Determination of ${\beta}$-sitosterol in Unsaponifiable Fraction of Zea mays and Related Drug Preparations by HPLC (HPLC에 의한 Zea mays 불검화추출물과 그의 함유제제 중 ${\beta}$-시토스테롤의 정량)

  • A high-performance liquid chromatographic method for the determination of ${\beta}$-sitosterol in the unsaponifiable fraction of Zea mays L. and its related drug preparations using a cholesterol as an internal standard was investigated. They were saponified with 20% methanolic KOH solution. Phytosterols in the reaction mixture were extracted with diethyl ether and separated on silica gel TLC plate with n-hexane-diethyl ether(40:60) as the solvent and then were scraped off. They were separated by reversed phase high perfomance liquid chromatography on Inertsil ODS-2 column with detection at 205nm. Cholesterol and ${\beta}$-sitosterol were resolved from interferences by adjusting the acetonitrile content in the MeOH-tetrahydrofuran-$H_2O$ eluent. The detection limit of ${\beta$-sitosterol was 0.43${\mu}$g.

  • A methanol extract of Cassia tora seeds was successively partitioned with diethyl ether, chloroform, ethyl acetate, and water, and the antitumor-promoting activity of the solvent fractions was determined by inhibition of Epstein-Barr virus early antigen (EBV-EA) activation induced by teleocidin B-4 in Raji cells. The diethyl ether (68.7%) and chloroform (91.2%) fractions and the hydrolysate (94.3%) of the ethyl acetate fraction had strong inhibitory activities. The chloroform and ethyl acetate fractions were chromatographed on silica gel and further purified by HPLC. Three active compounds, obtusifolin-2-glucoside (75.0%), chryso-obtusin-6-glucoside (56.8%), and norrubrofusarin-6-glucoside (39.4%), were obtained from the ethyl acetate fraction, and two active compounds, questin (97.9%) and chryso-obtusin (53.8%), were isolated from the chloroform fraction.

Anxiolytic Activities of Sanguisorbae Radix (지유(Sanguisorbae Radix)의 항 불안 활성)

  • To detect anxiolytic activity from Sanguisorbae Radix we used various animal models of fear or anxiety that are sensitive to known anxiolytic drugs. While diazepam showed significant anxiolytic activities in all five animal models empolyed in this study, $5-HT_3$ antagonist ondansetron and ethylacetate fraction of Sanguisorbae Radix did show anti-anxiety effects in social interaction and two compartment exploration tests. Ethylacetate fraction of Sanguisorbae Radix and 5-HT related drugs like ondansetron and buspirone, however, seem to have merit over diazepam in terms of not causing drowsiness. Among ten subfractions obtained from ethylacetate fraction of Sanguisorbae Radix by silica gel chromatography, subfraction I showed higher anxiolytic activities than subfraction DEF in two animal models, social interaction and two compartment exploration tests. There is growing evidence for the role of 5-HT in the control of anxiety. We hope that new compound(s) will be found from the active fractions of Sanguisorbae Radix as a potential anxiolytic agent in the future.

Antimicrobial and Antitumor Activity of Triterpenoids Fraction from Poria cocos Wolf (재배 복령(Poria cocos Wolf)의 Triterpenoids 분획의 항균 활성 및 항암 활성)

  • Seven fractions were separated by silica gel chromatography from the diethyl ether souble portion of the methanolic extract of the cultured hoelen(Poria cocos Wolf). Three fractions were separated from the Fr.II and Fr.IV by rechromatography, respectively. The most active fraction, Fr.II 3 and Fr.IV 3, were separated to 2 and 4 fractions by preparative HPLC. On the result of antimicrobial test, triterpenoids fractions showed weaker effect than benzoic acid but Fr.II 3 1, Fr.II 3 2 had an excellent antimi crobial activity. Triterpenoids fraction of hoelen(Poria cocos Wolf) showed a high inhibition activity on the growth of lung cancer, ovary cancer, skin cancer, central nerve cancer and rectum cancer cell, especially the activity of Fr.II 3 1 and Fr.II 3 2 was the highest.

