한국삼(韓國蔘)과 중국삼(中國蔘)의 품질 비교 연구 (A Comparative study on Characteristic of Ginseng Radix in Korea and China)

  • Objctive:To clarify the criterion, the characteristic of varaious age of ginseng radix cultivated in Korea and China were studied. Method:The surface of the transverse section of the specimen was made into a slid by the Paraffin Section method, and then dyed by Safranine Malachite Green method. The samples were observed at the power of 400 by an optic microscope(Olympus, Japan). The component and flavor of ginseng radix were analyzed by TLC(Thinlayer Chromatography) and electronic nose(FOX3000, France). Result:Ginseng radix according to the growing district and various age were comparative analyzed by optic microscope, TLC and electronic nose. The results were as followings. 1. The external form of Korean ginseng is longer and brightness then Chinese ginseng. 2. The internal form of Korean and Chinese ginseng are similar to each other. 3. The component of Korean and Chinese ginseng in TLC are similar to each other. 4. The fragrance of Korean and Chinese ginseng are clearly different. Conclusion:The results in this study demonstrate that morphology and component of Korean ginseng are similar to Chinese, on the other hand, fragrance of Korean and Chinese ginseng are different.

백삼,홍삼과 흑삼 추출물의 정성적 구별법에 관한 연구 (Study on the Qualitative Discrimination of White, Red, and Black Ginseng Extract)

  • 홍삼 추출물(5종)과 흑삼 추출물(2종)과 중국산 인삼 추출물(9종)의 말톨 정성, 지방산 조성비, 페놀성 화합물 함량을 분석하고, 이를 패턴 분석하여 특징적 요인을 정하고, 이를 통해 추출물의 정성 분석에 대한 가능성을 측정하였다. TLC를 이용한 maltol의 분석에서 백삼 추출물은 모두 불검출, 홍삼 흑삼 추출물은 모두 검출되어 말톨의 검출 여부는 정성 분석을 위한 특징적 요인으로 볼 수 있었다. 지방산 조성의 경우 palmitic과 linoleic acid가 인삼 추출물의 주요 지방산이었고, palmitic은 백삼, linoleic 은 홍삼 추출물이 높게 나타났다. 이 두 지방산의 조성을 비교한 비율Pal/Lin)의 경우 백삼 추출물에서 56.7~64.3%, 홍삼 추출물에서 32.0~38.5%로 두 값의 차이가 크게 나타나, 이 수치 역시 분석을 위한 특징적 요인으로 볼 수 있었다. 페놀성 화합물의 경우 추출물 속에서 maltol, caffeic acid, syringic acid, p-coumaric acid, ferulic acid와 cinnamic acid의 존재를 확인 할 수 있었다. 백삼 추출물은 페놀성 화합물이 비슷한 비율로 존재하는 패턴을 보였지만, 홍삼 추출물은 말톨의 함량비가 크게 높았다. 페놀성 화합물 중 maltol과 cinnamic acid의 비율을 측정한 결과, 백삼 추출물에서는 5이하의 수치가 나온데 비해 홍삼 흑삼 추출물에서는 53~289의 수치를 보여 이 역시 정성 분석을 위한 특징적 요인으로 판단되었다. 실험 결과에 의해 선정된 Pal/Lin 비율과 maltol/cinnamic acid 비율과 같은 특징적 요인을 통해 인삼 추출물에 대한 정성 분석의 가능성을 확인할 수 있었다.

Remarkable impact of steam temperature on ginsenosides transformation from fresh ginseng to red ginseng

  • Background: Temperature is an essential condition in red ginseng processing. The pharmacological activities of red ginseng under different steam temperatures are significantly different. Methods: In this study, an ultrahigh-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry was developed to distinguish the red ginseng products that were steamed at high and low temperatures. Multivariate statistical analyses such as principal component analysis and supervised orthogonal partial least squared discrimination analysis were used to determine the influential components of the different samples. Results: The results showed that different steamed red ginseng samples can be identified, and the characteristic components were 20-gluco-ginsenoside Rf, ginsenoside Re, ginsenoside Rg1, and malonyl-ginsenoside Rb1 in red ginseng steamed at low temperature. Meanwhile, the characteristic components in red ginseng steamed at high temperature were 20R-ginsenoside Rs3 and ginsenoside Rs4. Polar ginsenosides were abundant in red ginseng steamed at low temperature, whereas higher levels of less polar ginsenosides were detected in red ginseng steamed at high temperature. Conclusion: This study makes the first time that differences between red ginseng steamed under different temperatures and their ginsenosides transformation have been observed systematically at the chemistry level. The results suggested that the identified chemical markers can be used to illustrate the transformation of ginsenosides in red ginseng processing.

