In recent years bifidobacteria have drawn much scientific attention due to their use as live bacteria in numerous food products with various health-related claims. For this reason, these bacteria constitute a growing area of interest with respect to genomics, molecular biology, and genetics. Recent genome sequencing of a number of bifidobacterial species has allowed access to the complete genetic make-up of these bacteria. This review will focus how genomic data has allowed us to understand bifidobacterial evolution, while also revealing genetic functions that explains their presence in the particular ecological environment of the gastrointestinal tract.

Gastrointestinal tract of ruminants as well as monogastric animals are colonised by a variety of microorganisms including bacteria, fungi and protozoa. Gastrointestinal ecosystem, especially the rumen is emerging as an important source for enrichment and natural selection of microbes adapted to specific conditions. It represents a virtually untapped source of novel products (e.g. enzymes, antibiotics, bacteriocins, detoxificants and aromatic compounds) for industrial and therapeutic applications. Several gastrointestinal bacteria and fungi implicated in detoxification of anti-nutritional factors (ANFs) can be modified and manipulated into promising system for detoxifying feed stuffs and enhancing fibre fermentation both naturally by adaptation or through genetic engineering techniques. Intestinal lactobacilli, bifidobacteria and butyrivibrios are being thoroughly investigated and widely recommended as probiotics. Restriction endonucleases and native plasmids, as stable vectors and efficient DNA delivery systems of ruminal and intestinal bacteria, are increasingly recognised as promising tools for genetic manipulation and development of industrially useful recombinant microbes. Enzymes can improve the nutrient availability from feed stuffs, lower feed costs and reduce release of wastes into the environment. Characterization of genes encoding a variety of commercially important enzymes such as cellulases, xylanases, $\beta$-glucanases, pectinases, amylases and phytases will foster the development of more efficacious and viable enzyme supplements and enzyme expression systems for enhancing livestock production.
In this study, a 40% ethanol extract of Chinese yam flour (Dioscoreae rhizoma), containing $177{\pm}58{\mu}g/mL$ of dioscin, was tested in order to evaluate its pharmacological effects on the gastrointestinal tracts of Sprague-Dawley rats. Via the ingestion of the Chinese yam extract, the secretion of gastric acid was suppressed in the rats, and gastrointestinal motility increased by as much as 10%. The fecal quantity of rats fed on the Chinese yam extract also increased, by more than 40% as compared with that of the controls. The Chinese yam extract was found not to affect the growth of normal intestinal bacteria. However, a great deal of lactose-fermenting bacteria was observed in the fecal samples of rats fed for 6 weeks on 2% Chinese yam extract. This finding would appear to suggest that Chinese yam extract not only induces an improvement in digestive capability, but also affects the conversion of some intestinal flora to helpful bacteria. Our serochemical analyses indicated that serum glucose, neutral lipid, and total cholesterol levels were reduced to some degree by long-term feeding on Chinese yam extract. This finding bolsters the notion that Chinese yam extract may prove helpful as a digestion-aiding agent for patients suffering from hyperglycemia or hyperlipidemia.
The objective of this study was to investigate differences in the faecal microbial composition among Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire sows and to explore the possible link of the pig breed with the gut microbial community. Among the sows, the Meishan, Landrace, Duroc, and Yorkshire sows were from the same breeding farm with the same feed. Fresh faeces were collected from three sows of each purebred breed for microbiota analysis and volatile fatty acid (VFA) determination. Denaturing gradient gel electrophoresis (DGGE) analysis revealed that samples from Bama, Erhualian, and Xiaomeishan sows, which from different farms, were generally grouped in one cluster, with similarity higher than 67.2%, and those from Duroc, Landrace, and Yorkshire sows were grouped in another cluster. Principal component analysis of the DGGE profile showed that samples from the foreign breeds and the samples from the Chinese indigenous breeds were scattered in two different groups, irrespective of the farm origin. Faecal VFA concentrations were significantly affected by the pig breed. The proportion of acetate was higher in the Bama sows than in the other breeds. The real-time PCR analysis showed that 16S rRNA gene copies of total bacteria, Firmicutes and Bacteroidetes were significantly higher in the Bama sows compared to Xiaomeishan and Duroc sows. Both Meishan and Erhualian sows had higher numbers of total bacteria, Firmicutes, Bacteroidetes and sulphate-reducing bacteria as compared to Duroc sows. The results suggest that the pig breed affects the composition of gut microbiota. The microbial composition is different with different breeds, especially between overseas breeds (lean type) and Chinese breeds (relatively obese type).
