Profile of Non-Communicable Disease Risk Factors Among Employees at a Saudi University

  • Background: There is paucity of studies defining the prevalence of non-communicable disease (NCD) risk factors in Saudi Arabia despite the surging epidemic of obesity, change in dietary habits and sedentary lifestyle. Objectives: This cross-sectional study aimed to assess the prevalence of NCDs risk factors among employees at King Faisal University in Al Hassa, Saudi Arabia and to determine the possible correlates for clustering of NCDs risk factors among them. Materials and Methods: All employees were invited to participate; the World Health Organization STEPwise approach was used for data collection which consisted of a personal interview to collect socio-demographic characteristics, NCD history, tobacco use, vegetables and fruit consumption, and physical activity (PA), followed by anthropometric measurements namely weight, height and waist circumference and blood pressure measurements, subjects were finally subjected to biochemical tests with determination of fasting plasma glucose, serum triglycerides, cholesterol and high density lipoproteins. Results: Of the surveyed employees (n=691), daily current smokers accounted for 22.7%. 94.9%, 95.1% and 86% consumed < 5 servings per day of vegetables, fruits and both fruits and vegetables respectively, 73% were physically inactive, 64% were overweight or obese, 22.1% had hypertension, and 21.5% were diabetics. Elevated cholesterol levels were found in 36.6%, low high density lipoproteins in 36.8%, and elevated triglycerides in 36.1%. Only 3% had no NCD risk factors, and 57.6% had ${\geq}3$ factors. Multivariate logistic regression showed that gender (being male, adjusted odds ratio 'aOR'=1.51), aged ${\geq}50$ years (aOR=3.06), < college education (aOR=1.75), current smokers (aOR=2.37), being obese (aOR=6.96) and having a low PA level (aOR=4.59) were the significant positive predictors for clustering of NCD risk factors. Conclusions: Over fifty percent of the studied university's employees had multiple (${\geq}3$) NCD risk factors. Screening and health promotion initiatives should be launched at least targeting the modifiable factors to avert the excessive risk for cardiovascular disease, diabetes mellitus and several types of cancers.

  • Kefir is a probiotic food. Probiotics have shown to be beneficial to health, and are currently of great interest to the food industry. Hence, this study was carried out to evaluate the lactase activity of kefir-isolated lactic acid bacteria. Three strains, Lactobacillu kefiri DH5 isolated from the kefir grains and Bifidobacterium animalis subsp. Lactis and Bifidobacteria longum 720, commercial probiotic LAB, were fermented in 10% reconstituted nonfat dry milk suspensions and incubated at $37^{\circ}C$ for 48 h, and then analyzed for various saccarides by HPLC. The results showed that changes in the concentrations of lactose and galactose were significantly decreased and increased, respectively (p<0.05), but all 3 probiotic strains tested in this study showed no increase in glucose concentration during 48 h of incubation. Both DH5 and BL720 showed high lactase activities (p<0.05), whereas BLC exhibited the lowest activity. Additionally, all three lactic acid bacteria showed high tagatose, but did not show high xylose and sedoheptulose. Finally, DH5, a kefir-isolated LAB, may have similar characteristics and properties to typical Bifidobacterium spp. and showed higher lactase activity than commercial Bifidobacterium spp.

Anaerobiospirillum succiniciproducens에 의한 유기산 생성에 미치는 pH의 영향 (Effect of pH on Organic Acid Production by Anaerobiospirillum succiniciproducens)

  • Anaerobiospirillum succiniciproducens에 의한 유기산 생성에 미치는 pH의 영향을 조사하기 위하여 발효 배양액의 pH를 5.8, 6.0, 6.4, 6.8, 7.2로 유지하여 혐기 발효 실험을 수행하였다. 세포 농도는 여러 pH 조건에서 $1.0∼1.9g/\ell$로 다른 연구자들의 결과와 비교할 때 2~3 배 정도 더 높았으며 pH 5.8에서 가장 높았다. pH 6.0 ~ 6.8의 범위에서 기질 소모량은 pH가 높을수록 증가였으며 반면에 pH 5.8과 7.2에서의 당 소비 속도는 매우 느렸다. 유기산 생성으로 인한 pH 변화의 조절을 위 하여 첨가된 2M $Na_2C0_3$의 총량은 pH 6.8에서 최대였다. 여러 pH조건에서 발효액의 전기전도도의 변화는 첨가된 2M $Na_2C0_3$의 변화와 매우 유사한 경향을 보였고 따라서 발효액내의 유기산의 정량은 전기전도도를 측정 함으로 구할 수 있음을 알 수 있었다. pH 7.2의 조건에서 기질에 대한 lactatedml chleo todtks tndbfdms 64%, pH6.8의 경우 32% 였다.

