Identification of Benzidine Metabolites in Rats by Gas Chromatography/Mass Selective Detector and its Toxicity in vitro (Gas-Chromatography/Mass Selective Detector를 사용하여 쥐의 뇨시료 중 benzidine 대사체의 확인 및 in vitro 독성)

  • Metabolism study of the dye, benzidine, was performed by gas chromatography-mass selective detector (GC/MSD) in the urine of rats orally administered 100 mg/kg benzidine. Urine samples were collected in metabolic cages for 0-24, 24-48, and 48-72 hrs. Ten ml of the urine was extracted with XAD-2 resin and the XAD-2 column was eluted with methanol. After evaporation, benzidine and its metabolites were extracted with diethyl ether (for non-conjugated fraction). For conjugated metabolites, $\beta$-glucu-ronidase was added to the aqueous layer that was incubated for 1 hr at 5$0^{\circ}C$ and the aqueous layer was extracted as in non-conjugated fraction. Aliquot of trimethylsilylated derivatives was applied to the GC/MSD. The mutagenicity of benzidine and its acetylated metabolites was tested by histidine/reversion assay. Five metabolites observed and confirmed either by electron impact and chemical ionization modes of the GC/MSD, or authentic compounds were monoacetyl-, diacetyl-, hydroxyacetyl-, hydroxydiacetyl-, and hydroxy-benzidine. Monoacetyl-benzidine was more potent than benzidine in histidine/reversion assay. This data indicates that monoacetylation of benzidine may be one of the metabolites produced in metabolic activation process.

  • The combination of Gas chromatography (GC) for separation and Flame Ionization Detection (FID) for detection and identification of the components of a mixture of compounds is a fast and strongly proved method of analytic chemistry. The objective of this research was to design a combined High-speed miniature screening Gas chromatograph along with a Flame Ionization Detector for quick, quantitative and qualitative analysis of gas components. This combined GC-FID system is suitable to detect the volatile and semi-volatile hydrocarbons present in a gas mixture. The construction made it less expensive, easy to use and movable. The complete gas path was developed. On/off valves, temperature and flow sensors and their interface electronics were used for controlling purpose. A Microcontroller was programmed to measure the temperature and gas flow using the sensors and to control and regulate them using the electronics and valves. A pocket PC with its touch screen served as a user interface for the system. Software was developed for the pocket PC, which makes the communication possible with the Microcontroller. The system parameters can be indicated in the Pocket PC as simple text and also the analysis result can be displayed.

  • Some $\betha$-lactam antibiotics-penicillins and cephalosporins-were determined by gas chromatography (GC) with flame photometric detector (FPD) which was selective and sensitive to sulfur-containing compounds. Methyl ester derivatives of carboxyl group in $\betha$-lactam antibiotics were prepared using 0.5Mmethyl iodide in methylene chloride and were taken for gas chromatography with 0.9% or 0.6% QF-1 on Chromosorb WAW-DMCS. We have found that it is possible to determine methyl esters of $\betha$-lactam antibiotics by GC/FPD.

  • The current study was conducted to develop a portable composite gas detector allowing the detection of both $CO_2$ and $CH_4$ gases by means of the Non Dispersive Infra-Red (NDIR) method. The gas detector is configured to radiate infrared waves using infrared lamps, where the wavelength of the infrared light is reduced due to absorption throughout the chamber, and this reduction (absorption) is detected by the absorption detector, before being converted and amplified to a 3.5V~6V electrical signal, providing as accurate a measurement as possible. The conventional singe sensor method measures the relative measurement by absorbing only specified wavelengths of infrared radiation, which in the case of gas detection leads to problems with accuracy due to the lack of a reference sensor when detecting light with a wavelength of only $4.26{\mu}m$. The dual sensor employed in this study provides a comparative measurement between the reference value derived from the wavelength of $3.91{\mu}m$, which is not influenced by other gas sources, and the measurement value derived from the wavelength of $4.26{\mu}m$, in order to reduce the errors and enhance the reliability, thereby allowing low power consumption for portable devices and multi-gas detection for both $CO_2$ and $CH_4$ gases. The portable composite gas detector developed herein provides a measurement rage of 0ppm~5,000ppm for $CO_2$ gas, and 0.5%vol for $CH_4$, which allows the determination of whether the $CO_2$ and $CH_4$ contents in indoor air are less than 1,000ppm or not. The current study established that the composite gas detector can be interlinked with firefighting appliances through portable devices or home automation, and is anticipated to be very effective in fire prevention.

