생쥐 악하선으로부터 분리한 전 RNA를 poly(U)-sepharose chromatography와 sucrose linear density gradient centrifugation 방법으로 레닌 mRNA를 분리하여 in vitro translation과 immunoprecipitation에 의하여 레닌 mRNA를 확인하였다. 레닌 mRNA로부터 레닌 cDNA를 합성하여 EcoRI inker를 이용하여 pUC19에 삽입시켰고, Taq, polymerase를 이용한 PCR방법으로 합성한 레닌 cDNA는 pUC19의 Hinalll 부의에 삽입하여 재조합 plamid를 각각 작성하였다. 이것을 JM103에 형질전환시켜 레닌 유전자 발현을 유도하여 45,000의 분자량을 갖는 레닌을 얻었으며 이 레닌 단백은 토끼으 혈압을 850115mmHg에서 115-140mmHg로 증가시키는 생리 활성을 나타냈다. 토끼의 신장 DNA를 EMBL3 phage에 삽입시켜 genomic library를 작성한 후, 레닌 cDNA로부터 합성한 probe로 plaque hybridization시켜 genomic 유전자를 갖는 재조합 phage를 분리하였다.

The method of single-cell RNA sequencing has been rapidly developed, and numerous experiments have been conducted over the past decade. Their results allow us to recognize various subpopulations and rare cell states in tissues, tumors, and immune systems that are previously unidentified, and guide us to understand fundamental biological processes that determine cell identity based on single-cell gene expression profiles. However, it is still challenging to understand the principle of comprehensive gene regulation that determines the cell fate only with transcriptome, a consequential output of the gene expression program. To elucidate the mechanisms related to the origin and maintenance of comprehensive single-cell transcriptome, we require a corresponding single-cell epigenome, which is a differentiated information of each cell with an identical genome. This review deals with the current development of single-cell epigenomic library construction methods, including multi-omics tools with crucial factors and additional requirements in the future focusing on DNA methylation, chromatin accessibility, and histone post-translational modifications. The study of cellular differentiation and the disease occurrence at a single-cell level has taken the first step with single-cell transcriptome and is now taking the next step with single-cell epigenome.
카테콜아민 생합성에 관여하는 마지막 효소인 phenylethanolamine Nmethyltransferase는 norepinephrine 을 epinephrine으로 전환시키는 중요한 효소이다. PNMT효소의 발현은 epinephrine 신경세표의 발현에 필수적이다.따라서 PNMT 유전자를 크로닝하여 그 구조를 결정하고,유전자 발현연구를 하는 것은 상당히 중요한 일이다.그러나 최근에 저자가 bovine 및 human cDA 를 처음으로 분리하여 그 구조를 보고한 것 외에는 아직까지 인간과 백서 전체 genomic DNA 의 분리 보고는 없다.이에 저자들은 인간과 백서 PNMT유전자의 전체구조와 여러종(species)사이의 진화적인 관계를 규명하기 위해서 human 과 Rat genomic library 를 만들고,이 library 를 이용하여 bovine cDNA 를 probe로 13.1kb와 13.2kb길이의 인간과 백서의 genomic clone 을 분리 크리닝하는데 성공하여 유전자의 구조적 규명하였다.
Three clones containing mouse CD4 gene were prepared using AKR genomic cosmid library. The role of 6, 500 bp 5' flanking region of the first exon of the AKR CD4 gene in tissue or developmental stage specific expression of CD4 has been studied. The deletion constructs containing various amounts of CD4 5' flanking sequences were prepared, and they were transfected into the cell lines representing different cell types or developmental stages of CD4 expression. Study of the reporter gene expression revealed that at least 1, 700 bp of 5' flanking region did retain promoter activity for CD4 expression. This area did not seem to contain enhancer activity for a full expression of CD4. However, the putative promoter interacted with other tissue specific enhancer sequence and showed the tissue specificity of the enhancer element.
Enterobacter sp. B54 inhibited growth of the fungus Phytophthora capsici on potato dextrose agar (PDA). Three mutants with antifungal activities (denoted M54-47, M54-113, and M54-329) which were lost or increased, through Pl::Tn5 lac mutagenesis, were used to isolate genes responsible for fungal inhibition on PDA. Two clones were selected from the partially EcoR1-digested genomic library of the wild-type strain by probing with genomic flanking sequences of each mutant. We have isolated a 20-kb EcoR1 genomic DNA fragment from this strain that contains genes involved in hyphal growth inhibition of P. capsici on PDA. Subcloning and expression analysis of the above DNA fragment identified a 8-kb region which was necessary for antifungal activities. A 8-kb HindⅢDNA fragment covers three genomic loci inserted by Tn5 lac in each mutant. This suggested that all genes which are related to antifungal activities might be clustered in simple forms of at least 5-8 kb sizes.

Using the genomic library constructed at the downstream of the niiA promoter, which induces the over-expression of an inserted DNA fragment, we have attempted to screen the genes affecting growth or development by over-expression. The wild-type strain was transformed using the AMA-niiA(p) library and cultured on 1.2 M sorbitol media, in which asexual sporulation is induced, but sexual development is repressed. Over 100,000 strains transformed to $pyrG^+$ were analyzed with regard to any changes in phenotype. Consequently, seven strains were isolated for further analyses. These strains were designated NOT [niiA(p) over-expression transformants] stains. Four of the strains were of the inducible type, and the remaining strains were of the multi-copy suppression type. Two of the inducible-type strains, NOT 1 and NOT40, harbored genes which had been inserted in reverse direction, suggesting that the mutant phenotypes had been derived from an excess amount of anti-sense mRNA. Domain analyses of the deduced polypeptides from the DNA fragments rescued from the transformants revealed that NOT1, NOT40 and NOT6 harbored a LisH motif, a forkhead domain, and a $Zn(II)_2Cys_6$ binuclear zinc cluster, respectively.
