대전기영동법(帶電氣泳動法)에 의한 유청단백질(乳淸蛋白質)에 관(關)한 연구(硏究) (Studies on Milk Protein by Zone Electrophoresis)

  • 본연구(本硏究)에서는 한우유(韓牛乳)와 Holstein cow milk의 단백질(蛋白質)을 Zone electrophoresis에 의하여 분석(分析)하여 그 결과(結果)를 다음과 같이 얻었다. (1) 한우유(韓牛乳)와 Holstein cow milk의 이화학적(理化學的) 성질(性質)이 Table 1, Table 2에 나타나 있다. (2) 한우유(韓牛乳)의 유청단백질(乳淸蛋白質)의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.74mg %, Alpha-1 globulin 1.19mg%, Alpha-2 globulin 0.68mg %, Beta globulin 2.57mg %, Gamma globulin 0.93mg %이었다. (3) Holstein cow milk protein의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.23mg %, Alpha-1 globulin 1.93mg %, Alpha-2 globulin 0.86mg %, Beta globulin 2.56mg %, Gamma globulin 1.74mg %를 얻었다. (4) 한우(韓牛)(female)와 Holstein cow(male)와의 교잡종인 F-1인 유청단백질(乳淸蛋白質) 분포(分布)는 Lactoalbumin 1.30mg %, Alpha-1 globulin 1.58mg %, Alpha-2 globulin 2.46mg %, Beta globulin 1.39mg %이었고 F-2의 그것은 Lactoalbumin 1.57mg %, Alpha-1 globulin 1.74mg %, Alpha-2 globulin 2.19mg %, Beta globulin 1.17mg %, F-4에서는 Lactoalbumin 2.46mg %, Alpha-1 globulin 1.24mg %, Alpha-2 globulin 1.57mg %, Beta globulin 1.74mg %를 각각(各各) 얻었다.

Acyl화에 의한 녹두 Whole Globulin의 특성에 관한 연구 (Properties of Acylated Mungbean Whole Globulin)

  • The objective of this study was to improved the limited functional characteristics of mungbean whole globulin. The mungbean whole globulin was acylated with succinic and acetic anhydride, and the functional properties of acylated protein were investigated, The results obtained ware as follows. 1. The UV-absorption spectra of acylated whole globulins with that of the succinylated 74% whole globulin as large blue shift of the absorption maximum and minimum wavelength from 275 nm to 269 nm, respectively. 2. The mobility of acylated whole globulin were increased on PAGE pattern, and degree of mobility was particularly remarkable in case of succinylation, 3. The water absorption capacity of whole globulin was increased by acylation. The most increased rate of whole globulin was 174,02% from succinylated 74%. The oil absorption capacity of whole globulin was increased by acylation The most increased rate of whole globulin was 165.41% from acetylated 81.77%. 4. The bulk density of whole globulin was decreased by acylation. and the greater the extent of acylation, the smaller the bulk density. 5. The foaming capacity and stability of whole globulin was increased by acylation, and remarkably high in 74% succinylated whole globulin. In contrast, however, the foaming capacity and stability of native and acylated whole globulin were decreased by heat treatment.

스트레스 상태의 고양이에서 lipopolysaccharide 투여가 혈중 감마글로불린 생산에 미치는 영향 (Effect of lipopelysaccharide administration on the production of serum ${\gamma}$-globulin in stressed domestic cats)

  • The activated immune systems with lipopolysaccharide(LPS) were evaluates by ${\gamma}$- globulin levels in domestic cats. Especially, cats were in a stress condition, which was unfamiliar environmental change. Effect of continuous unfamiliar environmental stressor was a diminution of production of serum total globulin auld ${\gamma}$- globulin on feline infectious peritonitis(FIP) vaccination. The ${\gamma}$-globulin production on FIP vaccination more increased in LPS treated 77ups than non-LPS treated group. Also. the ${\gamma}$-globulin production on FIP vaccination was maintained in LPS treated group under stress condition. These results imply that that the ${\gamma}$-globulin can be produced under a stress condition by mitogen like as LPS.

