The liver is vulnerable to alcohol-related injury because it is the primary site of alcohol metabolism. Additionally, a number of potentially dangerous by-products are generated as alcohol is broken down in the liver. However, dietary supplements may prevent or relieve some of alcohol's deleterious effects. Therefore, this study was conducted to evaluate the prophylactic effect of aqueous extract of Sesamum indicum (SI) on ethanol induced toxicity in rats. Male Wistar albino rats were divided into control, ethanol, pre-treatment, simultaneous and post-treatment groups. In the prophylactic experiment, Sesamum indicum, (200 mg/kg body weight) was administered by oral gavage for 28 days; two hours before, simultaneously with or two hours after ethanol exposure. Toxicity was induced by administering 45% ethanol (4.8 g/kg bw) by oral gavage. Lipid peroxidation (TBARS) and reduced glutathione (GSH) levels and catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and gluthathione-S-transferase (GST) activities were then determined in the liver, serum triglyceride (TG) levels, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were monitored and histological examination was carried out. The results revealed that ethanol administration led to significant elevation of TBARS level while depleting in the level of GSH as well as CAT, GPx, SOD and GST activities. Similarly, TG level and ALT and AST activities were elevated. The SI pre-treated group significantly inhibited TBARS, restored GSH level, enhanced CAT, GPx, SOD and GST activities and significantly decreased the elevated level of serum TG, ALT and AST activities. SI treatment (simultaneously with ethanol) exhibited similar effects to those of the SI pre-treated groups, while the SI post-treated group did not show the same protection as the Pre-treated group. S. indicum possesses antioxidant and hepatoprotective properties, that eliminate the deleterious effects of toxic metabolites of ethanol.

This study was designed to investigate the effects of methionine(Met) on the activities of brain lipid peroxidation related enzymes in ethanol administrated rats of selenium(Se) deficiency. Male Sprague-Dawley rats were fed Se deficiency diets containing one of the three levels of Met (0, 3, 9g/kg diet) and ethanol(2.5g/kg of body weight) was administrated as 25v/v% ethanol treated groups orally. The rats sacrificed after 5 and 10 weeks of feeding periods. Alcohol dehydrogenase activity was increased in ethanol treated groups and was higher Met normal group than Met deficiency and excessive groups at 5 and 10 weeks dieting. Aldehyde dehydrogenase activity was decreased in ethanol treated groups and significantly decreased in Met deficiency group. Monoamine oxidase activity in brain was increased in ethanol treated groups and was predominently increased in Met deficiency groups. Superoxide dismutase and glutathione peroxidase activities were decreased in ethanol treated groups and tended to increase in proportion to level of dietary methionine. Glutathione S-transferase and catalase activities and lipid peroxide content were increased by ethanol administration and were higher Met deficiency group than normal and excessive groups.

This study was conducted to investigate the effects of dandelion leaf (Taraxacum officinale) extracts on hepatic antioxidative system in high cholesterol-fed rats. Four groups of rats were given high cholesterol diets containing 10 g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received a diet without dandelion leaf extract and the other three groups received dandelion leaf extracts, ie, water, ethyl acetate and ether extracts, respectively. There were no significant difference in cytochrome P-450 contents among four groups. Hepatic xanthine oxidase activity was significantly lower in water extract group than the other three groups. Superoxide dismutase activity was significantly lower in three dandelion leaf extract groups, but catalase activity was significantly higher in three dandelion leaf extract groups than control group. Glutathione peroxidase and glutathione S-transferase activities were significantly increased in water extract group than control group. Lipid peroxide content was decreased in water extract group than control group.

This study investigated the effect of Puerariae Flos (PF; flower of Puerariae plant) and Puerariae Radix (PR; root of Puerariae plant) water extracts on the activities on the activities of ethanol-metabolizing enzymes and free radical generating/scavenging enzymes of brain in ethanol-treated rats. Five groups of male Sprague-Dawley rats were orally administered ethanol (25%, v/v) 5 g/kg body weight/day, and sacrificed 5 weeks post treatment. PF and PR water extracts were supplemented in a diet based on 1.2g (I) or 2.4 g (II) raw PF or PR/kg body weight/day. Alcohol dehydrogenase activity of brain was significantly lowered in PF of PR groups, whereas aldehyde dehydrogenase activity was significantly higher in PR groups than those of control and PF groups. Cytochrome P-450 content, aminopyrine D-methylase and aniline hydroxylase activities were decreased in both PF and PR groups compared to control group. Aldehyde oxidase and xanthine oxidase activities tended to decrease by Puerariae plant extract supplemented goups and degree of decrease predominated in PRI. Superoxide dismutase and glutathione S-transferase activities were increased in PF or PR groups, whereas glutathione peroxidase and catalase activities were significantly decrased by Puerariae plant extracts supplement. These results indicated that supplementation of PF or PR lowers free radical generating enzymes activities. It was suggested that the activities of ethanol metabolizing emzymes and antioxidant enzymes in brain can be enhanced by PF or PR supplement in ethanol-treated rats.
