Antibiotic Spectrum and Mechanism of Centipedin (Centipede Scolopendra subspinipes multilans L. KOCH로부터 정제된 항균 물질 Centipedin의 항균 Spectrum 및 작용 Mechanism 연구)

  • A broad spectrum of antibiotic action was studied with Centipedin purified from centipede Scolopendra subspinipes multilans L. Koch aganist gram-positive, gram-negative bacteria and fungi. The minimal inhibitory concentrations(MICs) were determined in liquid medium. The significant antibiotic activity was obtained aganist gram-negative Klebsiella pneumoniae ATCC 8308 responsible for causing infection at lung and intestine. The MIC value against Klebsiella pneumoniae ATCC 8308 was $2{\mu}g/ml$, and this Centipedin was active against Proteus vulgaris NRRL B-123. In addition, it has been shown that Centipedin blocks procaryotic RNA transcription and a little of DNA replication system in vitro. Centipedin did not exhibit any significant cytotoxicity against animal cells such as human blood leukemia (HL-60) and mouse B lymphocyte myeloma cell.

A Study for Antimicrobial Susceptibility of Wetlands to Eliminate Toilet Bacteria

  • Purpose: The purpose of this study is to investigate whether wetland has antimicrobial activity on pathogenic bacteria in the toilet bowl. Research design, data and methodology: Air-dried mud obtained from "Jilmoe Bog" wetland was packed and dissolved in the autoclaved saline. Antimicrobial susceptibility was assessed against three Gram-negative bacteria using disk diffusion method and broth dilution method. Identification of specific bacterium presented in wetland supernatant was performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Results: Incubation of three Gram-negative bacteria with wetland supernatant inhibited bacterial growth of the bacteria, otherwise increased prevalence of specific bacterium. It was confirmed that Pseudomonas putida was presented in wetland supernatant. Conclusions: The results presented in this study might provide the possibility to utilize wetland supernatant as a bioremediation of toilet bowl bacteria.

  • The presence of slime in paper mills is practically universal. Many researches have been performed for many years to resolve the problem caused by the slime in pulp and paper mill. Many papers have been published to show the bacteria is a major cause of paper mill slime. Now that the recycling of the water has been increased and the regulations of a toxic chemical dosage have become more strengthen, the importance of the control of slime in pulp and paper mill recently has been more recognized. Therefore, to produce quality products at the lowest economic and environmental costs, a through study of the microbial ecology and the indentification of troublesome slime-forming bacteria is a quite necessary. The purpose of this paper is to indentify slime~forming bacteria isolated from the papermaking process. The samples were taken from four parts of making fine paper : machine chest, head box, wire part, white water tank. Machine chest showed the most numbers of bacteria, numbering $2.55{\times}10^7$. The different colony types were taken from the 105 dilution plate. Nine bacteria were identified u sing the Biolog system and the vitek system: 6 gram-negative bacteria, 3 gram-positive bacteria. They are Pseudomonas paucimobilis B., Staphylococcus sp., Acinetobacter calcoaceticus., Pseudomonas cepacia, Actinobaci1lus capsulatus, Acidovorax sp., Flavobacterium sp., and Staphylococcus auricularis in addition to one unidentified sp., Among them. Pseudomonas paucimobillis was found in all places where the samples were taken. And, each parts had the different predominant bacteria in it : Pseudomonas paucimobilis B. in machine chest, Acinetobactor calcoaceticus. in Wire Part and Staphylococcus sp. in head box.

Expression and Antibacterial Activity of a Bombus ignitus Apidaecin in Baculovirus-Infected Insect Cells

  • The apidaecins are highly active against Gram-negative bacteria. Here, we show the expression and antibacterial activity of the bumblebee, Bombus ignitus, apidaecin. We PCR-amplified 51 bp of the active domain sequence of the B. ignitus apidaecin gene and expressed the recombinant B. ignitus apidaecin active domain in baculovirus-infected insect cells. The recombinant B. ignitus apidaecin active domain shows bactericidal activity against Gram-negative bacteria, including Pseudomonas tolaasii, a serious pathogen in cultivated mushrooms, but not Gram-positive bacteria. This result suggests that the active domain of the B. ignitus apidaecin is a potential antibacterial agent for the control of bacterial brown blotch diseases.

