Effects of Germinated-Buckwheat on Blood Pressure , Plasma Glucose and Lipid levels of Spontaneously Hypertensive Rats (발아메밀이 본태성 고혈압쥐의 혈압, 혈당 및 혈중 지질수준에 미치는 영향)

  • The purpose of this study was to investigate the effect of germinated buckwheat on blood pressure, plasma glucose and lipid levels of spontaneously hypertensive rats(SHR). The male and female SHR were divided into two groups. After feeding the germinated buckwheat diet(50%, w/w) ad libitum for 6 weeks, the weight gain of the germinated buckwheat group in male rats was higher than those of control group. Systolic blood pressure of the germinated buckwheat group in male rats was significantly decreased at 6 weeks by 3%. The fasting glucose levels of the germinated buckwheat group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR of female SHR fed the germinated buckwheat diet were significantly increased compared to the control diet. Atherogenic index of the germinated buckwheat group was decreased in males(ns) and females(p<0.05) than those of the control group. From these results the germinated buckwheat might be useful for anti-hypertensive and anti-hyperlipidemic agents.

  • In this study, seven grain seeds(sorghum maize, buckwheat, soy bean, mung bean, red bean, and barngrass) and germinated seven grain seeds were examined the fibrinolytic activity through fibrin plate assay and SDS-PAGE. The results obtained were as follows : 1. In the fibrin plate assay, the extracts of maize, barngrass, sorghum and buckwheat showed fibrinolytic activity. Especially, Maize of them showed fibrinolytic activity that was almost similar to plasmin, fibrinolytic enzyme used as a positive control. 2. In the SDS-PAGE of seven grain seeds, fibrinolytic activity was remarkably shown in mung bean and red bean. 3. In the fibrin plate assay of germinated grain seeds, buckwheat(5 mm), buckwheat(10 mm) and soy bean(10 mm) showed a level of fibrinolytic activity that was about 0.3 fold than 1.0 unit of plasmin, Also maize(10 mm) of them showed a level of fibrinolytic activity that was about 0.5 fold than 1.0 unit of plasmin. As a result, maize of grain seeds was found that it has a strong fibrinolytic activity.

Nutritional Changes of Buckwheat During Germination (발아중 메밀의 영양성분의 변화)

  • To produce buckwheat sprouts, buckwheats were germinated at $23{\pm}2^{\circ}C$ up to 8 days in total darkness. In proximate analysis, moisture content increased from 13.87% of buckwheat groats to 93.75% of buckwheat sprouts on the 8th day of germination. On dry weight basis, lipid and ash contents increased like as protein content increased from 13.45% to 21.82% while carbohydrate content decreased due to enzyme hydrolysis. Amino acids were rich in glutamic acid, aspartic acids, and lysine, and also the ratio of the essential amino acids to total amino acids increased from 26.84% to 36.84%. Vitamin C did not exist in buckwheat groats but its content has continued to increase as far as 99.56 mg/100 g buckwheat sprouts. Rutin continued to increase by HPLC analysis from 4.71 mg/100 g buckwheat groats to 1,524 mg/100 g buckwheat sprouts. In analysis of organic acids, oxalic, maleic, and citric acids were commonly found in buckwheat sprouts. Fagopyrin was found almost none in buckwheat sprouts. In conclusion, the conversion of buckwheat seeds into sprouts through germination in darkness results in physically different final product with nutritional changes such as higher content of rutin, generation of vitamin C, abundance of the essential amino acids, and the existence of fagopyrin on very little.

Effect of LED Light Strength for Enhancing Rutin Content in Tatary Buckwheat Sprouts and Antioxidant Activity (타타리메밀싹의 루틴 함량 향상을 위한 LED 광량 효과와 항산화 활성)

  • This study aimed to enhance rutin contents by controlling germination condition for manufacturing buckwheat sprouts. Two kinds of buckwheat, a common buckwheat (Fagopyrum esculentum Moench) and a tartary buckwheat (Fagopyrum tataricum Gaertner) were used. By comparing the rutin content of two buckwheats, tartary buckwheat was 487 ppm, about 36 times higher than common buckwheat. Both common buckwheat and tartary buckwheat which germinated and grew under the light had higher rutin content relatively. In case of tartary buckwheat, rutin content of over 10 cm sprout was 4,579 ppm (without the light), and 5,160 ppm (with the light). Furthermore, tartary buckwheat was germinated and grew under different light strengths from 2,000 to 22,000 Lux. The rutin contents of tartary buckwheat sprout that was grown under the 22,000 Lux light was the highest. The rutin content was increased dramatically at 14,000 Lux of light. From 14,000 to 22,000 Lux, there was a little change on rutin content. Therefore, the condition of 14,000 Lux light was determined optimal for manufacturing tartary buckwheat sprouts. Also, rutin contents of extracts treated with 60, 70, 80 and $90^{\circ}C$ during different time had no significant difference. Therefore, rutin of tartary buckwheat sprout extract had thermostability up to $90^{\circ}C$.

