Isolation and Identification of Antimicrobial Compound from Dansam (Saliuia miltiorrhiza Bunge) (단삼으로부터 식품부패미생물에 대한 항균성 물질의 분리 및 동정)

  • Antimicrobial effect of Dansham (Saliuia miltiorrhiza Bunge) was investigated. Methanol extract of dried Dansham was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Chloroform fraction showed the highest inhibitory effect on the microorganisms such as B. subtilis, S. aureus, E. coli, L. monocytogenes and V. parahaemolyticus at 250 $\mu\textrm{g}$/disc. Chloroform fraction was further fractionated by silica gel column and thin layer chromatography (TLC). The antimicrobial compound was isolated from their fractions and its chemical structure was identified as a cryptotanshinone by GC-MS and $^1$H-NMR, $^{13}$ C-NMR.

Changes in Polygalacturonase during Softening of Persimmon and Jujube Fruits (감과 대추의 연화중 Polygalacturonase의 변화)

  • This work was carried out to investigate change of polygalacturonase activities and polygalacturonase patterns by gel filtration chromatography during softening of persimmon and jujube fruits. During softening of two kinds of fruit, polygalacturonase activities of water-soluble and salt-soluble proteins were increased, but that of cell wall-bound proteins was decreased. In water-soluble and salt-soluble proteins of persimmon fruits, two peaks of polygalacturonase activity were separated in mature stages, but one peak in soft stages. During softening of those fruits, the peaks of polygalacturonase activity in water-soluble and salt-soluble proteins appeared on the same fraction with the peaks of polygalacturonase activity in cell wall-bound proteins.

Chromatographic Fractionation of Protease Inhibitors from Fish Eggs (어류 알로부터 Protease Inhibitors의 크로마토그래피법에 의한 분획)

  • A protease inhibitor from fish eggs was fractionated using chromatographic methods. The fractionation efficiency was evaluated in terms of specific inhibitory activity (SIA, U/mg), purity (fold), total inhibitory activity (TIA, U), and recovery (%). The protease inhibitor (PI) from egg extracts of skipjack tuna (ST Katsuwonus pelamis), yellowfin tuna (YT Thunnus albacares) and Alaska pollock (AP Theragra chalcogramma) was fractionated using Sephadex G-50 gel filtration and DEAE-Sepharose CL-6B anion exchange chromatography based on protein size exclusion and net charge, respectively. Fractions exhibiting strong inhibitory activity were contained in the 30-50 kDa fraction on gel filtration and in the range of 0.4-0.7 M NaCl gradient fraction on anion exchange chromatography. The respective TIA and percent recovery of the fraction obtained with gel filtration toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 2,758.7 U and 29.6% for ST, 1,005.5 U and 25.6% for YT, and 1,267.5 U and 26.0% for AP. Gel filtration chromatography was more effective at fractionating PI than using ion exchange chromatography. These results suggest that fish eggs act as serine protease inhibitors and might be useful for protease inhibition in foodstuffs.

Characteristics of Bitter Peptides from a Cheese and a Soybean Paste (치즈 및 된장에서의 쓴 맛 펩타이드 특성)

  • To characterize bitter peptides in fermented protein foods, peptides were extracted with 2:1 (v/v) chloroform-methand from various samples and separated into fractions I, II, and III by Sephadex G-25 gel chromatography. Amino acid compositions of Mozzarella cheese, soybean paste, and each fraction from the two samples were analyzed to calculate the average hydrophobicity. All the solvent extracts of the food samples had strong bitter taste, although the original samples did not taste bitter. The yield of solvent extraction ranged from 0.08 to 62.50% of total nigrogen of food samples. The average hydrophobicity calculated from the amino acid composition of Mozzarella cheese was 1376 cal/mole, solvent extract 1,623 cal/mole, gel chromatography traction I, 1,797 cal/mole, fraction II, 2,454 cal/mole, and fraction III, 1,559 cal/mole. In the case of soybean paste, the average hydrophobicity of original sample, solvent extract, gel chromatography fraction I, II, and III wre 1,229, 1,654, 1,900,998 cal/mole, respectively. The important amino acids in bitter peptides were leucine, 2016, phenylalanine, proline, and voline.