  • DA-1131은 gram positive bacteria와 Pseudomonas aeruginosa를 포함한 gram negative bacteria에 대하여 광범위한 항균 spectrum과 우수한 항균력을 나타내었다. 각종 임상분리균주에 대한 항균력 시험 결과, gram positive bacteria 중 methicillin-resistant Staphylococcus aureus(MRSA)에 대하여는 DA-1131이 가장 우수한 항균력을 나타내었으며, methicillin-susceptible S. aureus(MSSA)에 대하여는 MEPM, CPR 및 CAZ보다 약 2-50배의 우수한 항균력을 나타내었으나 IPM/CS보다는 동등이하의 항균력을 나타내었다. Gram negative bacteria에 대하여는 IPM/CS, CAZ 및 CPR보다 우수한 항균력을 나타내어 0.2 $\mu\textrm{g}$/$m\ell$ 이하의 농도에서 91%의 Serratia marcescens, 89%의 Proteus mirabilis, 76%의 Morganella morganii 및 시험에 사용된 Enterobacteriaceae에 속하는 전균주의 생육이 억제되었다. P. aeruginosa 에 대하여 DA-1131은 1.56 $\mu\textrm{g}$/$m\ell$ 이하의 농도로 시험균주 전체의 생육을 저해하였으며, MEPM의 약 2배, IPM/CS의 약 4배의 강한 항균력을 나타내었다. CAZ에 내성인 Enterobacteriaceae 임상분리균주에 대한 DA-1131의 항균력은 CAZ 감수성균주에 대한 항균력과 동일한 것으로 나타났다.

Antimicrobial Effects of Lonicera japonica against Gram Positive and Gram Negative Anaerobic Bacteria

  • It has been shown that the butanol extract of Lonicera japonica has antimicrobial and other potentially useful biological activities. The purpose of this study was to determine the in vitro activity of Lonicera japonica compared to other antimicrobial agents against anaerobic bacteria. Specifically, the in vitro activity of the butanol extract was investigated against 104 clinical isolates of anaerobic bacteria using an agar dilution method and the results were compared to erythromycin, cefoxitin, imipenem, clindamycin, and metronidazole. It was found that Lonicera japonica and imipenem were the most active antimicrobial agents tested.

Impacts of Soil Organic Matter on Microbial Community of Paddy Soils in Gyeongnam Province

  • Agricultural management of paddy soil depends on the effects of soil microbial activities. The present study evaluated the soil microbial community of 25 paddy soils in Gyeongnam Province by fatty acid methyl ester (FAME). The average of microbial communities in paddy soils were 32.2% of total bacteria, 16.7% of Gram-negative bacteria, 12.9% of Gram-positive bacteria, 2.0% of actinomycetes, 14.9% of fungi, and 1.3% of arbuscular mycorrhizal fungi. The communities of total bacteria (34.9%) and Gram-negative bacteria (19.4%) in soils with $30{\sim}35g\;kg^{-1}$ of organic matter were significantly larger than those in soils with other organic matter levels. However, soils with $20{\sim}30g\;kg^{-1}$ of organic matter had significantly low ratio of cy17:0 to $16:1{\omega}7c$ and cy19:0 to $18:1{\omega}7c$ as compared with soils with $30{\sim}35g\;kg^{-1}$ of organic matter, indicating microbial stress decreased (p < 0.05). In principal component analyses of soil microbial communities, Gram-negative bacteria should be considered as a potential responsible factor for the obvious microbial community differentiation that was observed between the two different organic matter levels in paddy fields. Thus, soils containing $20{\sim}30g\;kg^{-1}$ of organic matter were responsible for strong effect on microbial biomass and stress in paddy fields.

