Plasma Stress Responses in Juvenile Red-Spotted Grouper (Epinephelus akaara) exposed to Abrupt Salinity Decrease

  • The objective of the current study was to determine acute plasma stress responses in two size groups of juvenile Epinephelus akaara (average body weight: $8.4{\pm}2.1$ and $3.3{\pm}0.6g$; 150 and 120 days after hatch, respectively) exposed to abrupt salinity drops (from 34 practical salinity unit, PSU seawater to 18, 10 PSU (experiment 1) or 26, 18, 10 PSU (experiment 2), respectively). Plasma glucose, glutamic oxalate transaminase, glutamic pyruvate transaminase, red blood cell counts, and gill histology were determined during 72 h exposure. Significantly increased plasma glucose, glutamic oxalate transaminase levels, and red blood cell counts were observed in fish exposed to 18 or 10 PSU. Histological changes, such as hyperplasia and lifting of epithelium in the gill secondary lamellae, were also observed in fish exposed to 18 or 10 PSU at 72 h post-drop. E. akaara exposed to sudden salinity drops to 18 or 10 PSU still seems to undergo the primary adjustment phase before fish reaches a new homeostasis, whereas fish exposed to 26 PSU seems to mount osmotic changes. Therefore, the no observed adverse effect levels for 72 h acute salinity challenge was 26 PSU in our study, and salinity drop to 18 PSU and below can possibly cause acute adverse effect, in which fish could be vulnerable to additional stresses such as a temperature changes or handling stress.

반순환여과시스템에서 오존 유래 잔류산화물 농도에 따른 넙치(Paralichthys olivaceus)의 성장과 생리학적 변화 (Growth Performance and Physiological Changes of Olive Flounder Paralichthys olivaceus by Concentration of Ozone Produced Oxidants in Semi-RAS)

  • This study investigated the effects of ozone-produced oxidants (OPO) on the growth, hematology, and histology of olive flounder Paralichthys olivaceus (average weight 500 g), raised in an ozonated semi-recirculating aquaculture system. The system was ozonated to maintained OPO concentrations of 0.004 (Control), 0.014 (OPO15), and 0.025 (OPO25) mg $Cl_2/L$ in culture tanksfor 26 days. The specific growth rate, feed conversion ratio, and survival rate did not significantly differ among the groups (P>0.05), while the daily feeding rate decreased OPO-dose-dependently (P<0.05). OPO appeared to affect the gill, hepatopancreas, and kidney tissues of fish from ozonated tanks. Hematologically, OPO affected some blood indices. The levels of chloride, glucose, glutamic oxaloacetic transaminase, and glutamic pyruvic transaminase were significantly increased in the ozonated groups, while the total cholesterol and cortisol decreased dose-dependently. These results imply that long-term exposure of olive flounder to an OPO concentration ${\geq}0.014mg\;Cl_2/L$ might result in damage to the gill, hepatopancreas, and kidney tissues and cause physiological stress, albeit with no apparent short-term effects on growth or survival.

Near Infrared Diffuse Reflectance Spectroscopy to Measure Pulmonary Edema

  • A non-invasive spectroscopic method is presented for the measurement of pulmonary edema. Both early diagnosis and quantitative edema estimates were investigated. The spectroscopic determination of pulmonary edema involved the acquisition of diffuse reflectance spectra in the near-infrared (NIR) region with change in water concentration - water is the main constituent of edema fluid. Pulmonary edema was induced into the excised perfused lungs of seven animals by elevating the hydrostatic pressure. Estimates of edema were ascertained from a partial least squares regression of the measured spectral response. Actual edema was determined from the change (increase) in total lung weight. Estimates in relative lung weight increases due to in vitro edema were made with the near infrared spectra. The results revealed that fluid accumulation produced spectral changes in the O-H and C-H absorptions as well as scattering changes in the spectra. Histology of the lung was used to verify the presence or absence of interstitial and alveolar edema. Results demonstrated that near infrared spectroscopy might provide a new tool for clinical assessment of pulmonary edema.

꼬막, Tegillarca granosa 아가미의 미세구조 (Gill Ultrastructure of the Granular Ark, Tegillarca granosa (Bivalvia: Acridae))

  • 광학현미경과 투과전자현미경을 이용하여 꼬막, Tegillarca granosa 아가미의 미세구조를 기재하였다. 꼬막의 아가미는 전형적인 사새형 아가미 구조를 가진다. 새엽은 다수의 측면섬모대와 정단섬모대를 가지며, 혈림프동을 둘러싸고 있는 새엽 상피층은 단층으로 상피세포, 섬모세포 그리고 분비세포들로 이루어져 있다. 상피세포들은 대부분 미세융모를 가진 편평형이다. 섬모세포는 원주형으로 두 종류(A와 B)로 구분되는데, A형 섬모세포는 B형 섬모세포에 비해 분포 비율이 높으며, 세포질의 전체적인 전자밀도는 낮다. 횡단면 표본에서 섬모 축사는 전형적인 "9+2" 구조를 나타냈으며, 기저 중심립은 "$2{\times}9$"의 구조를 보였다. 분비세포들은 새엽의 정단부에서 주로 관찰되며, 분비과립의 특징에 따라 세 종류 (A, B, C)로 구분할 수 있다. A형 분비세포는 다른 분비세포들에 비해 분포정도가 높고, 전자밀도가 낮은 분비과립을 함유한다. B형 분비세포는 막으로 싸여진 전자밀도가 높은 분비과립을 가지며, C형 분비세포들의 분비과립은 타원형으로 중심부의 균질한 물질을 과립상의 물질들이 싸고 있는 형태이다.

  • Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp(Carassius auratus) exposed to 0, 0.01, 0.1, 0.5mg/$\ell$ cadmium(Cd) concentrations for 25 days. TEM Observations of Chemosynthetic Bacteria in the Deep-sea Hydrothermal Vents and Seep Organisms

  • Symbiosis of chemoautrophic bacteria with the members of hydrothermal vent and cold seep communities in the deep-sea were examined by histology using transmission electron microscopy; Bathymodiolus spp. from Sagami Bay, the Iheya Ridge and the North Fiji Basin; and Ifremeria nautilei from the North Fiji Basin. Two species of Bathymodiolus, each from Sagami Bay and the Iheya Ridge harbored methane-oxidizing symbionts within their gill tissues. Vent gastropod Ifremeria nautilei from the hydrothermal vents of the North Fiji Basin housed two types of symbionts; one sulfur-oxidizing type and the other methane-oxidizing type. The occurrence of chemosynthetic symbionts in these organisms were expected before-hand based on the ecological observations of their habit. The other members of these groups from world oceans and the recent advances in the symbiosis of the vent and seep communities were reviewed.

인위적 수온 상승에 노출된 조피볼락(Sebastes schlegelii) 치어의 생존율 및 스트레스 반응 (Survival, Physiological Responses, and Histological Changes in Korean Rockfish (Sebastes schlegelii) Exposed to Artificial Increase of Water Temperature)

  • 본 연구는 조피볼락 치어를 대상으로 인위적인 수온 상승에 따른 생존율, 혈중 cortisol, glucose, GOT, GPT 농도 및 아가미와 간의 조직학적 변화를 조사하였다. 실험 수온은 26℃, 28℃, 30℃ 및 31℃로 설정하였고, 수온은 23℃ (initial)~28℃까지는 1일에 1℃씩, 28~31℃까지는 3일에 1℃씩 조절하였다. 실험은 각 실험 수온에서 7일 동안 수행하였고, 실험기간 동안 먹이는 공급하지 않았다. 그 결과, 26℃와 28℃는 폐사가 발생하지 않았으나, 30℃는 노출 4일째부터 폐사가 발생하여 실험종료 시 1.5%의 생존율을 보였고, 31℃는 노출 1일째부터 폐사가 발생하여 노출 2일째 전량 폐사하였다. 혈중 스트레스 관련 지표를 분석한 결과, 혈중 cortisol과 glucose 농도는 수온이 높을수록 증가하였고, 31℃에서 initial보다 유의하게 감소하였다(p<0.05). 혈중 GPT 농도는 모든 실험구 사이에 유의한 차이를 보이지 않았으나(p<0.05), 혈중 GOT 농도는 31℃에서 유의하게 증가하였다(p<0.05). 간과 아가미의 조직학적 관찰 결과, 간과 아가미는 모두 23℃ (initial) 및 26℃에서 정상적인 조직상을 보였으나, 28℃는 간세포의 비대화 및 아가미 새판 상피세포의 증식과 새판 상피의 탈락, 30℃는 간세포들이 응축 및 아가미 새판의 융합, 31℃는 간 조직 내 충혈 및 일부 간세포의 괴사 및 아가미 새엽과 새판의 괴사가 관찰되었다.

감성돔(Acanthopagrus schlegelii) 치어의 포르말린에 대한 내성 및 조직학적 반응 (Tolerance and Histological Responses to Formalin of Black Seabream Acanthopagrus schlegelii Juveniles)

  • Black seabream juveniles Acanthopagrus schlegelii held at $20^{\circ}C$ were exposed to formalin at concentrations of 50 to 400 ppm, and tolerance values were determined by calculating median lethal concentration values (LC50) through probit analysis. The 12-, 24, 48, 72- and 96-h LC50 values for formalin were 297, 233, 171, 162 and 157 ppm, respectively. The histological effects of formalin on gill and liver tissues in this fish were determined. No histological effects were observed in the control group. The intensity of cell damage increased with the concentration of, and duration of exposure to, formalin. Hyperplasia, separation and epithelial necrosis, epithelial lifting, lamellar synechiae and collapsed lamellae were observed in gill tissues exposed to formalin. Hepatic lesions in liver tissues of fishes exposed to formalin were characterized by cloudy swelling of hepatocytes, necrosis, cytoplasmic vacuolization, deposition of pigments, spongiosis hepatis, nuclear hypertrophy, dilation of sinusoids and bile stagnation. The LC50 values and histological results obtained in this study will aid in designing treatment regimens to minimize toxic side effects and increase efficacy.

침치료로 호전되지 않은 퇴행성 슬관절염 환자의 도침치료 1례 (Miniscalpel Acupuncture Treatment on a Knee Degenerative Osteoarthritis Patient, Who does not Responded to Acupuncture Treatment)

  • Objectives : This study was done to show the clinical effect of miniscalpel acupuncture treatment on osteoarthritis of the knee which is refractory to acupuncture treatment. Methods : A patient was treated with acupuncture for three weeks, non-treated(wash out period) for two weeks, and treated with miniscalpel acupuncture for three weeks. The effect of treatments were measured with Visual Analogue Scale(VAS), Range of Motion(ROM), Short form McGill pain questionnaire(SF-MPQ), Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC). Results : During the three weeks of acupuncture treatment, VAS, SF-MPQ and WOMAC improved, but after two weeks of the wash out period each score worsened. During the three weeks of miniscalpel acupuncture VAS, SF-MPQ, and WOMAC improved while ROM improved remarkably. Conclusion : These results suggest that miniscalpel acupuncture might be a therapeutic option for knee degenerative osteoarthritis patients who does not responded to acupuncture treatment.
