Antiulcer activity of Trichosanthes cucumerina linn. against experimental gastro-duodenal ulcers in rats

  • Trichosanthes cucumerina Linn. (cucurbitaceae) is widely used in Indian folk medicine for variety of disease conditions. The aim of present study was to evaluate the antiulcer activity of 50% ethanolic extract of fruits of Trichosanthes cucumerina Linn. (TCFE) using various experimental models of gastric and duodenal ulceration in rats. Oral administration of 50% ethanolic extract of fruits of Trichosanthes cucumerina Linn. was evaluated in rats against ethanol, aspirin and pylorus ligated gastric ulcers as well as cysteamine-induced duodenal ulcers. In all the models studied, the antiulcer activity of TCFE compared with that of cimetidine (100 mg/kg, p.o.), an $H_2$ receptor antagonist. TCFE showed significant antiulcer activity in ethanol-induced and aspirin-induced gastric ulcer models. In 19 h pylorus ligated rats, significant reduction in ulcer index, total acidity and pepsin activity was observed with TCFE, when compared with the control group. Mucosal defensive factors such as pH, mucin activity and gastric wall mucous content was found to be increased with TCFE. TCFE was also, afforded remarkable protection in cysteamine-induced duodenal lesions. The antiulcer activity of TCFE was comparable with that of cimetidine. Thus, TCFE possess significant antiulcer activity against both gastric and duodenal ulcers in rats. The antiulcer activity may be attributed to its cytoprotective action and inhibition of acid secretary parameters.

Effects of Pyeong yoe san Extract and Jo gi pyeong yoe san Extract on the Experimental Gastric Ulcer in Rats (평위산(平胃散)과 조기평위산(調氣平胃散)이 흰쥐의 위궤양(胃潰瘍)에 미치는 효과(效果))

  • In order to study on the effect of Volume of Gastric Juice Free acid, Total acid, and Anti-Ulceration, Jo gi pyeong yoe san Extract were administered to the Pylorus-Ligated Rats induced Shay's Ulcer. The results were as follows. 1. The effect upon Volume of Gastric Juice, Free Acid, and Total Acid of experimenting group with medicine, in contrast with Control Group and Pyeong yoe san Group, there was decreasing that it reveled Significant Value. 2. In the effect upon Shay's Ulcerlation, Jo gi pyeong yoe san Extract were decrease to the Ulcer Index that it reveled Significant Value. 3. Control Group and Pyeong yoe san Group were similar. According to the above results, Jo gi pyeong yoe san is assumed to have Anti-Ulcerlation.

Effects of Dietary Supplementation with Astaxanthin on Histamine Induced Lesions in the Gizzard and Proventriculus of Broiler Chicks

  • Astaxanthin (ASX) is a xanthophyll pigment isolated from crustaceans and salmonids. Owing to its powerful antioxidant activity, ASX has been reported to have the potential to protect against gastric ulcers and a variety of other illnesses. Histamine (His) is a dietary factor that causes gastric erosion and ulceration in young chicks. In this study, we examined whether ASX had protective effects on dietary histamine-induced lesions in the gizzard and proventriculus of broiler chickens. Four experimental treatment groups were planned: basal diet (BD), BD+His, BD+ASX, and BD+ASX+His, with four chicks (5 days old) in each group and three replications (i.e., a total of 12 chicks per group). The BD was supplemented with either 0.4% His or 100 ppm ASX. The birds were fed ad libitum for 3 weeks, and diets contained no antimicrobial compounds. Supplementing the diet with His significantly decreased body weight gain, but increased the weights of the gizzard and proventriculus of the chicks as compared with those of chicks in the BD group (p<0.05). ASX did not affect His-dependent changes in chick body weight or weights of the gizzard and proventriculus. The loss of gastric glands in the proventriculus, which was observed in His-treated chicks, was not prevented by ASX administration. The frequency of proventricular ulceration, however, was lowered by treatment with ASX, without significant differences between the two supplementation levels. In conclusion, our data showed that ASX might be helpful for alleviating structural damage to the digestive system in poultry under certain stressful conditions.

