Interaction of the Bacteriophage P2 Tin Protein and Bacteriophage T4 gp32 Protein Inhibites Growth of Bacteriophage T4

  • The growth of baceriophage T4 is inhibited by the presence of the tin gene product o bacteriophage P2. The interaction between purified Tin and gp32 proteins was observed using coimmunoprecipitation experiments. The in vivo interaction was confirmed by yeast two-hybrid experiments. A deletion analysis showed that the Asp 163 region of gp32 to DNA substrates was not affected by the presence of Tin, Thus, it would appear that the inhibition of 4 growth by Tin was due to a protein-protein interaction rather than affecting the DNA-binding ability of gp32.

Production and Characterization of Extracellular Polysaccharide Produced by Pseudomonas sp. GP32 (Pseudomonas sp. GP32에 의해 생산된 세포 외 다당류의 생산 및 특성)

  • A strain GP32 which produces a highly viscous extracellular polysaccharide was conducted with soil samples and identified as Pseudomonas species. The culture flask conditions for the production of extracellular polysaccharide by Pseudomonas sp. GP32 were investigated. The most suitable carbon and nitrogen source for extracellular polysaccharide production were galactose and (NH4)2SO4. The optimum carbon/nitrogen ratio for the production of extracellular polysaccharide was around 50. The optimum pH and temperature for extracellular polysaccharide production was 7.5 and 32℃, respectively. In batch fermentation using a jar fermentor, the highest extracellular polysaccharide content (15.7 g/l) was obtained after 70 hr of cultivation. The extracellular polysaccharide produced by Pseudomonas sp. GP32 (designated Biopol32) was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography. Biopol32, which has an estimated molecular weight of over 3×107 datons, is a novel polysaccharide derived from sugar components consisting of galactose, glucose, gulcouronic acid and galactouronic acid in an approximate molar ratio of 1.85 : 3.24 : 1.00 : 1.42. The solution of Biopol32 showed non-Newtonian characteristics. The viscosity of Biopol32 exhibited appeared to be higher at all concentration compared to that of zooglan from Zoogloea ramigera. An analysis of the flocculating efficiency of Biopol32 in industry wastewater (food, textile, and paper wastewater) revealed chemical oxygen demand (COD) reduction rates 58.4-67.3% and suspended solid (SS) removal rates 82.6-91.3%. Based on these results, Biopol32 is a possible candidate for industrial applications such as wastewater treatment.

  • To find the interacting protein with the cytoplasmic domain of HIV-1 gp41, the yeast two hybrid system was used for the expression cloning. Among the $1.4 \times 10^6 colonies, 20 colonies were selected as the final candidate for the interacting protein gene. The nucleotide sequencing revealed three kinds of protein, acidic ribosomal protein P0, beta tubulin, alpha catenin. These proteins interacted with the gp41 specifically in yeast system.

Flocculating Properties of Bioflocculant Biopol32 from Pseudomonas sp. GP32 (Pseudomonas sp. GP32가 생산하는 생물고분자응집제 Biopol32의 응집특성)

  • The flocculating properties of bioflocculant Biopol32 produced by Pseudomonas sp. GP32 were investigated for application in industrial wastewater treatment. The major flocculating substance of bioflocculant Biopol32 was identified as polysaccharide. Many anionic flocculants need a counter ion to induce higher flocculating activity. The flocculating activity of bioflocculant Biopol32 was markedly increased by the addition of cationic ions ($Ca^{2+}$, $Al^{3+}$). The flocculating activity of bioflocculant Biopol32 was the most effective when 7.0 mM $CaCl_2{\cdot}2H_2O$ as coflocculant was added. The flocculating activity on the effect of pH and the temperature of the bioflocculant Biopol32 was compared with anionic commercial flocculant (polyacrylamide) and bioflocculant (zooglan from Zoogloea ramigera). In kaolin suspension, the highest flocculating activity was obtained at the bioflocculant Biopol32 concentration of 1.5 mg/l. A high flocculating activity was observed in the pH range of 5.0 to 8.0. The flocculating activity of bioflocculant Biopol32 was sustained up to $60^{\circ}C$, but decreased rapidly at over $70^{\circ}C$. In the batch culture, the charge density of bioflocculant Biopol32 was compared with flocculating activity. The larger the anionic charge density and apparent viscosity of bioflocculant Biopol32, the higher the flocculating activity. Therefore, we confirmed that the flocculating activity and apparent viscosity of bioflocculant Biopol32 was closely related to the charge density of bioflocculant Biopol32.