당뇨병 환자의 혈청단백 전기영동 분획 비교 (Comparison of Protein Electrophoresis Fractions in Diabetes Patient Serum)

  • We compared the serum protein electrophoresis patterns in sera of diabetes patients (n=166) which showed more than 150 mg/dL of glucose. The concentrations of total protein, albumin, ${\alpha}1$-globulin, ${\alpha}2$-globulin, ${\beta}$-globulin and ${\gamma}$-globulin in sera of patients were $6.3{\pm}1.2g/dL$, $40.3{\pm}6.4%$, $4.9{\pm}2.3%$, $13.5{\pm}3.8%$, $15.5{\pm}3.9%$, and $25.9{\pm}5.8%$, respectively. The frequency of patients showed lower levels of total protein, albumin, ${\alpha}1$-globulin were 48.5%, 96.4%, and 33.7%. The frequency of patients showed higher levels of ${\alpha}2$-globulin, ${\beta}$-globulin, and ${\gamma}$-globulin were 56.6%, 47.0%, and 90.4%, respectively. This data suggests that the high concentration of glucose in blood may damage the kidney and liver. Also, the decrease of albumin and ${\alpha}1$-globulin and increase of ${\alpha}2$-globulin, ${\beta}$-globulin and ${\gamma}$-globulin shows that the concentration of glucose in blood will have an effect on the variation of serum protein electrophoretic patterns.

비색법에 의한 한우 혈청단백질의 분획정량 시험 (Studies on Separation and Determination of Korean Bovine Serum Protein by Colorimetric Method)

  • Serum Samples from adult of Korean cattles including 40 females and 20 males were analyzed by sodium salt precipitation and colorimetric method in the purpose of the determination of total serum protein, albumin, globulin, ${\alpha}$-globulin, ${\beta}$-globulin and ${\gamma}$-globulin. The results obtained arc summarized as follows: 1. Mean value of total serum protein showed a slight variation from 7.6%, and its regional and sex differences were not found to be significant. 2. Contents of albumin in serum showed lower level than that of globulin as low level of A/G ratio 0.4 in proportion. 3. Contents of Serum ${\alpha}$-globulin showed 1.4w/v% and $1.51{\pm}0.46$w/v% in each group of female, and $1.31{\pm}0.26$w/v%, in the group of male. 4. Contents of serum ${\beta}$-globulin showed 1.74w/v%, 1.95w/v%, in each group of female, and 1.82w/v% in the group of male. 5. Contents of serum ${\gamma}$-globulin showed 2.32w/v%, 2.30w/v% in each group of female, and 2.30w/v%, in the group of male.

Effect of Enzymatic Hydrolysis of 7S Globulin, a Soybean Protein, on Its Allergenicity and Identification of its Allergenic Hydrolyzed Fragments Using SDS-PAGE

  • This study was undertaken to investigate the effect of peptic and chymotryptic hydrolyses of 7S globulin, the major allergen of soybean protein, on its allergenicity, as measured by enzyme linked immunosorbent assay (ELISA), and to identify the allergenic hydrolyzed fragments of 7S globulin using SDS-PAGE. When 7S globulin was hydrolyzed by pepsin, the allergenicity was reduced by over 50%. However, the allergenicity of 7S globulin reduced by peptic hydrolysis was recovered in the sera from 5 out of 10 patients following sequential chymotryptic hydrolysis. Two fragments, with molecular weights 20-25 and 13-16 kDa, among the hydrolysate of 7S globulin by sequential pepsin and chymotrypsin showed reactivity with sera from 10 soybean-allergenic patients. As a result of the theoretical hydrolyses of ${\beta}$-conglycinin, which is a major protein of 7S globulin, it is suggested that the 20-25 kDa fragments were the fragments of the ${\alpha}$-subunit of ${\beta}$'-conglycinin and that the 10-16 kDa fragments were from the ${\alpha}$'-subunit.

