  • In the case of immobilizing of glucose oxidase (GOx) in polypyrrole (PPy) conducting polymer using electrosynthesis, the GOx obstructs charge transfer and mass transport during the film growth. This may lead to short chained polymer and/or make charge-coupling weak between the GOx and the backbone of the PPy. That is mainly due to insulating property and net chain of the GOx. Since being the case, it is useless to increase in amount of GOx mere than reasonable in the synthetic solution. We improved the amount of immobilized GOx into the PPy by adding a little ethanol in the synthetic solution without any more amount of GOx in the solution. We electrochemically analyzed an improvement in the immobilizing event. For the glucose sensing, when ethanol was added by 0.1 mol $dm^{-3}$ in the synthetic solution, the Michaelis constant of the resulting enzyme electrode was about 32 mmol $dm^{-3}$ and maximum current was about $146\mu A$.

  • In this study, we fabricated the catalysts for enzymatic biofuel cell anode with carbon nanotube (CNT), glucose oxidase (GOx) and various molecular weights branched poly(ethyleneimine)(bPEI) and terephthalaldehyde (TPA) as cross-linker. In case of GOx/bPEI/CNT using only physical entrapments for immobilization, the molecular weights of bPEI didn't affect to electrochemical performances and long term stability. but that of the catalysts cross linked via TPA (TPA[GOx/bPEI/CNT]) improved and the mass transfer of glucose to FAD was interrupted as increasing of the bPEI's molecular weights. Furthermore, it was confirmed that the optimum molecular weight of PEI for TPA [GOx/bPEI/CNT]) structure is 750k that showed marvelous high performance (maximum power density of $0.995mW{\cdot}cm^{-2}$).

  • In the case of immobilizing of glucose oxidase into polypyrrole (PPy) using electrosynthesis, the glucose oxidise (GOx) forms a coordinate bond with the polymers backbone. However, because of intrinsic insulation and net-chain of the enzyme, the charge transfer and mass transport are obstructed during the film growth. Therefore, the film growth is dull. We synthesized enzyme electrodes by electropolymerization added some organic solvent, such as ethanol and tetrahydrofuran (THF). The formative seeds of film growth was delayed by adding ethanol. The delay was induced by radical transfer between ethanol and pyrrole monomer. The radical transfer reactions shared the contribution of dopants between electrolyte anion and GOx polyanion. This led to increase amount of immobilized the enzyme in PPy. For the UV absorption spectra of synthetic solution before synthesis and after, in the case of ethanol added, the optical density was slightly decreased for the GOx peaks. It suggests amount of GOx in the solution was decreased and amount of GOx in the film was increased.

Glucose Oxidase-Coated ZnO Nanowires for Glucose Sensor Applications

  • Well-aligned Zinc oxide (ZnO) nanowires were synthesized on silicon substrates by a carbothermal evaporation method using a mixture of ZnO and graphite powder with Au thin film was used as a catalyst. The XRD results showed that as-prepared product is the hexagonal wurzite ZnO nanostructure and SEM images demonstrated that ZnO nanowires had been grown along the [0001] direction with hexagonal cross section. As-grown ZnO nanowires were coated with glucose oxidase (GOx) for glucose sensing. Glucose converted into gluconic acid by reaction with GOx and two electrons are generated. They transfer into ZnO nanowires due to the electric force between electrons and the positively charged ZnO nanostructures in PBS. Photoluminescence (PL) spectroscopy was employed for investigating the movements of electrons, and the peak PL intensity increased with the glucose concentration and became saturated when the glucose concentration is above 10 mM. These results demonstrate that ZnO nanostructures have potential applications in biosensors.

The radical transfer shares the contribution of dopant between small and fast electrolyte anion and big and slow GOx polyanion.

