Fermented Protaetia brevitarsis Larvae Ameliorates Chronic Ethanol-Induced Hepatotoxicity in Mice via AMPK and TLR-4/TGF-β1 Pathways

  • This study evaluated the hepatoprotective effect of fermented Protaetia brevitarsis larvae (FPB) in ethanol-induced liver injury mice. As a result of amino acids in FPB, 18 types of amino acids including essential amino acids were identified. In the results of in vitro tests, FPB increased alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activities. In addition, FPB treatment increased cell viability on ethanol- and H2O2-induced HepG2 cells. FPB ameliorated serum biomarkers related to hepatoxicity including glutamic oxaloacetic transaminase, glutamine pyruvic transaminase, total bilirubin, and lactate dehydrogenase and lipid metabolism including triglyceride, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol. Also, FPB controlled ethanol metabolism enzymes by regulating the protein expression levels of ADH, ALDH, and cytochrome P450 2E1 in liver tissue. FPB protected hepatic oxidative stress by improving malondialdehyde content, reduced glutathione, and superoxide dismutase levels. In addition, FPB reversed mitochondrial dysfunction by regulating reactive oxygen species production, mitochondrial membrane potential, and ATP levels. FPB protected ethanol-induced apoptosis, fatty liver, and hepatic inflammation through p-AMP-activated protein kinase and TLR-4/NF-κB signaling pathways. Furthermore, FPB prevented hepatic fibrosis by decreasing TGF-β1/Smad pathway. In summary, these results suggest that FPB might be a potential prophylactic agent for the treatment of alcoholic liver disease via preventing liver injury such as fatty liver, hepatic inflammation due to chronic ethanol-induced oxidative stress.

Effects of Dietary Kugija Lycium chinense Supplementation on Juvenile Korean Rockfish Sebastes schlegeli (치어기 조피볼락 사료내 구기자 첨가효과)

  • A 8-week feeding trial was conducted to investigate the effects of dietary supplementation of Kugija (Lycium chinense) on the growth and immunological response in juvenile Korean rockfish (Sebastes schlegeli). Six experimental diets were supplemented with Kugija at 0, 0.1, 0.5 1.0, 3.0 and 5.0% ($K_0,\;K_{0.1},\;Ko_{0.5},\;K_{1.0},\;K_{3.0},\;K_{5.0}$) on a dry-matter basis. After 2 weeks, triplicate groups of 30 fish initially averaging 3.36$\pm$0.2 g (mean$\pm$SD) were randomly distributed into the aquarium and were fed one of the experimental diets for 8 weeks. By the end of the 8-week feeding trial, fish fed the $K_{0.5}$ and $K_{1.0}$ diets exhibited a higher weight gain and specific growth rate than fish fed $K_0$ and $K_{0.1}$ diets (P<0.05). Feed efficiency of fish fed the $K_{0.5}$ diet showed significant higher value than that of fish fed the $K_0$, $K_{3.0}$ and $K_{5.0}$ diets (P<0.05). Hepatosomatic index of fish fed the $K_{0.5}$ diet was significantly higher than that of fish fed the $K_{0.1}$ and $K_{5.0}$ diets (P<0.05). Hematocrit of fish fed the $K_{0.5},\;K_{1.0}$ and $K_{5.0}$ diets was significantly higher than that of fish fed the $K_0$ diet (P<0.05). Glutamic oxaloacetic transaminase of fish fed the $K_{0.5},\;K_{1.0},\;K_{3.0}$ and $K_{5.0}$ diets was significantly lower than in fish fed the other diets (P<0.05). Glutamic pyruvic transaminase activity of fish fed the $K_{3.0}$ diet was significantly lower than those of fish fed the $K_0$ and $K_{0.1}$ diets (P<0.05). However, there was no significant difference in the activity of the transaminase in fish fed the $K_{0.5}\;K_{1.0},\;K_{3.0}$ and $K_{5.0}$ diets. Lysozyme activity of fish fed the $K_{0.5}$ and $K_{1.0}$ diets was significantly higher than that in fish fed the $K_0$ and $K_{0.1}$ diets (P<0.05). Respiratory burst activity of fish fed the $K_{3.0}$ diet was significantly higher than those of fish fed the $K_0$ and $K_{0.1}$ diets (P<0.05). However, there was no significant difference in respiratory burst activity of fish fed the $K_{0.5}\;K_{1.0},\;K_{3.0}$ and $K_{5.0}$ diets. Fish fed Kugija showed lower early mortality than fish fed a Kugija-free diet upon challenge with Vibrio ordalii. The results suggest that feeding of Kugija (Lycium chinense) enhances growth, non-specific immunity and disease resistance in juvenile Korean rockfish.

