The major aim of this work is the research of secondary metabolites isolated from the aerial parts of Petasites japonicus. The plant material is extracted with a polar solvent, which is 95% by volume methanol at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAC and n-BuOH fraction, two bakkenolides and two caffoylquinic acid were isolated using the Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies. According to the results of the results of physico-chemical and spectroscopic data including NMR, MS and UV. The chemical structures of the compounds were respectively determined as bakkenolide B (1), bakkenolide D (2), 1,5-dicaffeoylquinic acid (3), and 5-O-caffeoylquinic acid (4). These results suggest that the compounds isolated from the aerial parts of this plant were almost identical with known components of Petasites japonicus. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest area and time.
The heavy metal concentrations of seawater collected from the Onsan coastal zone in February and July 1996 and mussels(Mytilus edulis) in February 1997 were analysed. The concentrations of cadmium in seawater were in the range of 0.008-2.988 ${\mu}g/L$, while the ranges of copper and zinc concentrations were 0.08-2.55, and 0.21-35.12 ${\mu}g/L$, respectively. The metal concentrations decreased gradually with increasing distances from Daejeong stream, indicating that this stream was the major source of heavy metal input into the Onsan coastal zone. The concentrations of cadmium, copper and zinc in mussels were in the ranges of 1.40-25.09, 8.5-64.5, and 46.8-291.2 ${\mu}g/g$, respectively. The metal concentrations decreased gradually with increasing distances from Daejeong stream. Among organs of mussels, gill showed the highest concentrations of cadmium and the digestive gland showed the highest concentrations of copper and for zine the kidney showed the highest concentrations. The digestive gland and kidney revealed high proportion of cadmium in cytosolic fraction and the percentage of copper was high in the kidney and that of zine was high in the digestive gland. Metal-binding protein of mussels collected from the mouth of Daejeong stream was separated, using gel-filtration chromatography. In the kidney and gill of mussels, most of cadmium was associated with metal-binding protein. In contrast, most of the metal in the digestive gland and remaining tissues is bound to high molecular weight protein rather than metal-binding protein.

Proteins from sunflower seeds were hydrolyzed with Alcalase and Flavourzyme to isolate iron-binding peptides. The optimal hydrolysis conditions were determined. Hydrolysates were filtered under a 3 kDa membrane and iron-binding peptides separated from the hydrolysates using ion exchange and gel permeation chromatographic methods. A fraction with the highest iron-binding activity (Fe/peptide, 0.69), F22, was obtained. These results suggest that fractions isolated from sunflower seed protein hydrolysates can be applied toward the production of iron supplements.
In the course of searching for acetylcholinesterase inhibitors from crude drugs, it was found that total MeOH extract of Corydalis Tuber showed significant inhibitory effect on acetylcholinesterase. To isolate acetylcholinesterase inhibitors from Corydalis Tuber, total MeOH extract of the the crude drug was subjected to activity guided fractionation. The MeOH extract was suspended in water and fractionated with methylene chloride and subjected to acid-base fractionation. Silica gel column chromatography of the basic fraction which showed significant inhibitory effect on acetylcholinesterase was carried out and 5 subfractions (1-5) were obtained. From subtraction 4, compound I was isolated. The structure of isolated compound I was identified by spectroscopic parameters of $^1H-NMR$, $^{13}C-NMR$, EI-MS and FAB-MS. The compound I was identified as berberine. It was found from the Lineweaver-Burk plot that berberine was a reversible and specific inhibitor of acetylcholinesterase having 90% inhibitory effect at the concentration of $2.5{\mu}M$.
Colloidal coating solution was prepared to enhance the hydrophilic property of the film surface. Water and ethanol were used as the dispersion media and (glycidoxypropyl) trimethoxysilane (GPS) as a binder in the colloidal silica coatings. Ethylene diamine was added to the colloidal silica solution as the curing agent. The colloidal silica solution was regarded as a hard-sphere suspension model with low volume fraction of the silica particles. Rheological properties of the silica suspensions modified with GPS have been investigated as a function of pH and concentration. The acidic solution showed high viscosity change by fast hydrolysis reaction and adsorption of the organic binders on the surface of silica particles. However, the hydrolysis was slow at the basic condition and the binders combined with themselves by condensation. The viscosity change was smallest at pH 7. The viscosity increased with the curing time after adding ethylenediamine, and the increase of viscosity at low pH was higher than that at high pH. The hydrophilic properties of the coating film were investigated by the contact angle of water and film surface. The smallest contact angle was shown under the strong acidic condition of pH 2.
Effect of the cellulase treatment on the physicochemical properties of three varieties of rice (chucheongbyeo, chosengtongilbyeo and IR 36) and the texture of cooked rice were investigated. The swelling power and solubility of the rice flours were increased and amylographic viscosities, especially cold viscosities were decreased by cellulase treatment in all varieties. Gel chromatography of soluble carbohydrates from cellulase-treated rice flours on sepharose 2B-CL showed a singificant increase of low molecular weight $(10^{4})$ fraction which might be produced upon hydrolysis in endosperm cell wall constituents. The hardness of cooked rices prepared from cellulose-treated rices significantly decreased.
The fruits of Pourthiaea villosa were extracted with methanol and its extract was fractionated with n-hexane, methylene chloride, ethyl acetate and n-butanol. Repeated column chromatography of silica gel, sephadex LH-20 and HPLC led to the isolation of nine phenolic compounds from ethyl acetate soluble fraction. The chemical structures were elucidated as kaemferol-3-O-${\beta}$-D-glucopyranoside (astragalin) (1), isorhamnetin-3-O-${\beta}$-D-glucopyranoside (2), kaempferol-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\alpha}$-L-rhamnopyranoside (3), caffeic acid (4), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-a-L-rhamnopyranoside (5), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\beta}$-D-glucopyranoside (6), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\beta}$-D-galactopyranoside (7), quercetin-3-O-${\alpha}$-L-rhamnopyranoside (quercitrin) (8), and kaempferol-3-O-${\alpha}$-L-rhamnopyranoside (afzelin) (9) by spectroscopic techniques. These compounds were isolated from this plant for the first time.

