인삼 육성계통의 Fusarium sozani에 의한 근부병 저항성 비교 (Comparison of Resistance of Root Rot Caused by Fusarium solani in Ginseng Breeding Lines)

  • Root-rot of ginseng caused by Fusarium solani is one of the most obstacles to ginseng cultivation. We evaluated some inoculating techniques of ginseng with Fusarium solani, for selection of disease resistant breeding lines. The most effective inoculating techniques evaluated were inserting toothpicks colonized by F. solani into the seedling roots in laboratory test and dusting seedlings with vermiculite after dipping in conidial sllspension and then replanting method in field test. The resistance to diseased by F. solani was lines of 82022 and 82066 in laboratory test. 82920-1 and 78093 in field test.

Involvement of Growth-Promoting Rhizobacterium Paenibacillus polymyxa in Root Rot of Stored Korean Ginseng

  • Paenibacillus polymyxa is a plant growth-promoting rhizobacterium (PGPR) which can be used for biological control of plant diseases. Several bacterial strains were isolated from rotten roots of Korean ginseng (Panax ginseng C. A. Meyer) that were in storage. These strains were identified as P. polymyxa, based on a RAPD analysis using a P. polymyxa-specific primer, cultural and physiological characteristics, an analysis utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME), and the 16S rDNA sequence analysis. These strains were found to cause the rot in stored ginseng roots. Twenty-six P. polymyxa strains, including twenty GBR strains, were phylogenetically classified into two groups according to the ERIC and BOX-PCR analyses and 16S rDNA sequencing, and the resulting groupings systematized to the degrees of virulence of each strain in causing root rot. In particular, highly virulent GBR strains clustered together, and this group may be considered as subspecies or biovar. The virulence of the strains seemed to be related to their starch hydrolysis enzyme activity, but not their cellulase or hemicellulase activity, since strains with reduced or no starch-hydrolytic activity showed little or no virulence. Artificial inoculation of the highly virulent strain GBR-1 onto the root surfaces of Korean ginseng resulted in small brown lesions which were sunken and confined to the outer portion of the root. Ginseng root discs inoculated in vitro or two-year-old roots grown in soil drenched with the inoculum developed significant rot only when the inoculum density was $10^{6}-10^{7}$ or more colony-forming units (CFU) per ml. These results suggest that P. polymyxa might induce ginseng root rot if their population levels are high. Based on these results, it is recommended that the concentration of P. polymyxa should be monitored, when it is used as a biocontrol agent of ginseng, especially in the treatment of stored roots.

인삼의 연생, 생육시기 및 식물체 부위별 수용성과 불수용성 탄수화물 함량과 합성저장량 비교 (Comparative Analysis on Concentration and Synthetic Amount of Water Soluble and Water Insoluble Carbohydrates with Different Plant Tissues, Growth Stages and Years Old of Panax ginseng C. A. Meyer)

  • This study was carried out to have the basic and applied informations relating to develop the cultivation methods and to increase the productivity and quality of ginseng. 1 to 6 year old ginsengs of Jakyung cultivar were cultivated and the content and synthetic amount of carbohydrates were investigated with different plant tissues, growth stages, and years old. The concentration of total carbohydrates at six year old ginseng including water soluble and water insoluble carbohydrates was about 18.9%, 42.9%, and 43,6% in leaves, tap roots, and lateral roots, respectively. Water soluble carbohydrate of tap and lateral roots was slightly decreased from August until September, and then increased on November, whereas its water insoluble carbohydrate was increased from August to September and then decreased on November. Comparing with the content of carbohydrates of 1 to 6 year old ginsengs, it was continuously increased from one year old ginseng until five year old ginseng, however it was not increased much in six year old ginseng. The highest content of carbohydrates was at five year-old in all tissues of ginseng. Water soluble and water insoluble carbohydrates were significantly shown different in leaves, stems, tap roots, and lateral root at different growth stages and with different years old. The content of water soluble carbohydrate in the leaves was remarkedly higher compared to that of water insoluble carbohydrate, while in the root the content of water insoluble carbohydrate was clearly higher compared to the water soluble carbohydrate. Comparing with the synthetic amount of carbohydrates, water soluble carbohydrates was higher in the shoot than that in the root, whereas water-insoluble carbohydrates higher in the root than that in the shoot. Carbohydrates which would be utilized in ginseng tissues for short and long-term periods as major energy were appeared differently in between shoot and root, with different growth stages, and years old.

