Tobacco (Nicotiana tabacum cvs.Xanthi-nc and NC 82) plants infected with Tobacco mosaic virus (TMV) were examined ultrastructurally. Local lesions produced by TMV were sunken and withered. The plants were subjected to temperature shift (TS), a method to produce programmed cell death (PCD), by placing the infected plants initially at high temperature (35$^{\circ}C$) for 2 days and then shifting them to greenhouse temperature (22-27$^{\circ}C$). As a result, expanded lesions around the original necrotic lesions were produced. The expanded area initially had no symptoms, but it withered and became necrotic 15 h after TS. No ultrastructural changes related to PCD were noted at 0 h after TS in Xanthi-nc tobacco tissues as well as in healthy and susceptible tobacco tissues infected with TMV, At 6 h after TS, chloroplasts were convoluted and cytoplasm began to be depleted; however no necrotic cells were found. At 17 h after TS, ground cytoplasm of affected cells was completely depleted and chloroplasts were stacked together with bent cell wall or dispersed in the intracellular space. Necrotic cells were also observed, containing virus particles in the necrotic cytoplasm. There were initially two types of symptoms in the expanded lesions: chlorosis and non-chlorosis (green). Abundant TMV particles and X-bodies were only found in the chlorotic tissue areas. These results suggest that PCD by TMV infection may start with the wilting of cells and tissues before necrotic lesion formation.
The study was conducted to obtain the genetic informations for some useful characters in tobacco breeding. The eight parents and a set of 28 crosses of F1's and F2's were used as materials, and planted on two different cultivated systems, i. e., oriental's and burley systems, during 1986-1988 at taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. The observed characters were six agronomic characters which were plant height, number of leaves per plant, leaf length, leaf width, days to flowering, yield and five chemical components, nicotine, total nitrogen, protein nitrogen, total volatile base and pet, ether extract. The results obtaining are summarized as follows: 1. The higher heritabilities were found for days to flowering yield and nicotine in both generations, but values for number of leaves per plant, leaf length, total nitrogen and protein nitrogen were lower than other characters. 2. Genotypic correlation coefficients among all pairs of characters were slightly higher than the corresponding phenotypic correlations in F1's and F2's on two different cultivated systems. The relationship between leaf length and leaf width was the positive correlation, but that between number of leaves per plant and leaf width was negative. 3. From the genotypic correlations between yield and other characters, a conclusion that the yield was highly correlated with plant height, leaf length, leaf width and days to flowering was given. 4. Quality was positively correlated with number of leaves per plant and nicotine, but negatively with the other agronomic characters and chemical components.
A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Background: Panax notoginseng is a highly valued medicinal herb used widely in China and many Asian countries. Its root and rhizome have long been used for the treatment of cardiovascular and hematological diseases. Imaging the spatial distributions and dynamics of metabolites in heterogeneous plant tissues is significant for characterizing the metabolic networks of Panax notoginseng, and this will also provide a highly informative approach to understand the complex molecular changes in the processing of Panax notoginseng. Methods: Here, a high-sensitive MALDI-MS imaging method was developed and adopted to visualize the spatial distributions and spatiotemporal changes of metabolites in different botanical parts of Panax notoginseng. Results: A wide spectrum of metabolites including notoginsenosides, ginsenosides, amino acids, dencichine, gluconic acid, and low-molecular-weight organic acids were imaged in Panax notoginseng rhizome and root tissues for the first time. Moreover, the spatiotemporal alterations of metabolites during the steaming of Panax notoginseng root were also characterized in this study. And, a series of metabolites such as dencichine, arginine and glutamine that changed with the steaming of Panax notoginseng were successfully screened out and imaged. Conclusion: These spatially-resolved metabolite data not only enhance our understanding of the Panax notoginseng metabolic networks, but also provide direct evidence that a serious of metabolic alterations occurred during the steaming of Panax notoginseng.
The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.
