Characteristics of Malt Prepared with Covered Barley, Naked Barley and Wheat (쌀보리, 겉보리 및 밀을 이용한 엿기름의 특성)

  • Quality of Sikhe, the Korean conventional rice beverage depended on the characteristics of saccharifying activities of various amylases, intrinsic flavour, budding rate and so on. To improve the quality of Sikhe, characteristics of malt produced with wheat, covered barley and naked barley were evaluated. The germination rate of wheat was 82%, but those of naked and covered barley were 69% and 56% for 6 days, respectively. Malt prepared from germinated grains with 1.5~2.0 times length of buds had the highest saccharifying power. when the extraction of enzyme and reducing sugar was carried out at 5$0^{\circ}C$ for 4 hr, saccarifying power and reducing sugar contents were the highest. Malt of wheat had the highest saccharifying power. Malt of naked barley had higher saccharifying power than that of covered barley. The amylase types of wheat, covered barley and naked barley were similar to $\beta$-amylase.

Preparation of Sikhe with Brown Rice (현미를 이용한 식혜의 제조)

  • This study was conducted to investigate the possibility of using brown rice as a raw material for preparation of sikhe. The ${\alpha}-amylase$ activity of germinated brown rice was much lower than that of germinated barley; however, the extract and its sugar content of extract prepared from germinated brown rice was similar to those of germinated barley. The extract of sikhe prepared with brown rice at $60^{\circ}C$ for 4 hr was similar to that of milled rice for 1 hr. Sikhe prepared with brown rice did not differ from sikhe prepared from milled rice in sweetness, pH and acidity. Sensory evaluation of sikhe prepared with brown rice showed increase in aroma and decrease in texture compared with sikhe prepared with milled rice.

Effect of Red Light on Changes of Embryo Tissue of Barley during Germination (맥아제조시(麥芽製造時) 적색광(赤色光) 희사(熙射)에 의한 배유조직(胚乳組織)의 변화(變化))

  • This study was carried out to investigate the effect of the red light on embryo tissue of barley during germination. The solubility of starch in endosperm of germinated barley was different between dark and red treatments at the 3rd day of germination, but was increased by the red light from the 4th day of germination. Blue value of the starch in the germinated barley decreased rapidly from 0.42 at the 1st day to 0.13 at the 6th day in the dark, and same tendency was found in the red light, but blue value was lower in the red light than in the dark. Aleurone cell wall was swollen much faster in the red light than in the dark during germination. The cell wall was broken down more greatly in the red light than in the dark at the 5th day of germination.

Effect of Anaerobic Treatment on Carbohydrate-Hydrolytic Enzyme Activities and Free Amino Acid Contents in Barley Malt

  • Effects of acute anoxia on carbohydrate hydrolytic enzyme activities and free amino acid contents in malt were examined. Malts were prepared with barley grains germinated for 7 days which contained the highest levels of amylolytic and(1-3,1-4)-$\beta$-glucanase activities. $\alpha$-Amylase and $\beta$-amylase activities in malts were not significantly affected by anoxia for 5 or 10 h.(1-3,1-4)-$\beta$-Glucanase activity, however, decreased about 7 to 10% by anoxia for 5 or 10 h. Alanine and $\gamma$-aminobutyric acid content changed drastically. Alanine contents in malts increased by 2.2- and 2-fold, and $\gamma$-aminobutyric acid contents by 1.4- and 1.9-fold under anoxia for 5 and 10 h, respectively.

Varietal Differences of Germination and Emergence Capacity under Low Temperature Conditions in Barley (대맥에서 저온 발아 및 출현력의 품종간 차이)

  • Germination and emergence of 16 barley cultivars bred in Korea were studied under different low temperature conditions in laboratory at Suweon. This study revealed that the barley cultivars differed markedly in their germination rate index (GRI), emergence rate index (ERI), and dry matter weight. The seeds of Baekdong, Namhaebori, Gangbori and Buheung germinated better than others in $4^{\circ}C$ plot. Yeongsanbori, Kwangseong and Baekdong had very poor ERI values, but Buhobori, Namhaebori, Oweolbori, Albori and malting barley, Sacheon 6 had very good ERI values. Germination rate index were not correlated with emergence rate index under any temperature conditions. The correlation coefficient between ERI of $4^{\circ}C$ and $7^{\circ}C$ plots was very high. Those results showed that for screening the varieties or lines with good emergence capacity under low temperature conditions ERI, FS and dry matter weight would be available.

  • This study carried out to changes $\alpha$-amylase activity and isozymes in barley during germination in the dark and red light The specific activity of u-amylase was increased during the germination periods in the dark, giving 355.0 and 523.7 units/mg protein at 3 and 5 day, and the activity was increased by the red light up to 48 and 15% at 3 and 5 days of germination, respectively. The ratio of $\alpha$-amylase I and II was approximately 95:5 at both 3 and 5 days of germination in the dark while the different ratio was found by the red light i. e. 60 : 40 and 90 : 10 at 3 and 5 days of germination, respectively.