Morphological Characteristics of Korean Dried Ginseng Products

  • The purpose of this study was to determine a standard quality characteristic through the evaluation and statistical analysis of the morphological characteristics of dried ginseng (white ginseng) products. Samples of 4-year-old 30 and 50 piece grade curved ginseng and 5-year-old 25 and 15 piece grade straight ginseng products were collected from a market, with 5 to 10 packs of each product being collected annually over a 5-year period (2006-2010). Morphological characteristics, such as weight, length, diameter, and surface color, were measured and statistically analyzed to present a standard quality characteristic value using $mean{\pm}3SD$, a range that excluded outlier. The 4-year-old curved ginseng samples of 50 and 30 piece grade were 4.80 to 6.12 cm and 5.28 to 7.60 cm long, 0.22 to 1.70 cm and 0.21 to 2.07 cm wide, and weighed 5.28 to 7.40 g and 8.62 to 12.26 g, respectively. The 5-year-old straight ginseng samples of 25 and 15 piece grade were 9.66 to 15.47 cm and 10.66 to 16.80 cm long, 1.32 to 1.94 cm and 1.48 to 2.43 cm wide, and weighed 9.18 to 16.40 g and 15.89 to 24.82 g, respectively. The surface color of the different piece grades in the same type of dried ginseng product was similar, whereas the straight ginseng demonstrated a lower level of brightness, but the relative redness and yellowness were of higher levels, than that of curved ginseng.

홍삼제조과정 중 파낙사트리올계 진세노사이드의 물질균형 (The Mass Balance of Protopanaxtriol Ginsenosides in Red Ginseng Process)

  • This mass balance study about ginsenoside Rg1 and Re in Red ginseng processed from Fresh ginseng is useful to understand that herbal material sources of ginseng and raw material consumption in Red ginseng preparations. In our results, total molar amounts of ginsenoside Rg1, Re and their converts in Fresh ginseng, Red ginseng, and Red ginseng extract are substantially the same. The molar amounts of ginsenoside Rg1, Re (4.324, 2.880 μmol/g) as starting materials in Fresh ginseng are kept constant as total molar amounts (sum of starting and converts) in Red ginseng (4.264, 2.596 μmol/g) and Red ginseng extract (3.389, 3.129 μmol/g). This result means that protopanaxtriol type ginsenosides and their characteristic converts are not destroyed or inflowing in Red ginseng process. Therefore, it is important for quality assurance of Red ginseng preparations that the ratio between ginsenosides Rg1, Re and these converts is kept constant.

  • We studied the chloroplast rearrangement, short-term regulation depending on the light conditions in plants, and the characteristic of photosynthic rate as affected by in Panax ginseng C.A. Meyer. The chloroplast rearrangement of ginseng mesophyll cell was induced with the irradiation of blue light (400~500 nm) and through this process the rate of leaf transmittance increased 5~7.5%. The time to reach the maximum value of photosynthesis was shorter above 20 minutes with the blue light irradiation than that of the red light.

생물반응기에서 배양된 인삼의 품질 특성 (The Quality Characteristic of Ginseng Cultured in Bioreactor System)

  • 본 연구에서는 생물반응기에 배양된 인삼의 품질 특성 및 이를 이용한 액상차 제조에 관한 연구를 행하였다. 생물반응기에서 배양된 인삼 추출물에 대한 품질특성은 가용성 고형분 함량 및 조사포닌 함량이 각각 31.80%, 1.94%로 나타나 재배 인삼보다는 함량이 낮은 경향이었다. 농축액의 조사포닌 함량은 4.77%이었으며 ginsenoside Rc, Re 및 Rg$_1$은 7.36, 4.40 및 1.75 mg/g으로 나타났으며 재배인삼에 많은 함량을 차지하는 ginsenoside Rb$_1$과 Rb$_2$는 검출되지 않았다. 생물반응기에서 배양된 인삼의 활용방안으로 추출.농축액을 이용하여 액상차를 제조하여 최적 배합비를 설정하였다. 액상차제조의 최적 배합비 범위는 농축액 함량 9.0~10.4%, 사과식초 함량 6.8~8.1%, 과당 함량 40%로 예측되었다. 또한 배양 인삼 농축액과 재배인삼 농축액으로 제조된 액상차의 관능적 품질을 재배인산 농축액으로 제조된 액상차의 관능적 품질을 비교하고자 예측된 임의의 조건(농축액 10%, 사과 식초 7%, 과당 40%)으로 동일하게 제조하여 비교하였을 때 관능적 품질은 재배인삼이 생물반응기에 배양된 인삼보다 높은 관능평점을 나타내었다.

