Comparison of Proximate Composition, Mineral Nutrient, Amino Acid and Free Sugar Contents of Several Panax Species (인삼(Panax)속 식물의 일반성분, 무기성분, 아미노산 및 유리당 함량조성)

  • Chemical components of Panax (P) species were compared. p. species used were Korean white ginseng, Korean, Chinese and Japanese red ginseng (P ginseng), American and Canadian ginseng (P. quinquefolium) , and sanchl ginseng (P. notoginseng). No significant difference in the proximate contents was observed among P. species. Ash, crude lipld and total sugar contents in root of P. notoginseng were found to be relatively lower than those of P. ginseng and P. quinquefolium, but the contents of crude protein and crude fiber were similar among those ginsengs. Mineral nutrient con tents showed a little difference among ginseng species. Total nitrogen contents were slightly higher in P. ginseng than P. quinquefolium and P. notoginseng and Fe and Cu were lower in Chinese and Japanese red ginsengs. Kinds and compositions of amino acids were similar but contents of amino acids were different among ginseng species. Total amino acid contents were 76.3∼83.9 mg/g in P. ginseng 53.8∼60.4 mg/g in p. quinquefolium and 54.9 mg/g in P notoginseng. Free sugar contents were lower in P. notoginseng than P. ginseng or P. quinquefolium. Sucrose accounted for 90∼92% of total free sugar contents with relatively high content in white ginsengs, while sucrose and maltose were 32-36% and 55∼60%, respectively, in red ginseng.

Comparison study of Korean and Chinese ginsengs on the regulation of lymphocyte proliferation and cytokine production (한국 홍삼과 중국 홍삼의 경구 투여가 흰쥐의 림프구 증식과 Cytokine에 미치는 영향에 대한 비교 연구)

  • Red ginseng is a medicinal herb widely used in East-Asia for a long time. Recently there have been a lot of studies about the effect of red ginseng on the immune responses. We investigated the differences between Korean red ginseng and Chinese red ginseng in the lymphocyte proliferation and cytokine production. The rats were divided into 3 groups, KRG(Korean Red Ginseng) group, CRG(Chinese Red Ginseng) group, and the Control group. Experimental groups were administered with Korean and Chinese red ginsengs for three months respectively. Then we obtained the lymphatic cells from spleen and compared the ability of KRG on the lymphocyte proliferation and the cytokine production after mitogen-stimulated culture to CRG. The proliferation of lymphocyte and level of $IL-1{\alpha}$ were significantly increased only in KRG group. There were significant increases in the level of $INF-{\gamma}$ in both KRG and CRG groups. There were no significant differences in the level of IL-2 and $TNF-{\alpha}$. These results indicate that KRG can induce infection-relevant immune responses much faster and higher than CRG. Furthermore, functional activation of CD8+ T-cell may be activated by red ginsengs.

Monoclonal antibody-based enzyme-linked immunosorbent assay for quantification of majonoside R2 as an authentication marker for Nngoc Linh and Lai Chau ginsengs

  • Background: Recent years have witnessed increasing interest in the high amount of ocotillol-type saponin in Panax vietnamensis, particularly in relation to majonoside R2 (MR2). This unique 3%-5% MR2 content impart Ngoc Linh and Lai Chau ginsengs with unique pharmacological activities. However, in the commercial domain, unauthentic species have infiltrated and significantly hindered access to the authentic, efficacious variety. Thus, suitable analytical techniques for distinguishing authentic Vietnamese ginseng species from others is becoming increasingly crucial. Therefore, MR2 is attracting considerable attention as a target requiring effective management measures. Methods: An enzyme-linked immunosorbent assay (ELISA) was developed by producing monoclonal antibodies against MR2 (mAb 16E11). The method was thoroughly validated, and the potential of the immunoassay was confirmed by high-performance liquid chromatography with ultraviolet spectroscopy. Furthermore, ELISA was applied to the assessment of the MR2 concentrations of various Panax spp., including Korean, American, and Japanese ginsengs. Results and conclusions: An icELISA using mAb 16E11 exhibited linearity between 3.91 and 250 ng/mL of MR2, with detection and quantification limits of 1.53 and 2.50 - 46.6 ng/mL, respectively. Based on this study, the developed icELISA using mAb 16E11 could be a valuable tool for analyzing MR2 level to distinguish authentic Ngoc Linh and Lai Chau ginsengs from unauthentic ones. Furthermore, the analysis of the samples demonstrated that Ngoc Linh and Lai Chau ginsengs exhibit a notably higher MR2 value than all other Panax spp. Thus, MR2 might be their ideal marker compound, and various bioactivities of this species should be explored.

