양수 내에 존재하는 총 단백질을 이차원 전기영동을 이용하여 분리 분석하였고, gradient gel을 이용하여 양수 내에 소량으로 존재하는 미세 단백질까지 분리하였다. 양수 내에는 고농도로 존재하는 단백질이 있는데 이것이 serum albumin precursor임을 확인하였고, 8-18% gradient gel의 이용으로 분해능(resolution)이 향상되어 미세 단백질을 분리 분석할 수 있었다. 이차원 전기영동 후 MALDI-TOF를 이용하여 단백질을 identification하여 기존의 양수 protein database에 존재하는 단백질을 확인하였고, 존재하지 않는 새로운 단백질을 분리 분석하였다.
The microbial community during composting of gargage was analyzed using 16S rDNA PCR - DGCE (denaturing gradient gel electrophoresis). Pseudomonas spp. was found throughout the process, and thermophilic Bacillus spp. was dominated at the thermophilic stage. Six thermophilic bacteria were isolated and identified as B. caldoxylolyticus, B. thermoalkalophilus, and B. thermodenitrificans.
To detect the mutation in a given sequence, there are variety of methods developed by use of the gel electrophoresis. One of the methods, TGGE (Temperature Gradient Gel Electrophoresis), is a popular technique because it can detect mutations in DNA fragment with ease and at low cost. This study used 200 bp BamHI-digested DNA fragment containing the human $\varepsilon$-globin promoter which was mutated[$\varepsilon$ F1*(-141), GATA- I*(-163), and GATA-1* & $\varepsilon$F1]. This BamHI-digested DNA fragment was directly used to detect the mutated DNA fragment on 50% denaturant gel with temperature gradient of 45$^{\circ}C$ through $53^{\circ}C$. In agreement with the theoretical result of MELTSCAN program (Brossette and Wallet, 1994) the mobilities of mutated DNA fragments were shown to be nearly distinguished on the temperature gradient gel. In contrast to the above result the heteroduplex analysis under the temperature gradient condition was shown to detect the mutated DNA fragments through the heteroduplex formation between strands of mutated DNA and wild-type DNA.
현재 토양 생태에서 토양미생물은 유기물 분해, 질소 순환, 식물의 질소 이용 등 중요한 역할을 하고 있어, 토양 내 미생물 다양성을 분석하기 위한 연구는 지속적으로 진행되어 오고 있다. 본 연구에서는 논 토양의 미생물 생태 다양성을 조사하기 위한 효과적인 방법으로 denaturing gradient gel electrophoresis (DGGE)를 적용하고자 본 연구를 수행하였다. 논 토양 미생물의 DNA를 분리하기 위하여 lysis buffer method, skim milk bead method, sodium phosphate buffer method, Epicentre SoilMaster DNA extraction kit (Epicentre, USA), Mo Bio PowerSoil kit (Mo Bio, USA)를 이용하여 토양 내 gDNA 최적 추출방법을 확인하였다. 그 결과 Mo Bio PowerSoil kit를 사용하였을 때 Shannon 다양성지수가 세균 3.3870, 진균 3.6254으로 미생물 다양성 분석시에 가장 효과적이었다. DGGE 분석을 위한 조건은 세균의 경우 6% polyacylamide gel, 45-60% denaturing gradient였고, 진균의 경우 6% polyacrylamide gel, 45-80% denaturing gradient에서 최적 분석조건을 보였다. 위의 분석법을 적용하여 논 토양내의 미생물 군집의 변화를 살펴보면 시간의 변화 요인에 의해 미생물 변화가 일어나는 것을 알 수 있었다. 본 연구에서 사용된 DGGE 분석법을 통해 논토양 미생물의 분석 가능성을 제시 할 수 있었다.
본 실험은 보리품종의 구분에 가장 적합한 전기영동법과 효소를 구명하고자 6개 보리품종(알찬보리, 수원 216호, 조풍보리, 사천 6호, 향맥, 히노데하다가)의 종실 단백질을 7.5% polyacrylamide slab gel, 2-30% polyacrylamide porosity gradient tube gel, isoelectricfocusing(pH 4-9)과 starch gel을 사용 전기영동 후 protein band pattern과 esterase, acid phosphatase, malate dehydrogenase, glutamate dehydrogenase 및 leucine aminopeptidase의 동위효소 pattern을 관찰하였던 결과는 다음과 같았다. 1. 7.5% polyacrylamide slab gel에서 단백질 pattern과 2-30% polyacrylamide porosity gradient tube gel에서의 단백질과 esterase의 band pattern이 품종간 뚜렷한 차이를 보여 보리품종구분에 가장 적합하였다. 2. Acid phosphatase, malate dehydrogenase, glutamate dehydrogenase 및 leucine aminopeptidase 등의 동위효소 pattern은 모든 실험한 품종이 동일한 형태를 보여 품종구분에 이용하기에는 적합하지 않았다.