  • To provide the effective application scheme and basic information of buckwheat (Fagopyrum esculentum Moench), buckwheat was germinated at 10$^{\circ}C$ for 7 days and 100 kernels weight, germination rate, root length, chemical composition and mineral contents were examined at 24 hour Intervals. During the germination period, the 100 kernels weight increased approximately 0.3g per day. The germination rate increased sharply after 2 days and the root length increased greatly after 4 days. The crude protein contents increased with germination time, whereas the carbohydrate contents decreased. The crude ash and fat contents did not differ significantly during the germination period. The Ca contents Increased for the 4th day of gemination, but gradually decreased afterwords. The Na contents increased in the initial stage of germination, but then gradually decreased. However, there were no significant change In the Mg, K. Fe. Mn and Zn contents.

Hangover relieving effect of Sanghwang mushroom mycelium cultured in germinated buckwheat

  • The present study was performed to evaluate the hangover relieving effect of germinated buckwheat (GB) and Sanghwang mushroom mycelium cultured in GB (SGB). Both GB and SGB showed 1,1-diphenyl-2-picrylhydrazyl radical scavenging activities and significantly increased (p < 0.001) aldehyde dehydrogenase (ALDH) activities; up to 140% increase at concentrations of $16{\mu}L/mL$. Locomotor activity test results from alcohol-SGB and alcohol-GB groups showed improved motor activities over that of the alcohol-water group at 90 min post-administration. Both alcohol-GB and alcohol-SGB groups had significantly reduced (p < 0.001) alcohol ($40.02{\pm}33.38{\mu}g/mL$, $66.01{\pm}22.04{\mu}g/mL$, respectively) and aldehyde ($5.72{\pm}0.47{\mu}g/mL$, $6.72{\pm}1.70{\mu}g/mL$, respectively) concentrations in blood compared to those in the alcohol-water group ($199.75{\pm}33.83{\mu}g/mL$, $50.43{\pm}13.88{\mu}g/mL$, respectively) at 90 min post-administration. Based on cDNA microarray analysis, expressions of ALDH genes ALDH1a7 and ALDH18a1 and cytochrome P450 (CY450) gene CYP4a30b were upregulated in the alcohol-GB and alcohol-SGB groups compared to levels in the control group. Overall, the results suggest that both GB and SGB have hangover relieving effects by reducing blood acetaldehyde levels. The molecular mechanisms may involve ALDH activation and upregulated expression of alcohol metabolism-related genes such as ALDH and CYP450.

Monitoring on the Tea with Steaming and Drying Process of Germinated Buckwheat (메밀순의 증숙 및 건조에 따른 침출차 특성 모니터링)

  • To make the germinated buckwheat tea, soluble solid contents, total flavonoid contents and organoleptic properties were investigated under various steaming time and drying temperature. The optimum condition of soluble solid contents were 6.93 min of steaming time and 73.59$^{\circ}C$ of drying temperature. Total flavonoid contents were maximum under the condition of 5.22 min of steaming time and 79.05$^{\circ}C$ of drying temperature. The optimum condition of overall palatability was 6.00 min of steaming time and 77.33$^{\circ}C$ of drying temperature. The optimum ranges of soluble solid contents, total flavonoid contents and overall palatability of the tea were 5.4∼7.0 min of steaming time and 75∼8$0^{\circ}C$ of drying temperature. The values expected in the optimum ranges were also similar to the experimental values.