  • A lot of individual ginseng plants were selected in the farmer's fields to develop new ginseng varieties with desirable traits. Among them, a promising line has been developed through comparative cultivation of several lines selected with pure line separation of local raced. It was then designated as "KC (denotes Korean Ginseng)" and tested in the regional yield and adaptation trials for 10 years. KG lines grew vigorously after 4 years of age. Especially, KG 102 line arson운 them showed traits of multiple and short stems. As for the root characters, the length of taproot of KG 101 line was longer than that of local race, Jakyung-jong, or other lines ailed the root weight of KG 102 line waIn 15 % higher than that of local race. In general, KG 101, KG 103, KG 104, and Hwangsauk-jong had good root shape. Total amount of ginsenosides of ginseng taproot was the highest in KG 103 line than in local race and other lines. In these studies, we, elected three superior lines, KG 101, KG 102, and KG 103, having characteristic of good root shape, high yield, and large amount of ginsenoside, respectively.

Identification of mountain-cultivated ginseng and cultivated ginseng using UPLC/oa-TOF MSE with a multivariate statistical sample-profiling strategy

  • Background: Mountain-cultivated ginseng (MCG) and cultivated ginseng (CG) both belong to Panax ginseng and have similar ingredients. However, their pharmacological activities are different due to their significantly different growth environments. Methods: An ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS/MS)-based approach was developed to distinguish MCG and CG. Multivariate statistical methods, such as principal component analysis and supervised orthogonal partial-least-squares discrimination analysis were used to select the influential components. Results: Under optimized UPLC-QTOF-MS/MS conditions, 40 ginsenosides in both MCG and CG were unambiguously identified and tentatively assigned. The results showed that the characteristic components of CG and MCG included ginsenoside Ra3/isomer, gypenoside XVII, quinquenoside R1, ginsenoside Ra7, notoginsenoside Fe, ginsenoside Ra2, ginsenoside Rs6/Rs7, malonyl ginsenoside Rc, malonyl ginsenoside Rb1, malonyl ginsenoside Rb2, palmitoleic acid, and ethyl linoleate. The malony ginsenosides are abundant in CG, but higher levels of the minor ginsenosides were detected in MCG. Conclusion: This is the first time that the differences between CG and MCG have been observed systematically at the chemical level. Our results suggested that using the identified characteristic components as chemical markers to identify different ginseng products is effective and viable.

Chemical Diversity of Panax ginseng, Panax quinquifolium, and Panax notoginseng

  • The major commercial ginsengs are Panax ginseng Meyer (Korean ginseng), P. quinquifolium L. (American ginseng), and P. notoginseng (Burk.) FH Chen (Notoginseng). P. ginseng is the most commonly used as an adaptogenic agent and has been shown to enhance physical performance, promote vitality, increase resistance to stress and aging, and have immunomodulatory activity. These ginsengs contain saponins, which can be classified as dammarane-type, ocotillol-type and oleanane-type oligoglycosides, and polysaccharides as main constituents. Dammarane ginsenosides are transformed into compounds such as the ginsenosides $Rg_3$, $Rg_5$, and $Rk_1$ by steaming and heating and are metabolized into metabolites such as compound K, ginsenoside $Rh_1$, proto- and panaxatriol by intestinal microflora. These metabolites are nonpolar, pharmacologically active and easily absorbed from the gastrointestinal tract. However, the activities metabolizing these constituents into bioactive compounds differ significantly among individuals because all individuals possess characteristic indigenous strains of intestinal bacteria. To overcome this difference, ginsengs fermented with enzymes or microbes have been developed.
