Use of Polycarboxylic Acid to Inhibit Heat-and Moisture-Induced Yellowing of ECF/TCF Bleached Hardwood Kraft Pulp

  • The aim of this work was to inhibit the heat-and moisture-induced yellowing of ECF/TCF bleached hard wood kraft pulp (HBKP) retaining hexenuronic acid (HexA). We have already reported that one of the derivatives of HexA, 5-formyl-2-furancarboxylic acid (FFA), causes yellowing of HBKP in acidic paper, and that FFA might polymerize or react with pulp components to form new chromophoric groups (1). In this study, it was shown that the carboxylic base of FFA interacts with that of glucuronic acid or galacturonic acid, resulting in strong yellowing. Therefore, it seems that preventing the reaction of carboxylic bases could be effective in suppressing this yellowing. We have discovered that polycarboxylic acids (for example, citric acid, tartaric acid, etc.) are useful as a yellowing inhibitor.

Isolation of Polysaccharides Modulating Mouse’s Intestinal Immune System from Peels of Citrus unshiu (귤피로부터 분리한 마우스의 장관면역 활성 다당류의 검색)

  • Of solvent-extracts prepared from the 90 kinds of Korean traditional tea and rice gruel plants, cold-water extract from peels of Citrus unshiu (CUI-0) showed the most potent intestinal immune system modulating activity through Peyer’s patch whereas other extracts did not have the activity except for cold-water extracts of Laminaria japonica, Polygonatum japonicum, Poncirus trifoliata, and hot-water extracts of Gardenia jasminoides, Lycium chinense having intermediate activity. CUI-0 was further fractionated into MeOH-soluble fraction (CUI-1), MeOH insoluble and EtOH-soluble fraction (CUI-2), and crude polysaccharide fraction (CUI-3). Among these fractions, CUI-3 showed the most potent stimulating activity for the proliferation of bone marrow cells mediated by Peyer’s patch cells, and contained arabinose, galacturonic acid, galactose, glucose, glucuronic acid and rhamnose (molar ratio; 1.00:0.53:0.45:0.28:0.28:0.19) as the major sugars, and a small quantity of protein (9.4%). In treatments of CUI-3 with pronase and periodate (NaIO₄), the intestinal immune system modulating activity of CUI-3 was significantly reduced, and the activity of CUI-3 was affected by periodate oxidation particularly. The potently active carbohydrate-rich fraction, CUI-3IIb-3-2 was further purified by anion-exchange chromatography on DEAE-Sepharose FF, Sepharose CL-6B and Sephacryl S-200. CUI-3IIb-3-2 was eluted as a single peak on HPLC and its molecular weight was estimated to be 18,000 Da. CUI-3IIb-3-2 was consisted mainly of arabinose, galactose, rhamnose, galacturonic acid and glucuronic acid (molar ratio;1.00:0.54:0.28:1.45:0.63) in addition to a small amount of proteins (3.2%). In addition, CUI-3IIb-3-2 showed the activity only through Peyer’s patch cells, but this fraction did not directly stimulate proliferation of bone marrow cells. It may be concluded that intestinal immune system modulating activity of peels from C. unshiu is caused by pectic polysaccharides having a polygalacturonan moiety with neutral sugars such as arabinose and galactose.

Urinary Metabolites of p-Methoxycinnamic Acid (p-Methoxycinnamic Acid의 뇨대사물에 대하여)

  • The metabolism of p-methoxycinnamic acid (p-MCA) has been studied in man and rabbits. Possible compounds were examined for metabolites by crystalization and by paper chromatography, which excreted after adminstration of p-MCA by stomach tube and intravenous injection. p-Methoxyhippuric acid was isolated from urine. Although pure glucuronide was not crystallized from urine, product was obtained by basic lead salt method, which gave p-methodxybenzoic acid (p-MBA) on hydrolysis and gave an intese naphthoresorcinol reaction. No evidence for the demethylation of p-MCA was found. These results are indicating that p-MCA may be mainly converted to p-MBA by ${\betha}$-oxidationand excreted as its conjugates with glycine and glucuronic acid. Its oxidation does not appear to be dependent on intestinal micro-organisms.