Helicobacter pylori cag Pathogenicity Island cagL and orf17 Genotypes Predict Risk of Peptic Ulcerations but not Gastric Cancer in Iran

  • Background: Gastric cancer (GC) is the third most common cancer regarding mortality in the world. The cag pathogenicity island (PAI) of Helicobacter pylori which contains genes associated with a more aggressive phenotype may involve in the pathogenesis of gastrointestinal disease. We here aimed to examine the associations of cagH, cagL, orf17, and cagG genotypes of H. pylori cag PAI with severe gastrointestinal disease. Materials and Methods: A total of 242 H. pylori strains were genotyped. Histopathological examination and classification of subjects were performed. Results: The frequencies of the cagH, cagL, cagG, and orf17 genotypes were 40/54 (74.1%), 53/54 (98.1%), 38/54 (70.4%), and 43/54 (79.6%), respectively, in patients with peptidic ulceration (PU),while in the control group, the frequencies were 87/147 (59.6%) for cagH, 121/146 (82.9%) for cagL, 109/146 (74.7%) for cagG, and 89/146 (61.0%) for orf17. The results of simple logistic regression analysis showed that the cagL and orf17 genotypes were significantly associated with an increased risk of PU not GC; the ORs (95% CI) were 10.950 (1.446-82.935), and 2.504 (1.193-5.253), respectively. No significant association was found between the cagH and cagG genotypes and the risk of both the PU and the GC in Iran (P>0.05). Finally, multiple logistic regression analysis showed that the cagL genotype was independently and significantly associated with the age-and sex-adjusted risk for PU; the OR (95% CI) was 9.557 (1.219-17.185). Conclusions: We conclude that the orf17 and especially cagL genotypes of H. pylori cag PAI could be factors for risk prediction of PU, but not GC in Iran.

Development of Pneumoperitoneum after Endoscopic Retrieval of Gastric Foreign Body in a Dog (개에서 위내 이물의 내시경적 제거 후 발생한 기복증 증례)

  • An 1-year-old, intact female Yorkshire Terrier weighing 1.83 kg was referred for vomiting and anorexia. Survey abdominal radiographs revealed marked gastric distention. Ultrasonography showed a hyperechoic material with acoustic shadowing near the pylorus. A foreign body and ulceration were identified during gastroscopic examination. After endoscopic retrieval of gastric foreign body, unexpectedly abdominal distention was developed. Abdominal radiographs revealed pneumoperitoneum, but no leakage of contrast media was observed in gastrointestinal contrast study. Three days after hospitalization with supportive care, no abnormal findings were detected, and then the dog was discharged. This case describes the development of peumoperitoneum during endoscopic retrieval of gastric foreign body.

Effect of Recombinant Human Epidermal Growth Factor(DWP 401) on Gastric Secretion and Ulcers in Rats (재조합 인간 상피세포 성장인자(DWP 401)의 흰쥐 위액분비 및 궤양에의 작용)

  • The effects of human epidermal growth factor(EGF) which was produced by recombinant DNA technique was investigated on gastric secretion, gastric lesion and ulcer models in rats. The EGF showed significant inhibition of secretion of gastric juice and total acid output, at 0.4mg/kg, id and also inhibited Shay ulceration at 0.4mg/kg, id in rats. The lesion induced by absolute ethanol was significantly reduced by oral administration of EGF at 0.4mg/kg. Likewise, EGF caused significant inhibition of indomethacin induced gastric ulcer at oral doses of 0.2 and 0.4mg/kg. The EGF produced dose-dependent inhibition of gastric ulcer induced by acidified aspirin, but showed no significant inhibition at oral doses of 0.1, 0.2 and 0.4mg/kg. The chronic gastric ulcer induced by injection of 20% acetic acid solution was significantly reduced by oral doses of 0.1 and 0.4mg/kg of EGF. Duodenal ulcer induced by mepirizole was dose-dependently inhibited by oral doses of 0.1, 0.2 and 0.4mg/kg of EGF. These data suggest that EGF possesses pronounced inhibitory action in gastric ulcer and duodenal ulcer of rats.

Effect of Dioscorea Rhizoma on Gastritis by Acute Gastric Ulcer in Rats (산약이 흰쥐의 급성 위궤양에 의한 위장 장애에 미치는 효과)

  • In order to verify the effects of Dioscorea Rhizoma on gastrointestinal damages, we investigated the protective role of the crude extracts on induced gastric ulceration in rats. Rats administered of Dioscorea Rhizoma extracts showed gradually decreased congestion and hemorrhage, but control group did not show any symptom in gastric tissue. Moreover, Dioscorea Rhizoma extract had a role in lowering gastrin and histamine levels in gastric ulcer rats, thereby inhibiting the gastric tissue damages. Excessive production of malondialdehyde shown in gastric ulcer rats was declined in all rats administered with Dioscorea Rhizoma extract as well as the levels of SOD and GPX surged by acute gastric ulcer. Also, the increased activity of CAT showed an effect in activation of antioxidant enzyme to normal state. All data suggest that Dioscorea Rhizoma extract was verified to be highly effective resource in improving the gastrointestinal function of rats by preventing from gastric tissue damage in acute gastric ulcer and restoring the activities of plasma substances and antioxidant enzymes.