Prevalence of Multi-Antibiotic Resistant Bacteria Isolated from Children with Urinary Tract Infection from Baghdad, Iraq

  • Urinary tract infections (UTIs) are one of the most common infections in different age groups, including children. Bacteria are the main etiological agents of UTIs. The aim of the present study was to isolate, identify, and determine the antibiotic susceptibility of bacteria isolated from children with UTIs from Baghdad, Iraq. Three hundred and two urine samples were collected from children aged 6 months to 12 years. The samples were cultured on blood agar and MacConkey agar. The selected colonies were subjected to biochemical tests and antibiotic susceptibility analysis using the Vitek® 2 Compact automated microbial identification system. In this sample, 299 bacteria were identified, of which, 267 were gram-negative bacteria, and 32 were gram-positive bacteria. Escherichia coli (56%) was the most commonly isolated gram-negative bacteria, followed by Pseudomonas aeruginosa (14%), Enterobacter spp. (10.48%), Klebsiella pneumoniae (9.36%), Proteus spp. (7.8%), Acinetobacter baumannii (1.5%), and Morganella morganii (0.37%). Enterococcus faecalis (62.5%) was the most commonly detected gram-positive bacteria, followed by Staphylococcus aureus (37.5%). E. coli and P. aeruginosa were the most antibiotic-resistant bacteria. Among the tested antibiotics, meropenem showed 100% sensitivity, followed by imipenem (97.4%), amikacin (91.8%), and tobramycin (83.5%). In contrast, the high frequencies of resistance were observed with cefixime (93.2%), cefotaxime (78.7%), and ceftriaxone/cefotaxime (71.2%). In conclusion, carbapenems and aminoglycosides are highly recommended for the empirical treatment of UTIs, while, Quinolones, penicillins, and cephalosporins are not suggested. Frequent antibiotics susceptibility testing are warranted to determine the resistance pattern of UTI bacteria.

한국산 도꼬마리 추출물의 항균효과 및 분리 정제

  • Antimicrobial activity of various extracts of Xanthium strumarium L. was tested against 25 strains of bacteria, yeast and fungus. The crude ethylacetate extract exhibited strong growth inhibition to the tested strains with the exception of partial Gram-negative bacteria. The property of antimicrobial compound was very stable under heat treatment at $120^{\circ}C$, but it was unstable in acid (pH 3.0) and alkali (pH 10.0) treatment. The antimicrobial compounds were purified by boiling water extraction, ethylacetate extraction, charcoal column chromatography, silica gel column chro- matography and reverse phase HPLC. The purified compound A and B were detected in a single peak (each above 98% purity) through the HPLC analysis. The compound A and B showed a strong growth inhibition against Gram-negative and positive bacteria in the agar diffusion method. When tested by the FDA method using the esterase, compound A mainly inhibited the growth of bacteria and compound B showed the growth inhibition of both bacteria and yeasts.

Role of Amino Acid Residues within the Disulfide Loop of Thanatin, a Potent Antibiotic Peptide

  • Thanatin, a 21-residue peptide, is an inducible insect peptide with a broad range of activity against bacteria and fungi. It has a C-terminal disulfide loop, like the frog skin secretion antimicrobial peptides of the brevinin family. In this study, we tried to find the effect of a number of amino acids between the disulfide bond. Thanatin showed stronger antibacterial activity to Gram negative bacteria than other mutants, except Th1; whereas, the mutant peptides with deletion had higher activity to Gram positive bacteria than thanatin. An increase in the number of amino acid(s) using the alanine residue decreased the antibacterial activity in all of the bacteria. Th1 with deletion of threonine at position 15 ($Thr^{15}$) showed similar antibacterial activity against Gram-negative bacteria, but had higher activity against the Gram positive bacteria. In order to study the structure-function relationship, we measured liposome disruption by the peptides and CD spectra of the peptides. Th1 also showed the highest liposome leaking activity and α-helical propensity in the sodium dodecyl sulfate solution, compared with other peptides. Liposome disruption activity was closely correlated with the anti-Gram positive bacterial activity. All of the peptides showed no hemolytic activity. Th1 was considered to be useful as an antimicrobial peptide with broad spectrum without toxicity.