Streptanthus tortus 조직배양 세포에서 사공의 형성시기와 사공 영역과 사공의 크기 결정 (Determination of the Period of the Formation and Size of Sieve Element Area and Sieve Pore)

  • Streptanthus 조직배양 세포에서 유조직 세포로부터 사부 세포의 발달 동안 설탕 운반자는 유도되었고, 단당류인 포도당의 운반자는 사라졌다 (Cho 1998). Streptanthus 조직배양 세포에서 유도된 사부영역과 사공은 sieve endoplasmic reticulum (SER)과 p-protein 합성이 거의 완성되는 시기에 형성되며, SER이 새로 합성된 세포벽을 에워싼 후에 세포벽이 분해되기 시작하였다. 세포벽의 분해와 사공의 형성은 비교적 규칙적으로 진행되었다. 사공의 모양은 계란형이었고, 사공의 외부크기는 세로 1.2 $\mu\textrm{m}$~l.6 $\mu\textrm{m}$, 가로 0.8 $\mu\textrm{m}$~l.3 $\mu\textrm{m}$, 다양하고 사공의 내부크기는 별모양과 유사한 불규칙한 형태였다. 두 사부세포 사이에서 사공의 수는 $\mu\textrm{m}$$^2$당 2개~7개였고, 육상체를 포함한 사공벽에 두께는 0.05 $\mu\textrm{m}$~0.07 $\mu\textrm{m}$,의 두꺼운 벽으로 구성되었다. 사공영역과 사공의 형성에 필요한 에너지는 세포벽 가까이에 위치하고 있는 미토콘드리아에서 얻을 것으로 추측하며, 사공 형성에 대한 SER의 역할은 아직까지 설명할 수 없었다.

급성 중독환자에서 삼투압 계산식으로 추정된 에탄올 농도의 유효성 검증 (Discrepancies and Validation of Ethanol Level Determination with Osmolar Gap Formula in Patients with Suspected Acute Poisoning)

  • Purpose: Osmolar gap (OG) has been used for decades to screen for toxic alcohol levels. However, its reliability may vary due to several reasons. We validated the estimated ethanol concentration formula for patients with suspected poisoning and who visited the emergency department. We examined discrepancies in the ethanol level and patient characteristics by applying this formula when it was used to screen for intoxication due to toxic levels of alcohol. Methods: We retrospectively reviewed 153 emergency department cases to determine the measured levels of toxic ethanol ingestion and we calculated alcohol ingestion using a formula based on serum osmolality. Those patients who were subjected to simultaneous measurements of osmolality, sodium, urea, glucose, and ethanol were included in this study. Patients with exposure to other toxic alcohols (methanol, ethylene glycol, or isopropanol) or poisons that affect osmolality were excluded. OG (the measured-calculated serum osmolality) was used to determine the calculated ethanol concentration. Results: Among the 153 included cases, 114 had normal OGs (OG≤14 mOsm/kg), and 39 cases had elevated OGs (OG>14). The mean difference between the measured and estimated (calculated ethanol using OG) ethanol concentration was -9.8 mg/dL. The 95% limits of agreement were -121.1 and 101.5 mg/dL, and the correlation coefficient R was 0.7037. For the four subgroups stratified by comorbidities and poisoning, the correlation coefficients R were 0.692, 0.588, 0.835, and 0.412, respectively, and the mean differences in measurement between the measured and calculated ethanol levels were -2.4 mg/dL, -48.8 mg/dL, 9.4 mg/dL, and -4.7 mg/dL, respectively. The equation plots had wide limits of agreement. Conclusion: We found that there were some discrepancies between OGs and the calculated ethanol concentrations. Addition of a correction factor for unmeasured osmoles to the equation of the calculated serum osmolality would help mitigate these discrepancies.