Changes in Contents and Composition of Insoluble Dietary Fiber during Buckwheat Germination (메밀 발아 중 불용성 식이섬유 함량과 조성의 변화)

  • To provide the basic information of buckwheat insoluble dietary fiber (IDF) and efficient material for the buckwheat processed foodstuffs, buckwheat (Fagopyrum egculentum Moench) was germinated at 1$0^{\circ}C$ for 7 days and the contents and composition of the insoluble dietary fiber were examined at 24 hour intervals. The NDF and ADF contents in ungerminated seeds were 20.27%, 18.95% on dry weight basis, respectively. During the germination period, the NDF and ADF contents increased gradually. The cellulose, hemicellulose and lignin contents in ungerminated seeds were 11.10%, 1.32% and 7.85%, respectively. During the germination period, the cellulose and lignin contents increased. However, there was no significant change in the hemicellulose contents. The composition of cellulose and hemicellulose in the NDF decreased with germination time, whereas that of lignin increased. The IDF contents obtained by Prosky method were higher than the NDF contents obtained by Van Soest method. However, the IDF and NDF contents were closely correlated(r=0.9785, p<0.01). The cellulose(Xl), hemicellulose(X2), lignin(X3) and soluble dietary fiber (SDF) (BL) showed the significant regression equation(p<0.001) with the root length(Y). The multiple regression equation was Y = -12.1306 + 0.9213xl - 0.1615$\times$2 + 0.1071$\times$3 + 0.7397$\times$4 and R2(coefficient of determination) was 0.942.

Changes in Contents and Composition of Dietary Fiber during Buckwheat Germination (메밀 발아 중 식이섬유 함량과 조성의 변화)

  • To provide the efficient application scheme of buckwheat dietary fiber and basic information of seed germination, buckwheat(Fagopyrum esculentum Moench) was germinated at 10$^{\circ}C$ for 7 days and the contents and composition of the total dietary fiber(TDF), insoluble dietary fiber(IDF), soluble dietary fiber(SDF) wire examined at 24 hour intervals. The TDF content in ungerminated seeds was 24.86o on dry weight basis. It decreased for the 1st day of germination, but gradually increased for 7 days afterwords. The contents of IDF and SDF in ungerminated seeds were 22.05, 1.42% respectively. The IDF and SDF contents decreased in the initial stage of germination, but then gradually increased. The composition of the IDF and SDF in the TDF during the germination period showed different tendencies. The IDF decreased with germination time until 5 days and then increased. The SDV increased until 5 days and then decreased gradually. The TDF contents obtained by AOAC method were generally higher than those obtained by Prosky method. The TDF contents obtained by the two method, however, were very closely correlated (r=0.9966, p< 0.01) The IDF(X1) and SDF(X2) showed the significant regression equation(p<0.01) with the root length(Y). The equation was Y: -12.6681+0.5089${\times}$ 1 $\div$ 0.6022Xa and R2 was 0.968.

Antioxidative, Antimicrobial and Cytotoxic activities of Fagopyrum esculentum $M{\ddot{o}}ench$ Extract in Germinated Seeds (발아 메밀 추출물의 항산화.항균활성 및 세포독성)

  • This research was conducted to investigate the possibilities of usage of germinated-buckwheat (Fagopyrum esculentum $M{\ddot{o}}ench$) by examining antioxidative, antimicrobial and cytotoxic effects of extracts from different germinated root length of buckwheat. Antioxidant activity $(RC_{50})$ was shown higher in extracts of non-germinated seed $(50.41\;{\mu}g/mL)$ and root length 10 mm $(80.57\;{\mu}g/mL)$, 2 mm $(93.77\;{\mu}g/mL)$, 5 mm $(107.09\;{\mu}g/mL)$ than BHT $(163.96\;{\mu}g/mL)$ as a synthetic antioxidant. In antimicrobial activity, non-germinated and germinated seeds were formed inhibitory zone against S. aureus $(4{\sim}10\;mm)$, P. aeruginosa $(2{\sim}9\;mm)$ at the concentrations of $10{\sim}40\;mg/mL$ but B. subtilis, E. coli and S. typhimurium were not apparent antimicrobial activity. Extracts of germinated seed also decreased their antimicrobial activity compared to non-germinated seed extract. In addition, the growth of Calu-6 was inhibited of both 5 mm root length germinated and non-germinated seeds $(800\;{\mu}g/mL)$ as 95.12% and 87.15%, respectively, but these did not show any influence on cytotoxic effect against MCF-7 and Caco-2 cell lines. Extracts of 2 mm and 5 mm germinated seeds were also inhibited against Calu-6 and SNU-601 cell lines.
