This study was conducted to characterize the amylose and protein contents of 4,948 rice landrace germplasm using the NIRS model developed in the previous study. The amylose contents estimated by NIRS in the standard rice were Sinseonchal (6.881%) 4.994%, Chucheong (19.731%) 18.633%, Goami (23.246%) 20.548%. Protein contents were Sinseonchal (6.890%) 6.824%, Chucheong (6.350%) 6.869%, Goami (6.777%) 7.839%. The NIRS analysis showed that 1.1-2.7%point lower in amylose and 0.4-0.6%point higher in protein than standard contents. The average amylose content of the germplasm was 20.39% with a range of 3.97-37.13%. The average protein content was 8.17% with a range of 5.20-17.45%. Amylose contents with a range of 20.06-27.02% represented 62.20% of the germplasm. Protein contents with a range of 6.78-9.75% represented 81.60% of the germplasm. Korean landrace comprised 24.9% among the 4,948 germplasm collected from 41 countries. A specific range of amylose contents showed in Korea 16.58-20.06%, in Japan 20.06-23.25%, in North Korea 23.25-27.02% and in China 27.02-37.13%. Protein contents exhibited 5.20-17.45% evenly in the whole landraces, whereas Chinese landrace particularly observed with 6.78-8.27% and 9.75-17.45%. Fifty resources were selected with low and high amylose ranging from 3.97-6.66% to 30.41-37.13% respectively. Similarly fifty resources were selected with low and high protein ranging from 5.20-6.09% to 13.21-17.45% respectively. Landraces with higher protein should be adapted to practical utilization of food sources.

Buckwheat has gained scientists concern due to its nutritional and medicinal values in recent years, and many important bioactive compounds. The present study was performed to investigate the influence of the varietal performance and environmental conditions on the content of rutin and quercetin in Tartary buckwheat germplasm. A total of 44 foreign Tartary buckwheat germplasms were examined and compared their contents based on the collected countries, seed shape, and seed color. The highest number of germplasm (16) was found in ranged from 600 mg/100 g to 800 mg/100 g of rutin content. In case of quercetin content, the highest number of germplasm (19) was observed in ranged from 5 mg/100 g to 10 mg/100 g. However, the rutin content showed the highest value (1326.6 mg/100 g) from CBU408 and quercetin content showed the highest value (22.5 mg/100 g) from CBU456. The number of germplasm showed the highest value (23) in ranged from 3000 mg/100 g to 4000 mg/100 g of rutin content. The number of germplasm is the highest value (30) in ranged from 100 mg/100 g to 200 mg/100 g of quercetin content. When rutin and quercetin content in seed was compared according to collected countries, seed shape, and seed color respectively, no association was appeared with rutin and quercetin content. In sprout of whole Tartary buckwheat germplasm, mean of rutin content and quercetin content was 3362.9 mg/100 g and 14.2 mg/100 g respectively. In conclusion, the results observed from the present study suggest that highly rutin germplasm could be used more efficiently via breeding to develop a highly rutin content variety for using in sprout. No correlation was appeared in the case of quercetin. However, the quercetin content in sprout was 4~90 times more than seed. So, sprout could be used more efficiently than seed in the case of quercetin. Our results suggest that the varietal variation and qualitative characteristic differences of rutin and bioactive compounds may provide important nutrient sources and industrial application of Tartary buckwheat.