Effect of Starch Degradation Enzymes on the Retrogradation of a Korean Rice Cakes (떡노화에 대한 전분분해효소류의 효과)

  • In this study, enzymes were investigated as an antistaling agent for a Korean rice cake. Thermograms by a DSC demonstrated that the gelatinization-onset temperature of the Korean rice cake was at its lowest temperature of 71.1$^{\circ}C$ with the GP (glucoamylase+pullulanase) treatment, followed by $\beta$-amylase and $\alpha$-amylase. The gelatinization peak temperature of the Korean rice cake with enzyme treatment was relatively lower compared to the control. Furthermore, the Korean rice cake with GP treatment showed the lowest peak temperature. Melting enthalpy of the Korean rice cake increased with the enzyme treatment, with $\beta$-amylase, followed by $\alpha$-amylase and GP. Melting enthalpy of the Korean rice cake with GP treatment was significantly lower compared to the $\beta$- and $\alpha$-amylase treatment. Recrystallinity in the case of GP treatment was also significantly lower than control. The range of Avrami exponent (n) was 0.90 ∼ 1.20 and the time constant of retrogradation (1/k) of the Korean rice cake crystalline decreased in the following order: GP, $\beta$-, $\alpha$ -amylase and control. Textural characteristics of the Korean rice cake with enzyme treatment differed greatly from that of control. The L* values of all the Korean rice cakes made without $\beta$-amylase decreased and the a* values were significantly different at p<0.05. The GP treatment altered the b* value toward blue color, whereas $\beta$-and $\alpha$-amylase changed to the direction to yellow color. In sensory evaluation, the Korean rice cake with enzyme treatment showed higher evaluation compared to control.

Petrological characteristics on stone resources of granites in the Pocheon-Euijeongbu area (포천-의정부지역 화강암류 석재자원의 암석학적 특성연구)

  • The Jurassic granites, commercially called Yangu stone in the Pocheon-Euijeongbu area, have generally compact and coarse-grained textures, which could be classified into two types; grey granite(Gg) and light pink granite(Gp). Specific gravity, absorption ratio and prosity of Gg and Gp in physical property are 2.64 and 2.61, 0.32 % and 0.44 %, 0.86% and 1.13 %, respectively. These higher values of two latters of Gp than those of Gg are due to the more abundant microcracks in Gp. Compressive strength og Gg than those of Gg are due to more abundant microcracks in Gp. Compressive strength og Gg and Gp are 1,726 kg/cm2 and 1,717 kg/cm, respectively and bestrength has a positive proportion with Qz+Af+Pl(quartz+alkali feldspar+plagioclase) modes without trending with Bt+Ac(biotite+accessories). Tensile strength has the positive proportions with Qz+Af+Pl and Bt+Ac. While Bt+Ac has a negative trend with abrasive hardness, Qz+A+Pl shows a positive one. These may suggest Qz+Af+Pl mainly affects on strenghts potentional dimension stone than Gp.

  • The carrier phase measurements from GPS and GLONASS have different characteristics and therefore, have to be processed with different methods to provide precise positions or attitudes. In this contribution, at first, a measurement model is derived which can be used to not only GLONASS only applications but also both GPS and GLONASS applications. And then an error analysis of the proposed method performed using the derived model to derive analytic relationships between GDOP, PDOP and RGDOP. Finally, an integer ambiguity resolution method which was used in GPS is expanded to GPS and GLONASS. The proposed results can be directly applied to the design and analysis of GLONASS receiver and application programs. Furthermore, it is expected that the suggested method can also be effectively applied to combine the characteristically different measurements from the future satellite navigation systems such as GPS modernization, Galileo and QZSS.

Identification of pollutant sources using water quality and stable isotope ratios of inflow tributaries in the lower reaches of the Han-River

  • Despite the expansion of sewage treatment facilities to reduce pollutants in the tributaries of the Han River, water pollution accidents such as fish deaths continue to frequently occur. The purpose of this study was to identify the pollutant sources using water quality and stable isotope ratio (${\delta}^{15}N$, ${\delta}^{13}C$, ${\delta}^{15}N-NH_4$, ${\delta}^{15}N-NO_3$) analysis results in the three inflow tributaries (Gulpocheon (GP), Anyangcheon (AY) and Sincheon (SC)) of the Han River. Water quality was analyzed in June and October from 2013 to 2017, and the results showed that the concentrations of nutrients, such as T-N, $NO_3-N$, and T-P, were increased at GP4, AY3, SC3, and SC4, which lie downstream of sewage treatment facilities. The results of ${\delta}^{15}N$ for June 2017 indicated that the source of nitrogen was sewage or livestock excreta at GP4 and SC4, and organic fertilizers at AY3 and SC3. ${\delta}^{15}N-NO_3$ results suggested that the source of nitrogen was related to organic sewage, livestock or manure at GP4, AY3 and SC4. Therefore, GP4 and SC4 were more influenced by effluent from sewage treatment facilities than by their tributaries, AY3 and SC3 were considered to be influenced more by their tributary than effluent from sewage treatment facilities. With the results of this study, the source of contamination (sewage treatment facility effluent) of river inflow downstream of Han River could be confirmed using water quality and stable isotope ratio.
