Alteration of genetic make-up of the isolates and mono-sporidial strains of Tilletia indica causing Karnal bunt (KB) disease in wheat was analyzed using DNA markers and SDS-PAGE. The generation of new variation with different growth characteristics is not a generalized feature and is not only dependant on the original genetic make up of the base isolate/monosporidial strains but also on interaction with host. Host determinant(s) plays a significant role in the generation of variability and the effect is much pronounced in monosporidial strains with narrow genetic base as compared to broad genetic base. The most plausible explanation of genetic variation in presence of host determinant(s) are the recombination of genetic material from two different mycelial/sporidia through sexual mating as well as through parasexual means. The morphological and development dependent variability further suggests that the variation in T. indica strains predominantly derived through the genetic rearrangements.
An ethanol tolerant mutant was selected by successive transfers of Clostridium thermohydrosulfuricum ATCC 33223 into the media with progressively higher ethanol concentrations. The growth kinetics of the mutant were characterized under various growth conditions. Physiological differences such as enhanced growth, tolerance to various solvents, alteration of the optimum temperature and the ratio of end products during fermentation were noticed in the mutant.
The genetic toxicity of environmental pollutants, namely, nonylphenol (NP), bisphenol A (BPA) and chloropyriphos (CP) was investigated in aquatic sentinel species, freshwater crustacean, Daphnia magna, and larva of aquatic midge, Chironomus tentans, using Comet assay. Physiological effect of such pollutants was also investigated by studying the specimens' rates of reproduction, growth and survival. Acute toxicity results showed that, as expected, Daphnia was more sensitive than Chironomus to chemical exposure. The order of acute toxicity was CP > NP > BPA in D. magna and NP > CP > BPA in C. tentans. BPA may exert a genotoxic effect on D. magna and C. tentans, given that DNA strand breaks increased in both species exposed to this compound, whereas NP- and CP-induced DNA damage occurred only in C. tentans. In vivo genotoxic data obtained in aquatic sentinel species could provide valuable information for freshwater quality monitoring. The experiments with NP-exposed D. magna showed that the pollutant has long-term effects on reproduction, whereas no short-term effect on DNA integrity was found, being an example of a false-negative result from the biomarkers perspective. This result could be interpreted that other mechanism than genetic alteration might be involved in NP-induced reproduction failure in D. magna. False-positive results from the genotoxic biomarker obtained in BPA-exposed D. magna and in NP-exposed C. tentans make it difficult to use DNA integrity as an early warning biomarker. However, as the mere presence of genotoxic compounds, which are potentially carcinogenic, is of high concern to human and ecosystem health, it could also be important to rapidly and effectively detect genotoxic compounds in the aquatic system in ways that do not necessarily accompany a higher level of alteration. Considering the potential of D. magna and C. tentans as bioindicator species, and the importance of genotoxic biomarkers in ecotoxicity monitoring, DNA damage in these species could provide useful information for environmental risk assessment.
Food production is a major role of agriculture. It has been projected that the world population continues to increase by the middle of the 21st century, and the population growth results in raising a serious problem of food shortage. Thus we have to increase food as possible. A considerable amount of crops have been abandoned due to short-life after postharvest. Ethylene is a factor responsible for the postharvest loss in crops, especially horticultural crops. If we can reduce ethylene production or sensitivity by genetic engineering, we can develop, so called,“long-life crop”conferring long postharvest lives. During last two decades, intensive research for molecular dissection of ethylene biosynthesis has been carried out, and the researchers have succeeded in engineering ethylene productivity in some crops. On the other hand, after the successful isolation of Arabidopsis ethylene receptor gene ETR1, the homolog genes have been isolated in various plant species. Currently the characterization of these genes and alteration of ethylene sensitivity using the genes are in progress. This review summarizes current progress in the analysis of these genes, and discusses genetic engineering of ethylene sensitivity using these genes.