This study was conducted to obtain the breeding information for the leaf breakdown, and correlation coefficients between leaf breakdown and some agronomic characters in flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. NC 95 and NC 2326 were used as parents. The bulk method procedure was used from the F2 to the F4 generations, 54 plants were selected among F4, and 11 lines were selected among 54 F5 which based on high resistance to bacterial wilt compared with NC2326. The entries were composed of parents and 11 F6 advanced lines, and designed in randomized block with 3 replications. Plant seedlings were transplanted in 16 April. The number of breakdown leaf were investigated at flowering stage, 18 June. There were significant difference between NC 95 and NC 2326 for the number of breakdown leaf, number and weight of ground suckers, days to flower, yield and resistance to bacterial wilt, and similar results were appeared among lines. Estimated heritability in the broad-sense ranged from 59.3% to 87.4% for all characters. Most of genetic correlations were higher than phenotypic coefficients between characters. Number of breakdown leaf was positively correlated with days to flower, the number and weight of ground suckers, while it was negative to yield. One line among 11 had few breakdown leaf, low producted ground suckers and high resistance to bacterial wilt. Above results suggest that the number and weight of ground suckers can be use as a selection criterion for leaf breakdown even under the condition without leaf breakdown.
This study was conducted to obtain the breeding Information for varietal difference of leaf breakdown of flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute In 1988. The experiment was designed in randomized block with 3 replications. And data were analyzed as spilt plot design composed with varieties for main and growth stages for sub-plots. Among the 8 varieties (NC 95, SL 72, Sleight G-28, TC 518, NC82, NC 2326, NC 567 and TC 499), first half derived from NC 95, and 4 latter varieties were used for check Plants and not derived from NC 95. Plant seedlings were transplanted in 15 April. The number of breakdown leaf were investigated twice at peak growth stage, 5 June and at early harvest stage, 30 June. NC 95 and varieties derived from NC 95 had showed significantly different on the number of breakdown leaf compared to the latter 4 ones after heavy rainfall, but there were not significantly different at the two growth stages. It was suggested that NC95 and varieties derived from NC 95 had substance to induce leaf breakdown by conditional genes after water absorption to plant tissue. Among the varieties derived from NC 95 cultivar SC 72, Speight G-28 and TC 518 had appeared significantly different on the number of breakdown leaf, respectively. These results could appreciated analogically that character of leaf breakdown were governed by heteromeric genes In NC 95.
Kalopanax pictus is a long-lived woody species mostly distributed in East Asia. K. pictus has been regarded as medically and ecologically important species in Korea. Thornless castor aralia variant, local name 'Cheongsong' is an endemic to Cheongsong province in Korea. Random amplified polymorphic DNA (RAPD) was used to investigate the genetic variation and structure of Korean populations of two species. A high level of genetic variation was found in six K. pictus populations. Twelve primers revealed 49 loci, of which 29 were polymorphic (59.2%). Nei's gene diversity for K.pictus and K. pictus variant were 0.119 and 0.098, respectively. Mean of genetic diversity in K. pictus was higher than average values for species with similar life history traits. The asexual and sexual reproduction, perennial habitat, and longevity are proposed as possible factors contributing to high genetic diversity. An indirect estimate of the number of migrants per generation (Nm=0.857) indicated that gene flow was not extensive among Korean populations of K.pictus. It is suggested that the isolation of geographical distance and reproductive isolation between K.pictus and K.pictus variant populations may have played roles in shaping the population structure of this species.
과육색이 다르게 발현되는 사과(Malus domestica L.) 품종의 유전자 발현을 비교하기 위해 2개의 cDNA library를 제작하였다. 붉은 색 과육 품종인 'Redfield'와 백색 과육 품종인 'Granny Smith'의 유전자 발현 차이를 보기 위해 차세대 염기서열 분석(NGS) 기술을 사용하였고 두 품종으로부터 얻은 EST의 염기서열을 결정하고 기존에 보고된 유전자와의 상동성을 분석하였다. HRM 기술은 붉은 색 과육 품종 사과와 백색 과육 품종 사과의 짧은 PCR 증폭산물에서 한 개의 서로 다른 염기서열을 구분하여 분리해낼 수 있다. 'Redfield'와 'Granny Smith'의 EST database로부터 103쌍의 단일염기다형성(SNP) 분자표지를 선발하였고, 붉은 색 과육 품종 10개와 백색 과육 품종 11개를 구분할 수 있는 SNP 분자표지를 HRM 방법으로 분석하였다. 본 연구에서는 사과 EST database를 기반으로 HRM 분석 방법을 이용하여 사과 품종의 적육계와 백육계를 구분할 수 있는 효율적인 SNP 분자표지를 개발하였다. 이러한 SNP 분자표지는 사과육종에 유용하게 사용할 수 있으며 사과 품종의 다양한 색 변화에 관한 분자 기작 연구에 좋은 참고자료가 될 수 있을 것이다.
