은행 열매 추출물의 멜라닌 생성 저해효과 (Inhibitory Effect of Ginkgo biloba Extracts on Melanin Biosynthesis)

  • 본 연구는 은행 열매 오일의 멜라닌 생성 억제 효과를 확인한 것이다. 은행나무 열매 오일은 DPPH assay와 FRAP assay를 사용하여 라디컬 소거능을 시험하였다. 결과적으로 은행나무 열매 오일은 DMSO를 용매로 0.06% 녹였을 때, DPPH assay에서 9.96% 소거활성을 나타내었고 FRAP는 1.33 mM의 ferric sulfate ($FeSO_4$)를 생성하였다. 은행 열매 오일은 tyrosinase inhibition assay에서 37.72%의 억제력을 가졌고 B16/F10 세포에 멜라닌 생합성 실험을 통해 확인하였다. 은행 오일 0.06%에서 ${\alpha}$-MSH 처리 구에 비해 48.02%의 멜라닌 생성을 억제하였다. Tyrosinase, tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), microphthalmia-associated transcription factor (MITF)의 유전자 발현 수준은 control군에 비해 0.04%와 0.06% 농도 군이 크게 감소하였다. 결과적으로 은행 열매 오일 추출물이 멜라닌 생성을 억제하는 효과가 있는 것으로 확인되었다.

  • 은행나무 3년생 묘목에 각기 다른 수준의 질소, 인, 질소와 인 복합비료를 시비하고, 묘목의 생장 및 엽내 양분과 유용 추출물의 농도를 측정하였다. 시비 후 묘목의 잎, 줄기, 뿌리의 생체량은 고농도 질소와 인 복합비료 처리구 이외에서는 변화가 없었으며, 엽내 질소와 인 농도의 변화 정도는 시비량에 따라 차이가 있었다. 일반적으로 엽내 질소와 인의 농도는 시비량 증가에 따라 증가하나, 질소 시비량이 400kg/ha인 경우와 인 시비량이 100kg/ha에서는 오히려 감소하였다. 묘목 생장량과 엽내 양분 농도를 고려할 때 묘목은 질소와 인이 결핍된 상태에 있었던 것으로 볼 수 있었다. 시비 후 엽내 이차 대사 물질인 Ginkgo flavon glycosides와 terpene lactones의 농도는 감소하였으며, 은행나무 묘목에서 생체량과 이차 대사 물질량 사이에는 부의 상관관계 (r=-0.739, p=0.036)가 있는 것으로 나타났다.

Qualitative Anatomical Characteristics of Compression Wood, Lateral Wood, and Opposite Wood in a Stem of Ginkgo biloba L.

  • This study was conducted to understand and to compare the qualitative aspects of anatomical characteristics in compression wood (CW), lateral wood (LW), and opposite wood (OW) in a stem of Ginkgo biloba. The qualitative analysis was examined by optical microscopy and scanning electron microscopy. CW in Ginkgo biloba disks were dark brown in color, and the OW and LW were light brown in color. CW and OW showed abrupt transitions from earlywood to latewood, while LW showed a gradual transition. Cross sections of CW presented circular tracheids with angular outlines, many intercellular spaces, and varying sizes of lumens. Cross sections of LW and OW showed rectangular tracheids, fewer intercellular spaces, and varying sizes of lumens. In radial sections, CW showed an irregular arrangement of tracheids in earlywood, while LW and OW showed comparatively regular arrangements. Slit-like bordered pits on the tracheid cell wall, piceoid pits in the crossfield, and a few spiral checks were present in CW. LW and OW showed bordered pits with slightly oval-shaped apertures, as well as cuppresoid pits in the crossfields. Rays were primarily uniseriate, with few biseriate rays in the tangential sections of CW, LW, and OW. The tips of the tracheids were branched in CW but had a normal appearance in LW and OW.

은행나무의 몇가지 다형적(多形的) 동위효소(同位酵素)의 유전양식(遺傳樣式) 및 연관(連關) (Inheritance and Linkage of Some Polymorphic Isozymes in Ginkgo biloba L.)