The Concentration of Serum Lipids and Protein Electrophoresis Fractions in Thyroid Disease Patients

  • We analyzed and compared the concentration of total cholesterol (CHOL), high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol and triglyceride (TG) in serum and the serum protein electrophoresis fractions of thyroid disease patients. In comparison with the average of reference, our data showed that the average concentration of CHOL, LDL cholesterol and TG in hyperthyroidism patients were decreased significantly, but HDL cholesterol was increased significantly. In hypothyroidism patients, CHOL, HDL cholesterol, LDL cholesterol and TG were all increased significantly. In comparison of the concentration of lipids in each patient to reference range, 28.3% of hyperthyroidism patients showed abnormally low level of total cholesterol. In the patients with hypothyroidism, the percentage of patients showed abnormally high level of CHOL, HDL cholesterol, LDL cholesterol and TG were 37.7%, 10%, 68.8% and 49.1%, respectively. In our studies of serum protein electrophoresis, the average of ${\alpha}_2$-globulin and $\gamma$-globulin in hyperthyroidism patients were increased and $\beta$-globulin was decreased significantly. In hypothyroidism patients, the average of $\gamma$-globulin was increased and $\beta$-globulin was decreased significantly. In comparison of protein fractions of each patient to reference range, 38.3% and 50.0% of hyperthyroidism patients showed abnormally high levels of ${\alpha}_2$-globulin and $\gamma$-globulin, but 73.3% of patients showed abnormally low level of $\beta$-globulin. In hypothyroidism patients, 70.4% of patients were abnormally decreased in $\beta$-globulin and 63.9% of patients were abnormally increased in $\gamma$-globulin. These data suggest that the concentrations of CHOL, HDL cholesterol, LDL cholesterol and TG are not critical data for clinical interpretation of hyperthyroidism, but the levels of them are useful for interpretation of hypothyroidism patients. Our results of serum protein electrophoresis suggest that the concentration of serum protein electrophoresis fractions can be useful to understand the thyroid disease.

Interferon 및 gamma-globulin이 실험적 Pneumocystis carinii 폐염의 치료에 미치는 영향 (Study on the therapeutic effects of interferon and gamma-globulin in experimental Pneumocustis curinii pneumonia)

  • This study was performed to observe the therapeutic effects of interferon-gamma ($IFN-{\gamma}$) and gamma-globulin (${\gamma}-globulin$) in experimental Pneumocystis carinii pneumonia of immune suppressed mice. After 9 weeks, trimethoprim-sulfamethoxaBole(TMP-SMZ; 10~50 mg/mouse/day), mouse $IFN-{\gamma}(5{\times}10^4$ units/mouse/day) and mouse ${\gamma}-globulin$(20 mg/mouse/day) were administered to the mice for 3 weeks by the experimental group. The therapeutic efficacy was evaluated by body weights, histopatholo단ic and electron microscopic findings of the lungs, and number of p. carinii cysts by Gomori's methenamine silver stain. Body weights of the mice were significantly increased in the group of combination therapy of TMP-SMZ with $IFN-{\gamma}{\;}or{\;}{\gamma}-globulin$, and in the group of TMP- SMZ treatment(p<0.05), however, little effect was found in the group of T-globulin alone. Histopathologic 6ndings of p. carinii pneumonia were much improved in the group of combination therapy of TMP-SMZ with $IFN-{\gamma}$. Treatment with either TMP-SMZ or $IFN-{\gamma}$ significantly reduced the number of cysts in the p. carinii pneumonia, but {\gamma}-globulin alone was ineffective. In electron microscopic findings of p. carinii pneumonia, the number of trophozoites and cysts were reduced by treatment with either TMP-SMZ or $IFN-{\gamma}$, and most of the cysts were empty or containing one or two intracystic bodies. The present results suggested, that combination therapy of TMP-SMZ with $IFN-{\gamma}$ had synergistic effects in treatment of P carinii pneumonia in experi- mental mice.