개 정액의 냉동보존 시 κ-Carrageenan이 정자 성상에 미치는 영향 (Effect of κ-Carrageenan on Sperm Quality in Cryopreservation of Canine Semen)

  • This study was conducted to find out the effect that ${\kappa}-Carrageenan$ has on the properties of dog sperm when it was added to the cryoprotectant. Extender basically was contained 1.21 g Trizma base, 0.67 g citric acid, 0.4 g glucose, 0.03 g penicillin G, 0.05 g streptomycin sulfate. Extender1 was added with 0.1%, 0.2%, 0.3%, and 0.5% carrageenan, while extender2 was supplemented with glycerol. After freezing-thawing, the motility, viability, acrosome integrity, apoptosis, and ROS (reactive oxygen specifications) of sperm were measured to analyze the effects of the supplementation of carrageenan. Total Motile (TM), Rapid Progressive Motile (RPM), Medium Progressive Motile (MPM), and Immotile were measured through the CASA system after thawing in 37 degree water. Extender with 0.2% ${\kappa}-carrageenan$ ($64.26{\pm}0.49$) was significantly higher than control ($40.24{\pm}8.27$) (p < 0.05). RPMs of extender with 0.1%, 0.2% ${\kappa}-carrageenan$ ($57.64{\pm}6.34$, $56.47{\pm}1.35$) were significantly higher than the other groups (p < 0.05). Acrosome integrity was measured by dyeing to PSA-FITC with an epifluorescence microscope. Normal acrosome ratio of extender with 0.5% ${\kappa}-carrageenan$ ($61{\pm}8.03$) was higher than the other groups (p < 0.05). Apoptosis was measured with a FACSCalibur flow cytometer using FITC (FITC Annexin V Apoptosis Detection Kit). Treated groups of ${\kappa}-carrageenan$ of 0.1% ($0.81{\pm}0.05$), 0.2% ($0.85{\pm}0.05$) were significantly higer (p < 0.05) than control. Modified SYBR/PI staining was used for determination of viability and DCF staining was used for evaluation of ROS. Viability and ROS were not significantly different from other groups. In conclusion, adding a certain concentration of carrageenan to the extender of cryopreservation, carrageenan contributes to the improvement of the sperm motility, acrosome integrity and prevention of apoptosis.

해금강과 금강상류 남대천 및 금산천 주변으로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성 (Isolation and Determination of Microbiological Characteristics of Unrecorded Wild Yeasts from Waters and Soils of Haegeumgang in the Southern Sea, and from Namdaecheon and Geumsancheon Upstream of Geumgang, Korea)

  • 남해안과 금강상류 주변환경의 야생효모들의 효모 종 분포특성을 조사하고 국내 미보고된 효모들을 선별하여 이들의 균학적 특성들을 알아보고자 남해안의 해금강과 금강상류인 전북 무주의 남대천 및 금산의 금산천 주변의 물과 토양들로부터 야생효모들을 분리, 동정하였다. 해금강 주변 시료 35점에서 22종 52균주의 야생효모들을 분리, 동정하였고 무주의 남대천 주변 물과 토양 30점에서는 24 종 43균주의 야생효모들을, 금산의 금산천변 시료 60점에서 45종 79균주의 야생효모들을 분리, 동정하였다. 이들 지역에서 분리, 동정한 야생효모들중 Exobasidium rhododendri HHG10-5 (NNIBR2022633FG1), Udeniomyces pyricola NDC29-1 (NNIBR2022633FG2), Pichia scaptomyzae BAC2-3 (NNIBR2022633FG4), Diddensiella caesifluorescens GSC2-2 (NNIBR2022633FG5) 등 4균주들이 국내 미기록 야생효모들로 최종 선별되었다. 이들의 균학적 특성을 조사한 결과Udeniomyces pyricola NDC29-1 과Diddensiella caesifluorescens GSC2-2 균은 구형이었고 Exobasidium rhododendri HGG10-5 와 Pichia scaptomyzae BAC2-2 균들은 난형이었으며 Pichia scaptomyzae BAC2-3 균주만이 포자를 생성하였다. Udeniomyces pyricola NDC29-1 균주 외에는모두 vitamin-free배지에서도생육이아주양호하였고Pichia scaptomyzae BAC2-3, Diddensiella caesifluorescens GSC2-2 균주는 40% glucose 를함유한 YPD배지에서도생육하는내당성균이었다. Udeniomyces pyricola NDC29-1 균주를 제외한 나머지 균들은 Ca 등의 중금속등에 대하여 강한 내성을보였다.

