N-Acetyl-D-Glucosamine Kinase Promotes the Axonal Growth of Developing Neurons

  • N-acetyl-D-glucosamine kinase (NAGK) plays an enzyme activity-independent, non-canonical role in the dendritogenesis of hippocampal neurons in culture. In this study, we investigated its role in axonal development. We found NAGK was distributed throughout neurons until developmental stage 3 (axonal outgrowth), and that its axonal expression remarkably decreased during stage 4 (dendritic outgrowth) and became negligible in stage 5 (mature). Immunocytochemistry (ICC) showed colocalization of NAGK with tubulin in hippocampal neurons and with Golgi in somata, dendrites, and nascent axons. A proximity ligation assay (PLA) for NAGK and Golgi marker protein followed by ICC for tubulin or dynein light chain roadblock type 1 (DYNLRB1) in stage 3 neurons showed NAGK-Golgi complex colocalized with DYNLRB1 at the tips of microtubule (MT) fibers in axonal growth cones and in somatodendritic areas. PLAs for NAGK-dynein combined with tubulin or Golgi ICC showed similar signal patterns, indicating a three way interaction between NAGK, dynein, and Golgi in growing axons. In addition, overexpression of the NAGK gene and of kinase mutant NAGK genes increased axonal lengths, and knockdown of NAGK by small hairpin (sh) RNA reduced axonal lengths; suggesting a structural role for NAGK in axonal growth. Finally, transfection of 'DYNLRB1 (74-96)', a small peptide derived from DYNLRB1's C-terminal, which binds with NAGK, resulted in neurons with shorter axons in culture. The authors suggest a NAGK-dynein-Golgi tripartite interaction in growing axons is instrumental during early axonal development.

성인의 연령에 따른 건강기능식품 섭취실태와 정보 요구도 분석 (Intake Pattern and Needs Assessment for the Development of Web-Contents on Health Functional Foods according to Age of Adults)

  • This study was done to analyze the consumption patterns of health functional food (HFF) as well as to perform needs assessment for the development of web-contents on HFF according to age of adults. The subjects were 238 male and female adults, divided into 4 groups by their age. This study collected all information by self-administrated questionnaires. The awareness on HFF was high in the older adults. The younger adults showed more negative responses to reliability and safety on HFF. The main reason for the consumption of HFF was to supplement nutrients and to prevent diseases. The main types of HFF consumed by adults were nutritional supplementary food, red ginseng products, and glucosamine products. There was higher consumption of nutritional supplementary food in the younger adults and glucosamine products in the older adults. Internet users had low level of satisfaction, with tendency to complain poor contents, reliability, difficulties in searching as problems of the pre-existing HFF websites. As useful methods for provision of information on HFF, most adults wanted general information, articles written by experts and videos. They also wanted to know the safety and side effects of HFF. Requirement of contents composition were various in-depth information, clear indication of citation, fresh updated data while that of display composition was easily-findable, uncomplicated, allowing mutual exchange of communication through bulletin board. These results can be used as basic data that reflect the consumer's needs for developing HFF web-contents according to age of adults.

Glucosamine increases macrophage lipid accumulation by regulating the mammalian target of rapamycin signaling pathway

  • Elevated blood glucose is associated with an increased risk of atherosclerosis. Data from the current study showed that glucosamine (GlcN), a normal glucose metabolite of the hexosamine biosynthetic pathway (HBP), promoted lipid accumulation in RAW264.7 macrophage cells. Oleic acid- and lipopolysaccharide (LPS)-induced lipid accumulation was further enhanced by GlcN in RAW264.7 cells, although there was no a significant change in the rate of fatty acid uptake. GlcN increased acetyl CoA carboxylase (ACC), fatty acid synthase (FAS), scavenger receptor class A, liver X receptor, and sterol regulatory element-binding protein-1c (SREBP-1c) mRNA expression, and; conversely, suppressed ATP-binding cassette transporter A1 (ABCA-1) and ABCG-1 expression. Additionally, GlcN promoted O-GlcNAcylation of nuclear SREBP-1 but did not affect its DNA binding activity. GlcN stimulated phosphorylation of mammalian target of rapamycin (mTOR) and S6 kinase. Rapamycin, a mTOR-specific inhibitor, suppressed GlcN-induced lipid accumulation in RAW264.7 cells. The GlcN-mediated increase in ACC and FAS mRNA was suppressed, while the decrease in ABCA-1 and ABCG-1 by GlcN was not significantly altered by rapamycin. Together, our results highlight the importance of the mTOR signaling pathway in GlcN-induced macrophage lipid accumulation and further support a potential link between mTOR and HBP signaling in lipogenesis.