World-class magnesite deposits are developed in the Dashiqiao mineralized district of the Jiao-Liao-Ji Belt in China. This belt extends to the northern side of the Korean Peninsula and hosts major magnesite deposits in the Dancheon region of North Korea. Magnesite ores from the Pailou deposits in the Dashiqiao district is classified into pure magnetite, chlorite-magnetite, chlorite-talc-magnetite, and dolomite groups depending on the constituent minerals. According to the result of petrographic study, magnesite was formed by the alteration of dolomite, and, talc, chlorite, and apatite were produced as late-stage alteration minerals that replaced the magnesite. Fluid inclusions observed in magnesite are a liquid-type inclusion, with a homogenization temperature of 121-250 ℃ and a salinity of 1.7-22.4 wt% NaCl equiv. The chlorite geothermometer, indicating the temperature of hydrothermal alteration, is 137~293 ℃, slightly higher than the homogenization temperature of fluid inclusions, and the pressure is calculated to be less than 3.2 kb. For magnesite mineralization in the study area, the initially formed-dolomite was subjected to replacement by Mg-rich fluid to form a magnesite ore body, and then it was enriched through regional metamorphism and hydrothermal alteration. It seems that altered minerals such as talc were crystallized by Si and Al-rich late-stage hydrothermal fluids. These results are similar to the genetic environments of the Daeheung deposit, a representative magnesite deposit in North Korea, and it is believed that the two deposits went through a similar geological and ore genetic process of magnesite mineralization.
The purpose of statistical analyses of array-CGH experiment data is to divide the whole genome into regions of equal copy number, to quantify the copy number in each region and finally to evaluate its significance of being different from two. Several statistical procedures have been proposed which include the circular binary segmentation, and a Gaussian based local regression for detecting break points (GLAD) by estimating a piecewise constant function. We propose in this note a penalized spline regression and its simultaneous confidence band(SCB) approach to evaluate the statistical significance of regions of genetic gain/loss. The region of which the simultaneous confidence band stays above 0 or below 0 can be considered as a region of genetic gain or loss. We compare the performance of the SCB procedure with GLAD and hidden Markov model approaches through a simulation study in which the data were generated from AR(1) and AR(2) models to reflect spatial dependence of the array-CGH data in addition to the independence model. We found that the SCB method is more sensitive in detecting the low level copy number alterations.

Milyang pyrophyllite deposit which was formed by hydrothermal alteration occurs in Late Cretaceous andesitic tuff in the Milyang area, Gyeongsangnamdo. The wall rock alteration and genesis of the Milyang pyrophyllite deposit were studied. The ore minerals are composed dominantly of pyrophyllite accompanied by small amounts of quartz, kaolinite, pyrite, dumortierite and diaspore. The alteration halo of this deposit can be divided into three zones on the basis of mineral assemblage; pyrophyllite, sericite and chlorite zone. The common mineral assemblages of each alteration zone are as follows: (1) pyrophyllite zone; pyrophyllite-quartz-kaolinite-pyrite-dumortierite-diaspore, (2) sericite zone; sericite-quartz-pyrite-kaolinite, and (3) chlorite zone; chlorite-plagioclase-quartz. Major element chemistry shows that characteristic depletion in MgO, CaO, and $Na_2O$ and relative increase in FeO from less altered chlorite zone to extensively altered pyrophyllite zone corresponding to variation in mineral assemblages. The paragenesis of ore minerals, oxygen isotope data, chlorite and illite geothermometry suggest that ore deposit was formed at about $250{\sim}330^{\circ}C$. Both hydrogen and silica activities are high in pyrophyllite zone. Potassium activity increases in sericite zone while hydrogen activity becomes low in chlorite zone. The pyrophyllite zone was formed relatively higher temperature than those of sericite and chlorite zones. The ore fluid was considered to be magmatic water in origin derived from the residual granitic magma which interacted with meteoric water.