  • We have investigated air/200ppm NO$_2$ gas-detector characteristics of using CuTBP (Copper-tetra-tert-butylphthalocyanine) chemiresistor devices. The CuTBP films were made by Langmuir-Blodgett (LB) techniques. Sensitivity, response time, recovery time, and repoducibility of the devices were measured by current-voltage characteristics. To increase sensitivity, interdigital electrode was used. It was found that a conductance G increases monotonically as the number of interdigital electrode increases, and a Sensitivity, Reproducibility is stable. As far as a current is concerned, the current when N=25 is greater than that when N=1 by 70 or so. It indicates that the number of interdigital electrodes affects the current, sensitivity and stability We have also investigated applicability of the CuTBP chemiresistor device for a gas detector.

Multi-Pesticide Residue Method for Organopesticide Analysis (유기농약 분석을 위한 Multi-Pesticide Residue Method)

  • Pesticide residues were extracted with 70% acetone and transferred to dichloromethane. Extracts were applied to open-column chromatography with florisil and alumina-N. The final extract was analyzed by gas chromatography with electron-capture detector(GC /ECD) and nitrogen-phosphorus detector(GC/NPD). Recoveries of the 17 organochlorine pegticides were ranged from 60.8 to 84.9% and those of 15 organophosphate pesticides, from 70.5% to 100.0%(except phosmet and azlnphos-methyls. The minimum detectable levels of this analytical method were low(0.021-0.058mg/kg).

Detection of the Ultrasonic Signals due to Partial Discharges in a 154kV Transformer

  • We have developed an on-line ultrasonic detector to monitor partial discharge in an operating transformer. The ultrasonic sensor has 150[KHz] resonance frequency and contains a pre-amplifier with 60[㏈] gain. The on-line ultrasonic detector has 50~300[KHz] frequency band-pass filter to remove electrical and mechanical noises from the transformer. This detector has an ultrasonic signal discrimination algorithm which discriminates ultrasonic signals due to partial discharge in a transformer. A moving average method of ultrasonic signal number was employed to effectively monitor the increasing trend of the partial discharge. This paper describes an experience of partial discharge detection in a 154[㎸] operating transformer using an ultrasonic detector. With regards to gas analysis in oil, C2H2 gas was produced with a warning level in this transformer We detected ultrasonic signals on the transformer steel wall, and estimated the position of partial discharge. With further inspection, we found carbonized marks due to partial discharge on the supporting bolt which fastens the windings.

  • A plasma source with cylindrical microwave cavity was used as atomic emission detector for gas chromatography. Detection limits of several elements were determined for this system. Detection limits for bromine and sulfur were 0.46 pg/s and 0.51 ng/s, respectively. The plasma was stable at the range of flow rate of 10 to 20mL/min.

  • This paper describes NDIR $CO_2$ gas sensor that shows the characteristics of temperature compensation. It consists of novel optical cavity that has two elliptical mirrors and a thermopile detector that includes ASIC chip in the same metal package for the amplification of detector output voltage and temperature sensor. The newly developed sensor modules shows high accuracy (less than +/-40 ppm) throughout the measuring concentration of $CO_2$ gas from 0 ppm to 2,000 ppm. After implementing the calculation methods of gas concentration, which is based upon the experimental results, the sensor module shows high accuracy less than +/- 5 ppm error throughout the measuring temperature range $(15^{\circ}C\;to\; 35^{\circ}C)$ and gas concentrations.