Gelatin-Chondroitin-Glucosamine Scaffold에 접종한 인간지방조직-유래 중간엽 줄기세포의 연골형성 (Chondrogenesis of Human Adipose Tissue Derived Mesenchymal Stem Cells (ATMSCs) Seeded in Gelatin-Chondroitin-Glucosamine Scaffold)

  • 본 실험은 transforming growth factor-${\beta}1$(TGF-${\beta}1$)이 첨가된 chondrogenic induction medium(CIM)을 이용하여 인간지방조직에서 유래된 중간엽 줄기세포(human adipose tissue-derived mesenchymal stem cells, ATMSCs)의 연골형성능과 gelatin-chondroitin-glucosamine scaffold(GCG-scaffold)에 접종시킨 ATMSCs의 연골형성능을 알아보고자 수행하였다. ATMSCs와 생쥐 chondrocyte를 기본배양액과 TGF-${\beta}1$이 첨가되지 않은 CIM1 및 TGF-${\beta}1$이 첨가된 CIM2에서 배양하여 연골형성능을 비교하였다. ATMSCs의 연골형성은 gelatin scaffold(G-scaffold)와 GCG-scaffold를 사용하여 glycosaminoglycan(GAG) 합성과 조직화학적 염색으로 연골기질형성 여부를 확인하였다. 펠렛 배양에서 ATMSCs와 chondrocyte의 GAG합성은 대조군에서 14일의 배양기간 동안 약간 증가하였으나, CIM1과 CIM2배양군은 배양 14일에 대조군에 비해크게 증가하였으며, CIM2 배양군에서 가장 높았다. 그러나 연골기질은 배양 14일에 CIM2 배양군에서만 Safranin O와trichrome에 의해 염색되었다. Well plate에서 배양된 ATMSCs의 증식은 모든 배양군에서 배양 10일까지 계속적인 세포증식이 일어났으며, 대조군에 비해 CIM 배양군이 높았다. ATMSCs 접종 후 플라스크나 scaffold의 세포부착률은 배양시간이 길어짐에 따라 모든 배양군에서 증가하였고, 플라스크에 비해 scaffold에서 더욱 높게 나타났다. Scaffold에 ATMSCs 접종후 GAG 합성은 28일의 배양기간 동안 대조군에서는 별 변화가 없었으나, CIM1, CIM2 배양군은 대조군보다 GAG합성이 증가되었다. CIM2 배양군에서 GAG 합성이 매우 높게 나타났다. 또한 G-scaffold보다 GCG-scaffold에서 GAG 합성이 약간 높게 나타났다. 조직화학적으로 관찰한 ATMSCs의 연골기질형성도 CIM2 배양군의 GCG-scaffold에서 가장높게 나타났다. 본 실험의 결과들을 종합해 볼 때, 펠렛 배양에서 ATMSCs는 생쥐 chondrocyte보다 낮은 연골형성능을 보였고, CIM2 배양군에서만 연골기질이 형성된 것으로 보아 TGF-${\beta}1$이 연골분화에 중요한 요소로 작용한 것이라 사료된다. G-scaffold는 효과적인 연골분화 환경을 제공해 줌으로써 ATMSCs의 세포부착률과 GAG 합성을 증가시키는 것으로보여지며, G-scaffold보다 GCG-scaffold에서 많은 연골기질이 형성된 것은 GCG-scaffold에 첨가된 chondroitin과 glucosamine이연골기질 형성을 촉진시키는 물질로 작용한 것으로 사료된다.

A Docking Study of UDP-N-Acetylglucosamine Enolpyruvyl Transferase from Haemophilus influenzae in Complex with Inhibitors

  • UDP-N-acetylglucosamine enolpyruvyl transferase (MurA; EC 2.5.1.7) catalyzes the first committed step of peptidoglycan biosynthesis in bacteria, i.e., transfer of enolpyruvate from phosphoenolpyruvate to UDP-N-acetyl-glucosamine. Because the crystallization condition contained a high concentration of ammonium sulfate, our inhibitor binding studies were not successful. Therefore, we employed a docking approach to investigate the inhibitor binding. Our results will be useful in structure-based design of specific inhibitors of MurA for antibacterial discovery.