Ginseng authenticity testing by measuring carbon, nitrogen, and sulfur stable isotope compositions that differ based on cultivation land and organic fertilizer type

  • Background: The natural ratios of carbon (C), nitrogen (N), and sulfur (S) stable isotopes can be varied in some specific living organisms owing to various isotopic fractionation processes in nature. Therefore, the analysis of C, N, and S stable isotope ratios in ginseng can provide a feasible method for determining ginseng authenticity depending on the cultivation land and type of fertilizer. Methods: C, N, and S stable isotope composition in 6-yr-old ginseng roots (Jagyeongjong variety) was measured by isotope ratio mass spectrometry. Results: The type of cultivation land and organic fertilizers affected the C, N, and S stable isotope ratio in ginseng (p < 0.05). The ${\delta}^{15}N_{AIR}$ and ${\delta}^{34}S_{VCDT}$ values in ginseng roots more significantly discriminated the cultivation land and type of organic fertilizers in ginseng cultivation than the ${\delta}^{13}C_{VPDB}$ value. The combination of ${\delta}^{13}C_{VPDB}$, ${\delta}^{15}N_{AIR}$, or ${\delta}^{34}S_{VCDT}$ in ginseng, except the combination ${\delta}^{13}C_{VPDB}-^{34}S_{VCDT}$, showed a better discrimination depending on soil type or fertilizer type. Conclusion: This case study provides preliminary results about the variation of C, N, and S isotope composition in ginseng according to the cultivation soil type and organic fertilizer type. Hence, our findings are potentially applicable to evaluate ginseng authenticity depending on cultivation conditions.

Cytokinin signaling promotes root secondary growth and bud formation in Panax ginseng

  • Background: Panax ginseng, one of the valuable perennial medicinal plants, stores numerous pharmacological substrates in its storage roots. Given its perennial growth habit, organ regeneration occurs each year, and cambium stem cell activity is necessary for secondary growth and storage root formation. Cytokinin (CK) is a phytohormone involved in the maintenance of meristematic cells for the development of storage organs; however, its physiological role in storage-root secondary growth remains unknown. Methods: Exogenous CK was repeatedly applied to P. ginseng, and morphological and histological changes were observed. RNA-seq analysis was used to elucidate the transcriptional network of CK that regulates P. ginseng growth and development. The HISTIDINE KINASE 3 (PgHK3) and RESPONSE REGULATOR 2 (PgRR2) genes were cloned in P. ginseng and functionally analyzed in Arabidopsis as a two-component system involved in CK signaling. Results: Phenotypic and histological analyses showed that CK increased cambium activity and dormant axillary bud formation in P. ginseng, thus promoting storage-root secondary growth and bud formation. The evolutionarily conserved two-component signaling pathways in P. ginseng were sufficient to restore CK signaling in the Arabidopsis ahk2/3 double mutant and rescue its growth defects. Finally, RNA-seq analysis of CK-treated P. ginseng roots revealed that plant-type cell wall biogenesis-related genes are tightly connected with mitotic cell division, cytokinesis, and auxin signaling to regulate CK-mediated P. ginseng development. Conclusion: Overall, we identified the CK signaling-related two-component systems and their physiological role in P. ginseng. This scientific information has the potential to significantly improve the field-cultivation and biotechnology-based breeding of ginseng.

하우스에서 재배된 새싹인삼의 재배시기별 생육, 유효성분 및 물성의 변화 (Changes in Growth, Active Ingredients, and Rheological Properties of Greenhouse-cultivated Ginseng Sprout during its Growth Period)

  • Background: The ginseng ginsenosides, which have various physiological activities, are known to be more abundant in the leaves than in the roots, and the consumers' interest in ginseng sprout as a functional vegetable has been increasing. Methods and Results: The aim of this study was to investigate the effects of growth period on growth properties, active ingredients and rheology of ginseng sprouts cultivated in a non-heated greenhouse equipped with a shade net for 60 days, starting from the end of May to the middle of July. The chlorophyll content of the leaves decreased, but their length and width increased with increasing cultivation days. In particular, growth increased significantly until 40 days, but only slightly after 50 days. The stem length did not increase greatly from the 20 th to the 30 th day of cultivation, but increased significantly from the 30 th to the 40 th day, and then further increased gradually. The weight of the leaves, stems, and roots increased slightly, but not change significantly. After 40 days of cultivation, the total ginsenoside content increased by 1.07 times in the leaves and decreased by 0.80 times in the roots with increasing cultivation days. The leaf contents of ginsenosides $Rg_1$, Re, $Rb_1$, Rc, $F_3$ and $F_4$ increased with increasing cultivation days. The rheological properties of ginseng sprout showed the greatest influence on stem hardening with increasing cultivation days. Conclusions: Therefore, based on the growth characteristics, active ingredients and physical properties, 40 days after sowing was considered to be an appropriate harvesting time for ginseng sprouts.