Separation of Ether-Soluble Neutral Extract of a Commercial High-Grain Feed Stimulating Hay Intake in Cattle

  • Appetite stimulant ether-soluble neutral extract of a high-grain compound feed was separated into four fractions with an open column of silica gel chromatograph using four carrier solutions containing n-pentane and diethyl ether as 100:0 (Fraction A), 90:10 (Fraction B), 75:25 (Fraction C) and 0:100 (Fraction D). The stimulative effects of the fractions were examined by comparing the intake of hay (with or without the fractions) in two-choice test bioassays with cattle. The Fractions A, B and C of the neutral extract stimulated (p<0.05) hay consumption in cattle, whereas Fraction D had no effect on selective feeding when compared with the control fraction. Furthermore, Fractions A, B and C had higher (p<0.05) feeding stimulative indices (FSI) than that of the Fraction D. The results suggest that chemical stimulants to increase palatability of hay are present in the neutral Fractions A, B and C of high-grain concentrate.

Antioxidant Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항산화 효과)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-oxidant activity of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of Pyrus ussuriensis were extracted with acetone-$H_2O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The concentration of total phenolic compound of Pyrus ussuriensis acetate soluble fraction was high, 914 mg/g. The results obtained from the analyses of the anti-oxidanat effects of Pyrus ussuriensis acetate fraction can be summarized as follows: In the result of DPPH scavenging radical activity, Pyrus ussuriensis acetate soluble fraction showed more than 80% at 100 ppm. SOD-like activity of one of Pyrus ussuriensis acetate soluble fractions was 77% at 1000 ppm. Xanthine oxidase inhibition of Pyrus ussuriensis acetate soluble fraction was 38% at 100 ppm. From these results, we confirmed that acetate fraction of Pyrus ussuriensis has a great potential as a natural ingredients with a natural antioxidant and antimicrobial source.

Effect of Solidago altissima L. Extract on Forage Crop Germination

  • There are 28 families and 166 species of exotic weeds on agricultural land and among these, 23 families and 80 species of exotic weeds occur on pastures. Among them, the Solidago altissima is a perennial weed belonging to the asteraceae family and it is an exotic weed that spreads to the surrounding area using methods such as high seed production, vegetative propagation using underground rhizomes and allelochemical. Accordingly, in 2009, the Ministry of Environment designated it as an ecosystem-disrupting species. This study was conducted to obtain basic data about the effects of S.altissima derived allelochemicals on forage crops. The root of S.altissima was separated, dried in the shade and then pulverized to prepare an root powder. Powder was repeatedly extracted with methanol for 3 days and concentrated under reduced pressure to obtain an root methanol extract. Dissolve the extract in distilled water, dispense it in a separate-funnel and proceed with liquid-liquid extraction by adding equal amounts of n-haxane (Hex), chloroform (CHCI3), ethyl acetate (EtoAC), and butanol (BuOH) in order of increasing polarity. A seed-bioassay was performed using fractions for each solvent, followed by separation and purification by silica gel column chromatography. As a result of the fraction germination test for each solvent, the IC50 values using the fresh weight of each fraction were 898.3 mg L-1, 676.3 mg L-1, 1160 mg L-1 and 1360 mg L-1. CA, CB, and CC fractions were obtained through primary silica gel column chromatography that used CHCI3 fraction. As a result of seed-bioassay using each fraction, the IC50 values for the fresh weight of each fraction was 537.3 mg L-1, 1280 mg L-1 and 1947 mg L-1. Based on this, 5 fractions were obtained as a result of secondary silica gel column chromatography using the CA fraction. A seed-bioassay was performed, as a result, the lowest IC50 value was calculated as 226.7 mg L-1 in the CAE fraction. Based on this, the fraction was analyzed by GC-MS. The results of this study can be used as basic research data on the effects of weeds on forage crops and allelochemicals secreted from S. altissima.