4갈락토오즈 측정을 위한 전기화학적 바이오센서 개발 (Development of electrochemical biosensor for determination of galactose)

  • 본 연구에서는 갈락토오즈의 현장검사(point-of-care testing, POCT)를 위한 일회용 갈락토오즈 바이오센서 개발에 관해 논하고자 한다. Galactose oxidase(GAO)와 horseradish peroxidase(HRP) 두 효소를 0.05 M phosphate 완충용액에 용해시킨 후 스크린 프린팅(screen printing) 방법으로 제작한 전극위에 고정화하여 센서를 제작하였다. 이렇게 제작된 센서를 이용하여 $100{\mu}L$ 이하의 시료를 이용하여 갈락토오즈를 90초 이내에 측정하였다. 전극에서의 반응을 최적화하기 위하여 GAO 효소가 가장 우수한 활성을 나타내는 pH 7.0 완충용액을 이용하여 GAO와 HRP 효소를 1:1로 고정화하고, 1mM 1,1'-ferrocene-dimethanol 전자전달매개체를 도입하여 센서를 제작하였다. 유리 탄소전극의 경우 100 mV (vs Ag/AgCl), 스크린 프린트된 전극의 경우 -100 mV(vs Ag/AgCl)의 인가전압을 적용하였다. 본 연구에 의해 개발된 센서는 $0{\sim}400{\mu}M$의 갈락토오즈 농도에 대하여 우수한 직선성($r^2$ = 0.997, S/N = 3)을 나타내었고 낮은 인가전압을 적용하여 갈락토오즈를 측정하므로, ascorbic acid, uric acid 그리고 acetaminophen과 같은 방해물질의 영향을 최소화 할 수 있었다. 또한 갈락토오즈 표준 용액에 대하여 만족할 만한 재현성을 나타내었다(CV = 5%).

  • Observations were made on the blood picture of total 196 heads of healthy Korean cattles, including 98 males and females in the purpose of determination of blood chemical values and their sex differences and seasonal variations during one year period from December, 1963 to November, 1964. The blood sampling were scheduled by random in four different seasons and the sample size of both sex included in each season were designated to be same size. The ranges, averages or mean values of the blood glucose, total serum protein, serum globulin, serum albumin, total non-protein nitrogen, blood urea nitrogn, total serum cholesterol, serum inorganic phosphorus and serum calcium were determined in this studies and their respective standard deviation, standard error of means, sex differences and seasonal variations were as follows. 1. The blood glucose values for the male ranged from 32.8 to 70.0 mg/100cc. with a mean of $49.781{\pm}0.823mg/100cc$; for the female the range was 32.0 to 64.0mg/100cc. with a mean of $47.235{\pm}0.782mg/100cc$. Sex difference showed significant at 5% level and seasonal variation was highly significant at 1% level. 2. The total serum protein values for the male ranged from 5.61 to 8.83 gm/100cc with a. mean of $7.366{\pm}0.062gm/100cc$; for the female ranged from 5.53 to 8. 43 gm/100cc. with a mean of $6.832{\pm}0.063gm/100cc$. Sex difference and seasonal variation was not significant. 3. The serum globulin values for the male ranged from 2.97 to 4.78 gm/100cc. with a mean of $3.961{\pm}0.039gm/100cc$.; for the female ranged from 2.87 to 4.41 gm/100cc. with a mean of $3.699{\pm}0.037gm/100cc$. Sex difference showed highly significant at 1% level and seasonal variation was not significant. 4. The serum albumin values for the male ranged from 2.58 to 4.21 gm/100cc. with a mean of $3.405{\pm}0.029gm/100cc$.; for the female ranged from 2.39 to 4.10 gm/100cc. with a mean of $3.204{\pm}0.031gm/100cc$. Sex difference showed highly significant at 1% level and seasonal variation was not significant. 5. The total non-protein nitrogan values for the male ranged from 19.1 to 44.8 gm/100cc. with a mean of $31.166{\pm}0.582mg/100cc$.; for the female the range was 15.2 to 50.5 mg/100cc. with a mean of $28.89.6{\pm}0.673mg/100cc$. Sex difference showed significant at 5% level and seasonal variation was highly significant at 1 % level. 6. The blood urea nitrogen values for the male ranged from 6.4 to 28.3 mg/100cc. with a mean of $13.371{\pm}0.466mg/100cc$.; for the female the range, was 6.0 to 26.9 mg/100cc. with a mean of $13.631{\pm}0.321mg/100cc$. Sex difference was not significant and seasonal variation showed highly significant at 1 % level. 7. The total serum cholesterol values for the male ranged from 60.0 to 238.6 mg/100cc. with a mean of $140.897{\pm}2.826mg/100cc$.; for the female ranged from 50.0 to 243.0 mg/100cc. with a mean of $124.840{\pm}3.553mg/100cc$. Sex difference and seasonal variation showed highly significant at 1% level. 8. The serum inorganic phosphorus values for the male ranged from 3.5 to 7.8 mg/100cc. with a mean of $5.426{\pm}0.096mg/100cc$.; for the female ranged from 3.1 to 8.8 mg/100cc. with a mean of $5.570{\pm}0.128mg/100cc$. Sex difference and seasonal variation showed no significant. 9. The serum calcium values for the male ranged from 7.8 to 12.8 mg/100cc. with a mean of $10.761{\pm}0.102mg/100cc$.; for the female ranged from 8.0 to 13.0 mg/100cc. with a mean of 10. $756{\pm}0.097mg/100cc$. Sex difference was not significant and seasonal variation showed highly significant at 1% level. 10. The age of test group ranged from 2 years to 6 years in both sex and the averageage were, $4.45{\pm}0.114$ years in male and $4.50{\pm}0116$ years in female. Sex difference and seasonal variation of age were not found to be significant.