A total of twenty, 2-wk-old male broiler chickens were allotted into control diet(CON) or a diet supplemented with 1% ground grape seed(GGS). They had free access to feed and water for 3 wk. Growth performance and antioxidant markers in plasma, intestine and liver were determined. Dietary addition of 1% GGS did not affect weight gain, feed intake, feed conversion and organ weight in 35 day-old broiler chickens significantly. There was no difference in plasma levels of glucose, triglyceride, cholesterol, AST, ALT and LDH activity. However, total antioxidant status(TAS) in blood increased(P<0.05) in chickens fed the diet supplemented with 1% GGS compared to those fed the control diet. In addition, the specific activity of intestinal superoxide dismutase(SOD) increased(P<0.05) in birds fed the diet supplemented with GGS. However, the activities of intestinal gluthathione peroxidase(GSHPx) and gluthathione -S- transferase(GST) and hepatic SOD, GSHPx and GST were not affected by the dietary GGS. The levels of reduced glutathione and lipid peroxidation in the small intestine and liver were not different between the two groups. In conclusion, dietary supplementation of 1% GGS did not result in a negative effect on growth performance. In addition, some antioxidant indicators including blood TAS and intestinal SOD were markedly elevated in response to dietary GGS. Therefore, dietary addition of 1% GGS may be beneficial to improve antioxidant capacity in broiler chicken.
Objectives : This study was done to investigate the protective effects of Gagaminjakdowha-Tang on liver injury of rats induced by $CCl_4$ and d-galactosamine. Methods : All animals were divided into 5 groups, those were normal group(untreated), control group(treated with 0.9% Saline solution), sample I group(200mg/kg administrated), sample II group(400mg/kg administrated), Silymarin(200mg/kg administrated) group. Liver injury of rats were induced by $CCl_4$ and d-galactosamine, and then the serumtransaminase(ALT & AST) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities, Liver cytosol malondialdehyde(MDA), catalase, superoxide dismutase(SOD), glutathione S-transferase(GST) and glutathione-peroxidase(GPX) for enzyme activities were measured. Results : The inhibitory effects on the serum ALT activities were noted in both sample I and sample II group. The inhibitory effects on the serum AST activities were noted in only sample II group. The inhibitory effects on the serum ALP activities were noted in both sample I and sample II group. The inhibitory effects on the serum LDH activities were noted in only sample II group. The inhibitory effects on the liver cytosol malondialdehyde were noted in only sample II group. The decresed effects on the liver cytosol catalase activities were inhibited in only sample II group. The decresed effects on the liver cytosol superoxide dismutase activities were inhibited in only sample II group. The decresed effects on the liver cytosol GST activities were inhibited in only sample II group. The decresed effects on the liver cytosol GPX activities were inhibited in only sample II group. The inhibitory effects of the serum ALT activities were noted in both sample I and sample II. The inhibitory effects of the serum AST activities were noted in only sample II group. The inhibitory effects of the serum ALP activities were noted in only sample II group. The inhibitory effects of the serum LDH activities were noted in both sample I and sample II group. Conclusions : Gagaminjakdowha-Tang has protective effects against liver injury in rats induced by $CCl_4$ and d-galactosamine.
In order to investigate the protective effects of Gami Yugan-tang on liver damage in rats induced by $CCl_{4}$ and d-galactosamine, the serum transaminase(ALT & AST), alkaline phosphatase(ALP), lactic dehydrogenase(LDH), superoxide dismutase(SOD), catalase, glutathione S-transferase(GST), glutathione peroxidase(GPX) for enzyme activities, and lipid peroxidation were measured. All animals were divided into 4 groups: normal group (untreated), control group (treated with 0.9% saline solution), sample I group (treated with 740mg/kg Gami Yugan-tang), and sample II group (treated with 1,480mg/kg Gami Yugan-tang). The results were as follows : 1. The results of liver damage in rats induced by $CCl_4$ : The protective effects of ALT were displayed in sample I and sample II, and AST, ALP, LDH, SOD, catalase, GST, GPX, and lipid peroxidation were noted in sample II group. It showed slight necrosis of hepatic cell and pathologic changes, for example, inflammatory cells infiltration were improved in sample II group compared to the control group. 2. The results of liver damage in rats induced by d-galactosamine : The inhibitory effects of AST, ALT, LDH, and ALP activities were noted in both sample I and sample II groups. The findings from this experiment suggests that Gami Yugan-tang has protective effects against liver damage in rats induced by $CCl_{4}$ and d-galactosamine.