Comparisons of the Carcass Characteristics of Male White Mini Broilers, Ross Broilers and Hy-Line Brown Chicks under the Identical Rearing Condition (동일 조건에서 사육한 수컷 백세미, 브로일러 및 산란종 병아리에 있어서 도체 특성의 비교)

  • The present study was conducted to compare the carcass yields and meat characteristics of three types of commercial male chicks White mini broilers, Ross broilers and Hy-Line brown chicks under the identical feeding condition. One-hundred 1-d chicks of each type were randomly placed into four pens per group (25 chicks per pen) and fed corn-soybean meal based commercial diets for 35d, 18d or 49d, respectively. At the end of the feeding trial, the birds were sacrificed and subjected to carcass measurements. The dressing percentages of White mini broilers and Ross broilers were significantly higher (P<0.05) than that of Hy-Line brown cockerels. The rate of breast meat of Hy-Line brown cockerels was significantly lower (P<0.05) than those of White mini broilers and Ross broilers. However, Hy-Line brown cockerels showed higher (P<0.05) leg meats than the others. There were no significant differences in serum total cholesterol and the activities of glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase among the groups. The breast meats of White mini broilers presented highest lightness value. The yellowness of breast and redness of leg meats of White mini broilers and Ross broilers were significantly higher (P<0.05) than those of Hy-Line brown cockerels. There were no significant differences in the SOD-like activity and change of pH in edible meats among the groups. The meat color in White mini broilers was significantly higher than that of Hy-Line brown cockerels. No significant differences were observed in term of flavor, tenderness and overall acceptability. In conclusion, the physico-chemical properties and sensory characteristics of edible meats were not greatly affected by genotype if they were similar body weights and kept under the identical feeding condition. But the Hy-Line brown cockerels were less desirable as a meat-type strain due to lower carcass yields and inferior growth and feed conversion ratio.

Effect of Dietary Grape Pomace on Lipid Metabolism and Hepatic Morphology in Rats Fed a High Fat Diet (포도박이 고지방식이를 섭취한 흰쥐의 체내 지질대사와 간조직의 형태학적 변화에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary grape pomace on lipid metabolism and hepatic morphology of rats fed a high fat diet. The high fat diet contained additional 15% lard to AIN 93-based diet. Male Sprague-Dawley rats were fed experimental diets containing 5% grape pomace for 4 weeks. Serum activities of glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) were not changed by high fat and grape pomace feeding. Serum concentration of triglyceride in rats fed a high fat diet was decreased significantly by dietary grape pomace. Hepatic concentrations of total lipid, total cholesterol and triglyceride were reduced in grape pomace groups with a high fat diet. Fecal concentrations of total cholesterol and triglyceride were increased in grape pomace groups with a high fat diet. The fecal content of coprostanol was not different among the groups. Dietary grape pomace increased the fecal excretion of cholesterol and coprostanone in rats fed a high fat diet. The fecal excretion of bile acid was not affected by feeding grape pomace in rats fed a high fat diet. Light micrographs of liver tissue revealed lipid droplets were increased by a high fat diet, but dietary supplementation of grape pomace tended to alleviate such changes.

Effects of Human Milk Fortifier on Growth and Nutritional Status of Growing Rats Fed Infant Formula (모유강화제(HMF)가 흰쥐의 성장과 영양상태에 미치는 영향)

  • This study was conducted to evaluate the efficacy of human milk fortifier (HMF) on growth and nutritional status in growing rats fed infant formula supplemented with HMF. Three week-old male Sprague-Dawley rats were divided into three groups and fed regular formula (RF), premature formula (PF) and regular formula fortified with HMF (RF+HMF) diets for 3 weeks. There was no significant difference in weight gain among groups. However, a significant increase of food intake was observed in PF and RF+HMF groups compared with RF group. With increasing food intake, the intakes of carbohydrate and protein were significantly increased in PF and RF+HMF groups. The weight of perirenal fat was significantly increased in rats fed RF+HMF; however, the weights of liver, kidney and spleen were not significantly different among groups. Although total lipids, total cholesterol, HDL-cholesterol concentrations of serum were not significantly different among groups, triglyceride was significantly increased in PF group. The triglyceride and total-cholesterol of liver were significantly increased in rats fed regular formula fortified with HMF and PF compared with RF group. Glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT), creatinine (Cre) and blood urea nitrogen (BUN) in serum showed no significant difference among groups. The concentration of growth hormone was significantly increased in PF group compared with other groups. The concentration of hemoglobin was significantly increased in rats fed PF and RF+HMF. These results suggest that the supplementation of human milk fortifier in growing rats may promote growth as increasing food intake and lipid contents in tissues and prevent the anemia of infants.