Effect of 1-methylcyclopropene Treatment on Extension of Freshness and Storage Potential of Fresh Ginseng (1-methylcyclopropene 처리의 수삼의 신선도 유지 및 저장성 연장 효과)

  • Fresh ginseng has a limited storage life due to the quality change caused by microbial spoilage as well as physiological deterioration. The present study investigated the effects of 1-methylcyclopropene (1-MCP) treatment, an inhibitor of ethylene action, on the microbial growth and quality maintenance of fresh ginseng during storage. Harvested fresh ginsengs were treated with $1{\mu}L{\cdot}L^{-1}$ 1-MCP for 20 hours at $4^{\circ}C$ and then stored at room temperature (RT) for 18 days or low temperature ($4^{\circ}C$) for 160 days. After 18 days of storage at RT, the percentage weight loss in 1-MCP treated fresh ginseng (8.3%) is lower than that of control (10.1%). During long-term storage at $4^{\circ}C$, weight losses were increased slightly until 120 days without difference between non-treated and 1-MCP ginsengs. In contrast, after 120 days of storage at $4^{\circ}C$, higher increase in weight loss was observed in non-treated ginsengs than in 1-MCP treated ginsengs. Respiration rate and ethylene production of fresh ginseng were reduced by 1-MCP treatments at RT. The 1-MCP treatment also resulted in lower microbial population compared to those of non-treated ginsengs at RT. However, in ginsengs stored at $4^{\circ}C$ for short-term (45 days), no differences were noted in weight loss and microbial population between 1-MCP treated and non-treated ginsengs. Major ginsenosides was not changed by 1-MCP treatment during the 7 days of storage at RT. Results suggest that 1-MCP treatment can be used to maintain the freshness of ginseng at room temperature for short term storage and at low temperature for long term storage. 1-MCP treatment could be applied on fresh ginseng to avoid deleterious effect of exogenous ethylene during storage and shipping.

  • In the light of quality comparison the ratio of panaxadiol and panaxatriol contents was assayed in the extract of various ginseng products, whose origin of production and species of the original plants are different. Aglycone compositions of other ginsengs were not comparable with Korean ginseng in their ratio of panaxadiol and panaxatriol contents of dammarane glycosides.

  • Total polysaccharide contents in Panax ginseng roots were evaluated by a spectrophotometry, utilizing the complex formation of ginseng polysaccharide with alcian blue dye in 50 mM ammonium biphosphate, pH 4.2. The total polysaccharide content in red ginseng was about three times higher than that in fresh ginseng when both were extracted with water, and was increased about two times when red ginseng was extracted with an alkaline solution. The determination of total polysaccharide in various parts of ginseng revealed that main roots contained the component more than fine roots. Fresh ginseng sections stained by the dye showed polysaccharide mainly found in cortex and cambium but not in epidermis. Pattern-analysis on total and acidic polysaccharides from fresh and red ginsengs exhibited that the chemical compositions of the polysaccharides extracted from both ginsengs quite differed from each other.

A Comparative Biological Study of the Rhizome and Main Root from Red and White Ginsengs (홍삼 및 백삼의 뇌두, 동체에 대한 생물활성 비교)

  • Comparative biological activities of 70fr methanol extracts from the main roots and rhizomes of both red and white ginsengs were investigated using several in vitro experimental models. The main root of red ginseng and the rhizome of white ginseng strongly inhibited lipld peroxidation of hepatic microsomes induced by the non-enzymatic $Fe^{+}$ / Ascorbate system. The main root and rhizome of red ginseng markedly inhibited the release of G07, GPT and LDH by $CCl_4$-induced cytotoxicity in primary cultured rat hepatocytes as compared with those of white ginseng. And also, the main root of red ginseng showed a slight differentiating activity on HL-60 cancer cell line. The results suggest that the rhizome of ginseng have potential as a source of medicinal crude drug with possible pharmacolobica1 applications .

Gut microbiota-mediated pharmacokinetics of ginseng saponins

  • Orally administered ginsengs come in contact with the gut microbiota, and their hydrophilic constituents, such as ginsenosides, are metabolized to hydrophobic compounds by gastric juice and gut microbiota: protopanxadiol-type ginsenosides are mainly transformed into compound K and ginsenoside Rh2; protopanaxatriol-type ginsenosides to ginsenoside Rh1 and protopanaxatriol, and ocotillol-type ginsenosides to ocotillol. Although this metabolizing activity varies between individuals, the metabolism of ginsenosides to compound K by gut microbiota in individuals treated with ginseng is proportional to the area under the blood concentration curve for compound K in their blood samples. These metabolites such as compound K exhibit potent pharmacological effects, such as antitumor, anti-inflammatory, antidiabetic, antiallergic, and neuroprotective effects compared with the parent ginsenosides, such as Rb1, Rb2, and Re. Therefore, to monitor the potent pharmacological effects of ginseng, a novel probiotic fermentation technology has been developed to produce absorbable and bioactive metabolites. Based on these findings, it is concluded that gut microbiota play an important role in the pharmacological action of orally administered ginseng, and probiotics that can replace gut microbiota can be used in the development of beneficial and bioactive ginsengs.