  • 감자전분젤을 매개로 한 수평식 전기영동장치를 이용하여 은행나무(Ginkgo biloba L.)의 megagametophyte로부터 15개 동위효소의 변이가 분석되었다. 분석된 효소 가운데서 ADH, G6PD, IDH, MPI, UGPP의 5개 효소에서는 변이가 나타나지 않았으며, 나머지 10개 효소의 11개 동위효소 구역(ACON-A, FST-B, GDH-A, GOT-B, MDH-B, MDH-C, MNR-A, PGI-B, PGM-A, 6PGD-B, SKDH-B)에서 다형성이 관찰되었다. 이중 MDH-B를 제외한 모든 구역에서 관찰된 동위효소 변이들이 1 : 1의 독립적 분리비를 보임으로써, 이들이 단일 유전자좌에 의해 조절되는 공우성 대립유전자임을 추정할 수 있었다. 한편, 동위효소 유전자좌의 3가지 조합(ACON-A : MDH-B, GOT-B : PGI-B, MNR-A : SKDH-B)에서 약한 연관관계가 관찰되었으며, 이들의 재조합 비율은 0.38-0.40로 계산되었다 (p<0.05).

The Combined Effects of Ginkgo Biloba Extracts and Aspirin on Viability of SK-N-MC, Neuroblastoma Cell Line in Hypoxia and Reperfusion Condition

  • Objective: The purpose of this study is to investigate the combined effects of ginkgo biloba extract, ginkgolide A and B and aspirin on SK-N-MC, human neuroblastoma cell viability and mRNA expression of growth associated protein43 (GAP43), Microtubule-associated protein 2 (MAP2), B-cell lymphoma2 (Bcl2) and protein53 (p53) gene in hypoxia and reperfusion condition. Methods: SK-N-MC cells were cultured with Dulbecco's Modified Eagle's Medium (DMEM) media in $37^{\circ}C$, 5% $CO_2$ incubator. The cells were cultured for 8 hours in non-glucose media and hypoxic condition and for 12 hours in normal media and $O_2$ concentration. Cell survival rate was measured with Cell Counting Kit-8 (CCK-8) reagent assay. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to estimate mRNA levels of GAP43, MAP2, Bcl2, and p53 genes. Results: The ginkgolide A and B increased viable cell number decreased in hypoxic and reperfused condition. The co-treatment of ginkgolide B with aspirin also increased the number of viable cells, however, there was no additive effect. Although there was no increase of mRNA expression of GAP43, MAP2, and Bcl2 in SK-N-MC cells with individual treatment of ginkgolide A, B or aspirin in hypoxic and reperfused condition, the co-treatment of ginkgolide A or B with aspirin significantly increased GAP43 and Bcl2 mRNA levels. In MAP2, only the co-treatment of ginkgolide A and aspirin showed increasing effect. The mRNA expression of p53 had no change in all treating conditions. Conclusion: This study suggests that the combined treatments of Ginkgo biloba extracts and aspirin increase the regeneration of neuroblastoma cells injured by hypoxia and reperfusion.

  • This study was carried out to establish standard analytical methods for Ginkgo biloba leaf extract. Ginkgo flavonoids, terpene lactones, ginkgolic acids were employed as reference compounds for analytical method. Analytical method of US Pharmacopoeia was adopted for flavonoids and terpene lactones, and a new method was developed for ginkgolic acids. Analytical methods established in this study could be applied to a reasonable and unified quality control of G. biloba leaf extract.

Effect of Environmental Factors on Flavonol Glycoside Production and Phenylalanine Ammonia-lyase Activity in Cell Suspension Cultures of Ginkgo biloba

  • A study was carried out to elucidate the relation between the production of flavonol glycosides and the change of phenylalanine ammonia-lyase activity in cell suspension cultures of Ginkgo biloba by the unassisted and synergistic effects of various factors. The quercetin production showed a mixed-growth-associated pattern in cell suspension cultures. Fluorescent light and UV radiation increased phenylalanine ammonia-lyase (PAL) activity, and resulted in the increase of the production of quercetin and kaempferol ten- and four-fold, respectively, as compared to that obtained in the normal culture condition. The cell growth of Ginkgo biloba was enhanced .at higher temperatures whereas the quercetin production was at its maximum at low temperatures. Moreover, the quercetin production was increased by temperature change during the culture period. In particular, the quercetin production was at the highest level when the culture temperature was elevated from $10^{\circ}C\;to\;30^{\circ}C$. The addition of phenylalanine as a precursor in the culture medium stimulated an 8-fold increase in the production of quercetin; the addition of naringenin caused a l0-fold increase. The quercetin production was also greatly increased by feeding enzyme cofactors such as 2-ketoglutarate and ascorbic acid in the culture medium, but specific PAL activity was not increased except with phenylalanine feeding. The synergistic effect of UV radiation and naringenin feeding was observed, resulting in the increase of flavonol glycoside production at a rate higher than in any other case investigated.