Porphyromonas gingivalis, a Gram negative, anaerobic, asaccharolytic rod, is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. A 30 kDa (heated 24 kDa) hemin-binding protein whose expression is both hemin and iron regulated has recently been purified and characterized in this oral pathogen. This study has identified a hemin-binding P. gingivalis protein by expression of a P. gingivalis genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. A library of genomic DNA fragments from P. gingivalis was constructed in plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$ , and screened for recombinant clones with hemin-binding activity by plating onto hemin-containing agar. Of approximately 10,000 recombinant E. coli colonies screened on LB-amp-hemin agar, 10 exhibited a clearly pigmented phenotype. Each clone contained various insert DNA. The Hind III fragment transferred to the T7 RNA polymerase/promoter expression vector system produced a sligltly smaller (21 kDa) protein, a precursor form, immunoreactive to the antibody against the 24 kDa protein, suggesting that the cloned DNA fragment probably carried an entire gene for the 24 kDa hemin-binding protein.
탄저균(Bacillus anthracis)은 탄저(Antrax)의 원인균으로 사람은 물론, 초식동물인 소, 양, 말 등에서 급성의 폐사성 전염병을 일으킨다. 현재 사용되고 있는 탄저 치료 및 예방법은 항생제 치료와 약독화 백신주를 토대로 하고 있으나, 항생제 내성주의 출현 및 잔류 병원성이 문제시 되고 있는 실정이다. 따라서, 인체에 적용 가능하며 보다 안전한 탄저 치료제 및 백신 개발이 요구되고 있으며, 최근 탄저균 아포 및 영양세포, 그리고 탄저독소(Anthrax toxins)에 대한 동시 면역을 유도하는 다가백신 개발이 보고된 바 있다. 본 연구에서는, 향후 탄저균에 대한 새로운 다가백신 후보물질 발굴을 위하여, 탄저균에 대한 전장 유전자 발현 라이브러리(whole genomic expression library)를 구축하였다. 라이브러리 구축을 위하여, 탄저균(ATCC 14578) 게놈 DNA를 Sau3AI으로 부분 제한효소 처리였고, 유도 발현이 가능한 pET30abc 벡터에 접합시킴으로써, 총 $1{\times}10^5$개에 해당하는 대장균 BL21(DE3) 유래의 전장 유전자 발현 라이브러리를 구축하였다. 염기서열분석을 통한 중복성(redundancy) 확인 결과, 111개의 무작위 클론 중 56개(50.5%)가 탄저균 유전자로 확인되었으며, 17개(15.3%)는 벡터 유전자였고, 38개(34.2%)는 BLAST 탐색에서 일치하는 유전자를 찾지 못하였다. 또한 웨스턴 분석을 통하여 단백질 유도발현을 확인하였으며, 탄저균 항혈청에 대한 colony blot으로부터 양성반응을 보이는 일부 클론들을 확인할 수 있었다. 이러한 결과물들은, 구축된 전장 유전자 발현 라이브러리가 향후 탄저균에 대한 면역체(immunome) 분석을 위해 적용 가능함을 암시한다.
염색질을 구성하는 histone 단백질 lysine 잔기에 histone acetylase에 의하여 결합된 acetyl기를 제거하는 histone deacetylase (HDAC)는 진핵세포 생물의 염색질 안정 파 및 유전자 발현에 매우 큰 영향을 미친다. 국내에서 분리된 영지의 HDAC 유전자를 클로닝 하고자 cDNA 및 genomic DNA를 대상으로 PCR을 수행한 결과 470bp의 cDNA유전자와, 585 bp, 589 bp 및 630 bp길이의 genomic DNA유전자 조각을 클로닝 하였다. 이들의 염기서열을 근거로 아미노산 서열을 다른 균류의 HDAC와 비교한 결과 59-72%의 상동성을 보였다.
Medicago truncatula is a diploid legume plant related to the forage crop alfalfa. Recently, it has been chosen as a model species for genomic studies due to its small genome, self-fertility, short generation time, and high transformation efficiency. M. truncatula engages in symbiosis with nitrogen-fixing soil bacterium Rhizobium meliloti. M. truncatula mutants that are defective in nodulation and developmental processes have been generated. Some of these mutants exhibited altered phenotypes in symbiotic responses such as root hair deformation, expression of nodulin genes, and calcium spiking. Thus, the genes controlling these traits are likely to encode functions that are required for Nod-factor signal transduction pathways. To facilitate genome analysis and map-based cloning of symbiotic genes, a bacterial artificial chromosome library was constructed. An efficient polymerase chain reaction-based screening of the library was devised to fasten physical mapping of specific genomic regions. As a genomics approach, comparative mapping revealed high levels of macro- and microsynteny between M. truncatula and other legume genomes. Expressed sequence tags and microarray profiles reflecting the genetic and biochemical events associated with the development and environmental interactions of M. truncatula are assembled in the databases. Together, these genomics programs will help enrich our understanding of the legume biology.