Streptomyces coelicolor A3(2)로 부터 $\beta$-Glucosidase 유전자 클로닝 및 재조합 효소의 특성 (Cloning of $\beta$-Glucosidase Gene from Streptomyces coelicolor A3(2) and Characterization of the Recombinant $\beta$-Glucosidase Expressed in Escherichia coli)

  • Streptomyces coelicolor A3(2)의 $\beta$-glucosidase 유전자를 분리하여 대장균에서 발현하여 특성을 조사하였다. 최적 활성을 나타내는 온도는 pH 5에서는 $20^{\circ}C$, pH 6에서는 $60^{\circ}C$에서 높은 활성을 나타냈다. pH에 따른 활성은 pH 3 이하와 pH 9 이상의 범위에서는 낮은 활성을 나타냈으며 pH 7에서 가장 높은 활성을 나타냈다. $\alpha$-pNPG($\rho$-nitrophenyl-$\alpha$-D-glucopyranoside), $\beta$-pNPG ($\rho$-nitrophenyl-$\beta$-D-glucopyranoside), $\beta$-pNPF($\\rho$-nitrophenyl-$\beta$-D-fucopyranoside)는 pH 3-10까지 비슷한 활성을 나타냈으며, $\alpha$-pNPG가 pH 7에서 다소 높은 활성을 보였다. $\beta$-pNPGA는 pH 5-9까지 높은 활성을 나타냈으며, 특히 pH 9에서 3배 이상의 높은 활성을 나타냈다. 기질 $\alpha$-pNPG, $\beta$-pNPG, $\beta$-pNPF의 온도에 따른 활성변화는 $\beta$-pNPF의 활성이 $60^{\circ}C$에서 증가하였고, $\beta$-pNPGA는 $30-50^{\circ}C$까지 활성이 증가하여 $50^{\circ}C$에서 최대활성을 나타내었다. 당화 flavonoid를 이용한 기질특이성의 상대활성은 daidzin, glycitin, genistin, 순으로 나타났으며 esculin과 apigenin-7-glucose는 기질로 사용하지 않았다. $\beta$-Glucosidase 활성은 EDTA, DTT에 의해 억제되었으며, $MnSO_4$, $CaCl_2$, KCl, $MgSO_4$에 의해 증가하였고, 특히 Mn이온에 의해 증가하였다. $CuSO_4$, NaCl에 의해 효소활성이 저해되었으며, 특히 $ZnSO_4$의 경우 효소활성이 강하게 억제되었다.