Mineral assemblages, mineral chemistries and stable isotope compositions of altered rocks of the Ogmae, Seongsan, Haenam and Gusi mines near the Haenam volcanic field in the southwestern part of the Korea peninsula were studied. Characteristic hydrothermal alteration zones in these deposits occurring in the Cretaceous volcanics and volcanogenic sediments, acidic tuff, and rhyolite, were outlined. Genetic environment with particular reference to the spatial and temporal relationships for these deposits were considered. The alteration zones defined by a mineral assemblage in the Ogmae and Seongsan deposits can be classified as alunite, pyrophyllite, kaolinite or dickite, quartz, illite or illite/smectite. Alunite was not developed in the Gusi and Haenam deposits. Boundaries between the adjacent zones are always gradational except for vein-type alunite. Alteration zones are superimposed upon each other in some localities. These deposits formed $71.8{\pm}2.8{\sim}76.6{\pm}2.9$ Ma ago, which is the almost same age of later volcanic rocks $79.4{\pm}1.7{\sim}82.8{\pm}1.2$ Ma, the Haenam Group, corresponding to Campanian. It indicates that hydrothermal alteration of these deposits appeared to be related to felsic volcanism in the area. Consideration of the stability between kaolinite, alunite, pyrite and pyrophyllite, and the geothermometry based on the mineral chemistry of illite and chlorite suggests that the maximum formation temperature for alunite and pyrophyllite can be estimated at about $250^{\circ}C$ and $240{\sim}290^{\circ}C$, respectively. It also suggests that these deposits were formed by acidic sulfate solution with high aqueous silica and potassium activity in a shallow depth environment. Compositional variation of alunite also suggests that the physico-chemical conditions fluctulated considerably during alteration processes, indicating shallow depth environment. The Haenam deposit was formed at a relatively greater depth than the others. The sulfur isotope composition of alunite and pyrite indicates that sulfur probably had a magmatic source, and the oxygen isotope composition for kaolinite indicates that the magmatic hydrothermal solution was diluted by circulating meteoric water.
Aging is one of the most complicated biological processes in all species. A number of different model organisms from yeast to monkeys have been studied to understand the aging process. Until recently, many different age-related genes and age-regulating cellular pathways, such as insulin/IGF-1-like signal, mitochondrial dysfunction, Sir2 pathway, have been identified through classical genetic studies. Parallel to genetic approaches, genome-wide approaches have provided valuable insights for the understanding of molecular mechanisms occurring during aging. Gene expression profiling analysis can measure the transcriptional alteration of multiple genes in a genome simultaneously and is widely used to elucidate the mechanisms of complex biological pathways. Here, current global gene expression profiling studies on normal aging and age-related genetic/environmental interventions in widely-used model organisms are briefly reviewed.

In order to investigate genetic stability and gene expression profile after cloning procedure, two groups of cloned pigs were used for swine leukocyte antigen (SLA) gene nucleotide alteration and microarray analyses. Each group was consist of cloned pigs derived from same cell line (n=3 and 4, respectively). Six SLA loci were analyzed for cDNA sequences and protein translations. In total, 16 SLA alleles were identified and there were no evidence of SLA nucleotide alteration. All SLA sequences and protein translations were identical among the each pig in the same group. On the other hand, microarray assay was performed for profiling gene expression of the cloned pigs. In total, 43,603 genes were analyzed and 2,150~4,300 reliably hybridized spots on the each chip were selected for further analysis. Even though the cloned pigs in the same group had identical genetic background, 18.6~47.3% of analyzed genes were differentially expressed in between each cloned pigs. Furthermore, on gene clustering analysis, some cloned pigs showed abnormal physiological phenotypes such as inflammation, cancer or cardiomyopathy. We assumed that individual environmental adaption, sociality and rank in the pen might have induced these different phenotypes. In conclusion, the results of the present study indicate that SLA locus genes appear to be stable following SCNT. However, gene expressions and phenotypes between cloned pigs derived from the same cell line were not identical even under the same rearing conditions.