Helicobacter pylori is a well-known pathogen that is responsible for gastric disorders. Overcoming of the antibiotic-resistance is a main barrier to treat H. pylori infection. In our search for anti-H. pylori compounds from natural resources, bioactivity-guided isolation on the ethyl acetate fraction of Fraxinus mandshurica bark that had shown anti-H. pylori activity gave twelve compounds (1 - 12) of six coumarins, three phenylethanoids, two secoiridoids, and a lignan using silica gel column chromatography, Sephadex-LH 20, and recrystallization. The chemical structures were identified by spectroscopic data analysis, including 1D, 2D NMR, and mass spectrometry. Among them, compounds 2, 10, and 11 showed moderate growth inhibitory activity against three strains of H. pylori, compared with positive controls of quercetin and metronidazole. Compounds 5, 6, 8, and 12 exhibited the inhibitory activity against strains 26695 or 43504. This is the first report on the anti-H. pylori activity of this plant and the isolated compounds.

A 1.25 kb DNA fragment including the lscR gene, which encodes a levansucrase of Rahnella aquatilis ATCC 15552, was subcloned into a high-expression vector, pET-29b, and the recombinant enzyme was overexpressed in Escherichia coli. Most of the levansucrase activity was detected in the cytoplasmic fraction after induction with isopropyl ${\beta}-D-thiogalactoside$. The recombinant enzyme with a tag of six histidine residues at the C-terminus was purified 146-fold by affinity and gel-filtration chromatographies. The molecular mass of the purified LscR was approx. 49 kDa as determined by SDS-PAGE. The optimum pH and temperature of this enzyme for levan formation was pH 6.0 and $30^{\circ}C$, respectively. The optimum substrate concentration for levan formation was 300 mM sucrose. Levan formation was increased by the increase of the enzyme concentrations. Maxium yield of levan formation at optimum substrate concentration, pH, and temperature after 24 h of reaction was approximately 80%.
Components I and II (CI&II) are major phosphoproteins in the frog rod outer segments (ROS) of retina, whose phosphorylation is light- and cyclic nucleotide-dependent. Although it was reported that CI & II could be chemically cross-linked to ${\beta}{\gamma}-subunit$ of transducin (${\beta}{\gamma}_t$), it was not clear whether CI&II physically interact with ${\beta}{\gamma}_t$, under native conditions. CI&II extracted by hypotonic washing fo ROS membranes showed an overlapped migration with ${\beta}{\gamma}_t$, in sucrose density gradient centrifugation. The elution profile of CI&II in the peripheral membrane fractions from gel filtration chromatography also overlapped that of ${\beta}{\gamma}_t$. These hydrodynamic parameters indicate that the native molecular state of CI&II in the peripheral membrane fraction appears to be within a complex, most likely with ${\beta}{\gamma}_t$. CI&II coeluted with ${\beta}{\gamma}_t$, showed no phosphorylation by endogenous kinase which phosphorylates a serine of CI&II in other fractions. The purified CI&II were not able to inhibit trypsin-activated cGMP-phosphodiesterase, and CI&II were not recognized by a monoclonal antibody against the ${\gamma}-subunit$ of transducin, indicating that CI&II are not y-subunit of PDE or transducin. Thus, it is likely that native CI&II, which undergo a light-dependent phosphorylation/dephosphorylation cycle, can associate with ${\beta}{\gamma}$, in frog photoreceptor membranes, and the complex formation has an inhibitory effect on the endogenous phosphorylation of CI&II.