Characterization of a Novel Lipopolysaccharide Biosurfactant from Klebsiella oxitoca

  • The chemical, physical, and emulsifying properties of BSF-1, which is an extracellular lipopolysaccharide biosurfactant produced by Klebsiella oxytoca strain BSF-1, were studied. BSF-1 was found to be composed mainly of carbohydrate and fatty acids. The average molecular weight was $1,700{\sim}2,000 kDa$. The polysaccharide fraction contained L-rhamnose, D-galactose, D-glucose, and D-glucuronic acid at a molar ratio of 3:1: 1:1. The fatty acid content was 1.1 % (w/w) and consisted mainly of palmitic acid (C16:0), 3-hydroxylauric acid (3-OH-C12:0), and lauric acid (C12:0). In terms of thermal properties, BSF-1 was revealed to have inter- and intra-molecular hydrogen bonds. The hydrodynamic volume (intrinsic viscosity) of BSF-1 was 22.8dL/g. BSF-1 could be maintained as a stable emulsion for 48 h through a low-level reduction in surface tension. The optimal emulsification temperature was $30^{\circ}C$. Emulsification by BSF-1 was efficient at both acidic and neutral pH values.

  • Immunostimulating exopolysaccharides with anticancer activity produced by Enterobacter sp. SSYL(KCTC 0687BP), isolated from Chinese elm(Ulmus sp.) were investigated. The exopolysaccharide contains molecular weight 100,000 to 1,000,000 Da and total carbohydrates 43.0 to 70.8%, total uronic acid 7.1 7o 12.4%, and total proteins 15.4 to 20.6%. Compositions and contents of sugars in the exopolysaccharides are 10-30% glucose, less than 1% fructose, 10-15% galactose, 8-12% fucose, and 40-70% glucuronic acid. The anticancer immunostimulating activities were examined and proved with regard to both in vitro and in vivo bioassays. In vivo assay, the glycoprotein at the concentration of 0.3 mg/kg showed the best result that median survival time in creased to ca. 138.1% in contrast to control groups.

  • Hyaluronic acid(HA) is a naturally occurring mucopolysaccharide composed of the repeating disaccharide unit of D-glucuronic acid and N-acetyl-D-glucosamine. HA exists as a high molecular weight polymer in the extracellular matrix of many tissues in the body. HA has been widely used not only for osteoarthritis and ophthalmology but also for cosmetics for skin care. Recently HA has drawin much attention for use in health foods. Bioland Corporation started production of HA t1y biotechnological process in 1990 for cosmetical use. At present Bioland produces HA for cosmetic and food use. We have done many researches to evaluate the helpful effect for skin and safety of HA as food. In order to evaluate the effect of HA on the skin after its oral intake, we measured its bioavailability, skin-care effects on anti-wrinkle, moisturization elasticity and desquamation by clinical trial. The results showed that HA is very safe and could bs helpful to improve anti-wrinkling, moisturization, elasticity, and desquamation on the skin.

  • When ultrasonic wave with barium titanate as an ultrasonic transducer (96.84kc, 1.4kv, 240mA) was applied to a dilute glucose solution, glucose was oxidized to glucuronic acid. It was found that the lower the glucose concentration, the higher the oxidation rate. The aeration and the presence of ferrous sulfate as a catalyst were found to increase the rate. The optimal duration for applying the wave to the solution was found to be six hours.

  • Antitumor and antimicrobial activities of water soluble fractions of Duchesnea indica (Andr.) and Focke (Rosaceae), have been studied. The contents of total polysaccharide were 22% and its composition was identified by G.C.: L-arabinose(Rt: 7.71), D-xylose (Rt. 9.91), D-glucuronic acid (Rt. 12.77), D-fructose (Rt. 13.63), D-galactose (Rt. 14.68), D-glucose (Rt. 15.42). The antimicrobial activity of Duchesnea indica against four bacterial species, S. aureus, S. paratyphi, S. dysenteriae and Ps. aeruginosa was observed.

  • Hyaluronic acid is a disaccharide polymer that consists of glucuronic acid and N-acetylglucosamine, and is a glycosaminoglycan. It is widely distributed in animal connective tissue, coexisting with chondroitin sulfate. It forms viscous secretions or gel by bonding to protein in the body, and thus maintains and lubricates tissue structures. In addition, its antibiotic properties are applied to cosmetics and pharmaceuticals. Therefore, it has recently been an object of great interest at cosmetic and pharmaceutical companies. $^{125}I$ has a longer half-life (59.4 days) than many other radioisotopes, and is therefore useful to long-term studies. In this study, a facile and efficient procedure for the synthesis of $^{125}I$-labeled hyaluronic acid has been designed for in vivo imaging and pharmacokinetic studies of hyaluronic acid, which are required for the development of new drugs.