  • The effects of bioactive fractions containing protoberberine alkaloids from Coptis japonica on monoamine oxidase (MAO) activity were investigated. The MAO was obtained from the mitochondrial fraction of mouse brain. The butanol fraction from Coptis japonica was fractionated into separate bioactive fraction (Fr I-IV) by silica gel column chromatography. MAO activity was strongly inhibited by Fr III and IV, which mainly contain protoberberine alkaloids such as berberine, palmatine and coptisine. These results indicated that the protoberberine alkaloids from Coptis japonica had an inhibitory effect on MAO activity.

  • This study was undertaken to evaluate antioxidative activities of substances isolated from marine resources against human low density lipoprotein (LDL). Methanol-water extract(80 : 20, v/v) of Sargassum ringgoldianum had the highest antioxidant activity and the active substance was purified by silica gel column chromatography by eluting chlorform : methanol mixture (80 : 20 v/v). The active fraction was seperated to several spots on the TLC in chlorofrom : methanol (10 : 1, v/v) mixture. Antioxidative activity of band 4 of fraction 2 on TLC was highest than that of $\alpha$-tocopherol against human LDL oxidation by the method of thiobarbituric acid reactive substance (TBARS). The band 4 of fraction 2 inhibited the copper mediated oxidation of human LDL with almost completely at 1 or 2 mg/ml.

  • The antioxidant activity of Persicaria thunberii (Polygonaceae) was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-pocrylhydrazyl (DPPH) radical. The methanol extract of thunbergii showed strong radical scavenging activity at an I $C_{50}$ concentration of 12.2$\mu\textrm{g}$/ml, and thus fractionated by solvent extraction. The EtOAc soluble fraction was stronger than the others, and was further purified by silica gel and Sephadex LH-20 column chromatography, Antioxidant isorhamnetin, quercetin (2), quercitrin (3) and isoquercitrin (4) were isolated from the EtOAc soluble fraction. And isorhamnetin-3-sulfate (5) was isolated from the n-BuOH fraction. Compounds 2, 3 and 4 were found to have strong antioxidative potency.

  • The antioxidant activity of Juniperus chinensis (Cupressaceae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of J. chinensis heartwood showed the strong antioxidant activity. The antioxidant activity of n-BuOH soluble fraction was stronger than that of the others, and the fraction was subjected to purification by repeated silica gel and Sephadex LH-20 column chromatography. Quercetin, naringenin, taxifolin, aromadendrin and isoquercitrin were isolated from the n-BuOH fraction. Their structures were elucidated by physico-chemical and spectroscopic studies.

  • Phellinus pini (Hymenocaetaceae) has been used for the immunomodulating activity hypolipidemic effect, gastric cancer non-insulin dependant diabetes, diarrhea, and menstrual irregularity. From the screening of each fraction for the inhibitory activity of NO production in lipopolysaccaride (LPS) activated RAW 264.7 cells, methanol extract of Phellinus pini and hexane soluble fraction exhibited inhibition of NO production compared with LPS control without toxicity. The hexane soluble fraction showed dose dependent inhibition of NO production. According to activity guided fractionation, the active hexane fr. was repeatedly chromatographed over silica gel, ergosta-4,6,8(14),22- tetraen-3-one was isolated. The compound inhibited NOS activation (IC$_{50}$ = 29.7 uM) and NO production of activated macrophage at 30 uM.

Enhancement of Ethanol Productivity by Air Supplement in Immobilized Cell Reactor System (균체고정화 생물반응기에서 산소공급에 의한 에탄올 생산성 향상)

  • To achieve higher ethanol productivity in the fermentation system, a continuous ethanol production has been investigated with the air-supplement in a packed-bed immobilized cell reactor system. Yeast cells were immobilized using sodium alginate gel. The results showed that, when the feed medium was saturated with oxygen through aeration into the medium reservoir, the maximum ethanol productivity of the reactor was enhanced from 35 g/$\ell$-gel-hr to 55 g/$\ell$-gel-hr at the residence time of 10-20 min. and the residence time for the 90% conversion of substrate to ethanol was reduced from 40 min. to 25 min. In case of 18% glucose medium, the maximum productivity was increased from 35 g/$\ell$-gel-hr to 45 g/$\ell$-gel-hr and time required for 90% conversion was from 90 min to 70 min. This behavior of air-supplemented reactor system might be due to the fact that both growth and viable fraction of yeast within the Eel were increased during reactor operation.