Antigastritic and Antiulcer action of Effective Compounds from Propolis Extract (Propolis 유효성분의 항위염 및 항위궤양 작용)

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents(wax, flavonoids, phenolic compounds, etc.) and has been used in traditional medicines as early as 300 B.C. It was been demonstrated that ethanol, acetylsalicylic acid, ischemia reperfusion, non-steroidal antiin-flammatory drugs and stress induce gastric lesions by promoting the generation of reactive oxygen metabolites. Therefore, some drugs that are capable of scavenging or inhibiting the generation of reactive oxygen radicals might be expected to prevent the gastric mucosal injury. The aim of this study was 1) to examine the antiulcer effect of propolis, 2) to investigate the mechanism of action by determining gastric acid secretion, lipid per-oxidation, mucus content and proton pump ($H^+$/$K^+$-ATPase) activity on gastric mucus in varios experimental models, and finally, 3) to isolate and identify the pure compounds that exert antiulcer activity. Step 2-1 and 2-3 sub-sub fraction shoed a significant reduction of severity of gastirc damage at the dose of 25 mg/kg in various experimental models. We isolated 4 sub-sub-sub fractions by flash column chromatography of Step 2-1 sub-sub fraction and one sub-sub-sub fraction by recrystalization of Step 2-3 sub-sub fraction. The protective effects of propolis sub-sub-sub fraction manifested sifnificant effects in HCl-ethanol induced gastric erosion model and aspirin induced gastric ulcer model. These results showed that the gastric mucosal protective effect of propolis might result from the increase of mucus secretion, free radical scavenging effect as well as the reduction of acid secretion in accordance with the reduction of $H^+$/$K^+$-ATPase activitv. Three compounds were isolated and identified from sub-sub fraction of propolis which showed antiulcer effects. Subsequently, these compounds were identified as a flavonoid, namely, 2-acetoxy-5,7,-dihydroxy-flavanone, galangin and chrysin.

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents and has been used in traditional medicines as early as 300 B.C. Recently, it has been reported to possess many biological activities such as antibacterial, antiviral, fungicidal, local anaesthetic, immunostimulating, antiinflammatory and free radical scavenging properties. This study was performed to investigate the pharmacological effects of the propolis extract and fractions on the gastric lesion and ulcer. The ethanol extract was fractionated with hexane, toluene and ethyl acetate. Followed by bioassay on antigastric and antiulcer activity. Propolis ethanol extracts(500, 750, 1,000, 1,500 and 2,000 mg/kg) showed the protective effect on HCl·ethanol-induced gastric lesion and the antisecretory effect against Shay’s gastric secretion in pylorus-ligated rats in a dose related manner. In the animal models of HCl·ethanol, aspirin-induced gastric lesion and Shay’s gastric secretion, the hexane and toluene fraction of propolis significantly reduced the length of gastric lesion and the acid secretion. These data showed that the gastric protective effects of propolis might result from reduction of acid secretion through the inactivation of H+/K+ATPase activity.

Pathological Studies on the Esophagogastric Ulcers in Swine (돼지의 위궤양(胃潰瘍)에 관한 병리학적(病理學的) 연구(硏究))

  • Stomachs from 3,354 fattening pigs were examined at slaughter during the period from September 1980 to August 1981. Pigs of both sexes and $Landrace{\times}Hampshire$ crossbreds were included in the present studies, and they weighed about 60-120kg. Gross pathologic alterations of the stomach were classified as normal, epitnelial change, erosion, ulcer and scar formation. Representative tissue sections were taken from the stomach lesions at random and fixed in 10% buffered formalin. Cut sections were stained with hematoxylin and eosin, and examined histopathologically. The results obtained in the present studies were as follows. 1. In the seasonal prevalence of gastric ulceration, severe ulceration with erosion was shown during the Autumn and Winter, whereas mild ulceration was mainly shown during the Spring and Summer. 2. Of the 3,354 stomachs of the pigs, 20.8% were found to be normal. Of the rest, 40% had epithelial changes, followed by erosion (24.7%), ulcer (13.6%) and scar formation (0.9%), respectively. 3. In the prevalence of ulcers in the different regions of the stomach, the fundic region had the highest rate (61.6%) of ulceration, followed by cardiac (21.7%), esophageal (15.0%) and pyloric region (1.7%). 4. The principal gross changes were severe epithelial changes with keratotic proliferation in the esophageal region, and in the fundic region severe folding of the stomach wall was covered with bloody mucous exudates. 5. Main histopathological changes were inflammatory cell infiltrations in most cases, hemorrhages in acute ulcers and prominent proliferation of granulation tissues in chronic ulcers.