Varietal Variation in Structure and Physical Characteristics of Rice Endosperm Strach (쌀 배유전분구조 및 물리적 특성의 품종 변이)

  • Varietal difference in structural and physical characteristics of rice starch and interrelationships between these starch properties and the physicochemical components of rice grains were investigated for twenty four rice varieties, which were considerably different in amylose content, gelatinization temperature and gel consistency, to obtain the basic informations for diversifying the utility of rice grains and for developing various rice cultivars adaptable to rice food processing. Wave length of maximum absorbance for blue color reaction of starch-I$_2$ complex was 520nm for glutinous rice and 560~600nm for nonglutinous rice. The higher amylose rice showed the longer wave length but there was considerable difference of the wave length among rice materials with similar amylose content. Varietal difference in frequency distribution of debranched starch molecules by isoamylase hydrolysis was largest in amylose fraction, the longest fraction(Fr. I), and was next bigger in intermediate fraction(Int.Fr.) and the ratio between short and long glucose chain of amylopectin fraction(Fr. III /Fr. II). The Fr. III / Fr II ratio was higher in glutinous rice than in nonglutinous one but it was considerably different among nonglutinous rice varieties without any connection with amylose content. The lower alkali digestible rice showed the longer half-hydrolysis time in acid solution. Both soft and hard gel rice varieties in gel consistency test revealed significant varietal difference of 50% acid hydrolysis time. The harder rice in gel consistency exhibited the less frequency of short branch fraction of amylopectin (Fr. III) and the higher amylose content. Twenty four rice varieties were cleary classified by upper two principal components contracted from structural or physical traits of rice starch and some physicochemical properties of rice grain by principal component analysis. The 1st and 2nd principal components can be characterized to components related with amylose and amylopectin structure respectively.

Antioxidative Activity and Cytotoxicity on Human KB cell of Extracts from Rhododendron mucronulatum Turcz. Flower (진달래꽃 추출물의 항산화 효과 및 인체 KB cell에 대한 세포독성)

  • This study was carried out to investigate the antioxidative activity and cytotoxicity on human KB cell of extract from Rhododendron mucronulatum flower, and also to determine the contents of total polyphenol and flavonoid. The methanol extract of Rhododendron mucronulatum flower was fractionated with various solvents such as hexane, chloroform, ethyl acetate, butanol and water. The DPPH radical scavenging activities of ethyl acetate fraction had stronger activity than other fractions. So, antioxidative substances of ethyl acetate fraction were crude purified by silica gel column chromatography. The DPPH radical scavenging activities of crude purified fraction 2 and 3 were more than 90% at 40 ppm. In the presence of $100{\mu}g/mL$, growth inhibition on human KB cell by WST-1 assay showed 81.2% in chloroform fraction and 74.6% in hexane fraction. The contents of total polyphenol and flavonoid of ethyl acetate traction were 32.70% and 20.30%, respectively, The antioxidative activity showed correlation with total polyphenol and flavonoid contents.

Biological Activity of Phenol Compound from a Cactus Cheonnyuncho (Opuntia humifusa) in Korea (천년초선인장으로부터 분리한 페놀성 화합물의 생리활성 효과)

  • After Cheonnyuncho (Opuntia humifusa) was extracted by 70% ethanol and partitioned with hexane, chloroform, ethyl acetate, butanol, and water in order, ethyl acetate fraction with excellent antioxidant activities was acquired. Ethyl acetate fraction was conducted by TLC, and the third spot of 10 spots was selected due to its best antioxidant activities. When the third fraction further isolated on silica gel chromatography, the fourth fraction of 10 fractions had the best antioxidant activities and purified by HPLC. The molecular weight on EI-MS, and $^1H$- and $^{13}C$-NMR spectrum showed that the purified compound was taxifolin. In addition, antioxidant activities were analyzed, and the purified compound from Cheonnyuncho was found to be effective as much as $\alpha$-tocopherol, BHA. According to the analysis on antibacterial activities, the purified compound also showed more activity than benzoic acid against all pathogenic bacteria.