  • This study was intended to obtain the basic information on the germination status, changes of $\beta$-amylase activity and reducing sugar contents during seed germination of barley after an imposition of water stress by adding Polyethylene glycol solution. The effects of water deficit on germination were to be decreased in germination rate and velocity. Respiratory Quotient was increased as the osmotic potential in culture solution was decreased. But $\beta$-amylase activity and reducing sugar contents were decreased and the ratio of dry to fresh weight of germinated seeds was increased as the osmotic potential of the culture solution was decreased.

Effects of Dimethipin on α-amylase Activity of Barley Seeds (보리 종자의 α-아밀라아제 활성에 미치는 Dimethipin의 영향)

  • Effects of dimethipin on ${\alpha}$-amylase activity of barley seeds were investigated. In the treatments of $1{\mu}M\;and\;10{\mu}M$ dimethipin, the indexes of germination were reduced to 17% and 24 % respectively. After seed germination, dimethipin was added to germinated seedlings and then the seedlings were kept to measure seedling length under illumination for 7 days. In control, the length of seedling was 5.7 cm, but in the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, seedling lengths were 5.5 cm and 1.2 cm respectively. In the relationship between dimethipin concentrations and ${\alpha}$-amylase activities, there was a linear curve. The more dimethipin was added to the seeds, the more ${\alpha}$-amylase activities were inhibited. In the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, ${\alpha}$-amylase activities were reduced to 33% and 71% respectively. Dimethipin also inhibited ${\alpha}$-amylase activities increased by gibberellin and the content of soluble protein. Therefore, it could be suggested that dimethipin might inhibit directly the activities of hydrolysis enzymes including ${\alpha}$-amylase or the expression of ${\alpha}$-amylase genes as germination and seedling growth were severely disturbed.

Effect of Water Table Depth in Different Soil Texture on Quality of Barley and Wheat Grain (토성별 지하수위가 밀, 보리의 품질에 미치는 영향)

  • This experiment was performed to characterize the optimum water table level for the grain quality, seed germination and diastic power of barley(var. Olbori) and wheat(var. Grumil). Olbori and Grumil grew in the 550 liter plastic pot that filled with silt loam or sandy loam. During the whole growth period, the underground water level adjusted to be 20, 30, 40, 50 and 70cm. Filled grain ratio and specific gravity were not affected by soil texture and water table. Low level of water table caused the increase of 1,000 grain weight in wheat and barley, but soil texture didn't. Crude protein content tended to be high as the water table level was high, especially in wheat. Change in crude protein content was affected by underground water level more than soil texture. And the affection was slightly higher in sandy loam than silt loam, but the difference was small. The higher level of water table led to the lower crude lipid content in barley and wheat grain. Crude lipid content of both wheat and barley grain grown in sandy loam was higher than those grown in silt loam. As the water table level down, the ash content of barley and wheat grain tend to increase, especially in sandy loam. Wheat flour yield was not affected by soil texture. It was about 65% at 20cm of water level and above 67% at 40cm water level. The seed germination of wheat and barley was more than 95% when the seeds were placed at 2$0^{\circ}C$ for three days. Regardless of soil texture, the lowest germination was seen at 20cm of water table level. And the seed germination rate increased as the underground water level became low. Above 89% of barley grains were germinated within 48 hours except 20cm level of water table in sandy loam. Diastic power of germinated barley was the lowest at 20cm of water table level, and it was almost unchanged below 30cm of water table level. And also it was not affected by soil texture.

Dough Properties and Quality Characteristics of Breads added with Barley Flour (보릿가루 첨가 복합분의 반죽 물성 및 빵의 품질 특성)

  • This study was carried out to investigate the rheological properties of dough and the quality characteristics of breads with Saesalbori (non-waxy barley) and Saechalbori (waxy barley) flours added at concentrations of 10, 20, and 30% to wheat flour. The maximum viscosity increased in the Saesalbori flour mixtures and decreased in the Saechalbori flour mixtures. The dough stability of the 10% barley flour mixtures was equal to that of the control while that of the 30% barley flour mixtures decreased more. The extensibility of the dough decreased with the increasing level of barley flour in all the blends, but the resistance of the dough increased. As the ratio of barley flour increased, the loaf volume of the breads significantly decreased, but the bread weight increased. The 10% Saesalbori flour mixtures increased the hardness of the breads but did not cause any significant change in the other TPA parameters while the 10% Saechalbori flour mixtures did not cause any change in all the TPA parameters. The sensory evaluation results showed that the 10% barley flour mixtures had no significantly different overall acceptance from the control, and that the 20% substitution still resulted in acceptable sensory qualities. The results of the study also showed that the bread-making properties of Saesalbori were improved by germination, but those of Saechalbori declined.