인삼 재배시 생육기간 동안 환경적 요인인 미세기포수가 ginsenoside 생성에 미치는 영향 (The Effect of Environmental Fine Bubble on the Production of Ginsenoside during the Growth Period of Ginseng Cultivation)

  • 본 연구는 환경정화에 주로 사용되는 미세기포를 2년 근 인삼 지속적으로 처리하여 인삼이 성장하면서 변화되는 형태와 ginsenoside 변화를 조사하는 융합적 연구이다. 인삼 재배시 미세기포수를 적용하여 4개월 동안(120일) 재배한 후 인삼의 잎과 뿌리의 부위별 ginsenoside 함량과 조성을 분석하였다. 잎에 일반수를 처리한 결과 protopanaxatriol(PPT) 계열 Re 함량만 월등히 높게 나타났지만 미세기포수를 처리한 결과 protopanaxadiol(PPD) 계열 Rb1, RC, Rb2, Rd 성분도 같이 증가시키는 것을 확인하였다. 특히 Re, Rb1이 다량 증가함으로써 전체적인 total ginsenoside가 증가하는 요인이 되었다. 인삼의 부위별 PD/PT 비율은 미세기포수를 처리한 잎에서는 0.811으로 나타나고 뿌리는 1.28로 나타났다. 이것은 미세기포수 처리가 뿌리에서 ginsenoside의 합성을 유도하여 PD/PT 비율이 1과 가까운 결가를 가져와 유용성분의 증가 및 고른 분포 이루어졌다고 판단된다. 따라서 미세버블수를 사용한 고품질 인삼을 생산하는 재배 방법을 제시하고 인삼의 뿌리와 더불어 잎도 기능성 식품 소재로 활용할 수 있는 가능성을 제시하였다.

Characteristics of Transformed Panax ginseng C.A. Meyer Hairy Roots: Growth and Nutrient Profile

  • Ginseng (Panax ginseng CA. Meyer) hairy root cultures, which are established via the infection of ginseng root discs with Rhizobium rhizogenes, have been used to construct profiles of both biomass growth and nutrient consumption in flask cultures. In a 250 mL shake flask culture, the maximum biomass was observed on the 59th day of the culture period, at 216.8 g (fresh wt) per liter or 11.4 g (dry wt) per liter. The hairy roots were determined to have a growth rate of 0.355 g-DW/g cells/day during the exponential growth phase and a maximum specific growth rate on day 7. Total ginseng saponin and phenolic compound contents were noted to have increased within the latter portion of the culture period. Linear correlations between increases in biomass weight and nutrient uptake were used to imply the conductivity yield $2.60g-DW/(L{\cdot}mS)$ and carbon yield 0.45 g-DW/(g sugar) in the 250 mL flask cultures. The biomass yield when two different nitrogen sources were used (ammonia and nitrate) was shown to remain approximately constant. at $0.47g-DW/(L{\cdot}mM\;NH_4$) and $0.33g-DW/(L{\cdot}mM\;NO_3$); it remained at these levels for 16 days with the ammonia. and for 24 days with the nitrate. The biomass yield when a phosphate source was used was also shown to remain approximately constant for 9 days, at $3.17g-DW/(L{\cdot}mM\;PO_4$), with an $R^2$ of 0.99.

고려인삼중 다당체 성분이 암독소 호르몬-L의 지방분해 작용과 안지오텐신 변환효소의 활성에 미치는 영향 (Effect of Acidic Polysaccharide Components of Korean Ginseng on Lipolytic Action of Toxohormone-L and on Activity of Angiotensin Converting Enzyme)

  • This study was devised to observe in vitro, the inhibitory effects of acidic polysaccharide fractions from Korean red ginseng (KRG) and white ginseng (KWG) on the lipolytic action of loxohormone-L and on angiotensin converting enzyme (ACE, peptidyldipeptidase hydrolase, EC 3.4.15.1) . The crude acidic polysaccharides (CAP) extracted from main and lateral roots of KRG and KWG were separately purified through several procedures. The total inhibitory activities on the lipolytic action of toxohormone-L of CAP from main roots of KRG and KWG was higher than those of CAP from lateral roots of KRG and KWG, respectively, and that of CAP from main root of KRG was 3.1 times higher than that of CAP from main root of KWG. The specific activity of CAP from main root of KRG was measured as 5.40 units/mg, when one unit was defined as the amount giving 50% inhibition on toxohormone-L induced lipolysls. A subfraction named PG4 3 obtained by replanted chromatography on DEAE-TOYOPEARL 650M gave the specific activity of 24.4 units/mg. On the other hand, it was found that the total inhibitory activity on ACE of CAP from lateral root of KRG was the highest among the 4 kinds of CAP, but the specific activity of CAP from lateral root of KWG was the highest.
