Histological Studies of Gonad in the Hybrid Species Cobitis. sinensis-longicorpus Complex(Pisces, Cobitidae) (잡종기원의 Cobitis sinensis-longicorpus complex(Pisces, Cobitidae)에 대한 생식소의 조직학적 연구)

  • Cobitis sinensis-longicorpus complex considered as hybrid origin between C. sinensis and C. longicorpus coccurred ommonly in the upper streams of the Nakdong River, Korea. Histological examinations of their gonad were accompanied with 272 individuals of C. sinensis-longicorpus complex collected. Most of fishes collected were females, however, only 6 individuals were found males. The ovarian tissues of females are completely fertile undergoing normal oogenensis. In the male gonads, testicular lobule structure with abnormal vacuolar tissues were observed. Spermatogonia and spermatocytes were also observed of their testis however spermatids or sperms were not shown in their developmental stages. From these facts, we infer that female population of C. sinensis-longicorpus complex may be unique reproductive hierarchy accomplishing their reproduction with participation of males of their closely related bisexual species.

Risk Assessment of Nonylphenol using Sex Ratio, Sexual Maturation, Intersex and Lipofuscin Accumulation of the Equilateral Venus Gomphina veneriformis (Bivalvia: Veneridae) (대복 Gomphina veneriformis의 성비, 성 성숙, intersex 및 지방갈색소 침적을 이용한 nonylphenol의 위해성 평가)

  • Nonylphenol (NP) is an estrogen-mimicking compound or xenoestrogen. This study investigated the effects of nonylphenol on the reproductive status of the equilateral venus Gomphina veneriformis. The experiment lasted 24 weeks, Experimental groups consisted of a control and three nonylphenol exposures ($1.0,\;2.5,\;and\;5.0\;{\mu}g\;NP/L$). Mortality did not differ significantly between the control and the exposure groups. The sex ratio (F:M) was 1:1 in nature and 1:1.03 in the control group. However, it changed to 1:3.5 with $5.0\;{\mu}g\;NP/L$ exposure. Gonad maturity in females was higher in the nonylphenol exposure groups than in the control group. By contrast, in males, it was lower in the nonylphenol exposure groups. Intersex individuals constituted 0% in nature, 3.08% in the control group, and 23.6% in the group exposed to nonylphenol, with female characteristics more prevalent than male. As the concentration of nonylphenol increased, the accumulation of lipofuscin increased in the mid-gut gland.

Study of Feritilization and Developmental Rates by Hypoxia condition in Strongylocentrotue nudus (Strongylocentrotue nudus 성게에서 빈산소에 의한 배 발생률 및 수정률에 관한 연구)

  • 본 연구는 빈산소에 의한 둥근 성게 (Strongylocentrotue nudus) 수정란의 수정률 및 발생률에 관한 연구이다. 대조군 (normoxia)과 실험군 (hypoxia) 으로 나누어 수정률과 발생률의 변화를 관찰 하였으며 또한 gonad 세포의 유전자 발현의 차이를 봄으로써 스트레스 관련 유전자와 항산화 관련 유전자의 변화를 확인 할 수 있었다. 결과적으로 수정률에서는 큰 차이를 확인 할 수 없었던데 반해 발생률에 있어서 빈산소의 경우 전혀 발생이 진행되지 않는 것을 확인 할 수 있었으며 또한 빈산소에 노출된 gonad 세포의 경우도 스트레스 또는 항산화 유전자가 많이 발현 되는 것을 관찰 할 수 있었다. 앞의 실험을 토대로 빈산소 환경에서 유전자 발현량의 차이를 더욱더 수행함으로써 빈산소 상태에 따른 죽음의 바다의 증가 얼마나 위험한 것인지 더욱더 관찰 할 수 있을 것이다.

Reproductive Cycle and First Sexual Maturity of Sinonovacula constricta(Lamarck, 1818)(Bivalvia: Pharidae) in Western Korea

  • The gonad index, reproductive cycle and first sexual maturity of Sinonovacula constricta collected from Simpo, Kimje-gun, Korea were investigated by histological analysis. The gonad index(GI) in both sexes of S. constricta increased from April and reached a maximum in July when the water temperature rapidly increased. And then, the GI values gradually decreased by spawning from August through October. Monthly variations in the GI showed a close relationship with ovarian development. The reproductive cycle in females and males can be classified into five successive stages: early active stage(March to June), late active stage(May to July), ripe stage(July to September), partially spawned stage(August to October), spent/inactive stage(October to March). The percentage of first sexual maturations in female and male clams of 50.1-60.0 mm in shell length was over 50%, and for clams over 70.1 mm in shell length, it was 100%. Because harvesting clams < 50.1 mm in shell length could potentially cause a drastic reduction in recruitment, a measure including a prohibitory fishing size should be taken for adequate improved fisheries resource management.

  • A differential diagnosis between the true hermaphroditism (TH) and mixed gonadal dysgenesis (MGD) has important clinical implications for gender assignment and the decision for early gonadectomy; however, variable clinical and histological features frequently lead to the confusion of TH with MGD. A 17-month-old boy was presented with proximal hypospadias with chordee and right non-palpable testis in his scrotum. He also had right auricular anomaly including a separated tragus with skin tag. Left testis was well palpable in his left scrotum. Diagnostic right inguinal exploration showed Mullerian structures such as a gonad like an ovary and a fallopian tube with a uterus, which were removed. Repair of hypospadias and right auricular anomaly was also done. Following ultrasonography (USG) showed a normal looking testis in left scrotum. His chromosome was 45, XO/46, XY. We report a difficult case of mixed gonadal dysgenesis mimicking true hermaphroditism which combines ipsilateral congenital auricular anomaly.