Effects of Unripe Rubus coreanus Miquel Extract on Improvement of Lipid Metabolism in C57BL/6 Mice Fed a High-Cholesterol Diet (미숙과 복분자 열매 추출물이 고콜레스테롤 식이를 섭취시킨 C57BL/6 Mice의 지질대사 개선 효과)

  • Hypercholesterolemia is the presence of high levels of cholesterol in the blood, and it is regarded as a risk factor for cardiovascular disease due to the effects of cholesterol. The objective of this study was to investigate the effects of unripe Rubus coreanus Miquel (uRC) extract on lipid metabolism in hypercholesterolemic mice fed a high cholesterol diet (HC). uRC 50 (unripe R. coreanus 5% ethanol extract 50 mg/kg/day), uRC 100 (unripe R. coreanus 5% ethanol extract 100 mg/kg/day), uRC 300 (unripe R. coreanus 5% ethanol extract 300 mg/kg/day), and BO (borage seed oil containing minimum of 20% ${\gamma}$-linolenic acid 30 mg/kg/day) were orally administered for 60 days after HC. Oral administration of uRC 50, uRC 100, uRC 300, and BO significantly reduced serum total-cholesterol, triglyceride, LDL-cholesterol, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, atherogenic index, and cardiac risk factor levels. Similarly, uRC treatment elevated serum HDL-cholesterol levels. These results suggest that unripe R. coreanus extract could be established as a functional food for the improvement of lipid metabolism.

The Effects of Cordyceps Militaris on Lipid Metabolism, Protein Levels and Enzyme Activities in Rats Fed a High Fat Diet (동충하초 (Cordyceps Militaris)가 고지방식이를 섭취한 흰쥐의 지질대사, 단백질농도 및 효소활성에 미치는 영향)

  • The effects of Cordyceps militaris on weight gain, flood intakes, flood efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in adult male rats. Sprague-Dawley rats, 35 weeks old, were given four different types of diets for a succeeding period of five weeks: either a normal diet(5% com oil), a high fat diet(5% com oil + 15% lard). a CF diet (high fat diet + 3% Suiting body of cordyceps militaris), or a CM diet (high fat diet + 3% mycelium of cordyceps military). The body weight gains, hepatic weights and food efficiency ratios of the rats find (high fat) diets with CF and CM were significantly decreased compared with those find the high fat diet, but were similar to those find the normal diet. The concentrations of serum and hepatic triglycerides, and hepatic total lipids and cholesterol, of rats given CF or CM diets were significantly lower than those given the high fat diet. But the concentrations of total cholesterol, HDL-cholesterol, phopholipid and total protein in the serum of rats find the high fat, CF or CM diets were significantly higher than those find the normal diet. In the serum of the rats find the CF diet, the HDL-cholesterol/total cholesterol ratio was significantly higher but the atherogenic index was significantly lower than those fed the high fat diet. The alkaline phosphatase activity in the serum of the rats find the high fat, CF, and CM diets was more significantly decreased compared to rats on the normal dirt. No differences were noted in the weights of the pancreas, kidney and heart, the serum concentrations of glucose, hemoglobin and albumin, and the activities of GOT, GPT and ${\gamma}$-GTP, among the rats on all the experimental diets. In conclusion, the rats find the Cf or CM diets maintained normal body and hepatic weights. Despite of the high intake of fats in the CF and CM diets, the concentrations of total lipids, cholesterol and triglycerides were decreased in the liver, also, the concentration of triglycerides was decreased in the serum.

The Effect of Green Vegetable Drink Supplementation on Blood Pressure, Erythrocyte Antioxidant Enzyme Activities and Plasma Lipid Profiles of Korean Male Subjects (채소혼합음료의 섭취가 남자 성인의 혈압, 항산화 효소 및 혈장지질 양상에 미치는 영향)

  • The purpose of this study was to evaluate whether a daily regimen of green vegetable drink supplementation to male subjects improves the blood pressure and enhances other antioxidant enzyme status, lipid profiles and liver functionalities. Twenty nonsmokers and 19 smokers aged $23{\sim}60$ were given 240 mL of green vegetable drink every day for 8 weeks in addition to their normal diet, and blood samples were drawn before and after the intervention. The 8 weeks of green vegetable drink consumption resulted in a significant decrease of diastolic blood pressure in smokers (p<0.05) as well as in nonsmokers (p<0.01), and systolic blood pressure in smokers (p<0.05). Erythrocyte superoxide dismutase (SOD) activity was significantly increased in both nonsmokers and smokers after the trials. Plasma total cholesterol (TC) level was decreased in smokers and plasma TC and LDL-cholesterol (LDL-C) levels were decreased in nonsmokers while triglyceride (TG) and HDL-cholesterol (HDL-C) was not affected in both nonsmokers and smokers by green vegetable drink consumption. Plasma glutamic pyruvic transaminase (GPT), a marker of liver function was significantly reduced in both nonsmokers and smokers and plasma glutamic oxaloacetic transaminase (GOT) level was significantly reduced in nonsmokers after the supplementation. These results support the hypothesis that green vegetable drink exerts a blood pressure reducing effect and affects not only the activity of antioxidant enzymes, lipid profiles but also plasma GOT and/or GPT levels.