The nature of genetic variation and diversity among the 65 multivoltine silkworm genotypes was evaluated for 16 post cocoon characters. The components of genetic variation revealed higher PCV (60.487%) and GCV (44.56%) for evenness (variation 1) followed by cohesion (PCV=55.38%, GCV=40.36%) and non-broken filament length (PCV=32.05%, GCV=31.28%). The higher heritability ($h^2$ in broad sense) was observed for boil-off loss (95.6%) followed by non-broken filament length (95.22%). The both genotypic and phenotypic correlation indicated significant positive correlation of filament length with non-broken filament length, silk recovery, raw silk, neatness, and low neatness; and negative correlation with denier, renditta and silk waste. The principal component analysis (PCA) revealed 75.381 % of total variance from the five principal components extracted. On the basis of Mahalonobis' $D^2$ values (Ward's minimum variance), the sixty-five multivoltine silkworm genotypes were classified in to 9 clusters with substantial inter and intra cluster distances. Number of genotypes included in different clusters varied from 3 to 17. The results indicated that the optimum distance obtained in cluster VII (15.059) along with higher cluster mean values especially for filament length, non broken filament length, renditta, silk recovery, silk waste, and raw silk emphasized the utilization of these genotypes in the conventional silkworm breeding programme for improvement of multivoltine silk fibre quality. The possibility of exploiting genetic variation in post cocoon traits for efficient breeding programme is discussed.

The genotype characteristic of cultivars was investigated, along with varieties of both of the jute species, Corchorus olitorius and Corchorus capsularis, in the germplasm collection at the Bangladesh Jute Research Institute (BJRI). DNA fingerprinting was generated for 9 different varieties and 12 accessions of jute cultivars by using random amplified polymorphic DNA(RAPD). A total of 29 arbitrary oligonucleotide primers were screened. Seven primers gave polymorphism within the varieties, and 6 primers detected polymorphism within the accessions that were tested. A dendrogram was engendered from these data, and this gave a distinct clustering of the cultivated species of jute. Therefore, we generated RAPD markers, which are species-specific. These primers can distinguish between C. olitorius and C. capsularis. From the dendrogram that we generated between the various members of these two species, we found the existing genetic classification that agrees with our molecular marking data. A different dendrogram showed that jute accessions could be clustered into three groups. These data will be invaluable in the conservation and utilization of the genetic pool in the germplasm collection.
Heat shock proteins play an important role in plant stress tolerance and are mainly regulated by heat shock transcription factors (Hsfs). In this study, we generated transgenic rice over-expressing OsHsfA7 and carried out morphological observation and stress tolerance assays. Transgenic plants exhibited less, shorter lateral roots and root hair. Under salt treatment, over-expressing OsHsfA7 rice showed alleviative appearance of damage symptoms and higher survival rate, leaf electrical conductivity and malondialdehyde content of transgenic plants were lower than those of wild type plants. Meanwhile, transgenic rice seedlings restored normal growth but wild type plants could not be rescued after drought and re-watering treatment. These findings indicate that over-expression of OsHsfA7 gene can increase tolerance to salt and drought stresses in rice seedlings.