Chitosan이 사과 겹무늬썩음병균 Botryosphaeria dothidea의 생육에 미치는 영향

  • To examine the potential utilization of chitosan, the biodegradable natural polymer, as a control agent of apple white rot caused by Botryosphaeria dothidea in a new control measure by coating it on the diseased branches, the various antifungal activities of chitosan was investigated. Chitosan showed significant inhibitory effect on the mycelial growth of B. dothidea, along with the morphological changes including hyphal swelling and ultrastructural changes on solid PDA medium. In liquid PD broth medium, the chitosan showed more significant effect on the growth of B. dothidea also forming cell clusters indicating affection on the hyphal extension. The growth of B. dothidea was inhibited more than 90% at the concentration of 1.0 mg/ml. Chitosan also detained the spore germination and induced the morphological change of germ tubes. Glucosamine, monomer of chitosan, did not affect on the growth of B. dothidea indicating the antifungal activity was caused by chitosan polymer.

Isolation and Characterization of Epidermal Mucus from Hirudo nipponia

  • The epidermal organ of the leech contains a complex glycoprotein molecule of mucus. The mucus excreted from annelids plays Significant role in protection against desiccation and parasites. Mucus from the Korean native leech, H. nipponia, was investigated for biochemical characteristics for possible development of biomaterials of cosmetic and pharmaceutical agents. The leech skin mucus was heavily glycosylated mucin-like protein with a high molecular weight comprised 80% carbohydrate and 20% protein. Threonine, serine, and glycine were the major components of the isolated protein and these accounted for 50% of total amino acids. The carbohydrate portion contained glucosamine, galactosamine. galactose, glucose. mannose and sialic acid in oligosaccharide form linked with threonine and serine residues of the glycoprotein.

Purification and Characterization of Two Isolectins with Arginase Activity from the Lichen Xanthoria parietina

  • Two glycoproteins were purified and biochemically characterized from the lichen X. parietina. Both behaved as enzymes with arginase activity and haemaglutinins. Secreted arginase (SA) contained galactose and glucose in the saccharide moiety and an isoelectric point of 4.54. The algal binding-protein (ABP) had N-acetyl-glucosamine and glucose as glycosidic residues and an isoelectric point of 3.53. Both proteins had the same molecular mass (58.6 kDa) and the same qualitative amino acidic composition. The results allowed us to consider these glycoproteins as isolectins, which have significant physiological roles in the relationship between photobiont and mycobiont of symbiotic association.

Comparative lectin binding patterns of Cochlodinium polykrikoides Margalef

  • Four different FITC-conjugated lectins were used to visually evaluate lectin binding activity by optical staining quality using confocal laser scanning microscopy (CLSM) of Cochzodinium polykrikoides in nature (wild type) and culture (cultured type). Cells from the field and cultures treated with ConA fluoresced only at the outer cell wall, and the abundance and distribution of the fluorescent signal were similar. Treatment with PWM and HPA did not elicit fluorescence at the cell surface, but the wild type exposed to HPA showed greater binding than did the cultured cells, possibly due to greater concentrations of glucosamine. The wild type cells treated with LBL lectin showed a strong green fluorescence on the cell surface, whereas cultured cells did not. Signal intensity and abundance were greater than for any other lectins tested in this study. These results suggest that wild type and cultured type are significantly different based on surface sugar production. In particular, the wild type cells apear richer in galactosamine-like moieties. Neither glucose nor mannose-like moieties were present in either wild types or cultured cells.

  • Realizing the scope of utilizing by-products of silk cocoons by applying appropriate methods is the immediate crave to optimize returns. The nutritive value of pupae suits for human diet, feed for poultry, carps, fish, rabbits, piggery and dogs. The pupal skin, fat, oil, cocoon palade have applications in oleo chemical, soap, glycerin, cosmetic, artificial fibres, membranes and n-triacontanol isolation. The pupal proteins Chitin, Shinki fibroin, Serrapeptidase, glucosamine are latent precursors of post surgical, anti-carcinogenic, anti-inflammative, anti-bacterial, anti-histaminic, gastric, hepatitis, pancreatitis, leukocytopenia, neurological, ophthalmic, blood pressure, cardiac and diabetic medicines and for preparation of vitamins A, E and K. The silk and its proteins sericin and fibroin are potentially used for wound healing, diabetes, impotence, sinusitis, arthritis, edema, cystitis, epididymitis, tissue regeneration, cancer, post-surgical trauma and used as anti-oxidatives, bio-adhesives, ultra violet screens and bio-active textiles. The waste cocoons can be used in making art crafts like garlands, carpets, overcoats, decoratives and greeting cards. The in-depth research towards utility optimization and make aware this reality to sericulturists, reelers, weavers, traders, entrepreneurs, policy makers etc., is the upright want of the today's Sericulture industry.