신생아 분변으로부터 장내세균 127균주를 분리하였다. 각각의 균주들의 배양균체를 검정종양세포들에 대해 항종양활성을 측정한 결과, 항종양활성을 나타내는 분리균주는 27균주였고, 그 중 비교적 강한 항종양활성을 보이는 균주는 5균주였으며, 5균주중 위암세포주, SNU-1에 선택적으로 강한 항종양활성을 나타내는 2B4-1 균주를 최종분리하였다. API 20 S system kit를 이용하여 분리 균주 2B4-1을 동정한 결과, Enterococcus faecalis와 99.9%의 % ID value를 나타내었다. 동정된 분리균주 2B4-1과 type strain, Enterococcus faecalis NCTC 775와의 생리생화학적 특성들을 비교해 본 결과, 온도별대 성장률, 내염성, 내산성, arabinose등의 당 이용성 및 통성혐기 상태에서의 생육 등에서는 일치된 결과를 나타내었으나 hippurate hydrolysis test의 음성반응 및 $\beta$-hemolysis test의 음성반응을 나타낸 것으로 보아 Enterococcus faecalis NCTC 775와 2가지의 다른 특성을 나타내었다. 따라서 분리균주 2B4-1은 Enterococus faecalis NCTC 775와는 약간의 특성이 다른 변종으로 확인되어 Enterococcus faecalis 2B4-1으로 동정하였다.

Intestinal bacteria comprise one-third of the contents of the large intestine in humans. Their interactions with the gastrointestinal immune system induce characteristic immunological responses which stimulate or suppress the host's defense system. RAW 264.7 murine cell line was used as a macrophage model to assess the effects of the exposure to the isolated human intestinal bacteria, Bacteroides, Bifidobacterium, Eubacterium, Streptococcus, and E. coli, on NO (nitric oxide), $H_2O_2$(hydrogen peroxide), and cytokines IL (interleukin)-6 and TNF (tumor necrosis factor)-a production. RAW 264.7 cells were cultured in the presence of heat-killed bacteria for 24 h at concentrations of 0-$50\mu$g/ml. Our results showed that Bacteroides and E. coli stimulated IL-6, TNF-$\alpha$, NO, and $H_2O_2$production at high levels even at $1\mu$g/ml, whereas Bifidobacterium, Eubacterium, and Streptococcus showed a low level of stimulation at $1\mu$g/ml, and a gradual increase as the cell concentration increased up to $50\mu$g/ml. This result suggests that gram-negative Bacteroides and E. coli are better able to stimulate macrophage than gram-positive Bifidobacterium, Streptococcus, and Eubacterium. The in vitro approaches employed here should be useful in further characterization of the effects of intestinal bacteria on gastrointestinal and systemic immunity.
최근 우리나라는 서구화된 식생활과 생활양식의 변화로 다양한 대사성 질환과 만성 질환이 나타나고 있다. 이러한 질환들은 지속적으로 약물을 복용해야 한다. 또한 많은 사람들이 건강한 삶을 유지하기 위해 건강기능식품과 각종 비타민, 영양제를 복용한다. 하지만 복용하는 약물들이 장내 세균에는 어떠한 영향을 주는 지에 대한 연구는 많이 이뤄지지 않고 있다. 이에 본 연구에서는 흔하게 사용되며 장기간 복용하는 약이 항균활성시험으로 위장 관련 박테리아에 영향을 미치는 지를 조사하였다. 그 결과 비타민 및 미네랄, 중추신경계에 작용하는 약들이 장내 세균과 유산균에 대해 항균력을 나타냈으며, 그 중 중추신경계에 작용하는 약 중 항현훈제의 dexibuprofen는 장내 유익균 인 Lactobacillus casei과 Lactobacillus rhamnosus와 장내 유해균 인 Staphylococcus aureus에 대해 항균력이 높게 작용하였다. 또한 심혈관계, 조혈기계에 작용하는 약 중 항이뇨제 의 fenofibric acid는 장내 유익균인 Lactobacillus casei에 대해 항균력이 나타났다. 비타민 및 미네랄은 대부분의 장내 균에서 항균력을 보이지만, 특히 Vitamin B-Complex/with C와 vitamin C가 유익균인 Bifidobacterium infantis과 장내 유해균 인 Escherichia coli, Enterobacter aerogenes, Shigella flexneri, Salmonella Typhimurium, Staphylococcus aureus에서 높게 나타났다. 따라서 본 연구결과 장내 유해균과 유익균에 다양하게 복용되는 약들이 대체적으로 항균활성을 보이는 것으로 판단된다.