In this paper, the effect of Ganopoly(extracts of Ganoderma lucidum) and Ganopoly/C+(70% Ganopoly + 30% chitosan) on cisplastin-induced nephrotoxicity was investigated in Sprague-Dawley rats. A single dose of cisplastin(5 ㎎/㎏) kg) was administered intraperitoneally after pretreatment of saline, Ganopoly and Ganopoly/C+ for 7 days. The nephrotoxicity and renal function were manifestated by the changes of body weight, blood pressure, biochemical changes and solute in urine and plasma. After the treatment of CDDP(cis-dichlorodiamineplatinum), a significant elevation of kidney weight, serum urea, cretinine, urine volume for 24 hours, urine magnesium, and a severe or significant decrease in body weight, blood pressure, creatinine clearance, urine osmolarity, serum albumin, etc. The nephrotoxicity was further confirmed by a significant decrease in glutathione S-transferase(GSH) in urine and kidney homogenate, GSH, glutathione peroxidase(GSH-Px) and catalase in kidney tissue. And also the lipid peroxidation was significantly increased in kidney homogenate. These signs of nephrotoxicity was ameliorated by the pretreatment and consecutive administration of Ganopoly and Ganopoly/C+ for 14 days after the Lp. injection of CDDP on 7th day after pretreatment of Ganopoly and Ganopoly/C+. The amelioration of nephrotoxicity was evidenced by significant reduction in serum urea and creatinine concentration, and improvement of other index of renal function. And The activity of antioxidant enzymes were partially recovered in kidney tissue of rats treated by CDDP and the administration of Ganopoly and Ganopoly/C+. These results indicate the cispastin induced nephrotoxicity is due to an impairment of tubular reabsorption systems enhanced by necrosis of proximal tubule, and the Ganopoly and Ganopoly/C+ has a partial protective effect on nephrotoxicity induced by CDDP. The polysacchride of Ganoderma lucidum may improve the therapeutic index of nephrotoxicity induced by CDDP. However, it is needed to elucidate the mechanism for confirming the therapeutic effect.
Bisphenol A, an everywhere chemical, is applied as a plasticizer in polycarbonate plastics, which often used in our everyday products and in epoxy resins as protective coatings and linings for food and beverage cans for decades. Human exposure to BPA may lead to adverse effects by interfering with oestrogen receptors. Our present study was conducted to investigate the protective effects of selenium (Se) and vitamin E (Vit E) on BPA-induced damage in the liver of male rats. Animals were randomly divided into four groups: the first group received olive oil and served as control. The second group received both (Se + Vit E) (0.5 mg/kg diet; 100 mg/kg of diet). The third one treated orally by (10 mg/kg b.w.) of BPA. The last group received (Se + Vit E) (0.5 mg/kg diet; 100 mg/kg of diet) concomitantly with (10 mg/kg b.w.) BPA. Exposure to BPA for three weeks engendered a hepatic disorder. An increased AST and ALT enzymatic activity was noticed in BPA-treated group as compared to other groups. Furthermore, a change in glucose, cholesterol, LDL-C, HDL-C, albumin, and bilirubin level was remarkable. Moreover, exposure to BPA increased malondialdehyde levels while reduced gluthatione content was decreased in the liver homogenate. A decrease in glutathione peroxidase, glutathione s-transferase and catalase activities was observed in the same group. Administration of selenium and vitamin E through the diet in BPA treated rats ameliorated the biochemical parameters cited above. In addition, an improvement in activities of liver enzymes was recorded. The histological findings confirmed the biochemical results. The model of this study that we employed characterized the relationships between BPA-induced hepatotoxicity and its alleviation by natural antioxidants like selenium and vitamin E.
The mode of safening action and potency of the 1, 8-naphthalic anhydride(NA) were investigated in corn(Zea mays) and soybean(Glycine max) treated with herbicide bensulfuron[2-{{{{{(4,6-dimethoxy-2-pyrimidinyl)amino}carbonyl}amino}sulfonyl}methyl}benzoic acid] and imazaquin[2-{4,5-dihydro-4-methyl-4-(1-methylethyl)-5-oxo-1H-imidazol-2-yl} 3-quinolinonecarboxylic acid]. Seed dressing with 0.2%(w/w) NA showed successful protection in corn against injury from herbicide bensulfuron and imazaquin but not in soybean. Safening factors of NA against bensulfuron and imazaquin were 10.2 and 5.0, respectively, in corn, while they were both 1.3 in soybean. In vivo, Glutathione-S-transferase(GST) activity of NA-treated corn and soybean increased 1.8-and 1.3-fold, respectively, but the activity was not affected by the herbicides in vitro. Acetolactate synthase(ALS) levels of NA-treated corn was increased 1.3-fold, but not changed in soybean. Tolerance of ALS activity to the herbicides was slightly greater in ALS obtained from NA-treated corn than that from the untreated, whereas the difference was not found in soybean. A significant increase of ACCase due to NA occurred in corn, but not in soybean. The herbicides did not affect in vitro ACCase activity.