집 토끼의 Blood Cholesterol Level에 미치는 몇가지 식물성(植物性) 식용유(食用油)의 영향 (The Effects of Dietary Vegetable Oils on the Blood Cholesterol Level of Rabbit)

  • 집토끼를 일정한 조건 하에서 기본 식이에 식물성 식용유를 첨가하여 콜레스테롤에 어떠한 영향을 주는가를 조사하였다. 1. 체중 변화에 있어서 실험군의 평균 체중의 변화는 대조군(對照群)이 16.3g/day 이었고 들깨기름 급여군은 18.2g/day로 가장 성장률이 좋았다. 2. 간장의 무게는 대조군(對照群)이 30.35g이고 들깨 기름 및 미강유 급여군의 무게는 각각 38.25과 39.54g으로 나타났다. 3. 혈청 콜레스테롤 값은 대조군(對照群)이 71.55mg/100 ml 이었고 참깨기름 급여군은 112.0mg/100 ml로 가장 높게 나타났다. 4. 간장 콜레스테롤 값은 대조군(對照群)과 들깨 기름 급여군이 각각 255.0 mg/100 ml 이었고 콩기름 급여군이 317.5 mg/100 ml로 가장 높게 나타났다. 5. 혈청 단백질에 있어서 총(總) 단백질(蛋白質)은 큰 차이가 없었으나 alpha-1-globulin에 있어서 대조군(對照群)은 0.60%인데 콩기름 및 미강유 급여군은 각각 0.35와 0.32%로 상당히 낮게 나타났고 alpha-2-globulin에 있어서는 대조군(對照群)이 0.68%인데 미강유 급여균이 0.26%로 나타났다. 6. 혈당은 대조군(對照群)이 40.34mg/100 ml인데 들깨 기름 급여군만이 77.0mg/100ml 로서 가장 높았고 다른군은 모두 낮았다. 7. 혈청(血淸) 콜레스테롤 값과 alpha-1-globulin 및 간(刊) 콜레스테롤 값 과 gamma-globulin 사이에 정 상관성을 보였고 혈당과는 별 상관이 없게 나타났다.

대두(大豆) 단백질(蛋白質)에 관(關)한 연구(硏究) 제2보[第二報]-7S Globulin중의 복합단백질(複合蛋白質)의 분리(分離) 및 그 구성(構成) Subunit에 대하여 (Studies on Soybean Protein [Part ll]-Isolation and Subunit Composition of Multiple 7S Globulins-)

  • 대두(大豆)(Glycine max cultivar Gwang-gyo)의 각 성숙시기(成熟時期)에 나타나는 7S globulin을 분리하여 Davis 방법(方法)에 의한 전기영동과 PAWU용매에 의해서 유리(遊離)되는 그들의 subunit를 전기영동한 결과 7S globulin중의 복합단백질간(複合蛋白質間)에는 그 구성(構成) subunit에 유사성(類似性)이 있음을 시사하였다. 7S globulin의 복합단백질(複合蛋白質)을 DEAE-Sephadex A-50으로 크로마토그라피하여 분리하였다. 이때 pH 7.6의 인산완충액(燐酸緩衝液)에서 NaCl의 농도구배(濃度句配)가 0.28M부터 0.40M 사이에서 두 개의 분획(分劃)으로 분리되었다. 이들 명(名) 단백질(蛋白質)의 subunit를 5M urea와 1% SDS로 유리(遊離)시켜 7.5% acrylamide-PAWU gel과 5.6% acrylamide-SDS gel에서 전기영동하였다. 그 결과 subunit의 하전량(荷電量)에 의해서 분리되는 PAWU gel전기영동에서 7S globulin이 5개의 주 분리대로 분리되고 그중 2개의 분리대가 7S-A globulin과 7S-B globulin에 공유(共有)되어 있었다. 또 subunit의 분자량(分子量)에 따라서 분리되는 SDS gel 전기영동에서는 7S globulin이 7개의 주 분리대를 나타내는데 그 중에서 3개의 분리대가 7S-A와 7S-B 분획에 공유(共有)되어 있었다. 따라서 7S globulin의 복합단백질간(複合蛋白質間)에는 구성(構成) subunit간(間)에 유사성(類似性)이 있는 것으로 나타났다.