인간 미생물 균총은 장, 구강, 피부와 같이 체내외 다양한 부위에 존재하는 박테리아, 균류, 바이러스 등을 포함하는 미생물 집단이다. 16s ribosomal RNA에 대한 대사체 분석 및 차세대 염기서열 분석기술의 개발과 함께, 살아있는 유기체 내에 존재하는 미생물 균총에 대한 많은 연구가 진행되었다. 이에 따라, 미생물 균총이 숙주의 대사 및 면역과정과 복잡하게 연관되어 있음이 확인되었다. 공생균(commensal microbiota)이라 불리는 미생물 균총의 특정 박테리아가 필수 영양소를 생성하거나 다른 병원성 미생물로부터 숙주를 보호하여 긍정적으로 영향을 미치고 있는 반면, 비정상적인 미생물 균총의 조성을 의미하는 미생물 균총의 불균형(dysbiosis)에 의해 체내 항상성 유지를 방해하여 다양한 종류의 질병을 발생시키기도 한다. 최근, 미생물 균총 중에서도 구강과 장내 존재하는 박테리아가 위장관암의 발암과정과 항암제의 치료효과에 상당한 영향을 미치고 있음이 여러 논문을 통해 보고되고 있다. 미생물 균총-암-면역계 사이의 복잡한 연관성과 미생물 균총 기반 발암 메커니즘에 대한 규명은 암에 대한 이해와 새로운 항암제 개발에 중요한 단서를 제공할 것으로 기대된다. 본 리뷰는 미생물 균총의 박테리아가 위장관암과 항암제에 어떤 영향을 미치고 있는지에 대해 초점을 맞추고 있는 논문들을 요약하고 있으며, 나아가 기존 항암제의 치료효과를 개선하기 위해 복합제로써 미생물 균총의 잠재력과 도전과제에 대해 논의한다.
The study aimed to investigate the effects of gynosaponin on in vitro methanogenesis under different forage-concentrate ratios (F:C ratios). Experiment was conducted with two kinds of F:C ratios (F:C = 7:3 and F:C = 3:7) and gynosaponin addition (0 mg and 16 mg) in a $2{\times}2$ double factorial design. In the presence of gynosaponin, methane production and acetate concentration were significantly decreased, whereas concentration of propionate tended to be increased resulting in a significant reduction (p<0.05) of acetate:propionate ratio (A:P ratio), in high-forage substrate. Gynosaponin treatment increased (p<0.05) the butyrate concentration in both F:C ratios. Denaturing gradient gel electrophoresis (DGGE) analysis showed there was no apparent shift in the composition of total bacteria, protozoa and methanogens after treated by gynosaponin under both F:C ratios. The real-time polymerase chain reaction (PCR) analysis indicated that variable F:C ratios significantly affected the abundances of Fibrobacter succinogenes, Rumninococcus flavefaciens, total fungi and counts of protozoa (p<0.05), but did not affect the mcrA gene copies of methanogens and abundance of total bacteria. Counts of protozoa and abundance of F.succinogenes were decreased significantly (p<0.05), whereas mcrA gene copies of methanogens were decreased slightly (p<0.10) in high-forage substrate after treated by gynosaponin. However, gynosaponin treatment under high-concentrate level did not affect the methanogenesis, fermentation characteristics and tested microbes. Accordingly, overall results suggested that gynosaponin supplementation reduced the in vitro methanogenesis and improved rumen fermentation under highforage condition by changing the abundances of related rumen microbes.