  • Sexual maturation and gametogenic cycle of the hard shelled mussel, Mytilus coruscus were investigated by means of histological method. Specimens were collected monthly in Hansan Bay of south coast, Korea from January to December 2002. Sex of the clam was dioecious. The gonads are composed of a number of gametogenic follicles. The gonad index (GI) was reached the maximum in February (3.40), and the minimum in August (1.18). The monthly variation of condition index (CI) coincides with gonad index. The main spawning occurs between February and March. The gametogenic cycle of the clam could be divided into six stages: resting (August to November), multiplicative (November to December), growing (December to January), maturation (January to February), partially spent (February to April) and completely spent/degenerative stage (April to July). Sex ratio (♀/♂) was 1:0.96.

Reproductive Biology of the Pen Shell, Atrina(Servatrina) pectinata on the Boryeong Coastal Waters of Korea

  • The gonad index, condition index, reproductive cycle and spawning of the pen shell Atrina (Servatrina) pectinata were investigated using samples from the subtidal zone of Nokdo on the Boryeong coastal waters of Korea. Samples were collected monthly by SCUBA divers for one year from January to December, 2001. A. (Servatrina) pectinata is dioecious and oviparous. The spawning season of this species occurred once a year from June to August, with the main spawning occurring between June and July when the seawater temperature was around $20^{\circ}C$. Ripe oocytes were about 60-65 ${\mu}m$ in diameter. The reproductive cycle of this species could be classified into five successive stages; early active stage (November to March), late active stage (February to May), ripe stage (April to July), partially spawned stage (June to August), and spent/inactive stage (August to October). Monthly changes in the gonad index reached a maximum (4.6) in May (ripe stage), thereafter, the GI values gradually decreased from June to August when spawning occurred continuously. Therefore, monthly changes in the GI values showed a similar pattern to the gonadal phase. The condition index (CI) of the meat part without the posterior adductor muscle reached the maximum in June (ripe and partially spawned stage) and the minimum in September (spent/inactive stage), Accordingly, monthly changes in the condition indice of the meat part without the posterior adductor muscle coincided with the gonadal phases.

Ultrastructure of Germ Cells during Spermatogenesis and the Reproductive Cycle in Male Meretrix petechialis on the West Coast of Korea

  • Gonad index, spermatogenesis and the reproductive cycle of Meretrix petechialis were investigated by cytological, histological observations. Monthly changes in the gonad index coincides the gonadal development. The morphology of the spermatozoon had a primitive type and is similar to that of other bivalves having a short mid-piece with five to six mitochondria surrounding the centrioles. The morphology of the sperm nucleus type and the acrosome shape of this species were cylindrical type and cap-like shape, respectively. The spermatozoon was approximately 40-45 ${\mu}m$ in length including the sperm nucleus length (about 1.50 ${\mu}m$), acrosome length (0.60 ${\mu}m$) and tail flagellum. The axoneme of the tail flagellum consisted of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail showed 9 + 2 microtubular arrangement. The spawning period was from June to September and the main spawning occurred from July to August when seawater temperatures were higher than $20^{\circ}C$. The reproductive cycle of this species could be categorized into five successive stages: early active stage (February to March), late active stage (February to May), ripe stage (April to July), partially spawned stage (June to September), and spent/inactive stage (September to February).

Characterization of Fetal Gonad-Derived Cells by Stem Cell Markers (줄기세포 Marker를 이용한 돼지 태아 생식선 유래 세포의 특성화)

  • In mammals, male and female germline stem cells are derived from primodial germ cells. Despite many efforts to identify stem cells from gonads, there has been little successe to identify germline stem cells yet. In this study, we isolate and characterized porcine germline stem cells using only stem cell markers that are prevalently expressed in various tissues. Gonadal cells derived from both male and female formed colonies and showed AP activities and different lectin binding properties. Pluripotency of germline stem cells was also identified by positive signals against putative stem cells markers such as SSEA-1 and SSEA-3. In addition, nestin was also found in primary gonad cells that have a similar morphology to the AP-positive cells. The nestin expression suggests that the germline stem cells may have similar expression of the prevalent stem cell markers found in other tissues. The demonstration of nestin expression together with pluripotent cell markers calls further investigation of the possible differentiation of nestin-positive cells into neurons.

  • The suitability of any fish species for successful aquaculture requires basic information on its reproduction and growth. This study investigated some facets of the reproductive biology of Synodontis shall (Mochokidae) from River Siluko in Nigeria. Fish samples were collected forth-nightly for a duration of fourteen (14) months-March 2015 to April, 2016 with the assistance of artisanal fishermen. Fishes were identified using taxonomic guides and standard techniques were used for determination of sex ratio, gonad maturation and fecundity. Linear regression method was used to define the correlation between fecundity and fish length, body weight and ovary weight. Results showed that sex ratio did not indicate a significant divergence (p > 0.05) from the 1 male to 1 female distribution ratio (1:1.41). Gonad morphology revealed paired gonads. Testes and ovaries were classified into four maturity stages: immature, resting, ripening and ripe. Gonadosomatic index ranged from 0.04 to 5.68 (males) and 0.03 to 20.19 (females). Absolute fecundity ranged from 1,014 to 4,520 eggs (mean = 2,592 eggs) and did not correlate significantly (p > 0.05) to ovary weight. This study has contributed to existing data on the biology of freshwater fish species in Nigeria and provided valuable information for fishery management tools in the conservation and utilization of this valuable freshwater fish species.