Performance Improvement of Glucose Sensor Adopting Enzymatic Catalyst bonded by Glutaraldehyde (글루타알데하이드에 의해 결합된 효소촉매를 이용한 글루코스 센서의 성능향상)

  • In this study, we synthesized a biocatalyst consisting of glucose oxidase (GOx), polyethyleneimine (PEI) and carbon nanotube (CNT) with addition of glutaraldehyde (GA)(GA/[GOx/PEI/CNT])for fabrication of glucose sensor. Main bonding of the GA/[GOx/PEI/CNT] catalyst was formed by crosslinking of functional end groups between GOx/PEI and GA. Catalytic activity of GA/[GOx/PEI/CNT] was quantified by UV-Vis and electrochemical measurements. As a result of that, high immobilization ratio of 199% than other catalyst (with only physical adsorption) and large sensitivity value of $13.4{\mu}A/cm^2/mM$ was gained. With estimation of the biosensor stability, it was found that the GA/[GOx/PEI/CNT] kept about 88% of its initial activity even after three weeks. It shows GA minimized the loss of GOx and improved sensing ability and stability compared with that using other biocatalysts.

Dietary inclusion of glucose oxidase supplementation to corn-wheat-based diet enhance growth performance, nutrient digestibility, blood profile of lactating sows

  • The objective of this study is to investigate the effect of dietary glucose oxidase (GOx) supplementation on the reproductive performance, litter performance, total tract digestibility, and blood profile of lactating sows fed corn- wheat-based diet. A total of twenty multiparous sows (Landrace × Yorkshire) were allocated into one of four treatments with five replicates per treatment. The dietary treatments were as follows: CON (Basal diet), GO1 (basal diet + 200 U GOx/kg), GO2 (basal diet + 300 U GOx/kg), GO3 (basal diet + 400 U Gox/kg). Dietary GOx supplementation did not affect lactating sow's reproduction performance as well as body weight, backfat thickness, and body condition score during pre and post farrowing, and at weaning (p > 0.05). However, after farrowing to weaning period lactating sow's fed GOx supplement has linearly (p = 0.0196) decreased the bodyweight loss. While, there were no effects (p > 0.05) observed on sows backfat thickness loss, average daily feed intake, and estrus interval among treatment groups. Dietary supplementation of GOx has linearly improved the body weight gain (p = 0.049) and average daily gain (p = 0.040) of suckling piglets. The total tract digestibility of dry matter and nitrogen was linearly increased with the graded level of GOx supplement. Also, a linear effect was observed on the glucose and superoxide dismutase of blood profile with the dietary inclusion of GOx. In summary, our finding indicates that the dietary inclusion of GOx supplement with corn- wheat-based diet had a beneficial effect on the nutrient digestibility and blood profile of lactating sows and improved the growth performance of suckling piglets.

Performance Enhancement of Biofuel Cell by Surface Modification of Glucose Oxidase using Ferrocene Carboxylic acid (페로신카르복시산을 이용한 글루코스 산화효소의 표면개질에 의한 바이오 연료전지 성능향상)

  • In this study, we synthesized a mediator immobilized biocatalyst([FCA/GOx]/PEI/CNT) by surface modification using ferrocene carboxylic acid(FCA), and evaluated its performance as anode catalyst for biofuel cell. Through the application of FCA on glucose oxidase (GOx), the free amine groups on the lysine residue of GOx surface reacted with carboxylic acid of FCA and make amide bond between GOx and FCA. As the result of that, the electron transfer of catalyst was increased up to 1.91 times($0.468mA{\cdot}cm^{-2}$) than the catalyst without surface modification (GOx/PEI/CNT), and high maxium power density of $1.79mA{\cdot}cm^{-2}$ was gained.

Correlative Production of Mn-Peroxidase and Glucose Oxidase Depending on the Culture Condition of Schizopora paradoxa (좀구멍버섯균의 배양조건에 따른 Mn-Peroxidase와 Glucose Oxidase의 생성조절)

  • White-rot fungus, Schizopora paradoxa did not produce Mn-peroxidase and glucose oxidase without manganese. But, in high concentration of manganese (40 ppm), the activities of both enzymes were higher than those in basal concentration of manganese (11.15 ppm). Unlike the activities of the enzymes, mycelial mass was the same level as the control culture (11.15 ppm manganese) through out the culture period, depending on the concentration of manganese. The same experiments were carried out for the effect of copper and veratryl alcohol added to the culture. The results were not consistent dependent on the concentration of copper and veratryl alcohol, respectively. The involvement of cAMP in the correlative production of MNP and GOX was investigated. In this study, addition of atropine to the culture resulted in a concomitant inhibition of production of MNP and GOX, depending on the concentration of inhibitor added.