These experiments were carried out to develop new techniques for in vitro separation of x-and Y-bearing spermatozoa. The results obtained in these experiments were summarized as follows: 1. Following centrifugation of discontinuous percoll density gradient, populatin of spermatozoa increased progressively from low to high density. The highest concentration of spermatozoa was observed at the 4th fraction which included 36.6% of spermatozoa. 2. As increasing percoll concentration, the higher motility index was obtained and the highest motility index(74.2) was obtained at the 5th fraction. 3. The percentage of B-body bearing spermatozoa following percoll density gradient centrifugation was decreased from 39.7% to 25.6%. 4. The sperm population following chromatography by sephadex gel and percoll density gradient centrifugation was decreased in 1st, 5th and 6th fractions but the reverse was turn for 2nd, 3rd and 7th fractions, and the highest sperm concentration was observed at the 7th fraction which included 37.4% of spermatozoa. 5. Motility index of spermatozoa was increased from 77.6 to 79.4 after the sephadex gel filtration, however it was decreased at all fractions after percoll density gradient centrifugation. The lowest motility index(33.2) was obtained from the 7th fraction. 6. The rate of B-body bearing spermatozoa was shown the trend to decrease by the sephadex gel filtration and the trend was accelerated by the percoll density gradient centrifugation. The lowest percentage of B-body bearing spermatozoa, 12.0% was obtained from the 5th fraction.
Vaginal lactic acid-producing bacteria of 80 pre-menopausal women were studied by isolation on Blood and DeMan-Rogosa-Sharpe agar, PCR with group-specific primers for Lactobacillus-denaturing gradient gel electrophoresis (DGGE), and PCR with specific primers for V3 region in 16S rRNA-temporal temperature gel electrophoresis (TTGE). Conventional isolation method on media detected only one lactobacillus (Lactobacillus brevis) while TTGE detected only Lactobacillus sp. DGGE detected seven Lactobacillus species; L. coleohominis, L. crispatus, L. iners, L. reuteri, L. rhamnosus, L. vaginalis, and Leuconostoc lactis. L. acidophilus and L. gasseri, which are prevalent in Western women, were not detected in Korean women. Furthermore, L. rhamnosus, Leuc. lactis, L. coleohominis, and Weissella cibaria, which were not previously reported in the vaginal microbiota of Korean women, were detected. The five most prevalent LABs in vaginal microbiota in Korean women were L. iners, Enterococcus faecalis, L. crispatus, Leuc. lactis, and W. cibaria.
A radioprotective ginseng protein fraction was obtained from Korean white ginseng powder by the following isolation and purification procedures: Tris-HCI buffer extraction, 70% ammonium sulfate fractionation, CM-rellulosr column chromatography, heat inactivation and Sephadex G-75 column chromatography. This fraction was further purified by Sepharose 4B and Sephadex G-150 column chromatographies. Three fractions obtained were subjected to Native-PAGE and SDS-PAGE using gradient gels and the silver staining method. Molecular weights of the native proteins and their subunits were estimated.
Saccharomyces cerevisiae로 부터 glucose-6-phosphate dehydrogenase을 신속하고 간편하게 정제하는 과정을 affinity chromatography를 이용하여 개발하였다. 이 효소를 정제하는데 적절한 affinity medium을 조사해 본 결과, $NADP^+ -agarose$와 Affi-gel Blue(Cibacron Blue F3GA)가 Affi-gel Red(Procion Red HE-3B), AMP-agarose, ATP-agarose, 그리고 $NADP^+ -agarose$보다 유용함이 밝혀졌다. 이 두가지 affinity media에 흡착된 효소를 분리 하는데 가장 적합한 elution 조건을 조사하였는데 KCI gradient( (0-1.OM)가 효소의 순도 및 수회율을 가장 높일 수 있는 적합한 방법이었다. 특히 Affi-gel Blue를 사용할 경우, KCI gradient로 효소를 용출시키기 전에 NAD-(15mM)로NAD+에 친화역을 갖는 효소들을 제거하는 것이 enzyme의 순도를 높이는데 매우 효과적이었다. 그 결과 glucose-6-phosphate dehydrogenase를 baker's yeast로 부터 기존의 간단한 정제 과정과 affinity chromatography를 병행한 방식을 샤용하여 분리 하였는데, affinity medium으로 Affi-gel Blue를 사용했을 때는 180배 정도, NADP+-agarose를 사용했을 때는 2,000배 정도로 정제 되었다. 대량으로 glucose-6-phosphate dehydrogenase를 정제하는 경우, Affi-gel Blue를 사용하던 효소의 순도는 NADP+-agarose보다 낮으나, 효소의 회수율은 훨씬 더 높았다. 또한 G-6-P dehydrogenase에 대한 affinity medium의 capacity도 Affi-gel Blue가 NADP+-agarose보다 5배정도 높았으며 더우기 Affi-gel Blue는 여러번 반복적으로 사용될 수 있고, 그 제조 과정도 NADP+-agarose보다 간단하며 경비도 적게 들었다.
바이로이드 RNA 분자를 포함한 저분자량의 식물성 RNA 분자에 대해서 0M부터 8M 까지의 요소구배를 가진 폴리아크릴아미드켈을 이용하여 전기영동적 이동성을 조사하였다. 막대모양의 바이로이드 RNA 분자는 요소구배 겔에서 전반적으로 다른 저분량의 식물성 RNA들과 마찬가지로 요소구배에 수직인 방향으로 비슷한 수준의 전기영동적 이동성을 보여주었다. 이와 달리 고리모양의 바이로이드 RNA 분자는 요소구배 겔에서 고농도의 요소구배 부분에서 보다도 저 농도의 요소구배 부분에서 더 빠르게 이동하였다. 따라서 고리모양의 바이로이드 RNA 분자는 요소구배 겔의 중간부분에서 막대모양의 바이로이드 분자의 밴드를 가로 지르는 기울기를 가진 특이한 전기영동적 이동을 나타내었다. 결과적으로 고리모양의 바이로이드 RNA 분자의 전기영동적 이동성은 0M 8M의 요소구배를 가지는 바이로이드 RNA 분자의 변성겔에서 주로 요소의 농도에 따라서 영향을 받는 것으로 보인다.