Hepatic Protective Effect and Single-dose Toxicity Study of Water Extract of Cordyceps militaris Grown upon Protaetia dreujtarsis (굼벵이 유래 밀리타리스 동충하초 열수추출물의 간기능개선 효과 및 단회독성 평가)

  • This study was designed to evaluate the single dose toxicity and the protective effect of water extract of Cordyceps militaris grown upon Protaetia dreujtarsis (CMPD extract) on liver damage on carbon tetrachloride ($CCl_4$)- induced acute hepatotoxicity in Sprague-Dawley (SD) rats. The CMPD extract was once administered orally to both sexes of rats at dose of 2,000, 1,000 and 500 mg/kg body weight, the recommended maximum limit dose for acute toxicity. Neither significant toxic signs nor death was observed during the observation period. These results indicate that $LD_{50}$(lethal dose of 50%) of CMPD extract is greater than 2,000 mg/kg body weight in SD rats. To investigate also the effect of hepatoprotection of CMPD extract, SD rats were orally treated with CMPD extract (50, 25 and 12.5 mg/kg body weight) or silymarin (25 mg/kg body weight) before and after administration of $CCl_4$ (2 mL/kg body weight, 20% $CCl_4$ in olive oil). Treatment with CMPD extract or silymarin could decrease the GPT (glutamic-pyruvic transaminase) and GOT (glutamic-oxaloacetic transaminase) levels in serum when compared with $CCl_4$-treated group. Therefore, the results of this study show that CMPD extract can be proposed to protect the liver against $CCl_4$-induced hepatic damage in rats.

Effect of oral administration of Injinhotang with bile extract of bear on carbon tetrachloride $(CCl_4)-induced$ hepatic cirrhosis rat (인진호탕가웅담(茵蔯蒿湯加熊膽)의 경구투여(經口投與)가 $CCl_4$ 투여(投與)로 유발(誘發)된 간경변(肝硬變)에 미치는 영향(影響))

  • In order to determine the effects of annexing bile extracts of bears on the anti-fibrotic effect of Injinhotang. Mix compound of Injinhotang and bile extracts of bears were administered to the carbon tetrachloride ($CCl_4$)-induced cirrhotic rats during 20 days and the changes of serum levels of GOT (glutamic-oxalacetic transaminase), GPT (glutamic pyruvic transaminase), LDH (lactate dehydrogenase), ALP (alanine phosphatase), GGT (gamma glutamyl transpeptidase) and T-BIL (total bilirubin) were monitored with comparison to the results of Injinhotang administered group. The results were summarized as follows. 1. A significant (p<0.01) increase of serum GOT levels were observed in control group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang and Injinhotang with Fel Ursi-administered group. In addition, a significant (p<0.05) increase were also detected In Injinhotang with Fel Ursi-administered group compared to that of Injinhotang-administered group. 2. A significant (p<0.01) increase of serum GPT levels were observed in control group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang and Injinhotang with Fel Ursi-administered group. Although significances were not recorded, increase of serum GPT levels were also detected in Injinhotang with Fel Ursi-administered group compared to that of Injinhotang-administered group. 3. A significant (p<0.01) increase of serum LDH levels were observed in control group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang and Injinhotang with Fel Ursi-administered group. Although significances were not recorded, increase of serum LDH levels were also detected in Injinhotang with Fel Ursi-administered group compared to that of Injinhotang-administered group. 4. A significant (p<0.01 or p<0.05) increase of serum ALP levels were observed in control group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang and Injinhotang with Fel Ursi-administered group. In addition, a significant (p<0.05) increase were also detected in Injinhotang with Fel Ursi-administered group compared to that of Injinhotang-administered group. 5. A significant (p<0.01) increase of serum GGT levels were observed in control and Injinhotang-administered group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang with Fel Ursi-administered group. 6. A significant (p<0.01) increase of serum T-BIL levels were observed in control group compared to those of normal group but these increased levels were dramatically decreased in Injinhotang and Injinhotang with Fel Ursi-administered group. Although significances were not recorded, increase of serum T-BIL levels were also detected in Injinhotang with Fel Ursi-administered group compared to that of Injinhotang-administered group. In conclusion, it is considered that bile extract of bears has some additional effect to the anti-fibrotic effect of Injinhotang but to know the exact mechanism of suitable dose and duration of administration, further studies such as pharmacokinetics and dose-dependent pharmacological studies were needed
