Haemophilus influenzae, Synechocystis sp., Mycobacterium tuberculosis, Streptococcus pneumoniae, S. pyogenes, and Clostridium acetobutylicum, which are currently investigating for their physiological functions.

  • Tylopeptins A and B are novel peptides from methanol extract of fruit body of mushroom, Tylopilus neofelleus, These novel peptides were isolated by reverse-phase HPLC. And they have been shown to consist of one acetylated N -terminal residue, fourteen amino acids, and leucinol, C-terminal amino alcohol. Sequencial determination and complete 1H and 13C resonance assignments were based on positive ion F AB mass spectroscopy and two dimensional NMR techniques using HOHAHA, ROSEY, HMQC, and HMBC. These compounds are active against some Gram-positive bacteria, but inactive against phathogenic fungi and Gram-negative bacteria.

  • Antimicrobial activity of GL (the aqueous extract from the carpophores of Ganoderma lucidum ($F_RK_{KARST}$) was tested in vitro aginst Gram positive and Gram negative bacteria by serial broth dilution method, and the antimicrobial activity was expressed by minimal inhibitory concentration (MIC). Among fifteeen species of bacteria tested, the natimicrobial activity of GL was of antimicrobial combinations of GL with four kinds of antibiotics (ampicilin, cefazolin, oxytet-racycline and chloramphenicol), the fractional inhibitory concentraction index (FICI) was determined by checkerboard assy for each stain. The antimicrobial combinations of GL with four antibiotics resulted in additive effect in most instances, synergism in two instances, and antagonism in two instances. Synergism was obversed when GL was combined with cefazolin against Bacillus subtilis and Klebsiella oxytoca.

  • 6$\beta$-(trans-3-Aryl-5-oxo-pyrrolidin-2-yl)acetamidopenicillanic acid(7a~7c) and 7$\beta$-(trans-3-Aryl-5-oxo-pyrrolidin-2-yl) acetamidocephalosporanic acid(8a~8c) were synthesized and tested in vitro antibacterial activity. Of these new penicillins exhibited good antibacterial activity against Gram-positive bacteria whereas none of the compounds possessed the activity against Gram-negative bacteria at the concentration tested.

Cystocin, a Novel Antibiotic, Produced by Streptomyces sp. GCA0001: Biological Activities

  • Cystocin belongs to the class of nucleoside antibiotics from Streptomyces sp. GCA0001. Cystocin showed good activity against Gram-positive bacteria, but showed less activity against the Gram-negative bacteria. Cystocin exhibited about two to four folds higher activity than puromycin. Especially, cystocin shows relatively strong activity against Streptococcus strains. Cystocin shows quite potent antitumor activity against all of the cells tested showing $IC_{50}$ values of 0.10 to 0.14 $\mu$ g/mL. This in vitro result indicates that the cytotoxocity of cystocin is two ten folds more active than puromycin s.

  • Celluloytic bacteria, -Gram positive, Gram negative and actionmycetes-were used to study their catabolic pathways of carbohydrate. It was found that Embden-Meyerhof-Parnas(EMP) pathway and hexose monophosphate(MHP) shunt were operated in Cellulomonase sp. CS1-1, C. flavigena, and Pseudomonas fluorescens subsp. cellulosa when they were cultured in a glucose containing medium, whilst gluconate was catabolised mainly via Entner-Doudoroff(ED) pathway, and to some extend through HMP shunt. Enzymes of ED pathway in the orgamisms were induced by gluconate. On the other hand Nocardia cellulans catabolised glucose and gluconate via EMP pathway and HMP shunt. The growth rate of N. Cellulans on gluconate were much slower than that on glucose.

Independent regulation of antigen processing and presentation on induction of antibody responses to various bacterial antigens in C3H/He mice

  • Induction of antibody production in C3H/He mice by bacterial infection is regulated through the processing exerted by antigen presenting cells. From the studies with Psudomonas aeruginosa, Salmonella typhimurium, and Micrococcus luteu, lipopolysaccharides (LPS) in Gram negative bacteria, which are known to be T-cell independent B cell mitogen, seem to be the major factor stimulating immune responses via activation of macrophages. Activation of macrophage, however, does not seem to correlate with antibody production. M. luteus was easily eliminatd by activated macrophages, while the processed antigens were immediately releasedd into culture medium before presentation. Nevertheless, antigens from Gram positive bacteria, Staphylococcus aureus and Bacillus subtilis, were very very active in chemotaxis and activation of periotoneal macrophages as well as in antien presnetation, while the very nature of the antigens is not yet clearly understood.

Bacteriocin을 생산하는 Lactococcus sp. HY 449의 분리와 항균 특성 (Isolation of Bacteriocin-Producing Lactococcus sp.HY 449 and Its Antimicrobial Characteristics)

  • A bacteriocin-producing lactic acid bacteria was isolated from contaminated milk products, which was identified by using the API50 CH kit as Lactococcus lactis subsp. lactis with reliability of 98%. Fatty and analysia of the cell membrane showed that this strain contained same fatty acids profiles as type strain, Lactococcus lactis subsp. lactis ATCC 19435. The bacteriocin of Lactococcus sp. HY 449 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacteria such as Escherichia coli and maintained the inhibitory activity between pH2.0 and pH9.0 The thermostability of this bacteriocin was higher in acidic solution than in distilled water and was stable at 60$\circ $C for 1 hour.

정유의 항균 성분을 이용한 항균 라이너지 개발 (Development of Antibacterial Corrugating Liner Using Essential Oils Extracted from Chamaecyparis obtusa)

  • Essential oils extracted from Chamaecyparis obtusa were applied to make antibacterial corrugating liner. The quantitative analysis of the essential oils was carried out by GC-MS, which showed that the monoterpenes as a basic component of the essential oils extracted from Chamaecyparis obtusa in the domestic mountain were greater in amount than those in China. The antibacterial activity was then examined with Tyromyces palustris and gram-negative/positive bacteria. The papers treated with the essential oils desplayed great resistance against T. Palustris and gram bacteria but the best results were obtained with the handsheet formed by softwood bleached kraft pulp rather than the liner by OCC. The low antibacterial activity of the liner was considered to be due to starch components which could be readily attacked by the fungi.