Wheat is one of the most important crops in production and consumption. Despite increasing of importance, the self-sufficiency of wheat is less than 2% in Korea. To improve yield potential and broaden the genetic pool of common wheat in Korea, introduction of alien germplasms into the Korean wheat breeding program is suggested. For effective utilization of the germplasm, we introduced total 1,195 germplasms from the world, which were provided by National Plant Germplasm System (NPGS, USDA) and evaluated the yield, field performances and agronomic traits for 8 years. Among 55 countries, germplasms from Canada, Ethiopia, Mexico and United States accounts for 78%, especially germplasms collected from United States accounts for 50%. Yield comparison of germplasms and collected region analysis indicate that the high yielding germplasms are collected from countries or states of particular range of latitude. The combination analysis of the yield and agronomic traits and the geographical information of collected region will be utilized for improving Korean wheat breeding programs.
Safflower (Carthamus tinctorius L.) is a herb primarily distributed throughout in the world. We have used the inter-simple sequence repeats (ISSR) technique to investigate the phylogenetic relationships and genetic diversity of C. tinctorius. Of all germplasms, 88.7% were polymorphic among all germplasms. Mean genetic diversity within germplasms was very low (0.048). The Turkey germplasm had the highest expected diversity (0.082) and Australia germplasm was the lowest (0.020). These values indicate that most of the genetic diversity of safflower is found among germplasms and there is a high among-germplasm differentiation. We found eight phenetic bands for determining the specific marker of germplasm with SCAR markers. The regions of the Mediterranean Sea and India may be the most probable candidates for the origin of safflower. The tree showed four major clades: (1) European germplasms, (2) Azerbaijan, Egypt, and Ethiopia, (3) Australia, and (4) America.
A strain of the cyanobacterium Arthrospira was isolated from Lake Chahannaoer in northern China and was characterized according to microscopic morphology, photosynthetic oxygen-evolving activity, growth rate, and nutritional profile. Compared with thermophilic Arthrospira species occurring naturally in tropical and subtropical lakes, this isolate is mesophilic and grows optimally at ${\sim}20^{\circ}C$. The total protein, fatty acid, phycocyanin, carotenoid, and chlorophyll a contents were 67.6, 6.1, 4.32, 0.29, and 0.76 grams per 100 grams of dry weight, respectively. The strain is rich in polyunsaturated fatty acids (PUFAs). An essential omega-3 fatty acid, docosahexaenoic acid (DHA), was detected, and ${\gamma}$-linolenic acid (GLA) and DHA accounted for 28.3% of the total fatty acid content. These features of this newly isolated strain make it potentially useful in commercial mass culture in local areas or as a biofuel feedstock. It is also an alternative resource for studying the metabolic PUFA pathways and mechanisms of cold stress tolerance in cyanobacteria.

Pears (Pyrus spp.) have been grown worldwide as a kind of important economical fruits. Over 1,500 accessions collected from countries have been preserved in National Institute of Horticultural and Herbal Science, Rural Development Administration in Korea. However, redundancies and misidentification are happening in the germplasm preservation due to same cultivars which have different names in various localities (synonyms) and different cultivars which have same names (homonyms). That can lower germplasm management efficiency. The object of this study is to identify synonyms and homonyms in pear germplasms by analyzing genetic variation with four microsatellite markers: CH03d12, CH03g07, CH02b10, and EMPc117. PCR amplification with above 4 microsatellite markers was done for the 31 pear accessions, and the products were analyzed by agarose gel electrophoresis. As a result, 7 synonyms and 9 homonyms were identified among 31 pear accessions. We'll compare these genotypes with phenotypes of each pear accessions, and reduces the redundancy and misidentification in pear germplasm collection for the reliable management.
The objective of this study was to analyze the genetic diversity using SSR marker and investigate the fatty acid composition of perilla (P. frutescens var. frutescens) germplasm. Genetic diversity among 95 accessions, which consisted of 29 weedy types and 66 landrace accessions, was evaluated based on 12 SSR markers carrying 91 alleles. The mean values of observed ($H_O$) and expected heterozygosities ($H_E$) were 0.574 and 0.640, respectively, indicating a considerable amount of polymorphism within this collection. A genetic distance-based phylogeny grouped into two distinct groups, which were the landrace, moderate and weedy type, genetic distance (GD) value was 0.609. The physicochemical traits about crude oil contents and fatty acid compositions were analyzed using GC. Among tested germplasm, the total average oil contents (%) showed a range from 28.57 to 49.67 %. Five fatty acids and their contents in the crude oils are as follows: ${\alpha}$-linolenic acid (41.12%-51.81%), linoleic acid (15.38%-16.43%), oleic acid (18.93%-27.28%), stearic acid (2.56%-4.01%), and palmitic acid (7.38%-10.77%). The average oil content of wild types was lower than landrace, and the oil content of middle genotype accessions was higher than other germplasm, but no significant variation between landrace and wild types was shown. Nevertheless, IT117174, landrace of Korea, was highest in crude oil content (47.11%) and linolenic acid composition (64.58%) among the used germplasm. These traits of the selected accessions will be helped for new functional plant breeding in perilla crop.
